Im Rahmen der vorliegenden Dissertation wurde die molekulare Evolution von Globinen in Amphibien und Teleostiern untersucht und Analysen zur Genexpression ausgewählter Globine durchgeführt. Die bisher besonders für die neueren Mitglieder der Superfamilie der Globine – Neuroglobin und Cytoglobin – schwerpunktmäßig in Mammaliern erbrachten Daten sollten durch die Analyse in Amphibien und Teleostiern auf ihre generelle Gültigkeit für Vertebraten überprüft werden. Die Analysen zur Genexpression wurden sowohl in silico, basierend auf genomischen wie EST-Daten, als auch experimentell durch qualitative und quantitative RT-PCR-Nachweise durchgeführt. Die mRNA-Lokalisation wurde durch in situ-Hybridisierungen an Gewebeschnitten beziehungsweise durch Whole mount in situ-Hybridisierung an ganzen Embryonen detektiert. In einem ersten Teil der Arbeit wurde das Globin-Repertoire von Xenopus tropicalis umfassend analysiert. Die Expressionsanalyse der gefundenen Globine umfasste nicht nur adulte Tiere, sonder erstmals auch detailliert die Entwicklungsstadien eines Vertebraten. Dabei wurde festgestellt, dass die vorwiegend neuronale Expression des streng konservierten Neuroglobins ein generelles Charakteristikum aller Tetrapoden ist und bereits in der frühembryonalen Entwicklung auftritt. Auch für das als Einzelkopie im Amphibiengenom vertretene Cytoglobin konnte eine strenge Sequenzkonservierung gezeigt werden. Das Expressionsmuster des Amphibien-Cytoglobins stimmte mit dem aus Mammaliern bekannten überein und zeigte konservierte Charakteristika dieses Globins bei Tetrapoden auf. Die Analyse des Xenopus-Genoms ergab zudem, dass Krallenfrösche nicht über Myoglobin verfügen. Genomische Vergleiche syntäner Genregionen ließen auf Rearrangements in diesem Genombereich im Verlauf der Evolution schließen, in deren Folge das Myoglobingen in den Krallenfröschen deletiert wurde. Die Hämoglobine wurden in Xenopus tropicalis erstmals in einem Amphibium umfassend analysiert. Die Gene zeigten demnach eine geclusterte Anordnung: der tropische Krallenfrosch verfügte über je ein funktionelles α- bzw. β-adultes und sieben bzw. vier α- bzw. β-larvale Hämoglobine, die während der Entwicklung bzw. in adulten Tieren charakteristisch exprimiert wurden. Die Analyse der Hämoglobine hinsichtlich ihrer Lage in einem Cluster, ihrer phylogenetischen Relation zueinander und nicht zuletzt ihres Expressionsmusters ließen Rückschlüsse auf ihre Evolution zu. Zusätzlich zu diesen bereits bekannten Globinen konnte im Rahmen dieser Dissertation das Globingen-Repertoirs von Xenopus um zwei weitere, bisher unbekannte Globine erweitert werden. Diese wurden entsprechend ihrer bisher unbekannten Funktion als GlobinX und GlobinY bezeichnet. Während GlobinY bisher ausschließlich in Amphibien nachgewiesen werden konnte, wurde GlobinX zudem in Teleostiern detektiert und repräsentiert damit ein auf Anamnia beschränktes Globin. Die rekombinante Proteinexpression von Neuroglobin, Cytoglobin, GlobinX und GlobinY des tropischen Krallenfrosches zeigte ein hexakoordiniertes Bindungsschema dieser Globine in ihrem Deoxy-Zustand. In einem zweiten Teil dieser Dissertation wurden Neuroglobin und Cytoglobin in Teleostiern untersucht und die Analyse für diese zwei Gene somit über die Tetrapoden hinaus auf den gesamten Stammbaum der Vertebraten ausgedehnt. Dabei wurde deutlich, dass die vorwiegend neuronale Expression des seit 420 Millionen Jahren streng konservierten Neuroglobins ein generelles Merkmal dieses Globins in allen Vertebraten ist. Der in Amphibien und Teleostiern erbrachte und mit Ergebnissen in Mammaliern übereinstimmende Nachweis von Neuroglobin in neuronalen Geweben mit einem hohen Stoffwechsel lässt derzeit eine Funktion dieses Globins im Sauerstoffmetabolimus als wahrscheinlich erscheinen. Ob Neuroglobin dabei als kurzzeitiger Sauerstoffspeicher, O2-Transoprter oder aber in der Detoxifikation reaktiver Sauerstoff- bzw. Stickstoffspezies agiert, bleibt zu untersuchen. Für Cytoglobin konnte eine offenbar alle Teleostier betreffende Genduplikation nachgewiesen werden. Phylogenetische Analysen zeigen die Monophylie der Vertebraten-Cytoglobine. Der Vergleich der paralogen Cytoglobine der Teleostier mit dem syntänen Genombereich des humanen Cytoglobins zeigte die wahrscheinliche Entstehung der Fisch-Cytoglobine durch eine Genomduplikation in einem Vorfahren aller Teleostier vor etwa 300-450 Millionen Jahren. Die paralogen Cytoglobine zeigten in Danio rerio und Tetraodon nigroviridis differierende, charakteristische Expressionsmuster, die mit der Theorie der Subfunktionalisierung von Genen in Folge eines Duplikationsereignisses kompatibel sind. Die Analyse zeigte, dass Cygb-1 prädominant in Gehirn und Herz exprimiert wurde, Cygb-2 hingegen bevorzugt in Gehirn und Auge. Dies bestätigte indirekt die Hypothese, nach der das Cytoglobin der Mammalier zwei unterschiedliche Funktionen in differenten Geweben wahrnimmt. Die rekombinante Expression von Cygb-1 des Zebrabärblings zeigte zudem, das auch dieses Globin in seiner Deoxy-Form über ein hexakoordiniertes Bindungsschema verfügt.

