Contagious caprine pleuropneumonia (CCPP) caused by Mycoplasma capricolum subsp. capripneumoniae is a severe epidemic affecting mainly domestic Caprinae species but also affects wild Caprinae species. M. capricolum subsp. capripneumoniae belongs to the "Mycoplasma mycoides cluster." The disease features prominently in East Africa, in particular Kenya, Tanzania, and Ethiopia. CCPP also endangers wildlife and thus affects not only basic nutritional resources of large populations but also expensively built-up game resorts in affected countries. Here, we report the complete sequences of two M. capricolum subsp. capripneumoniae strains: the type strain F38 and strain ILRI181 isolated druing a recent outbreak in Kenya. Both genomes have a G+C content of 24% with sizes of 1,016,760 bp and 1,017,183 bp for strains F38 and ILRI181, respectively.

Mycobacterium avium subsp. avium (Maa) is an intracellular pathogen belonging to the Mycobacterium avium-intracellulare complex (MAC). Reservoirs of MAC are the natural environment, wildlife and domestic animals. In adult bovine, MAC infections are typically caused by Mycobacterium avium subsp. paratuberculosis (Map). Maa infections in bovine are rarely reported but may cause clinical disease and pathological lesions similar to those observed in paratuberculosis or those induced by members of the Mycobacterium tuberculosis complex (MTBC). Therefore, differentiation of MAC from MTBC infection should be attempted, especially if unusual mycobacterial lesions are encountered. Four veal calves from a fattening farm dying with clinical signs of otitis media, fever, and weight loss were submitted for necropsy. Samples from affected organs were taken for histologic investigation, bacteriologic culture, and bacterial specification using PCR. Macroscopic thickening of the intestinal mucosa was induced by granulomatous enteritis and colitis. Intracytoplasmic acid-fast bacteria were detected by Ziehl-Neelsen stains and PCR revealed positive results for Mycobacterium avium subsp. avium. Clinical and pathological changes of Maa infection in veal calves had features of Mycobacterium avium subsp. paratuberculosis and the MTBC. Therefore, Mycobacterium tuberculosis complex infection should be considered in cases of granulomatous enteritis in calves.

Aeromonas salmonicida subsp. salmonicida is the causal agent of furunculosis in salmonids. We recently identified a group of genomic islands (AsaGEI) in this bacterium. AsaGEI2a, one of these genomic islands, has almost exclusively been identified in isolates from North America. To date, Aeromonas salmonicida subsp. salmonicida JF3224, a strain isolated from a wild brown trout (Salmo trutta) caught in Switzerland, was the only European isolate that appeared to bear AsaGEI2a. We analyzed the genome of JF3224 and showed that the genomic island in JF3224 is a new variant of AsaGEI, which we have called AsaGEI2b. While AsaGEI2b shares the same integrase gene and insertion site as AsaGEI2a, it is very different in terms of many other features. Additional genomic investigations combined with PCR genotyping revealed that JF3224 is sensitive to growth at 25°C, leading to insertion sequence-dependent rearrangement of the locus on the pAsa5 plasmid that encodes a type three secretion system, which is essential for the virulence of the bacterium. The analysis of the JF3224 genome confirmed that AsaGEIs are accurate indicators of the geographic origins of A. salmonicida subsp. salmonicida isolates and is another example of the susceptibility of the pAsa5 plasmid to DNA rearrangements.

Members of the Mycoplasma mycoides cluster' represent important livestock pathogens worldwide. Mycoplasma mycoides subsp. mycoides is the etiologic agent of contagious bovine pleuropneumonia (CBPP), which is still endemic in many parts of Africa. We report the genome sequences and annotation of two frequently used challenge strains of Mycoplasma mycoides subsp. mycoides, Afadé and B237. The information provided will enable downstream 'omics' applications such as proteomics, transcriptomics and reverse vaccinology approaches. Despite the absence of Mycoplasma pneumoniae like cyto-adhesion encoding genes, the two strains showed the presence of protrusions. This phenotype is likely encoded by another set of genes.

