Stemming from in vitro and in vivo pre-clinical and human models, tissue-engineering-based strategies continue to demonstrate great potential for the regeneration of the pulp-dentin complex, particularly in necrotic, immature permanent teeth. Nanofibrous scaffolds, which closely resemble the native extracellular matrix, have been successfully synthesized by various techniques, including but not limited to electrospinning. A common goal in scaffold synthesis has been the notion of promoting cell guidance through the careful design and use of a collection of biochemical and physical cues capable of governing and stimulating specific events at the cellular and tissue levels. The latest advances in processing technologies allow for the fabrication of scaffolds where selected bioactive molecules can be delivered locally, thus increasing the possibilities for clinical success. Though electrospun scaffolds have not yet been tested in vivo in either human or animal pulpless models in immature permanent teeth, recent studies have highlighted their regenerative potential both from an in vitro and in vivo (i.e., subcutaneous model) standpoint. Possible applications for these bioactive scaffolds continue to evolve, with significant prospects related to the regeneration of both dentin and pulp tissue and, more recently, to root canal disinfection. Nonetheless, no single implantable scaffold can consistently guide the coordinated growth and development of the multiple tissue types involved in the functional regeneration of the pulp-dentin complex. The purpose of this review is to provide a comprehensive perspective on the latest discoveries related to the use of scaffolds and/or stem cells in regenerative endodontics. The authors focused this review on bioactive nanofibrous scaffolds, injectable scaffolds and stem cells, and pre-clinical findings using stem-cell-based strategies. These topics are discussed in detail in an attempt to provide future direction and to shed light on their potential translation to clinical settings.

The nanostructured materials over the last decade have been increasing the variety of studies and research applications in many industries. From the understanding and manipulation of nanoscale is possible to obtain high-performance materials. One method, which has been very effective in obtaining of nanostructured composites, is the electrospinning, a technique that uses electrostatic forces to produce fibers from a polymer solution. By understanding and controlling of process conditions, such as solution viscosity, working distance, the velocity of the collector, applied voltage and others conditions, it is possible to obtain fibers in many different morphologies. This work aims to obtain nanostructured composites from polysulfone (PSU) a thermoplastic polymer with high oxidation resistance and good mechanical strength at high temperatures and carbon nanotubes (CNTs) that are excellent reinforcements for polymer materials, their mechanical resistance is greater than that of all known materials; using the electrospinning process via polymer solution. Were used polysulfone solutions, n,n-ndimetil acetamide (PSU / DMAc) and this same solution added of CNTs in order to obtain the nanofibers. In both cases were analyzed the effectiveness of the process from the analysis of fiber diameters, rheological behavior and infrared spectroscopy. The results obtained confirmed the efficiency of the electrospinning process to obtain polymeric fibers

The aims of this study were (1) to synthesize and characterize random and aligned nanocomposite fibers of multi-walled carbon nanotubes (MWCNT)/nylon-6 and (2) to determine their reinforcing effects on the flexural strength of a dental resin composite.Nylon-6 was dissolved in hexafluoropropanol (10 wt%), followed by the addition of MWCNT (hereafter referred to as nanotubes) at two distinct concentrations (i.e., 0.5 or 1.5 wt%). Neat nylon-6 fibers (without nanotubes) were also prepared. The solutions were electrospun using parameters under low- (120 rpm) or high-speed (6000 rpm) mandrel rotation to collect random and aligned fibers, respectively. The processed fiber mats were characterized by scanning (SEM) and transmission (TEM) electron microscopies, as well as by uni-axial tensile testing. To determine the reinforcing effects on the flexural strength of a dental resin composite, bar-shaped (20 x 2 x 2 mm(3)) resin composite specimens were prepared by first placing one increment of the composite, followed by one strip of the mat, and one last increment of composite. Non-reinforced composite specimens were used as the control. The specimens were then evaluated using flexural strength testing. SEM was done on the fractured surfaces. The data were analyzed using ANOVA and the Tukey's test (alpha=5%).Nanotubes were successfully incorporated into the nylon-6 fibers. Aligned and random fibers were obtained using high- and low-speed electrospinning, respectively, where the former were significantly (p<0.001) stronger than the latter, regardless of the nanotubes'presence. Indeed, the dental resin composite tested was significantly reinforced when combined with nylon-6 fibrous mats composed of aligned fibers (with or without nanotubes) or random fibers incorporated with nanotubes at 0.5 wt%. Introduction: Antibiotic-containing polymer-based nanofibers (hereafter referred to as scaffolds) have demonstrated great potential for their use in regenerative endodontics from both an antimicrobial and cytocompatibility perspective. This study sought to evaluate in vitro the effects of ciprofloxacin (CIP)-containing polymer scaffolds against Enterococcus faecalis biofilms. Methods: Human mandibular incisors were longitudinally sectioned to prepare radicular dentin specimens. Sterile dentin specimens were distributed in 24-well plates and inoculated with E. faecalisfor biofilm formation. Infected dentin specimens were exposed to 3 groups of scaffolds, namely polydioxanone (PDS) (control), PDS + 5 wt% CIP, and PDS + 25 wt% CIP for 2 days. Colony-forming units (CFU/mL) (n = 10) and scanning electron microscopy (SEM) (n -= 2) were performed to quantitatively and qualitatively assess the antimicrobial effectiveness, respectively. Results: PDS scaffold containing CIP at 25 wt% showed maximum bacteria elimination with no microbial growth, differing statistically (P < .05) from the control (PDS) and from PDS scaffold containing CIP at 5 wt%. Statistical differences (P < .05) were also seen for the CFU/mL data between pure PDS (5.92-6.02 log CFU/mL) and the PDS scaffold containing CIP at 5 wt% (5.39 5.87 log CFU/mL). SEM images revealed a greater concentration of bacteria on the middle third of the dentin specimen. after 5 days of biofilm formation. On scaffold exposures, SEM images showed similar results when compared with the CFU/mL data. Dentin specimens exposed to PDS + 25 wt% CIP scaffolds displayed a practically bacteria-free surface. Conclusions: On the basis of the data presented, newly developed antibiotic-containing electrospun scaffolds hold promise as an intracanal medicament to eliminate biofilm/infection before regenerative procedures.

