The use of transposable elements (TEs) as genetic drive mechanisms was explored using Drosophila melanogaster as a model system. Alternative strategies, employing autonomous and nonautonomous P element constructs were compared for their efficiency in driving the ry(+) allele into populations homozygous for a ry(-) allele at the genomic rosy locus. Transformed flies were introduced at 1%, 5%, and 10% starting frequencies to establish a series of populations that were monitored over the course of 40 generations, using both phenotypic and molecular assays. The transposon-borne ry(+) marker allele spread rapidly in almost all populations when introduced at 5% and 10% seed frequencies, but 1% introductions frequently failed to become established. A similar initial rapid increase in frequency of the ry(+) transposon occurred in several control populations lacking a source of transposase. Constructs carrying ry(+) markers also increased to moderate frequencies in the absence of selection on the marker. The results of Southern and in situ hybridization studies indicated a strong inverse relationship between the degree of conservation of construct integrity and transposition frequency. These finding have relevance to possible future applications of transposons as genetic drive mechanisms.

The gerbil (Meriones unguiculatus) is a rodent native of the and regions of Mongolia and China. Because the gerbil can be easily bred in laboratory conditions, this species has been largely used as an experimental model in biomedical research. However, there is still little information concerning the testis structure and function in the gerbil. In this regard, we performed a detailed morphofunctional analysis of the gerbil testis and estimated the spermatogenic cycle length utilizing H-3-thymidine as a marker for germ cell progression during their evolution through the spermatogenic process. The stage frequencies of the XII stages characterized according to the acrosome formation and development were (I-XII) 13.8, 10.1, 8.1, 7.8, 4.0, 11.2, 7.5, 7.1, 5.9, 7.6, 8.1, and 8.9. The mean duration of each seminiferous epithelium cycle was determined to be 10.6 +/- 1.0 days and the total duration of spermatogenesis, based on 4.5 cycles, was approximately 47.5 days. The volume density of tubular and interstitial compartments was approximately 92% and 8%, respectively. Based on the volume occupied by seminiferous tubules in the testis and the tubular diameter, about 9 and 18 m of seminiferous tubules were found per testis and per gram of testis, respectively. Twelve primary spermatocytes were formed from each type A1 spermatogonia. The meiotic index was 2.8, indicating that 30% of cell loss occurs during meiosis. The number of Leydig and Sertoli cells per gram of the testis was 28 million and each Sertoli cell was able to support approximately 13 spermatids. The daily sperm production per gram of testis (spermatogenic efficiency) was 33 million. Taken together, these data indicate that, mainly due to the high seminiferous tubule volume density and Sertoli cell support capacity for germ cells, the gerbil presents high spermatogenic efficiency compared with other mammalian species already investigated. The data obtained in the present study might provide the basis for future research involving the reproductive biology in this species.

Hrp1p is a heterogeneous ribonucleoprotein (hnRNP) from the yeast Saccharomyces cerevisiae that is involved in the cleavage and polyadenylation of the 3'-end of mRNAs and mRNA export. In addition, Hrp1p is one of several RNA-binding proteins that are posttranslationally modified by methylation at arginine residues. By using-functional recombinant Hrp1p, we have identified RNA sequences with specific high affinity binding sites. These sites correspond to the efficiency element for mRNA 3'-end formation, UAUAUA. To examine the effect of methylation on specific RNA binding, purified recombinant arginine methyltransferase (Hmt1p) was used to methylate Hrp1p. Methylated Hrp1p binds with the same affinity to UAUAUA-containing RNAs as unmethylated Hrp1p indicating that methylation does not affect specific RNA binding. However, RNA itself inhibits the methylation of Hrp1p and this inhibition is enhanced by RNAs that specifically bind Hrp1p. Taken together, these data support a model in which protein methylation occurs prior to protein-RNA binding in the nucleus.

