The retinoid orphan-related receptor-alpha (RORalpha) is a member of the ROR subfamily of orphan receptors and acts as a constitutive activator of transcription in the absence of exogenous ligands. To understand the basis of this activity, we constructed a homology model of Rill using the closely related TRbeta as a template. Molecular modeling suggested that bulky hydrophobic side chains occupy the RORa ligand cavity leaving a small but distinct cavity that may be involved in receptor stabilization. This model was subject to docking simulation with a receptor-interacting peptide from the steroid receptor coactivator, GR-interacting protein-1, which delineated a coactivator binding surface consisting of the signature motif spanning helices 3-5 and helix 12 [activation function 2 (AF2)]. Probing this surface with scanning alanine mutagenesis showed structural and functional equivalence between homologous residues of RORalpha and TRbeta. This was surprising (given that Rill is a ligand-independent activator, whereas TRbeta has an absolute requirement for ligand) and prompted us to use molecular modeling to identify differences between Rill and TRbeta in the way that the All helix interacts with the rest of the receptor. Modeling highlighted a nonconserved amino acid in helix 11 of RORa (Phe491) and a short-length of 3.10 helix at the N terminus of AF2 which we suggest i) ensures that AF2 is locked permanently in the holoconformation described for other liganded receptors and thus 2) enables ligand-independent recruitment of coactivators. Consistent with this, mutation of RORa Phe491 to either methionine or alanine (methionine is the homologous residue in TRbeta), reduced and ablated transcriptional activation and recruitment of coactivators, respectively. Furthermore, we were able to reconstitute transcriptional activity for both a deletion mutant of Ill lacking All and Phe491 Met, by overexpression of a GAL-AF2 fusion protein, demonstrating ligand-independent recruitment of AF2 and a role for Phe491 in recruiting AF2.

In order to understand the earthquake nucleation process, we need to understand the effective frictional behavior of faults with complex geometry and fault gouge zones. One important aspect of this is the interaction between the friction law governing the behavior of the fault on the microscopic level and the resulting macroscopic behavior of the fault zone. Numerical simulations offer a possibility to investigate the behavior of faults on many different scales and thus provide a means to gain insight into fault zone dynamics on scales which are not accessible to laboratory experiments. Numerical experiments have been performed to investigate the influence of the geometric configuration of faults with a rate- and state-dependent friction at the particle contacts on the effective frictional behavior of these faults. The numerical experiments are designed to be similar to laboratory experiments by DIETERICH and KILGORE (1994) in which a slide-hold-slide cycle was performed between two blocks of material and the resulting peak friction was plotted vs. holding time. Simulations with a flat fault without a fault gouge have been performed to verify the implementation. These have shown close agreement with comparable laboratory experiments. The simulations performed with a fault containing fault gouge have demonstrated a strong dependence of the critical slip distance D-c on the roughness of the fault surfaces and are in qualitative agreement with laboratory experiments.

Telemedicine promises to revolutionize medical care delivery in rural and remote areas. The ability to accurately evaluate physical impairment via the Internet is important to the possible future provision of Internet-based physiotherapy. This study evaluated the reliability and validity of assessing knee range of motion via the Internet. Two therapists evaluated knee angle on a single subject via two methods of assessment: the Internet and the traditional method (face-to-face). Nine random positions of the knee were chosen with the principal examiner performing 20 face-to-face and two sets of 20 Internet measures in each position (n=540). The secondary therapist performed Internet assessments only. The Internet connection was established at a readily available speed of 17 kbit/s. The Internet-based goniometer was found to be a valid tool for measuring both knee flexion and extension angles. It was shown to possess both high intra and inter-rater reliability. Difference average plots of the scores verified the consistency of measurement between both modes of assessment. The successful evaluation of the physical outcome measure of knee range of motion via the Internet assists the further development of Internet-based physiotherapy applications. Skin-friction measurements are reported for high-enthalpy and high-Mach-number laminar, transitional and turbulent boundary layers. The measurements were performed in a free-piston shock tunnel with air-flow Mach number, stagnation enthalpy and Reynolds numbers in the ranges of 4.4-6.7, 3-13 MJ kg(-1) and 0.16 x 10(6)-21 x 10(6), respectively. Wall temperatures were near 300 K and this resulted in ratios of wall enthalpy to flow-stagnation enthalpy in the range of 0.1-0.02. The experiments were performed using rectangular ducts. The measurements were accomplished using a new skin-friction gauge that was developed for impulse facility testing. The gauge was an acceleration compensated piezoelectric transducer and had a lowest natural frequency near 40 kHz. Turbulent skin-friction levels were measured to within a typical uncertainty of +/-7%. The systematic uncertainty in measured skin-friction coefficient was high for the tested laminar conditions; however, to within experimental uncertainty, the skin-friction and heat-transfer measurements were in agreement with the laminar theory of van Driest (1952). For predicting turbulent skin-friction coefficient, it was established that, for the range of Mach numbers and Reynolds numbers of the experiments, with cold walls and boundary layers approaching the turbulent equilibrium state, the Spalding & Chi (1964) method was the most suitable of the theories tested. It was also established that if the heat transfer rate to the wall is to be predicted, then the Spalding & Chi (1964) method should be used in conjunction with a Reynolds analogy factor near unity. If more accurate results are required, then an experimentally observed relationship between the Reynolds analogy factor and the skin-friction coefficient may be applied.

