The present study provides the first epidemiological data on infection with Babesia bovis in cattle raised in the southwestern Brazilian Amazon. Blood clot samples were filtered through nylon cloth before being submitted to DNA extraction. PCR and nested-PCR were applied to assess the frequency of infection with B. bovis in calves with ages from 4 to 12 months bred in 4 microregions each in the states of Rondônia and Acre. After the DNA was extracted from the samples, the infection in cattle was investigated by amplification of the rap1 gene from B. bovis. The DNA amplification results revealed a frequency of infection with B. bovis of 95.1% (272/286) in the samples from Rondônia and 96.1% (195/203) in those from Acre. The high frequency of B. bovis infection in the animals with ages from 4 to 12 months indicates a situation of enzootic stability in the regions studied. The infection rates are comparable to those detected by immunodiagnostic techniques in other endemic regions of Brazil. © 2012 Elsevier GmbH.

Ninety-five specimens from 13 species of lizard collected during a herpetofaunal monitoring programme of the Faxinal II power plant, municipality of Aripuanã, state of Mato Grosso, Brazil (southern Amazon region) were examined for helminths. A total of 21 helminth species (16 Nematoda, 1 Cestoda and 4 Trematoda) were recovered, with an overall prevalence of 67.37%. Seventeen new host records and seven new locality records are reported. A low number of specialists and core helminth species were found. Lizard body size was positively correlated with both the total number of helminth species and individuals. Active foragers exhibited higher helminth diversity. However, sit-and-wait foragers, especially Plica plica, had similar diversity values as active foragers and harboured more helminth species. The degree of similarity in helminth fauna was higher among closely related host species. Copyright © 2011 Cambridge University Press.

Hemiodus iratapuru, a new species of the Hemiodontidae from the Rio Iratapuru, a left bank tributary of the Rio Jari, Amazon Basin, Brazil, is described. The new species is diagnosed from other species of Hemiodus by modifications in the ectopterygoid, tooth form, scale counts, dorsal-fin form and colour pattern. Studies to determine suitable levels of intensification are essential for developing sustainable aquaculture. The objective of this study was to evaluate the quality of effluents discharged from ponds stocked with 10 (D10), 20 (D20), 40 (D40), and 80 (D80) postlarvae of Macrobrachium amazonicum/m2. Intake and effluent water samples were taken throughout a 5.5-mo grow-out cycle. In that study, twelve 0.01-ha earthen ponds were stocked postlarvae with 0.01g. Average water exchange rate was 15%/d; water was discharged from the bottom of the ponds. Prawns were fed a commercial feed with 38% crude protein according to their biomass (3-10%) and the concentration of dissolved oxygen (DO). In our research, temperature, turbidity, total suspended solids, conductivity, DO, pH, biochemical oxygen demand (BOD), chemical oxygen demand (COD), N-ammonia, N-nitrite, N-nitrate, N-Kjeldahl nitrogen, total phosphorus, and soluble orthophosphate were measured every 15d throughout the experiment in the early morning (0630 to 0730h). Turbidity was lower in D10 than in D20 and D40 and total phosphorus was higher in D80 than in D10 and D20. An analysis of principal components comparing treatments and intake water showed three groups: intake, D10 and a cluster of D20, D40, and D80. On the basis of the water characteristics found in our study it appears that the farming of M. amazonicum is likely to have a low environmental impact, at least up to a stocking density of 80prawns/m2. © by the World Aquaculture Society 2013.

In the present study, a simple, rapid and sensitive method was developed for the determination of mercury concentrations in the muscle tissue of fish from the Brazilian Amazon using graphite furnace atomic absorption spectrometry (GFAAS) following acid mineralization of the samples in an ultrasonic cold water bath. Using copper nitrate as a chemical modifier in solution and sodium tungstate as permanent modifier, we were able to attain thermal stabilization of the mercury up to the atomisation temperature of 1600 °C in the GFAAS assay. The calculated limits of detection (LOD) and quantification (LOQ) were 0.014 and 0.047 mg kg-1, respectively. © 2013 Elsevier Ltd. All rights reserved.

Here we describe the first case report of an hemangiosarcoma in a male Blue-fronted amazon parrot (Amazona aestiva). The mass was localized near the inferior portion of the right orbit. A fine needle aspiration biopsy (FNAB) was performed and a presuntive diagnosis of hemangioma/hemangiosarcoma was established. Further histopathological evaluation revealed a mesenchymal neoplasm composed basically by well formed vascular channels lined by pleomorphic endothelial cells. Thus, the final diagnosis was a well-differentiated cutaneous hemangiosarcoma.

Contents: The osteopontin gene may influence the fertility of water buffaloes because it is a protein present in sperm. The aim of this work was to identify polymorphisms in this gene and associate them with fertility parameters of animals kept under extensive grazing. A total of 306 male buffaloes older than 18 months, from two farms, one in the state of Amapá and the other in the state of Pará, Brazil were used in the study. Seven SNPs were identified in the regions studied. The polymorphisms were in gene positions 1478, 1513 and 1611 in the region 5′upstrem and positions 6690, 6737, 6925 and 6952 in the region amplified in intron 5. The SNPs were associated with the traits, namely scrotal circumference, scrotal volume, sperm motility, sperm concentration and sperm pathology. There were significant SNPs (p < 0.05) for all the traits. The SNP 6690 was significant for scrotal circumference, sperm concentration, sperm motility and sperm pathology and the SNP 6737 for scrotal volume. The genotype AA of SNP 6690 presented the highest averages for scrotal circumference, sperm concentration and motility and the lowest total number of sperm pathologies. For the scrotal volume trait, the animals with the largest volume were correlated with the presence of the genotype GG of SNP 6737. These results indicate a significance of the osteopontin gene as it seems to exert a substantial influence on the semen production traits of male buffaloes. © 2013 Blackwell Verlag GmbH.

HLA-G has an important role in the modulation of the maternal immune system during pregnancy, and evidence that balancing selection acts in the promoter and 3′UTR regions has been previously reported. To determine whether selection acts on the HLA-G coding region in the Amazon Rainforest, exons 2, 3 and 4 were analyzed in a sample of 142 Amerindians from nine villages of five isolated tribes that inhabit the Central Amazon. Six previously described single-nucleotide polymorphisms (SNPs) were identified and the Expectation-Maximization (EM) and PHASE algorithms were used to computationally reconstruct SNP haplotypes (HLA-G alleles). A new HLA-G allele, which originated in Amerindian populations by a crossing-over event between two widespread HLA-G alleles, was identified in 18 individuals. Neutrality tests evidenced that natural selection has a complex part in the HLA-G coding region. Although balancing selection is the type of selection that shapes variability at a local level (Native American populations), we have also shown that purifying selection may occur on a worldwide scale. Moreover, the balancing selection does not seem to act on the coding region as strongly as it acts on the flanking regulatory regions, and such coding signature may actually reflect a hitchhiking effect.Genes and Immunity advance online publication, 3 October 2013; doi:10.1038/gene.2013.47.
