This thesis has two major parts. The first part of the thesis will describe a high energy cosmic ray detector -- the High Energy Isotope Spectrometer Telescope (HEIST). HEIST is a large area (0.25 m2sr) balloon-borne isotope spectrometer designed to make high-resolution measurements of isotopes in the element range from neon to nickel (10 ≤ Z ≤ 28) at energies of about 2 GeV/nucleon. The instrument consists of a stack of 12 NaI(Tl) scintilla tors, two Cerenkov counters, and two plastic scintillators. Each of the 2-cm thick NaI disks is viewed by six 1.5-inch photomultipliers whose combined outputs measure the energy deposition in that layer. In addition, the six outputs from each disk are compared to determine the position at which incident nuclei traverse each layer to an accuracy of ~2 mm. The Cerenkov counters, which measure particle velocity, are each viewed by twelve 5-inch photomultipliers using light integration boxes.

HEIST-2 determines the mass of individual nuclei by measuring both the change in the Lorentz factor (Δγ) that results from traversing the NaI stack, and the energy loss (ΔΕ) in the stack. Since the total energy of an isotope is given by Ε = γM, the mass M can be determined by M = ΔΕ/Δγ. The instrument is designed to achieve a typical mass resolution of 0.2 amu.

The second part of this thesis presents an experimental measurement of the isotopic composition of the fragments from the breakup of high energy 40Ar and 56Fe nuclei. Cosmic ray composition studies rely heavily on semi-empirical estimates of the cross-sections for the nuclear fragmentation reactions which alter the composition during propagation through the interstellar medium. Experimentally measured yields of isotopes from the fragmentation of 40Ar and 56Fe are compared with calculated yields based on semi-empirical cross-section formulae. There are two sets of measurements. The first set of measurements, made at the Lawrence Berkeley Laboratory Bevalac using a beam of 287 MeV/nucleon 40Ar incident on a CH2 target, achieves excellent mass resolution (σm ≤ 0.2 amu) for isotopes of Mg through K using a Si(Li) detector telescope. The second set of measurements, also made at the Lawrence Berkeley Laboratory Bevalac, using a beam of 583 MeV/nucleon 56FeFe incident on a CH2 target, resolved Cr, Mn, and Fe fragments with a typical mass resolution of ~ 0.25 amu, through the use of the Heavy Isotope Spectrometer Telescope (HIST) which was later carried into space on ISEE-3 in 1978. The general agreement between calculation and experiment is good, but some significant differences are reported here.

The Barton laboratory has established that octahedral rhodium complexes bearing the sterically expansive 5,6-chrysene diimine ligand can target thermodynamically destabilized sites, such as base pair mismatches, in DNA with high affinity and selectivity. These complexes approach DNA from the minor groove, ejecting the mismatched base pairs from the duplex in a binding mode termed metalloinsertion. In recent years, we have shown that these metalloinsertor complexes also exhibit cytotoxicity preferentially in cancer cells that are deficient in the mismatch repair (MMR) machinery.

Here, we establish that a sensitive structure-activity relationship exists for rhodium metalloinsertors. We studied the relationship between the chemical structures of metalloinsertors and their effect on biological activity for ten complexes with similar DNA binding affinities, but wide variation in their lipophilicity. Drastic differences were observed in the selectivities of the complexes for MMR-deficient cells. Compounds with hydrophilic ligands were highly selective, exhibiting preferential cytotoxicity in MMR-deficient cells at low concentrations and short incubation periods, whereas complexes with lipophilic ligands displayed poor cell-selectivity. It was discovered that all of the complexes localized to the nucleus in concentrations sufficient for mismatch binding; however, highly lipophilic complexes also exhibited high mitochondrial uptake. Significantly, these results support the notion that mitochondrial DNA is not the desired target for our metalloinsertor complexes; instead, selectivity stems from targeting mismatches in genomic DNA.

