We report the cloning and characterization of a long interspersed nucleotide element (LINE) fi-om a cichlid fish, Oreochromis niloticus, and show the distribution of this element, called CiLINE2 for cichlid LINE2, in the chromosomes of this species. The identification of an open reading frame in CiLINE2 with amino acid sequence similarity to reverse transcriptases encoded by LINE-like elements in Caenorhabditis elegans, Platemys spixii, Schistosoma mansoni, Gallus gallus (CRI), Drosophila melanogaster (I factor), and Homo sapiens (LINE2), as well as the structure of the element, suggest it is a member of this family of non-long terminal repeat-containing retrotransposons. Search of a DNA sequence database identified sequences similar to CiLINE2 in four other fish species (Haplotaxodon microlepis, Oreochromis mossambicus, Pseudotropheus zebra, and Fugu rubripes). Southern blot hybridization experiments revealed the presence of sequences similar to CiLINE2 in all Tilapiini species analyzed from the genera Oreochromis, Tilapia, and Sarotherodon, and gave an estimated copy number of about 5500 for the haploid genome of O. niloticus. Fluorescent in situ hybridization showed that CiLINE2 sequences were organized in small clusters dispersed over all chromosomes of O. niloticus, with a higher concentration near chromosome ends. Furthermore the long arm of chromosome 1 was strikingly enriched with this sequence. The distribution of LINE2-related elements might underlie the difference in chromosome banding patterns observed between cold-blooded vertebrates and mammals.

This study aimed to evaluate the infection by the Austrodiplostomum compactum metacercariae in fishes from the Nova Avanhandava Reservoir, low Tiete river, São Paulo State, Brazil. The parasites were collected from eye (aqueous and vitreous humor), fixed in AFA solution and stained with carmine. The morphometric analysis was performed using a computerized system for analysis of images QWin Lite 2.5 (Leica). Prevalence, mean intensity of infection and abundance of infected fish were calculated. of the 22 species of fish registered, five were infected by metacercariae: Hoplias malabaricus, Metynnis maculatus, Plagioscion squamosissimus, Satanoperca pappaterra and Schizodon imparts. of the 627 fish evaluated, 34% were infected. A higher prevalence was observed in P. sqnamosissionis and S. pappaterra. Schizodon nasutus and M. maculatus are new hosts reported for A. compactum metacercariae.

Cytogenetic studies performed on 17 specimens (11 females and six males) of Astyanax scabripinnis paranae from the Cascatinha stream showed that this population has 2n = 50 chromosomes (8M + 22SM + 10ST + 10A), two chromosome pairs with NORs and conspicuous C-band positive blocks in the terminal position of the long arm of five chromosome pairs. Three females presented 2n = 51 chromosomes and the extra chromosome was a large metacentric similar in size and morphology to the first chromosome pair in the karyotype. This accessory chromosome was entirely heterochromatic in C-banded metaphases, which permitted its classification as a supernumerary chromosome. Some aspects related to the morphology of such macro B-chromosomes are discussed.

In the present study, we describe the cloning and characterization of a new SINE-like element from O. niloticus (ROn-2) and show the distribution of this SINE and a previously isolated SINE, ROn-1, in the chromosomes of O. niloticus. The ROn-2 element is 359 base pairs (bp) in length, contains short direct terminal repeats, a tRNA-related region similar to tRNA Val and tRNA Arg, a tRNA-unrelated region, and a poly-A tail. Analysis of the chromosomal distribution of ROn-1 and ROn-2 by fluorescent in situ hybridization showed that both SINE sequences are present in all chromosomes of tilapia, and organized in small clusters. The only exception was a large cluster of ROn-1 repeats found in the middle of the long arm of chromosome 1. In view of our data we discuss the hypothesis that the absence of large clusters of SINE sequences and the structural composition of these sequences may explain the absence of base-specific fluorochrome bands in the chromosomes of tilapia.

The present study shows the distribution of alien fish species in tropical lakes in the middle Doce river basin, southeastern Brazil, obtained from a rapid assessment program. The causes for their introductions were sport-fishing improvement in some specific lakes and aquaculture in the studied basin. Presently, these species have a wide distribution occurring in 41 of the 54 lakes studied, representing an actual threat to regional native fish community. The natural connection among lakes and streams during the rainy season and the dispersal mediated by local people are the main invasion agents for alien fishes. The success of these invaders is probably due to absence of pre-existing effective competitors or top-predators in the invaded communities. We consider that the eradication of alien fishes by means of the available management tools may be very difficult due to the large number of lakes invaded and to the wide spectrum of lake conditions and resources exploited by these alien species. We recommend the use of environmental education as a tool to stop the human-mediated dispersion of aliens and to improve conservation of native fish community in lakes where these alien species are not present yet. © 2004 Instituto de Ciências Biológicas - UFMG.