For dynamic simulations to be credible, verification of the computer code must be an integral part of the modelling process. This two-part paper describes a novel approach to verification through program testing and debugging. In Part 1, a methodology is presented for detecting and isolating coding errors using back-to-back testing. Residuals are generated by comparing the output of two independent implementations, in response to identical inputs. The key feature of the methodology is that a specially modified observer is created using one of the implementations, so as to impose an error-dependent structure on these residuals. Each error can be associated with a fixed and known subspace, permitting errors to be isolated to specific equations in the code. It is shown that the geometric properties extend to multiple errors in either one of the two implementations. Copyright (C) 2003 John Wiley Sons, Ltd.

Esta dissertação apresenta o desenvolvimento de uma plataforma multimodal de aquisição e processamento de sinais. O projeto proposto insere-se no contexto do desenvolvimento de interfaces multimodais para aplicação em dispositivos robóticos cujo propósito é a reabilitação motora adaptando o controle destes dispositivos de acordo com a intenção do usuário. A interface desenvolvida adquire, sincroniza e processa sinais eletroencefalográficos (EEG), eletromiográficos (EMG) e sinais provenientes de sensores inerciais (IMUs). A aquisição dos dados é feita em experimentos realizados com sujeitos saudáveis que executam tarefas motoras de membros inferiores. O objetivo é analisar a intenção de movimento, a ativação muscular e o início efetivo dos movimentos realizados, respectivamente, através dos sinais de EEG, EMG e IMUs. Para este fim, uma análise offline foi realizada. Nessa análise, são utilizadas técnicas de processamento dos sinais biológicos e técnicas para processar sinais provenientes de sensores inerciais. A partir destes, os ângulos da articulação do joelho também são aferidos ao longo dos movimentos. Um protocolo experimental de testes foi proposto para as tarefas realizadas. Os resultados demonstraram que o sistema proposto foi capaz de adquirir, sincronizar, processar e classificar os sinais combinadamente. Análises acerca da acurácia dos classificadores utilizados mostraram que a interface foi capaz de identificar intenção de movimento em 76, 0 ± 18, 2% dos movimentos. A maior média de tempo de antecipação ao movimento foi obtida através da análise do sinal de EEG e foi de 716, 0±546, 1 milisegundos. A partir da análise apenas do sinal de EMG, este valor foi de 88, 34 ± 67, 28 milisegundos. Os resultados das etapas de processamento dos sinais biológicos, a medição dos ângulos da articulação, bem como os valores de acurácia e tempo de antecipação ao movimento se mostraram em conformidade com a literatura atual relacionada.