Contagious caprine pleuropneumonia (CCPP) is a highly contagious disease caused by Mycoplasma capricolum subsp. capripneumoniae that affects goats in Africa and Asia. Current available methods for the diagnosis of Mycoplasma infection, including cultivation, serological assays, and PCR, are time-consuming and require fully equipped stationary laboratories, which make them incompatible with testing in the resource-poor settings that are most relevant to this disease. We report a rapid, specific, and sensitive assay employing isothermal DNA amplification using recombinase polymerase amplification (RPA) for the detection of M. capricolum subsp. capripneumoniae. We developed the assay using a specific target sequence in M. capricolum subsp. capripneumoniae, as found in the genome sequence of the field strain ILRI181 and the type strain F38 and that was further evidenced in 10 field strains from different geographical regions. Detection limits corresponding to 5 × 10(3) and 5 × 10(4) cells/ml were obtained using genomic DNA and bacterial culture from M. capricolum subsp. capripneumoniae strain ILRI181, while no amplification was obtained from 71 related Mycoplasma isolates or from the Acholeplasma or the Pasteurella isolates, demonstrating a high degree of specificity. The assay produces a fluorescent signal within 15 to 20 min and worked well using pleural fluid obtained directly from CCPP-positive animals without prior DNA extraction. We demonstrate that the diagnosis of CCPP can be achieved, with a short sample preparation time and a simple read-out device that can be powered by a car battery, in <45 min in a simulated field setting.

Contagious bovine pleuropneumonia (CBPP) is a serious respiratory disease of cattle caused by Mycoplasma mycoides subsp. mycoides. Current vaccines against CBPP induce short-lived immunity and can cause severe postvaccine reactions. Previous studies have identified the N terminus of the transmembrane lipoprotein Q (LppQ-N') of M. mycoides subsp. mycoides as the major antigen and a possible virulence factor. We therefore immunized cattle with purified recombinant LppQ-N' formulated in Freund's adjuvant and challenged them with M. mycoides subsp. mycoides. Vaccinated animals showed a strong seroconversion to LppQ, but they exhibited significantly enhanced postchallenge glomerulonephritis compared to the placebo group (P = 0.021). Glomerulonephritis was characterized by features that suggested the development of antigen-antibody immune complexes. Clinical signs and gross pathological scores did not significantly differ between vaccinated and placebo groups. These findings reveal for the first time the pathogenesis of enhanced disease as a result of antibodies against LppQ during challenge and also argue against inclusion of LppQ-N' in a future subunit vaccine for CBPP.

Classical quorum-sensing (autoinduction) regulation, as exemplified by the lux system of Vibrio fischeri, requires N-acyl homoserine lactone (AHL) signals to stimulate cognate transcriptional activators for the cell density-dependent expression of specific target gene systems. For Pantoea stewartii subsp. stewartii, a bacterial pathogen of sweet corn and maize, the extracellular polysaccharide (EPS) stewartan is a major virulence factor, and its production is controlled by quorum sensing in a population density-dependent manner. Two genes, esaI and esaR, encode essential regulatory proteins for quorum sensing. EsaI is the AHL signal synthase, and EsaR is the cognate gene regulator. esaI, DeltaesaR, and DeltaesaI-esaR mutations were constructed to establish the regulatory role of EsaR. We report here that strains containing an esaR mutation produce high levels of EPS independently of cell density and in the absence of the AHL signal. Our data indicate that quorum-sensing regulation in P. s. subsp. stewartii, in contrast to most other described systems, uses EsaR to repress EPS synthesis at low cell density, and that derepression requires micromolar amounts of AHL. In addition, derepressed esaR strains, which synthesize EPS constitutively at low cell densities, were significantly less virulent than the wild-type parent. This finding suggests that quorum sensing in P. s. subsp. stewartii may be a mechanism to delay the expression of EPS during the early stages of infection so that it does not interfere with other mechanisms of pathogenesis.

Se realizó un estudio para conocer características relacionadas con el crecimiento inicial de la especie Chysolaena flexuosa (Sims) H. Rob., Asteraceae nativa de las sierras bonaerenses con potencial ornamental gracias al colorido de sus capítulos durante la floración y a su inflorescencia. Se empleó para el ensayo simiente de C. flexuosa recolectada en la Sierra La Barrosa (Partido de Balcarce) y se analizó la evolución de las plantas en invernáculo durante un ciclo de crecimiento. Los estadios reproductivos de C. flexuosa comprendieron las estaciones de primavera, verano e inicios de otoño. En todas las plantas se observaron, a mediados de la etapa reproductiva, capítulos con flores violáceas conjuntamente con capítulos fructificados. A principios de otoño todas las plantas poseían capítulos fructificados que exhibían sus papus de color blanco, situación que entrega a la planta otro interesante atributo. Por su rusticidad, establecida sobre la base de la germinación y supervivencia de sus plántulas, y por el atractivo de la especie desde primavera a inicios del otoño, C. flexuosa se destaca como promisoria para la obtención de cultivares ornamentales para uso como ejemplar aislado, en macetas, borduras o como flor de corte para la realización de arreglos florales secos.