In heterogeneous catalysis, numerous elements such as titanium and iron have been studied as nanoscale catalysts, but little is known about the use of niobium in nanocatalysis. The nanostructured particles have intrinsic and different physicochemical characteristics with great potential for use in industrial scale. Brazil having the largest known worldwide niobium reserve has the great challenge of creating pioneering technologies with the metal. Biodiesel is an alternative fuel and renewable substitute for regular diesel. Being biodegradable, non-toxic and have CO2 emissions lower than regular diesel, it contributes to the environment and to the independence from oil. The aim of this work was initially synthesize nanoscale particles of niobium pentoxide (Nanospheres, nanorods, nanofibers, nanocubes) from the sol-gel technique. The characterization of different nanoscale structures obtained was performed using different analytical techniques such as x-ray diffraction (XRD) and Scanning Electron Microscopy (SEM). The synthesized nanometer niobium oxide will be used as a heterogeneous catalyst in biodiesel synthesis from commercial soybean oil, checking in detail what the effect of morphology is presented (Nanospheres, nanorods, nanofibers, nanocubes) in the yield of biodiesel synthesis, comparing these results with those already described in literature for the amorphous niobium oxide and other oxide catalysts. The aim of this study was to evaluate the behavior of reinforced composites with polyamide 6 fibers aligned (6000 rpm) and alignment (120 rpm) with or without CNT using the flexural strength test. After preparation of nanofibers aligned nylon 6 (6000 rpm) and alignment (120 rpm) with and without incorporation of nanotube carbon by the method of electrospinning, were performed one control group (n = 10) and 4 experimental groups (n = 40) G1: Control (just resin Charisma - Heraeus Kulzer) ;G2 Resin + N6 aligned (6000 rpm) + CNT; G3:Resin + N6 alignment (120 rpm) + CNT; G4: Resin + aligned ( 6000 rpm) N6. G5: Resin + N6 alignment (120 rpm). The fibers were cut to the dimensions of 0,3 x 15 mm and were applied an adhesive at the surface (Single Bond 2) for 5 min and cured. In the matrix, was added resin in the proximal box (Charisma A2, Heraeus Kulzer) and cured for 40 s. (power 1100 mW / cm²). A first layer of resin and on the resin was deposited. The resin layers specimens were light irradiated with three overlapping exposures delivered. For each resin layer were light irradiated for 40 sec. The samples were tested with a cross-speed of 1 mm / min, and a 50 Kgf at Universal testing machine (EMIC mod.DL2000). The Dunnet test showed that only the nanotube group was significantly different from the control group. The ANOVA two-way indicates that the nanotube factor was statistically significant (p < 0.05) and there is no interaction between factors and orientation nanotube. The presence of nanotube showed lower fracture resistance values for aligned and unaligned groups. The results of this study showed that the orientation of the fibers does not influence the strength of composite resins and the incorporation of nylon nanofibers with carbon nanotubes decreased the fracture resistance values. Natural fibers have been highlighted as a renewable material that can replace materials from oil and its derivatives. In this context, Brazil becomes the perfect setting because of the diversity of fibers found in its territory, such as sugarcane, sisal, rice, cotton, coconut, pineapple, among others. The paineiras (Chorisia speciosa St. Hil) are typically Brazilian trees, which produce paina as fruit. These fruits are still little studied as a source of lignocellulose by research groups. This project aimed obtaining and characterization of cellulose nanofibers from the fibers from the paina fibers. Obtaining nanocellulose is practically made through simplified chemical processes. First, was performed out pre-treatments to removal of waxes, lignin and hemicellulose. The first stage of pre-treatment was carried out by alkaline aqueous solution of sodium hydroxide (NaOH) at 5wt%, where the fibers were under constant agitation for 1h at 70°C. Through alkali treatment it was possible to remove most of the lignin, hemicellulose, waxes and extractives. After the alkaline treatment was done bleaching with an aqueous solution of sodium hydroxide (NaOH) to 4wt% and hydrogen peroxide (H2O2) to 24wt% 1:1 during 2h with constant stirring to 50 °C. Through bleaching was possibe to remove residual lignin, and got cellulose with 72% of crystallinity. Nanocellulose of paina fibers was extracted using different conditions of acid hydrolysis with sulfuric acid (H2SO4) to 50wt%. After acid hydrolysis, the suspensions were centrifuged during 30 min and dialyzed in water to remove excess acid until neutral pH (6-7). Then the suspensions were passed by ultrasonification in an ultrasound 20 kHz during 1h in an ice bath.
