O crescente consumo de bebidas com elevado teor de cafeína pode resultar no aparecimento de sintomas provenientes do transtorno de ansiedade induzida por essa droga. Atualmente, tem-se utilizado a cafeína como um indutor farmacológico do comportamento tipo ansiedade e essa indução pode facilitar a melhor compreensão da relação entre alterações comportamentais e os mecanismos de ação envolvidos nesse efeito, portanto o presente trabalho propôs que a via nitrérgica poderia ser um mecanismo chave para explicar os efeitos comportamentais produzidos pela cafeína e que esses efeitos poderiam ser revertidos por um antioxidante, logo, no presente trabalho nós tivemos como objetivo avaliar o possível efeito do L-NAME e do α-tocoferol no comportamento tipo ansiedade ampliado pela cafeína nos testes de preferência claro/escuro (PCE) e distribuição vertical eliciada pela novidade (DVN) em Daniorerio. Foram utilizados peixes da espécie Daniorerio(n=178) subdivididos nos seguintes grupos experimentais: SAL – salina 0,9%; CAF – cafeína 100 mg/kg; DMSO – dimetilsulfóxido 0,1%; L-NAME - (N -Nitro-L-arginina-metil éster hidrocloreto) 10 mg/kg; TF – α-tocoferol 1 mg/kg (receberam apenas uma injeção por i.p); SAL + SAL; DMSO + SAL; SAL + CAF; L-NAME + SAL; L-NAME +CAF; TF + CAF (receberam duas injeções seguidas, uma injeção de cada substância na forma de cotratamento, por i.p). Os animais foram submetidos ao teste de preferência claro/escuro e de distribuição vertical eliciada pela novidade. Todos os testes foram filmados e os vídeos foram avaliados utilizando o X-PLO-RAT. Os dados foram expressos em média ± erro padrão. Foi aplicado o teste de normalidade utilizando o teste Shapiro-Wilk e o teste paramétrico ANOVA de uma via com pós-teste Tukey, considerando significativos valores com p<0,05. Nós demonstramos que o α-tocoferol na dose de 1 mg/kg reverteu todos os parâmetros do comportamento tipo ansiedade ampliado pela cafeína nos testes de PCE e do DVN e esse efeito foi semelhante ao observado quando administrado um inibidor da enzima óxido nítrico sintase (NOS), L-NAME. Portanto, o presente trabalho demonstrou pela primeira vez que o efeito comportamental ampliado pela cafeína no teste escotáxico e no DVN pode ser modulado pelo sistema nitrérgico e que o α-tocoferol reverte esse efeito comportamental induzido pela cafeína de forma total.

In conservation agriculture, mainly under direct seeding, maintaining the vegetation ground cover is essential, since this serves as a reservoir of nutrients which are slowly released to plants by microorganisms. Some authors have sought to study increases in the amount of straw in the soil, in addition to slowing down the process of decomposition, with hormesis being one of the techniques used. This technique states that all chemical substances are both poisonous and nonpoisonous, with only the dosage determining whether they are lethal or not. This study aimed to evaluate the dry weight and agronomic characteristics of a crop of black oat subjected to hormesis. The experimental design was of randomised blocks, with 12 treatments and 4 replications, giving a total of 48 experimental lots. The treatments were: Haloxyfop-R Methyl Ester at dosages of 0.625, 1.25 and 2.50 g ha(-1); Glyphosate at dosages of 12.50, 25.00 and 50.00 g ha-1; 2,4-D dimethylamine salt at dosages of 100.00, 200.00 and 300.00 g ha(-1); Alterbane at a dosage of 500.00 g ha(-1); Salicylic acid at a dose of 100 g ha(-1); and a control. It was concluded that for the subdosages under test, the herbicides 2,4-D at medium dosage and Verdict at low dosage were shown to be the best treatments for conserving straw as ground cover under direct seeding.