In this paper a comparative analysis of the environmental impact caused by the use of natural gas and diesel in thermoelectric power plants utilizing combined cycle is performed. The objective is to apply a thermoeconomical analysis in order to compare the two proposed fuels. In this analysis, a new methodology that incorporates the economical engineering concept to the ecological efficiency once Cardu and Baica [1, 2], which evaluates, in general terms, the environmental impacts caused by CO2, SO2, NOx and Particulate Matter (PM), adopting as reference the air quality standards in vigour is employed. The thermoeconomic model herein proposed utilizes functional diagrams that allow the minimization the Exergetic Manufacturing Cost, which represents the cost of production of electricity incorporating the environmental impact effects to study the performance of the thermoelectric power plant [3,4], It follows that it is possible to determine the environmental impact caused by thermoelectric power plants and, under the ecological standpoint, the use of natural gas as a fuel is the best option compared to the use of the diesel, presenting ecological efficiency values of 0.944 and 0.914 respectively. From the Exergoeconomic point of view of, it was found out that the EMC (Exergetic Manufacturing Cost) is better when natural gas is used as fuel compared to the diesel fuel. The energy efficiency of buildings should be a goal at the pre-design phase, though the importance of the design variables is often neglected even during the design process. Highlighting the relevance of these design variables, this research studies the relationships of building location variables with the electrical energy consumption of residential units. The following building design parameters are considered: orientation, story height and sky view factor (SVF). The consideration of the SVF as a location variable contributes to the originality of this research. Data of electrical energy consumption and users' profiles were collected and several variables were considered for the development of an Artificial Neural Network model. This model allows the determination of the relative importance of each variable. The results show that the apartments' orientation is the most important design variable for the energy consumption, although the story height and the sky view factor play a fundamental role in that consumption too. We pointed out that building heights above twenty-four meters do not optimize the energy efficiency of the apartments and also that an increasing SVF can influence the energy consumption of an apartment according to their orientation.

At this time, each major automotive market bares its own standards and test procedures to regulate the vehicle green house gases emissions and, thus, fuel consumption. Hence, much are the ways to evaluate the overall efficiency of motor vehicles. The majority of such standards rely on dynamometer cycle tests that appraise only the vehicle as a whole, but fail to assess emissions for each component or sub-system. Once the amount of work generated by the power source of an ICE vehicle to overcome the driving resistance forces is proportional to the energy contained in the required amount of fuel, the power path of the vehicle can be straightforwardly modeled as a set of mechanical systems, and each sub-system evaluated for its share on the total fuel consumption and green house gases emission. This procedure enables the estimation of efficiency gains on the system due to improvement of particular elements on the vehicle's driveline. In this work a simple systematic mechanical model of an arbitrary smallsized hatch back was assembled and total required energy calculated for different regulatory cycles. All the modeling details of the energy balance throughout the system are presented. Afterward, each subsystem was investigated for its role on the fuel consumption and the generated emission quantified. Data from a multibreed commercial flock located at Mid-West of Brazil, supported by Programa de Melhoramento Genético de Caprinos e Ovinos de Corte (GENECOC), were used to estimate genetic parameters of traits related to ewe productivity by Average Information Restricted Maximum Likelihood method applied to an animal model. The analyzed traits were litter weight at birth (LWB) and at weaning (LWW), ewe weight at weaning (EW) and ewe production efficiency, estimated by WEE=LWW/EW 0.75. The heritabilities were 0.26±0.05, 0.32±0.06, 0.37±0.03 and 0.10±0.02 for LWB, LWW, EW and WEE, respectively. Significant effects for direct heterosis were observed for LWW and EW. Recombination losses were important for EW and WEE. Genetic correlations of LWB with LWW, EW and WEE were 0.68, 0.37 and 0.15, respectively; of LWW with EW and WEE were 0.30 and 0.34, respectively; and between EW and WEE was -0.25. Even though it is a low heritability trait, WEE can be indicated as a selection criteria for improving the ewe productivity without increasing the mature weight of animals due to its genetic correlations with LWW and other traits. © 2011 Elsevier B.V.

This paper presents a mixed-integer linear programming model to solve the problem of allocating voltage regulators and fixed or switched capacitors (VRCs) in radial distribution systems. The use of a mixed-integer linear model guarantees convergence to optimality using existing optimization software. In the proposed model, the steady-state operation of the radial distribution system is modeled through linear expressions. The results of one test system and one real distribution system are presented in order to show the accuracy as well as the efficiency of the proposed solution technique. An heuristic to obtain the Pareto front for the multiobjective VRCs allocation problem is also presented. © 2012 Elsevier Ltd. All rights reserved.

This course will be designed for the officers within government departments who have responsibilty for guiding the country’s energy policy and energy management framework. Other stakeholders also will include private sector representatives who have interest in providing energy efficiency equipment and renewable energy solutions to the market towards advancing improvements in both energy efficiency and meeting renewable energy targets. The course will provide insight into all aspects of energy management with specific emphasis on energy efficiency as well as renewable energy. Emphasis will be placed on highlighting issues and challenges that countries face in pursuing energy efficiency and renewable energy strategies. International and regional best practices will be highlighted as a means of showcasing how various countries have overcome the barriers to advancing renewable energy targets and increasing energy efficiencies towards meeting national energy goals. The curriculum is divided into five modules and is designed to be covered over a 3-day period. The course will be designed to ensure practical application of the learning. The course also is designed to enable the Caribbean to demonstrate leadership in energy efficiency practices and the adoption of renewable energy strategies, serving as a model for other small island developing states.