La tesi esamina il codice musicale Gr. Rés Vm7 676 della Biblioteca Nazionale di Parigi, che rappresenta una fonte di grande interesse per lo studio della musica vocale italiana tra Quattro e Cinquecento. Compilato nel 1502, il codice è stato oggetto di analisi da parte di vari studiosi, che ne hanno preso in esame singoli brani o intere sezioni, allo scopo di attestare procedimenti compositivi particolari (Torrefranca) o caratteri stilistici locali, in particolare relativi alla frottola mantovana e ferrarese (Prizer). Un’accurata ricognizione sul repertorio è stata effettuata da Nanie Bridgman in un saggio degli anni Cinquanta del secolo scorso, ma non è mai stato realizzato uno studio organico sul manoscritto. Pertanto la ricerca si è proposta di riconsiderare l’intero repertorio italiano tramandato dal codice, per proporre un plausibile inquadramento stilistico nella cultura della poesia per musica coeva. La trascrizione dei testi e delle musiche, supportata dal confronto con le fonti manoscritte e a stampa, letterarie e musicali, ha consentito di formulare alcune ipotesi in merito alla circolazione del repertorio tramandato e all’ambiente di produzione del documento. L’inconsueta varietà di forme musicali riscontrate nel codice consente inoltre di assumere questo manoscritto come una delle principali fonti della tradizione musicale che precede immediatamente la ‘sistemazione’ del repertorio frottolistico effettuata da Ottaviano Petrucci, a partire dal 1504, con la pubblicazione dei suoi undici libri di frottole (1501-1514).

Salivary gland proteins of Culicoides spp. have been suggested to be among the main allergens inducing IgE-mediated insect bite hypersensitivity (IBH), an allergic dermatitis of the horse. The aim of our study was to identify, produce and characterize IgE-binding salivary gland proteins of Culicoides nubeculosus relevant for IBH by phage surface display technology. A cDNA library constructed with mRNA derived from C. nubeculosus salivary glands was displayed on the surface of filamentous phage M13 and enriched for clones binding serum IgE of IBH-affected horses. Ten cDNA inserts encoding putative salivary gland allergens were isolated and termed Cul n 2 to Cul n 11. However, nine cDNA sequences coded for truncated proteins as determined by database searches. The cDNA sequences were amplified by PCR, subcloned into high level expression vectors and expressed as hexahistidine-tagged fusion proteins in Escherichia coli. Preliminary ELISA results obtained with these fusions confirmed the specific binding to serum IgE of affected horses. Therefore, the putative complete open reading frames derived from BLAST analyses were isolated by RACE-PCR and subcloned into expression vectors. The full length proteins expressed in Escherichia coli showed molecular masses in the range of 15.5-68.7 kDa in SDS-PAGE in good agreement with the masses calculated from the predicted protein sequences. Western blot analyses of all recombinant allergens with a serum pool of IBH-affected horses showed their ability to specifically bind serum IgE of sensitized horses, and ELISA determinations yielded individual horse recognition patterns with a frequency of sensitization ranging from 13 to 57%, depending on the allergen tested. The in vivo relevance of eight of the recombinant allergens was demonstrated in intradermal skin testing. For the two characterized allergens Cul n 6 and Cul n 11, sensitized horses were not available for intradermal tests. Control horses without clinical signs of IBH did not develop any relevant immediate hypersensitivity reactions to the recombinant allergens. The major contribution of this study was to provide a repertoire of recombinant salivary gland allergens repertoire from C. nubeculosus potentially involved in the pathogenesis of IBH as a starting basis for the development of a component-resolved serologic diagnosis of IBH and, perhaps, for the development of single horse tailored specific immunotherapy depending on their component-resolved sensitization patterns.