Visando a aplicação do aço UNS S32304 em embalados para transporte de material radioativo, amostras soldadas por processo TIG, com diferentes gases de proteção, foram submetidas a tratamentos térmicos nas temperaturas de 475°C, 600°C e 750°C por 8 horas, seguidas de resfriamento ao ar, a fim de analisar o efeito de temperaturas críticas no perfil de tensões residuais e microestrutura. A difratometria de raios X foi utilizada para determinação das tensões residuais, em diferentes condições (amostras como recebidas e apenas tratadas termicamente) e o perfil de tensões residuais total das amostras soldadas é apresentado para cada fase (austenita e ferrita). A rotação de culturas é uma prática essencial, no cultivo de hortaliças, para controle de pragas e doenças e tem sido usada, também, visando ao aproveitamento dos resíduos de adubação. O presente trabalho teve por objetivo avaliar o efeito residual da adubação orgânica, aplicada na cultura do quiabo, sobre o rendimento de espigas de milho-verde em cultivo subsequente. O quiabeiro foi plantado com duas populações de plantas e adubado com biofertilizante líquido de suínos, nas doses: 0, 6, 12, 24, e 48 m³ ha-1. Após a retirada dos restos culturais do quiabeiro, sementes de milho híbrido AG 1051 e da variedade UFVM 100 foram semeadas, em linhas duplas, no espaçamento de 1,0 m x 0,40 m e 0,25 m entre plantas após o desbaste. O delineamento experimental foi em blocos casualizados com quatro repetições, no esquema fatorial 2 x 5, sendo dois cultivares de milho e resíduo de cinco doses de biofertilizante. O resíduo da adubação com biofertilizante de suínos em quiabeiro não foi suficiente para nutrir as plantas de milho-verde; consequentemente obteve-se baixa produtividade comercial. O plantio do milho-verde em sucessão ao quiabeiro, visando à rotação de culturas e aproveitamento de resíduos orgânicos, é promissor, sendo necessária adubação complementar de cobertura, para suprimento de N e K. O híbrido AG 1051 apresentou maior altura de plantas, número e produtividade de espigas comerciais despalhadas e peso médio de espigas comerciais. A variedade UFVM100 apresentou maior teor de P e K nas folhas, número de espigas com palha, número e produtividade de espigas não comerciais despalhadas.

Laparoscopy is a surgical procedure on which operations in the abdomen are performed through small incisions using several specialized instruments. The laparoscopic surgery success greatly depends on surgeon skills and training. To achieve these technical high-standards, different apprenticeship methods have been developed, many based on in vivo training, an approach that involves high costs and complex setup procedures. This paper explores Virtual Reality (VR) simulation as an alternative for novice surgeons training. Even though several simulators are available on the market claiming successful training experiences, their use is extremely limited due to the economic costs involved. In this work, we present a low-cost laparoscopy simulator able to monitor and assist the trainee’s surgical movements. The developed prototype consists of a set of inexpensive sensors, namely an accelerometer, a gyroscope, a magnetometer and a flex sensor, attached to specific laparoscopic instruments. Our approach allows repeated assisted training of an exercise, without time constraints or additional costs, since no human artificial model is needed. A case study of our simulator applied to instrument manipulation practice (hand-eye coordination) is also presented.

Recent progresses in the software development world has assisted a change in hardware from heavy mainframes and desktop machines to unimaginable small devices leading to the prophetic "third computing paradigm", Ubiquitous Computing. Still, this novel unnoticeable devices lack in various capabilities, like computing power, storage capacity and human interface. Connectivity associated to this devices is also considered an handicap which comes generally associated expensive and limited protocols like GSM and UMTS. Considering this scenario as background, this paper presents a minimal communication protocol introducing better interfaces for limited devices. Special attention has been paid to the limitations of connectivity, storage capacity and scalability of the developed software applications. Este trabalho foi desenvolvido em campo, com o objetivo de avaliar a atividade residual de herbicidas recomendados para a cultura da cana-de-açúcar, quando aplicados ao solo na época seca. Os tratamentos foram organizados em esquema de parcelas subdivididas, em que, no âmbito das parcelas, foram aplicados seis herbicidas e, no âmbito das subparcelas, foram considerados três períodos de permanência dos produtos no solo (130 dias, 70 dias e testemunha, sem aplicação). Os tratamentos herbicidas foram (g ha-1): amicarbazone a 1260; clomazone a 1250; hexazinone a 450; imazapic a 147; isoxaflutole a 187,5; e sulfentrazone a 900. Além da infestação natural de plantas daninhas, para avaliação da atividade residual dos produtos, semeou-se pepino na área, considerado como bioindicador. A mesma molécula herbicida teve avaliação diferenciada em razão da espécie bioindicadora considerada. Os herbicidas amicarbazone, hexazinone, isoxaflutole e sulfentrazone têm propriedades positivas para aplicação ao solo em época seca, tais como elevada solubilidade, baixo Kow e baixa fotodegradação. A aplicação de imazapic em pré-emergência tem eficácia consistente em época seca, controlando adequadamente o pepino e a comunidade natural de plantas daninhas. O herbicida clomazone não controlou adequadamente o pepino e a comunidade natural de plantas daninhas.
