Telecentric optical computed tomography (optical-CT) is a state-of-the-art method for visualizing and quantifying 3-dimensional dose distributions in radiochromic dosimeters. In this work a prototype telecentric system (DFOS-Duke Fresnel Optical-CT Scanner) is evaluated which incorporates two substantial design changes: the use of Fresnel lenses (reducing lens costs from $10-30K t0 $1-3K) and the use of a 'solid tank' (which reduces noise, and the volume of refractively matched fluid from 1 ltr to 10 cc). The efficacy of DFOS was evaluated by direct comparison against commissioned scanners in our lab. Measured dose distributions from all systems were compared against the predicted dose distributions from a commissioned treatment planning system (TPS). Three treatment plans were investigated including a simple four-field box treatment, a multiple small field delivery, and a complex IMRT treatment. Dosimeters were imaged within 2 h post irradiation, using consistent scanning techniques (360 projections acquired at 1 degree intervals, reconstruction at 2mm). DFOS efficacy was evaluated through inspection of dose line-profiles, and 2D and 3D dose and gamma maps. DFOS/TPS gamma pass rates with 3%/3mm dose difference/distance-to-agreement criteria ranged from 89.3% to 92.2%, compared to from 95.6% to 99.0% obtained with the commissioned system. The 3D gamma pass rate between the commissioned system and DFOS was 98.2%. The typical noise rates in DFOS reconstructions were up to 3%, compared to under 2% for the commissioned system. In conclusion, while the introduction of a solid tank proved advantageous with regards to cost and convenience, further work is required to improve the image quality and dose reconstruction accuracy of the new DFOS optical-CT system.

Both lamotrigine and 5-(2,3,5-trichlorophenyl)-2,4-diaminopyrimidine (code name BW1003C87), the subject of the present study, are anticonvulsant as well as neuroprotective in models of brain ischaemia and in a model of white matter ischaemia. BW1003C87 is a sodium channel blocker which also reduces the release of the neurotransmitter glutamate. The three dimensional structures reported here form part of a newly developed data base for the detailed investigation of members of this drug family and their biological activities.

Drugs based on 5-phenyl-2,4 diamino pyrimidine and 6-phenyl-1,2,4 triazine derivatives are well known for their effects on the central nervous system. The study presented here provides detailed crystal structures of two pyrimidine derivatives which have neuroprotective properties in models of both grey and white matter ischemia. Recently published studies suggest that the compounds lamotrigine (a triazine derivative), and the two pyrimidines BW1003C87 (I) and sipatrigine (II) mediate their primary in vivo mode of action by inhibiting voltage-gated Na+ channels. The X-ray crystal structures will contribute valuable data for applications involving binding and modelling studies of the biological actions of these drugs.

Absolute cross sections have been measured for single and double charge exchange and x-ray line emission for highly charged ions of C, N, 0, and Ne colliding with He, H-2 CO2, and H2O at collisions energies of 7q keV. Present results of charge exchange in He and H-2 compare favorably with previous results. For CO2 and H2O, where prior work is scarce, the classical overbarrier model is found to overestimate results by up to a factor of 3. An analysis of the relative intensities of the observed Lyman x-ray transitions indicates that capture into l states is not statistical, as collision velocities are insufficient to populate the highest angular-momentum states. The importance of autoionization following multiple capture is highlighted, and enhanced radiative stabilization following double capture is observed and compared to other studies. Present results are also discussed in terms of mechanisms likely to generate x-ray emission in comets.

We have performed short-pulse x-ray scattering measurements on laser-driven shock-compressed plastic samples in the warm dense matter regime, providing instantaneous snapshots of the system evolution. Time-resolved and angularly resolved scattered spectra sensitive to the correlation effects in the plasma show the appearance of short-range order within a few interionic separations. Comparison with radiation-hydrodynamic simulations indicates that the shocked plastic is compressed with a temperature of a few electron volts. These results are important for the understanding of the thermodynamic behavior of strongly correlated matter for conditions relevant to both laboratory astrophysics and inertial confinement fusion research.

Double laser pulses of duration similar to 75 ps and short laser pulses similar to 1 ps superimposed on longer duration background pulses have been shown to efficiently pump lasing in Ne-like and Ni-like ions. For the 75 ps pumping, X-ray laser output without travelling wave pumping is shown to be well-described by a model of ASE output. With I ps pumping, the X-ray laser output with different velocity travelling wave pumping is well-fitted with an extension to the ASE model allowing for travelling wave excitation of the gain along the plasma line. The model is used to investigate the production of short (<1 ps) x-ray laser pulses and the effects of non-ideal travelling wave velocities on the X-ray laser output. Resonance line spectra of emission perpendicular to the plasma line are measured and simulated. It is shown that an accurate opacity model for the more intense Ne-like ions is needed to correctly simulate the spectra.

Cellular response to radiation damage is made by a complex network of pathways and feedback loops whose spatiotemporal organization is still unclear despite its decisive role in determining the fate of the damaged cell. The single-cell approach and the high spatial resolution offered by microbeams provide the perfect tool to study and quantify the dynamic processes associated with the induction and repair of DNA damage. The soft X-ray microbeam has been used to follow the development of radiation induced foci in live cells by monitoring their size and intensity as a function of dose and time using yellow fluorescent protein (YFP) tagging techniques. Preliminary data indicate a delayed and linear rising of the intensity signal indicating a slow kinetic for the accumulation of DNA repair protein 53BP1. A slow and limited foci diffusion has also been observed. Further investigations are required to assess whatever such diffusion is consistent with a random walk pattern or if it is the result of a more structured lesion processing phenomenon. In conclusion, our data indicates that the use of microbeams coupled to live cell microscopy represent a sophisticated approach for visualizing and quantifying the dynamics changes of DNA proteins at the damaged sites.