In the Monte Biogeographic Province, located in the arid region of Argentina, the presence of Prosopis flexuosa DC. produces spatial heterogeneity through edaphic modifications and microclimate changes. This results in vegetation patches differing in species composition and abundance. However, this interaction can be modified by the occurrence of gradients of biotic stress or disturbance intensity. In particular, grazing has been observed to enhance or reduce vegetation heterogeneity. Such complex of interactions could determine forage availability for cattle in one of the driest areas of the Monte Desert. We assessed the effect of Prosopis on understory species and analyzed whether the outcomes of this interaction differed with distance to watering points, as a proxy of grazing intensity, in the Northeast of Mendoza Province, Argentina. We used a two-way factorial design including the following factors: 1) microsite (under the cover of P. flexuosa trees and in intercanopy microsites) and 2) distance to watering points ("near the watering point", 500-700 m away, and "far from the watering point", 3-4 km away). Cover of each species, total cover, bare soil, and litter were recorded, and plant diversity, richness, and evenness were estimated with the modified Point Quadrat method. Results showed that P. flexuosa cover, distance from watering points, and the interaction between them determined species composition, abundance and spatial distribution of understory species, and were, consequently, a determining factor for forage availability. The presence of P. flexuosa enhances carrying capacity by supporting higher abundance of grasses under its canopy. Near watering points, high grazing intensity appears to disrupt the patches formed under P. flexuosa canopies, reducing the differences between microsites.

El ordenamiento forestal de los bosques del país es esencial para el manejo sostenible de los mismos. Para que esto sea posible, es necesario inventariar y conocer las características de los bosques nativos. En zonas áridas, donde las tasas de regeneración de los bosques son lentas, es necesario conocer la estructura forestal, estado sanitario, estado de conservación, etc. a la hora de proponer pautas de manejo. Bajo este marco se están llevando a cabo importantes estudios sobre los algarrobales del Monte, con resultados significativos para su conservación y ordenamiento. Este trabajo tiene como objetivo principal conocer la estructura y estado de conservación de los algarrobales de Prosopis chilensis y Prosopis flexuosa de la depresión del Río Bermejo, en el departamento de Jáchal (San Juan), aportando información de base para la conservación y manejo de estos bosques. Se efectuó un inventario que tuvo en cuenta parámetros dasonómicos, el estado de conservación y la forma de los árboles. Además, se generó un mapa de bosques, a través del procesamiento y clasificación de una imagen LANDSAT TM 5 (14/03/2005). A partir del mapeo, se obtuvieron cuatro tipos de bosques. Se realizó un Análisis de Componentes Principales, para verificar la separación entre sitios pertenecientes a los diferentes tipos de bosques y justificar dicha clasificación. Se describieron dos algarrobales de Prosopis chilensis (La Ciénaga y Cauces) y dos algarrobales de P. flexuosa (Huaco y Monte Grande). Los bosques de P. chilensis, presentaron mayor diversidad de especies que los de P. flexuosa y se encontraron sobre los cauces de ríos temporales. El Bosque La Ciénaga (93,1 árboles ha-1) presentó mayor densidad que el Bosque Cauces (30,6 árboles ha-1), y mayor cantidad de renovales (clase de regeneración). La densidad del total del Bosque Huaco fue considerablemente mayor (420,1 árboles ha-1) que el Bosque Monte Grande (82,5 árboles ha-1). Estos dos, presentaron diferencias en densidad de renovales, porcentaje de cobertura de Prosopis, y demás parámetros analizados, siendo el Bosque Huaco un bosque con buen estado sanitario, buena regeneración y mayor porcentaje de cobertura, a diferencia del Bosque Monte Grande, un bosque con un estado sanitario pobre, muy baja regeneración y alto grado de disturbio. Los datos analizados en este estudio sugieren que debido a la estructura forestal de los bosques, no es posible la extracción de productos forestales de alto valor económico. En todos los bosques se encontraron signos de uso por parte de los pobladores locales, siendo el Bosque Monte Grande el de mayor presión de uso. Conocer la estructura forestal y demás variables es indispensable a la hora de proponer pautas de manejo. En base a los resultados obtenidos, se revisaron las categorías de conservación del inventario de Bosques Nativos de San Juan. Se propone recategorizar los Bosques La Ciénaga y Huaco.