We have also explored the potential for metalloinsertors to be developed into more complex structures with multiple functionalities that could either enhance their overall potency or impart mismatch selectivity onto other therapeutic cargo. We have constructed a family of bifunctional metalloinsertor conjugates incorporating cis-platinum, each unique in its chemical structure, DNA binding interactions, and biological activity. The study of these complexes in MMR-deficient cells has established that the cell-selective biological activity of rhodium metalloinsertors proceeds through a critical cellular pathway leading to necrosis.

We further explored the underlying mechanisms surrounding the biological response to mismatch recognition by metalloinsertors in the genome. Immunofluorescence assays of MMR-deficient and MMR-proficient cells revealed that a critical biomarker for DNA damage, phosphorylation of histone H2AX (γH2AX) rapidly accumulates in response to metalloinsertor treatment, signifying the induction of double strand breaks in the genome. Significantly, we have discovered that our metalloinsertor complexes selectively inhibit transcription in MMR-deficient cells, which may be a crucial checkpoint in the eventual breakdown of the cell via necrosis. Additionally, preliminary in vivo studies have revealed the capability of these compounds to traverse the complex environments of multicellular organisms and accumulate in MMR-deficient tumors. Our ever-increasing understanding of metalloinsertors, as well as the development of new generations of complexes both monofunctional and bifunctional, enables their continued progress into the clinic as promising new chemotherapeutic agents.

The propagation of cosmic rays through interstellar space has been investigated with the view of determining what particles can traverse astronomical distances without serious loss of energy. The principal method of loss of energy of high energy particles is by interaction with radiation. It is found that high energy (1013-1018ev) electrons drop to one-tenth their energy in 108 light years in the radiation density in the galaxy and that protons are not significantly affected in this distance. The origin of the cosmic rays is not known so that various hypotheses as to their origin are examined. If the source is near a star it is found that the interaction of electrons and photons with the stellar radiation field and the interaction of electrons with the stellar magnetic field limit the amount of energy which these particles can carry away from the star. However, the interaction is not strong enough to affect the energy of protons or light nuclei appreciably. The chief uncertainty in the results is due to the possible existence of general galactic magnetic field. The main conclusion reached is that if there is a general galactic magnetic field, then the primary spectrum has very few photons, only low energy (˂ 1013 ev) electrons and the higher energy particles are primarily protons regardless of the source mechanism, and if there is no general galactic magnetic field, then the source of cosmic rays accelerates mainly protons and the present rate of production is much less than that in the past.

Background: Two distinct trends are emerging with respect to how data is shared, collected, and analyzed within the bioinformatics community. First, Linked Data, exposed as SPARQL endpoints, promises to make data easier to collect and integrate by moving towards the harmonization of data syntax, descriptive vocabularies, and identifiers, as well as providing a standardized mechanism for data access. Second, Web Services, often linked together into workflows, normalize data access and create transparent, reproducible scientific methodologies that can, in principle, be re-used and customized to suit new scientific questions. Constructing queries that traverse semantically-rich Linked Data requires substantial expertise, yet traditional RESTful or SOAP Web Services cannot adequately describe the content of a SPARQL endpoint. We propose that content-driven Semantic Web Services can enable facile discovery of Linked Data, independent of their location. Results: We use a well-curated Linked Dataset - OpenLifeData - and utilize its descriptive metadata to automatically configure a series of more than 22,000 Semantic Web Services that expose all of its content via the SADI set of design principles. The OpenLifeData SADI services are discoverable via queries to the SHARE registry and easy to integrate into new or existing bioinformatics workflows and analytical pipelines. We demonstrate the utility of this system through comparison of Web Service-mediated data access with traditional SPARQL, and note that this approach not only simplifies data retrieval, but simultaneously provides protection against resource-intensive queries. Conclusions: We show, through a variety of different clients and examples of varying complexity, that data from the myriad OpenLifeData can be recovered without any need for prior-knowledge of the content or structure of the SPARQL endpoints. We also demonstrate that, via clients such as SHARE, the complexity of federated SPARQL queries is dramatically reduced.