Pain is one of the most common reasons for patients to seek medical care. Bee Apis mellifera venom (AMV) has traditionally been used to treat inflammatory diseases and the alleviation of pain. Herein, we aimed to investigate the visceral antinociceptive potential of A. mellifera bee venom and its possible mechanism of action. Acetic acid-induced writhing assay was used in mice to determine the degree of visceral antinociception. Visceral antinociceptive activity was expressed as the reduction in the number of abdominal constrictions. Mice received an intraperitoneal injection of acetic acid after administration of AMV (0.08 or 0.8 mg/kg; intraperitoneally (i.p.)). In mechanistic studies, separate experiments were realized to examine the role of α2-receptors, nitric oxide, calcium channels, K+ATP channel activation, TRPV1 and opioid receptors on the visceral antinociceptive effect of AMV (0.8 mg/kg), using appropriate antagonists, yohimbine (2 mg/kg), L-NG-Nitroarginine methyl ester (L-NAME, 10 mg/kg), verapamil (5 mg/kg), glibenclamide (5 mg/kg), ruthenium red (3 mg/kg) or naloxone (2 mg/kg). AMV presented visceral antinociceptive activity in both doses tested (0.08 and 0.8 mg/Kg). Visceral antinociceptive effect of AMV was resistant to all the antagonists used. Mice showed no significant alterations in locomotion frequency, indicating that the observed antinociception is not a consequence of motor abnormality. Although AMV efficient diminished the acetic acid-evoked pain-related behavior, its mechanism is unclear from this study and future studies are needed to verify how the venom exerts its antinociceptive action.

Zebrafish are currently used at various stages of the drug discovery process and can be a useful and cost-effective alternative to some mammalian models. Nitric oxide (NO) plays an important role in physiology of zebrafish. The availability of appropriate analytical techniques to quantify the NO is crucial for studying its role in physiological and pathological conditions. This work aimed at establishing a high-performance liquid chromatography method for determination of NO levels in zebrafish larvae. Attempts were also made to assess the normal levels of NO at the first days postfertilization and the possible changes under pathological conditions. The method validation was quantitatively evaluated in terms of sensitivity, specificity, precision, accuracy, linearity, and recovery. NO levels from zebrafish larvae at the first days postfertilization and larvae challenged to N(G)-nitro-L-arginine methyl ester, sodium nitroprusside, Escherichia coil lipopolysaccharide, and copper sulfate were analyzed. The samples were derivatized with 2,3-diaminonaphthalene, and fluorescence detection was used for the indirect determination of NO. The method showed a good performance for all validation parameters evaluated and was efficient to monitor changes in NO concentration under physiological and pathophysiological conditions. Araucaria angustifolia, a unique species of this genus that occurs naturally in Brazil, has a high socio-economic and environmental value and is critically endangered of extinction, since it has been submitted to intense predatory exploitation during the last century. Root-associated bacteria from A. angustifolia were isolated, selected and characterized for their biotechnological potential of growth promotion and biocontrol of plant pathogenic fungi. Ninety-seven strains were isolated and subjected to chemical tests. All isolates presented at least one positive feature, characterizing them as potential PGPR. Eighteen isolates produced indole-3-acetic acid (IAA), 27 were able to solubilize inorganic phosphate, 21 isolates were presumable diazotrophs, with pellicle formation in nitrogen-free culture medium, 83 were phosphatases producers, 37 were positive for siderophores and 45 endospore-forming isolates were antagonistic to Fusarium oxysporum, a pathogen of conifers. We also observed the presence of bacterial strains with multiple beneficial mechanisms of action. Analyzing the fatty acid methyl ester (FAME) and partial sequencing of the 16S rRNA gene of these isolates, it was possible to characterize the most effective isolates as belonging to Bacillaceae (9 isolates), Enterobacteriaceae (11) and Pseudomonadaceae (1). The combination of solid-phase microextraction (SPME) with comprehensive two-dimensional gas chromatography is evaluated here for fatty acid (FA) profiling of the glycerophospholipid fraction from human buccal mucosal cells. A base-catalyzed derivatization reaction selective for polar lipids such as glycerophospholipid was adopted. SPME is compared to a miniaturized liquidliquid extraction procedure for the isolation of FA methyl esters produced in the derivatization step. The limits of detection and limits of quantitation were calculated for each sample preparation method. Because of its lower values of limits of detection and quantitation, SPME was adopted. The extracted analytes were separated, detected, and quantified by comprehensive two-dimensional gas chromatography with flame ionization detection (FID). The combination of SPME and comprehensive two-dimensional gas chromatography with FID, using a selective derivatization reaction in the preliminary steps, proved to be a simple and fast procedure for FA profiling, and was successfully applied to the analysis of adult human buccal mucosal cells.