Este artigo apresenta um modelo para o fomento da autonomia dos aprendentes, mostra alguns resultados alcançados na aplicação desse modelo e discute desafios ainda a serem enfrentados. O modelo comporta a investigação de áreas problemáticas do processo individual de aprendizagem de cada sujeito, a identificação de seus estilos preferenciais de aprender, o uso de ferramentas tecnológicas para melhorar a autonomia na aprendizagem, o desenvolvimento de um leque maior de estratégias de aprendizagem de línguas e a implementação de rotinas de auto-monitoramento e auto-avaliação. Este modelo tem sido aplicado nos últimos três anos com alunos de Letras cursando Licenciaturas em Alemão, Francês ou Inglês. Três ordens de resultados emergem dos dados da pesquisa: primeiramente, o modelo provou sua eficácia em prover um andaime para a aprendizagem autônoma de línguas dos alunos; em segundo lugar, as experiências de aprendizagem autônoma vividas pelos futuros professores de línguas poderão ser espelhadas em suas vidas profissionais futuras com seus próprios alunos; finalmente, os dados emanados dos participantes da pesquisa podem lançar uma luz sobre a variedade de maneiras pelas quais as pessoas aprendem no contexto local.

This paper presents an assessment of an educational game for teaching the efficient use of electricity. Developed with Adobe Flash (R), it is a virtual board game where participants choose a car that starts the path and reaches the same final goal, going through a number of track steps defined in terms of a dice that each player rolls in turn. The car moves if the participant is able to correctly answer a question that is randomly generated by the software. The objective of the game is to answer questions related to energy efficiency promoting a healthy and attractive learning process for participants on concepts related to energy efficiency such as: the rational use of energy, the basic concepts of forms of energy generation, among others. The main objective of this paper is to assess the impact of the application of this virtual game in the teaching and learning process of high school students. Therefore, the game was applied in the discipline of physics in a class of junior high public school in the state of Sao Paulo. Initially, the class that had 43 students was divided into 10 groups of 4 students, and 1 group of 3 students. Each student group competed with one another. The idea was that each of them should indicate a student who was the representative of this group until only 4 group leaders were selected for the finals. At this stage, each student could interact with a group of up to ten students that acted as advisers. The adopted assessment process is based on the model proposed by Savi [7]. Then, at the end of the game, the students answered a prepared questionnaire based on the model proposed by Savi. According to Savi, although there are significant studies that show the importance of educational games for the process of cognitive development and learning concepts of students, there are few papers that present forms of assessing the potential of these resources. Thus, the assessment criteria proposed by Savi are based on the model of training evaluation by Kirkpatrick [3], taken as a reference to measure the efficiency of processes of continuing education courses for professionals. The authors assert that the metric of the evaluation proposed to assess the game is based on the first level of the model proposed by Kirkpatrick.

This paper presents a mixed-integer quadratically-constrained programming (MIQCP) model to solve the distribution system expansion planning (DSEP) problem. The DSEP model considers the construction/reinforcement of substations, the construction/reconductoring of circuits, the allocation of fixed capacitors banks and the radial topology modification. As the DSEP problem is a very complex mixed-integer non-linear programming problem, it is convenient to reformulate it like a MIQCP problem; it is demonstrated that the proposed formulation represents the steady-state operation of a radial distribution system. The proposed MIQCP model is a convex formulation, which allows to find the optimal solution using optimization solvers. Test systems of 23 and 54 nodes and one real distribution system of 136 nodes were used to show the efficiency of the proposed model in comparison with other DSEP models available in the specialized literature. The purpose of this study is to describe the development of a model to predict the digestible lysine requirements of broilers using the factorial approach, and to evaluate the model using as reference the model presented in Brazilian Tables for Poultry and Swine. The model partitions the requirement for maintenance and growth for feather-free body protein and feather protein, in which the inputs are body and feather protein weight and the daily rates of protein deposition in the feather-free body and feathers. The parameters that express the lysine requirement for maintenance were obtained in metabolism trials with roosters, and those for the efficiency of lysine utilization in experiments with broilers from 1 to 42 d. Based on these results the model proposed was: Lys (mg/d) = [(151xBP(m)(-0.27)xBP(t)) + (0.01xP(t)x18)] + [(75xBPD/0.77) + (18xFPD/0.77)], where Lys = digestible lysine requirement (mg/d), BPm=body protein weight at maturity (kg), BPt=body protein weight at time t (kg), FPt=feather protein weight at time t (kg), BPD=body protein deposition (g/d), FPD = feather protein deposition (g/d). The model yields sensible predictions of the digestible lysine requirements of broilers of different strains and ages growing at their potential, and suggests a lower lysine requirement after 27 d than does the Brazilian model. The proposed model is the first step in the development of a simulation model that predicts the food intake of a broiler and hence the dietary amino acid content that would optimise performance.