A presente tese investiga as poéticas do trágico nas obras teatrais de Eurípides (Grécia, c. 484 406 a.C.), Johann Wolfgang von Goethe (Alemanha, 1749 1832) e Federico García Lorca (Espanha, 1898 1936) e defende que, nos três autores, as concepções estéticas do trágico se constituem principalmente sobre a representação poética do filicídio materno. A antinomia trágica engendrada, metaforicamente, no assassinato da criança pelas mãos daquela que lhe deu a vida é tema de Medeia (431 a.C.), Tragédia de Margarida (1790) e Yerma (1934), obras fundamentais para compreender a visada trágica dos três poetas aqui, em respectivo, estudados. O conflito com o sagrado e com a razão, ao apontar para a afirmação trágica do corpo e do feminino, frequenta as três obras. O paralelismo entre as dimensões política e estética é, por conseguinte, patente nos três dramas, ao mesmo tempo em que cada um dos autores, com contexto e assinatura próprios, configura uma ideia estética acerca do trágico inteiramente singular. O diálogo entre literatura e filosofia, ou entre intuição e conceito, atravessa, nesta tese, a leitura do trágico na metáfora do filicídio. Sob tal perspectiva, a Medeia de Eurípides impõe-se no centro do debate entre socráticos e sofistas, e aborda temas, como o domínio das paixões sobre a razão, que também aliciaram autores como Platão, Aristóteles e Nietzsche. A Gretchentragödie, de Goethe, apresenta-se, por sua vez, como obra poética aonde convergem as mais calorosas discussões estéticas do moderno pensamento alemão, como as questões do sublime (Kant, Schiller) e da vontade (Schopenhauer). Yerma, de García Lorca, será também uma obra de convergência filosófica, expressando a nueva manera espiritualista que marca a última fase da produção lorquiana: a perspectiva trágica de Nietzsche, na afirmação do corpo como grande razão, assim como o diálogo de Lorca com o pensamento de Miguel de Unamuno sobre El sentimiento trágico de la vida, caracteriza uma espécie de tragédia às avessas, que nega o sagrado e afirma o trágico como síntese libertária do eu, do corpo e do feminino

Ao entrarmos em contato com a obra de Gilles Deleuze, não podemos ignorar a influência de Antonin Artaud e da idéia de Crueldade no desenvolvimento de sua filosofia. Assim, cabe a pergunta: Qual é a importância de Antonin Artaud e do Teatro da Crueldade na constituição da Filosofia Contemporânea? A presente dissertação não pretende responder esta pergunta em toda sua complexidade, a sua pretensão é mais modesta, ela visa traçar algumas linhas que possam nos orientar nas relações complexas entre o pensamento de Artaud e a Filosofia, entre o Teatro da Crueldade e o Teatro Filosófico. Assim, partimos da idéia de uma Metafísica da Crueldade, tal como nos é apresentada por Camille Dumoulié, para uma Metafísica Cruel, uma metafísica que já é ela mesma uma experimentação. Pensar a crueldade como um teatro metafísico põe em crise o Sistema da Representação: tarefa do pensamento contemporâneo. Dessa forma, compreender a Filosofia e a Arte Contemporâneas torna Antonin Artaud uma passagem obrigatória.