Context. The X-ray lines between 10.9 and 11.2 Å have attracted little attention but are of interest since they enable an estimate of the coronal abundance of Na to be made. This is of great interest in the continuing debate on the nature of the FIP (first ionization potential) effect. Aims. Observations of the lines with the Solar Maximum Mission Flat Crystal Spectrometer and a rocket-borne X-ray spectrometer are used to measure the Na/Ne abundance ratio, i.e. the ratio of an element with very low FIP to one with high FIP. Methods. New atomic data are used to generate synthetic spectra which are compared with the observations, with temperature and the Na/Ne abundance ratio as free parameters. Results. Temperature estimates from the observations indicate that the line emission is principally from non-flaring active regions, and that the Na/Ne abundance ratio is 0.07 ± 50%. Conclusions. The Na/Ne abundance ratio is close to a coronal value for which the abundances of low-FIP elements (FIP < 10 eV) are enhanced by a factor of 3 to 4 over those found in the photosphere. For low-temperature (Te 1.5 MK) spectra, the presence of lines requires that either a higher-temperature component is present or a revision of ionization or recombination rates is needed.

Recent progress using the VULCAN laser at the Rutherford Appleton Laboratory to pump X-ray lasing in nickel-like ions is reviewed. Double pulse pumping with similar to 100 ps pulses has been shown to produce significantly greater X-ray laser output than single pulses of duration 0.1-1 ns. With double pulse pumping, the main pumping pulse interacts with a pre-formed plasma created by a pre-pulse. The efficiency of lasing increases as there is a reduced effect of refraction of the X-ray laser beam due to smaller density gradients and larger gain volumes, which enable propagation of the X-ray laser beam along the full length of the target. The record shortest wavelength saturated laser at 5.9 nm has been achieved in Ni-like dysprosium using double pulse pumping of 75 ps duration from the VULCAN laser. A variant of the double pulse pumping using a single similar to 100 ps laser pulse and a superimposed short similar to 1 ps pulse has been found to further increase the efficiency of lasing by reducing the effects of over-ionisation during the gain period. The record shortest wavelength saturated laser pumped by a short similar to 1 ps pulse has been achieved in Ni-like samarium using the VULCAN laser operating in chirped pulse amplified (CPA) mode. Optical source model for the 23.2-23.6 nm radiation from the multielement germanium soft X-ray laser
Resumo:
Distributions of source intensity in two dimensions (designated the source model), averaged over a single laser pulse, based on experimental measurements of spatial coherence, are considered for radiation from the unresolved 23.2/23.6 nm spectral lines from the germanium collisional X-ray laser. The model derives from measurements of the visibility of Young slit interference fringes determined by a method based on the Wiener-Khinchin theorem. Output from amplifiers comprising three and four target elements have similar coherence properties in directions within the horizontal plane corresponding to strong plasma refraction effects and fitting the coherence data shows source dimensions (FWHM) are similar to 26 mu m (horizontal), significantly smaller than expected by direct imaging, and similar to 125 mu m (vertical: equivalent to the height of the driver excitation). Through the use of time-integrated space-resolved keV spectroscopy, we characterize line plasmas showing gain in Ne-like Zn with prepulsed irradiation to explain the enhanced performances of x-ray lasers using the prepulse technique. It is observed that the value of the electron temperature does not vary significantly with prepulse level, nor does its spatially resolved profile along the line. The lateral width and density of the Ne-like region in the plasma are seen to increase with the prepulse level. (C) 1995 Optical Society of America

Many studies have shown that with increasing LET of ionizing radiation the RBE (relative biological effectiveness) for dsb (double strand breaks) induction remains around 1.0 despite the increase in the RBE for cell killing. This has been attributed to an increase in the complexity of lesions, classified as dsb with current techniques, at multiply damaged sites. This study determines the molecular weight distributions of DNA from Chinese hamster V79 cells irradiated with X-rays or 110 keV/mu m alpha-particles. Two running conditions for pulsed-field gel-electrophoresis were chosen to give optimal separation of fragments either in the 225 kbp-5.7 Mbp range or the 0.3 kbp to 225 kbp range. Taking the total fraction of DNA migrating into the gel as a measure of fragmentation, the RBE for dsb induction was less than 1.0 for both molecular weight regions studied. The total yields of dsb were 8.2 x 10(-9) dsb/Gy/bp for X-rays and 7.8 x 10(-9) dsb/Gy/bp for a-particles, measured using a random breakage model. Analysis of the RBE of alpha-particles versus molecular weight gave a different response. In the 0.4 Mbp-57 Mbp region the RBE was less than 1.0; however, below 0.4 Mbp the RBE increased above 1.0. The frequency distributions of fragment sizes were found to differ from those predicted by a model assuming random breakage along the length of the DNA and the differences were greater for alpha-particles than for X-rays. An excess of fragments induced by a single-hit mechanism was found in the 8-300 kbp region and for X-rays and alpha-particles these corresponded to an extra 0.8 x 10(-9) and 3.4 x 10(-9) dsb/bp/Gy, respectively. Thus for every alpha-particle track that induces a dsb there is a 44% probability of inducing a second break within 300 kbp and for electron tracks the probability is 10%. This study shows that the distribution of damage from a high LET alpha-particle track is significantly different from that observed with low LET X-rays. In particular, it suggests that the fragmentation patterns of irradiated DNA may be related to the higher-order chromatin repealing structures found in intact cells.