El objetivo de esta investigación fue generar un modelo de distribución potencial de los bosques de Prosopis flexuosa, en la provincia Biogeográfica del Monte, desde Catamarca hasta Mendoza, para luego comparar con distribuciones actuales y poder inferir acerca de los principales procesos y cambios en la distribución que pudieron haber sufrido estos bosques ya sea por causas naturales o antrópicas. Para obtener el mapa modelo se utilizó el software Maxent, el cual utiliza como datos los registros de presencia (georreferenciados) de la especie y las variables ambientales que se consideran relevantes para su distribución. Para correr el modelo, se utilizaron las variables bioclimáticas (obtenidas del sitio Worldclim) y las variables altura, pendiente, orientación, subórdenes de suelos y freática. Como registros, se utilizaron puntos de presencia de bosques (tomados con GPS) en diferentes zonas del Monte. El mapa de distribución potencial resultante concuerda con la ubicación de los principales valles, bolsones y llanuras del Monte, en donde se conoce la existencia actual de los bosques de Prosopis flexuosa. En comparación con estas distribuciones reales, existe una importante coincidencia pero, en general, el área abarcada por los bosques potenciales es superior a lo determinado por las imágenes actuales, como así también existen casos particulares en los cuales las probabilidades de ocurrencia de bosques son muy bajas y las imágenes nos muestran existencia de los mismos. A su vez, al comparar con fuentes y registros históricos, con documentos oficiales (cartas IGM) y estadísticas del ferrocarril se pueden inferir posibles procesos de desmontes, que estarían determinando la ausencia de bosques actuales en sitios donde nuestro mapa potencial presenta altas probabilidades.

Nearly complete Paleogene sedimentary sequences were recovered by Leg 114 to the subantarctic South Atlantic. Silicoflagellate assemblages from the Paleogene and immediately overlying lower Neogene from Sites 698 (Northeast Georgia Rise), 700 (East Georgia Basin), 702 (Islas Orcadas Rise), and 703 (Meteor Rise) were examined. The described assemblage from Hole 700B represents the most complete yet described from the Paleocene, encompassing planktonic foraminifer Zones Plb (upper part) through P4 and Subchrons C25N to C23N. All lower Eocene sediments are barren as a result of diagenesis, except for a single sample from Hole 698A. Middle Eocene silicoflagellates described from Hole 702B range in age from early middle Eocene (P10) to late Eocene (PI5), with correlations to Subchrons C21N to C18N. Hole 703A contains late Eocene through early Miocene assemblages, with paleomagnetic control from Subchrons C16R to C6AAN. Leg 114 biosiliceous sequences contain exceptionally diverse assemblages of silicoflagellates. Approximately 155 species and separate morphotypes are described from the Paleogene and earliest Neogene. New taxa described from Leg 114 sediments include Bachmannocena vetula n. sp., Corbisema animoparallela n. sp., Corbisema camara n. sp., Corbisema constricta spinosa n. subsp., Corbisema delicata n. sp., Corbisema hastata aha n. subsp., Corbisema praedelicata n. sp., Corbisema scapana n. sp., Corbisema triacantha lepidospinosa n. subsp., Dictyocha deflandreifurtivia n. subsp., Naviculopsis biapiculata nodulifera n. subsp., Naviculopsis cruciata n. sp., Naviculopsis pandalata n. sp., Naviculopsis primativa n. sp., and Naviculopsis trispinosa eminula n. subsp. Taxonomic revisions were made to the following taxa: Corbisema constricta constricta emended, Corbisema disymmetrica crenulata n. comb., Corbisema jerseyensis emended, and Distephanus antarcticus n. comb. Silicoflagellate assemblages from the Paleogene and earliest Neogene of Holes 698A, 699A, 700B, 702B, and 703A are the basis of a silicoflagellate zonation spanning the interval from 63.2 to 22.25 Ma. Silicoflagellate zones recognized in this interval include the Corbisema hastata hastata Zone, Corbisema hastata aha Zone, Dictyocha precarentis Zone, Naviculopsis constricta Zone, Naviculopsis foliacea Zone, Bachmannocena vetula Zone, Dictyocha grandis Zone, Naviculopsis pandalata Zone, Naviculopsis constricta-Bachmannocena paulschulzii Zone, Bachmannocena paulschulzii Zone, Naviculopsis trispinosa Zone with subzones a and b, Corbisema archangelskiana Zone, Naviculopsis biapiculata Zone, Distephanus raupii Zone, Distephanus raupii-Corbisema triacantha Zone, and Corbisema triacantha mediana Zone.