Esta tese se propõe a explorar as possibilidades de lidar com o trabalho enquanto atividade, compreendendo-o, a partir da perspectiva ergológica (SCHWARTZ, 2000), portando um paradoxo ou um tipo de dialética entre o Registro 1 das normas antecedentes que marcam fortemente a antecipação desse trabalho; e o Registro 2 das dimensões singulares do encontro de encontros, que se produz em todo trabalhar. A partir da permanente dialética entre esses dois registros, a perspectiva ergológica vai buscar métodos que sejam capazes de dar visibilidade à dimensão gestionária que atravessa todo trabalhar, para que os protagonistas da atividade possam se apropriar de sua atividade, individual e coletivamente, ampliando seus níveis de autonomia frente ao trabalho e à vida. O termo ergo(trans)formação afirma uma prática de pesquisa-intervenção que se propõe transformadora da realidade, apostando numa intervenção micropolítica no seio da experiência social, conforme Rocha e Aguiar (2003). Este modo de compreensão de pesquisa e de afirmação da produção coletiva do conhecimento pauta-se numa abordagem construtivista onde conhecer não é representar uma realidade pré-existente, mas é um processo de invenção de si e do mundo (KASTRUP, 1999), em um acoplamento direto com a matéria. Enfrentamos o seguinte problema: o trabalho pode ser pensado como invenção de si e do mundo, entrando em curto-circuitagem com as formas de produzir em conformidade com organização formal do trabalho, afirmando o primado do agir próprio do trabalhador como atividade, frente ao acontecimental do trabalho? Para dar conta deste problema, propomos desenvolver uma cartografia, enquanto método de análise da atividade de trabalho, que possibilite acompanhar a curto-circuitagem entre as formas de produzir em conformidade com organização do trabalho e o agir próprio do trabalhador. São apresentadas duas experimentações de ergo(trans)formação desenvolvidas em dois territórios distintos dos mundos do trabalho: o desenvolvimento de uma Oficina de Vendas e a formação de educadores internos, em empresa financeira pública e a possibilidade de formação educacional e profissional de jovens com transtorno mental grave em uma rede de restaurantes que se desdobra na cartografia do trabalho coletivo e do coletivo de trabalho, tendo um jovem portador de síndrome de Down como protagonista da atividade. As escolhas metodológicas se fizeram em duas perspectivas distintas e complementares: (1) o método cartográfico elaborado a partir de Deleuze e Guattari (1995), Guattari e Rolnik (1986), assim como Passos, Kastrup e Escóssia (2009); e (2) o que propõe Schwartz (1999), o Dispositivo Dinâmico de Três Polos (DD3P), incorporando a reformulação conceitual proposta por Athayde e Brito (2003) denominada Comunidade Ampliada de Pesquisa. Como conclusão, refletimos se é possível pensar a atividade inventiva de trabalho, a partir das experimentações de ergo(trans)formação que apresentamos, considerando suas condições e efeitos e, mesmo, se podemos nomeá-las assim, levando-se em conta as diferenças de atividades envolvidas. Estas experimentações nos impuseram pensar o trabalho (enquanto atividade) em sua indissociabilidade com a formação humana e com a produção de subjetividade

Many of British rivers hold stocks of salmon (Salmo salar L.) and sea trout (Salmo trutta L.) and during most of the year some of the adult fish migrate upstream to the head waters where, with the advent of winter, they will eventually spawn. For a variety of reasons, including the generation of power for milling, improving navigation and measuring water flow, man has put obstacles in the way of migratory fish which have added to those already provided by nature in the shape of rapids and waterfalls. While both salmon and sea trout, particularly the former, are capable of spectacular leaps the movement of fish over man-made and natural obstacles can be helped, or even made possible, by the judicious use of fish passes. These are designed to give the fish an easier route over or round an obstacle by allowing it to overcome the water head difference in a series of stages ('pool and traverse' fish pass) or by reducing the water velocity in a sloping channel (Denil fish pass). Salmon and sea trout make their spawning runs at different flow conditions, salmon preferring much higher water flows than sea trout. Hence the design of fish passes requires an understanding of the swimming ability of fish (speed and endurance) and the effect of water temperature on this ability. Also the unique features of each site must be appreciated to enable the pass to be positioned so that its entrance is readily located. As well as salmon and sea trout, rivers often have stocks of coarse fish and eels. Coarse fish migrations are generally local in character and although some obstructions such as weirs may allow downstream passages only, they do not cause a significant problem. Eels, like salmon and sea trout, travel both up and down river during the course of their life histories. However, the climbing power of elvers is legendary and it is not normally necessary to offer them help, while adult silver eels migrate at times of high water flow when downstream movement is comparatively easy: for these reasons neither coarse fish nor eels are considered further. The provision of fish passes is, in many instances, mandatory under the Salmon and Freshwater Fisheries Act 1975. This report is intended for those involved in the planning, siting, construction and operation of fish passes and is written to clarify the hydraulic problems for the biologist and the biological problems for the engineer. It is also intended to explain the criteria by which the design of an individual pass is assessed for Ministerial Approval.