Bryconamericus turiuba, new species, is described from the upper Rio Paraná system in Brazil. It differs from all congeners in the following combination of characters: 15-20 branched anal-fin rays; four or five scales in a transverse series from the dorsal-fin origin to the lateral line; shallow body (depth 23.6-30.6% of SL); 37-43 perforated lateral line scales; first, third, and fifth or, more rarely, first and fourth outer premaxillary tricuspid teeth projecting anteriorly; two to five tricuspid to pentacuspid maxillary teeth; dentary with three or four tricuspid or pentacuspid large teeth, followed by five to seven smaller conical to tricuspid teeth; a conspicuous dark, vertically elongate humeral spot, extending to below the lateral line; a dark dorsal stripe extending from the supraoccipital spine to the caudal peduncle with a gap at the adipose-fin base; caudal-fin lobes without conspicuous markings, uniformly pigmented along rays; mature males lacking hooks on the pelvic- and anal-fin rays. The new species is compared with the Bryconamericus species described from the basins of the Rio Paraná, Rio Paraguay, Rio São Francisco, and coastal Brazilian rivers. © 2005 by the American Society of Ichthyologists and Herpetologists.

The Nile tilapia fish (Oreochromis niloticus) has a high potential to be used as a model in neuroscience studies. In the present study, the preference of the Nile tilapia between a gravel-enriched (GEE), a shelter-enriched (SEE) or a non-enriched (NEE) environment was determined, for developing a place preference model. Nile tilapia had an initial preference for GEE, but after 1 day of observation, the fish stabilized their frequency of visits among compartments. Hence, any stimulus motivating tilapia increase in compartment visiting indicates a positively reinforcing effect. This feature is very useful for the development of new behavioural paradigms for fish in tests using environmental discrimination, such as the conditioning place preference test. © 2006 Elsevier GmbH. Although there are almost thirty-thousand species of fish living in a great variety of habitats and utilizing vast reproductive strategies, our knowledge of morphofunctional and quantitative aspects of testis structure and spermatogenesis is still incipient for this group of vertebrates. In this review, we discuss aspects that are important to better understanding of testis structure and function, and of the development of germ cells (GC) during spermatogenesis. To achieve this, we have recently completed a number of studies presenting morphometric and functional data related to the numbers of GC and Sertoli cells (SC) per each type of spermatogenic cyst, the number of spermatogonial generations, the SC efficiency, and the magnitude of GC loss that normally occurs during spermatogenesis. We also investigated SC proliferation and the relationship of this important event to early spermatogenic cysts. The available data strongly suggest that SC proliferation in sexually mature tilapia is the primary factor responsible for the increase in testis size and for determination of the magnitude of sperm production. The influence of temperature on the duration of spermatogenesis in tilapia was also evaluated and we have used this knowledge to deplete endogenous spermatogenesis in this teleost, in order to develop an experimental system for GC transplantation. This exciting technique results in new possibilities for investigation of spermatogenesis and spermatogonial stem cell biology, creating also an entirely new and promising scenario in biotechnology - transgenic animal production and the preservation of the genetic stocks of valuable animals or endangered species. © Springer Science+Business Media B.V. 2008.

This study presents new information on feeding habits of Guiana dolphins, Sotalia guianensis, in south-eastern Brazil, together with new regression equations to evaluate the weight and length of fish from otoliths, showing an overview on the knowledge about this species-diet in this area. Eighteen stomach contents had been analysed and compared to 180 samples collected in another eight feeding studies. The analysed specimens were either incidentally caught in gillnets used in coastal waters by the fleet based in the Cananéia main harbour (25°00âS 47°55âW), south of São Paulo State, or found dead in inner waters of the Cananéia estuary between 2003 and 2009. Based on the index of relative importance analysis, the most important fish species were the banded croaker, Paralonchurus brasiliensis. Doryteuthis plei was the most representative cephalopod species. Stellifer rastrifer was the most important fish species observed in dolphins in inner estuarine waters and P. brasiliensis in recovered dolphins from coastal waters. Loliguncula brevis is the only cephalopod species reported from dolphins found in inner estuarine waters up to date. Doryteuthis plei was the most important cephalopod species observed in coastal dolphins. When considering other feeding studies, the most representative fish family in the diet of S. guianensis was Sciaenidae, which is mainly represented by demersal fishes. The main preys of S. guianensis are abundant in the studied areas, which may indicate an opportunistic feeding habit. The majority of them are not the most important target species by the commercial fishery in south-eastern Brazil. © 2012 Marine Biological Association of the United Kingdom.

Chilodonelids are small ciliated protozoans found worldwide and can be dangerous in culture conditions. This study presents morphometric data on the ciliate Chilodonella that is found in cultured Nile tilapia (Oreochromis niloticus), native bait fish tuvira (Gymnotus aff. inaequilabiatus) and native pacu (Piaractus mesopotamicus) and includes a histopathological assessment of the changes that occur in the pacu. For parasitic diagnosis, skin and gill samples were scraped onto slides, dried at room temperature, stained with Giemsa or impregnated with silver nitrate, and the measurements were obtained from photomicrographs. In the diseased pacu, the first gill arch was collected and fixed in a 10% buffered formalin solution for histopathological analysis. Parasite specimens from the different collection sites were identified morphologically as C. hexasticha Kiernik (1909). Diseased fish exhibited depigmentation, skin ulceration, scale loss, excessive mucus production and gill lesions. Histopathological analysis of pacu gills displayed epithelial proliferation with mononuclear inflammatory infiltrate, hemorrhages, and scattering necrosis. In Brazilian-farmed fish this is the first record of C. hexasticha, which has great pathogenic potential in cultured freshwater species. In addition, two new hosts are presented. © 2012 Elsevier B.V.