QUESTIONS UNDER STUDY: Vocal cord dysfunction (VCD) is characterised by recurrent attacks of paradoxical adduction of the vocal cords during inspiration, accompanied clinically by dyspnoea attacks with inspiratory stridor lasting between minutes and hours. The aim of the study is to evaluate the self-perception of patients suffering from VCD and how they deal with aggressivity. METHODS: The Giessen Test (GT) and the Picture Frustration Test (PFT) were used on 6 patients with VCD. RESULTS: Five variables show significant differences between the patients with VCD and the values of the normative groups. VCD-patients show an idealized image of themselves, ie relaxed, open, sociable, and capable of devotion. They do not reject accusations against them by others and rarely make self-criticism. In conflicts they behave passively, thus blocking their expression of aggressivity. CONCLUSIONS: In many cases organic disposition and trigger stimuli are thought to be responsible for provoking dyspnoea attacks in VCD. However, the self-perception of patients and their way of dealing with aggressivity strongly suggests that psychosocial factors play a great role in the development of VCD. This requires not only symptom-orientated therapy but also psychological counselling.

OBJECTIVE: To evaluate the effects of a single preoperative dose of steroid on thyroidectomy outcomes. BACKGROUND: Nausea, pain, and voice alteration frequently occur after thyroidectomy. Because steroids effectively reduce nausea and inflammation, a preoperative administration of steroids could improve these thyroidectomy outcomes. METHODS: Seventy-two patients (men = 20, women = 52) undergoing thyroidectomy for benign disease were included in this randomized, controlled, 2 armed (group D: 8 mg dexamethasone, n = 37; group C: 0.9% NaCl, n = 35), double-blinded study (clinical trial number NCT00619086). Anesthesia, surgical procedures, antiemetics, and analgesic treatments were standardized. Nausea (0-3), pain (visual analog scale), antiemetic and analgesic requirements, and digital voice recording were documented before and 4, 8, 16, 24, 36, and 48 hours after surgery. Patients were followed-up 30 days after hospital discharge. RESULTS: Baseline characteristics were similar among the 2 treatment groups. Nausea was pronounced in the first 16 hours postoperatively (scores were <0.3 and 0.8-1.0 for group D and C, respectively (P = 0.005)), and was significantly lower in group D compared with group C during the observation period (P = 0.001). Pain diminished within 48 hours after surgery (visual analog scale 20 and 35 in group D and C, respectively (P = 0.009)). Antiemetic and analgesic requirements were also significantly diminished. Changes in voice mean frequency were less prominent in the dexamethasone group compared with the placebo group (P = 0.015). No steroid-related complications occurred. CONCLUSION: A preoperative single dose of steroid significantly reduced nausea, vomiting, and pain, and improved postoperative voice function within the first 48 hours (most pronounced within 16 hours) after thyroid resection; this strategy should be routinely applied in thyroidectomies.

Maternal antibodies protect newborns whilst they are immunologically immature. This study shows that maternal antibodies can also shape the B cell repertoire of the offspring long after the maternal antibodies themselves become undetectable. V(H)DJ(H) gene-targeted (VI10) mice expressing a heavy chain specific for vesicular stomatitis virus (VSV) produce a 20-fold increased spontaneous titer of VSV-neutralizing antibodies. When transferred from mother to offspring, these antibodies prevented accumulation of Ag-specific transitional type 2 and marginal zone B cells with an activated phenotype and favored selection to the B cell follicles. This effect was B cell-intrinsic and lasted up to adulthood. The pups nursed by mothers producing specific antibodies developed higher endogenous antibody titers of this specificity which perpetuated the effects of specific B cell selection into the mature follicular compartment, presumably by blocking auto-Ag-dependent development of transitional type 2 B cells in the spleen. This repertoire change was functional, as following infection of adult mice with VSV, those pups that had received specific maternal antibodies as neonates had increased pre-immune titers and mounted strong early IgG neutralizing antibodies.

BACKGROUND: Specificities for carbohydrate IgG antibodies, thought to be predominantly of the IgG2 subclass, have never been broadly examined in healthy human subjects. OBJECTIVE: To examine commercial intravenous immunoglobulin (IVIG) preparations for their ability to recognize a wide range of glycans and to determine the contribution of IgG2 to the binding pattern observed. METHODS: We used a glycan microarray to evaluate IVIG preparations and a control mix of similar proportions of human myeloma IgG1 and IgG2 for binding to 377 glycans, courtesy of the Consortium for Functional Glycomics Core H. Glycans recognized were categorized using public databases for their likely cellular sources. IgG2 was depleted from IVIG by using immunoaffinity chromatography, and depletion was confirmed by using nephelometry and surface plasmon resonance. RESULTS: Nearly half of the glycans bound IgG. Some of the glycans with the greatest antibody binding can be found in structures of human pathogenic bacteria (eg, Streptococcus pneumoniae, Mycobacterium tuberculosis, Vibrio cholera) and nonpathogenic bacteria, including LPS and lipoteichoic acid, capsular polysaccharides, and exopolysaccharides. Surprisingly, depletion of IgG2 had only a modest effect on anticarbohydrate recognition patterns compared with the starting IVIG preparation. Little to no binding activity was detected to human endogenous glycans, including tumor-associated antigens. CONCLUSIONS: This novel, comprehensive analysis provides evidence that IVIG contains a much wider range than previously appreciated of anticarbohydrate IgG antibodies, including those recognizing both pathogenic and non-pathogen-associated prokaryotic glycans.