This monograph on the ecology of Atlantic white cedar wetlands is one of a series of U.S. Fish and Wildlife Service profiles of important freshwater wetland ecosystems of the United States. The purpose of the profile is to describe the extent, components, functioning, history, and treatment of these wetlands. It is intended to provide a useful reference to relevant scientific information and a synthesis of the available literature. The world range of Atlantic white cedar (Chamaecyparis thyoides) is limited to a ribbon of freshwater wetlands within 200 km of the Atlantic and Gulf coasts of the United States, extending from mid-Maine to mid-Florida and Mississippi. Often in inaccessible sites and difficult to traverse, cedar wetlands contain distinctive suites of plant species. Highly valued as commercial timber since the early days of European colonization of the continent, the cedar and its habitat are rapidly disappearing. This profile describes the Atlantic white cedar and the bogs and swamps it dominates or codominates throughout its range, discussing interrelationships with other habitats, putative origins and migration patterns, substrate biogeochemistry, associated plant and animal species (with attention to those that are rare, endangered, or threatened regionally or nationally), and impacts of both natural and anthropogenic disturbance. Research needs for each area are outlined. Chapters are devoted to the practices and problems of harvest and management, and to an examination of a large preserve recently acquired by the USFWS, the Alligator River National Wildlife Refuge in North Carolina.

Esta dissertação tem como objetivo identificar e analisar os principais debates no contexto histórico que antecedeu a votação da primeira lei de direitos autorais no Brasil, em meados de 1898. Devido à abrangência do tema, a pesquisa debruçou-se sobre o direito autoral literário. Esse debate se processou de forma intermitente, ao longo da segunda metade do século XIX, envolvendo alguns nomes da política e da literatura nacional. Nesse sentido, o pilar central do trabalho pautou-se na análise de projetos de lei e debates registrados em Anais da Câmara dos Deputados, ou seja, de iniciativas e atos de fala formulados sobre autoria e suas circunstâncias morais e econômicas. A fim de contribuir para a contextualização do debate político sobre direitos autorais, foi analisada brevemente a relação entre o escritor de textos literários e o editor. Recorrendo a crônicas, contratos de edição, recibos e processos judiciais, pretendeu-se enriquecer a discussão sobre posicionamentos e práticas dessas duas personagens históricas. O viés interpretativo, embasado na análise dessas fontes, possibilitou pensar a dimensão que a discussão sobre os direitos autorais assumiu no respectivo período. Longe de ter sido um debate restrito ao mundo literário e editorial, ele ganhou o espaço político, trazendo à tona experiências, expectativas e contradições.

Boat wakes in the Atlantic Intracoastal Waterway (AIWW) of North Carolina occur in environments not normally subjected to (wind) wave events, making sections of AIWW potentially vulnerable to extreme wave events generated by boat wakes. The Snow’s Cut area that links the Cape Fear River to the AIWW is an area identified by the Wilmington District of the U.S. Army Corps of Engineers as having significant erosion issues; it was hypothesized that this erosion could be being exacerbated by boat wakes. We compared the boat wakes for six combinations of boat length and speed with the top 5% wind events. We also computed the benthic shear stress associated with boat wakes and whether sediment would move (erode) under those conditions. Finally, we compared the transit time across Snow’s Cut for each speed. We focused on two size classes of V-hulled boats (7 and 16m) representative of AIWW traffic and on three boat speeds (3, 10 and 20 knots). We found that at 10 knots when the boat was plowing and not yet on plane, boat wake height and potential erosion was greatest. Wakes and forecast erosion were slightly mitigated at higher, planing speeds. Vessel speeds greater than 7 knots were forecast to generate wakes and sediment movement zones greatly exceeding that arising from natural wind events. We posit that vessels larger than 7m in length transiting Snow’s Cut (and likely many other fetch-restricted areas of the AIWW) frequently generate wakes of heights that result in sediment movement over large extents of the AIWW nearshore area, substantially in exceedance of natural wind wave events. If the speed, particularly of large V-hulled vessels (here represented by the 16m length class), were reduced to pre-plowing levels (~ 7 knots down from 20), transit times for Snow’s Cut would be increased approximately 10 minutes but based on our simulations would likely substantially reduce the creation of erosion-generating boat wakes. It is likely that boat wakes significantly exceed wind wave background for much of the AIWW and similar analyses may be useful in identifying management options.

Shear layers shed by aircraft wings roll up into vortices. A similar, though far less common, phenomenon can occur in the wake of a turbomachine blade. This paper presents experimental data from a new single stage turbine that has been commissioned at the Whittle Laboratory. Two low aspect ratio stators have been tested with the same rotor row. Surface flow visualisation illustrates the extremely strong secondary flows present in both NGV designs. These secondary flows lead to conventional passage vortices but also to an intense vortex sheet which is shed from the trailing edge of the blades. Pneumatic probe traverse show how this sheet rolls up into a concentrated vortex in the second stator design, but not in the first. A simple numerical experiment is used to model the shear layer instability and the effects of trailing edge shape and exit yaw angle distribution are investigated. It is found that the latter has a strong influence on shear layer rollup: inhibiting the formation of a vortex downstream of NGV 1 but encouraging it behind NGV 2.

Many types of oceanic physical phenomena have a wide range in both space and time. In general, simplified models, such as shallow water model, are used to describe these oceanic motions. The shallow water equations are widely applied in various oceanic and atmospheric extents. By using the two-layer shallow water equations, the stratification effects can be considered too. In this research, the sixth-order combined compact method is investigated and numerically implemented as a high-order method to solve the two-layer shallow water equations. The second-order centered, fourth-order compact and sixth-order super compact finite difference methods are also used to spatial differencing of the equations. The first part of the present work is devoted to accuracy assessment of the sixth-order super compact finite difference method (SCFDM) and the sixth-order combined compact finite difference method (CCFDM) for spatial differencing of the linearized two-layer shallow water equations on the Arakawa's A-E and Randall's Z numerical grids. Two general discrete dispersion relations on different numerical grids, for inertia-gravity and Rossby waves, are derived. These general relations can be used for evaluation of the performance of any desired numerical scheme. For both inertia-gravity and Rossby waves, minimum error generally occurs on Z grid using either the sixth-order SCFDM or CCFDM methods. For the Randall's Z grid, the sixth-order CCFDM exhibits a substantial improvement , for the frequency of the barotropic and baroclinic modes of the linear inertia-gravity waves of the two layer shallow water model, over the sixth-order SCFDM. For the Rossby waves, the sixth-order SCFDM shows improvement, for the barotropic and baroclinic modes, over the sixth-order CCFDM method except on Arakawa's C grid. In the second part of the present work, the sixth-order CCFDM method is used to solve the one-layer and two-layer shallow water equations in their nonlinear form. In one-layer model with periodic boundaries, the performance of the methods for mass conservation is compared. The results show high accuracy of the sixth-order CCFDM method to simulate a complex flow field. Furthermore, to evaluate the performance of the method in a non-periodic domain the sixth-order CCFDM is applied to spatial differencing of vorticity-divergence-mass representation of one-layer shallow water equations to solve a wind-driven current problem with no-slip boundary conditions. The results show good agreement with published works. Finally, the performance of different schemes for spatial differencing of two-layer shallow water equations on Z grid with periodic boundaries is investigated. Results illustrate the high accuracy of combined compact method.
