Background: Several studies seek biological markers that give diagnostic and degree of tumor development. The aim of this study was to validate the determination of plasma DNA using nanotechnology (Nanovue™-NV) in samples of 80 patients with prostate cancer. Methods. Blood samples of 80 patients of the Urology Ambulatory of Faculdade de Medicina do ABC with prostate cancer confirmed by anatomical-pathology criteria were analyzed. DNA extraction was performed using a GFX TM kit (Amersham Pharmacia Biotech, Inc, USA) following the adapted protocol. Plasma was subjected to centrifugation. Results: There was a big difference between the first and the second value obtained by NanoVue Only two samples had no differences between duplicates. Maximum difference between duplicates was 38 μg/mL. Average variation between 51 samples was 10.29 μg/mL, although 21 samples had differences above this average. No correlation was observed between pDNA obtained by traditional spectrophotometry and by nanotechnology. Conclusion: Determination of plasma DNA by nanotechnology was not reproducible. © 2013 Moreno et al; licensee BioMed Central Ltd.

This paper is concerned with an overview of upwinding schemes, and further nonlinear applications of a recently introduced high resolution upwind differencing scheme, namely the ADBQUICKEST [V.G. Ferreira, F.A. Kurokawa, R.A.B. Queiroz, M.K. Kaibara, C.M. Oishi, J.A.Cuminato, A.F. Castelo, M.F. Tomé, S. McKee, assessment of a high-order finite difference upwind scheme for the simulation of convection-diffusion problems, International Journal for Numerical Methods in Fluids 60 (2009) 1-26]. The ADBQUICKEST scheme is a new TVD version of the QUICKEST [B.P. Leonard, A stable and accurate convective modeling procedure based on quadratic upstream interpolation, Computer Methods in Applied Mechanics and Engineering 19 (1979) 59-98] for solving nonlinear balance laws. The scheme is based on the concept of NV and TVD formalisms and satisfies a convective boundedness criterion. The accuracy of the scheme is compared with other popularly used convective upwinding schemes (see, for example, Roe (1985) [19], Van Leer (1974) [18] and Arora & Roe (1997) [17]) for solving nonlinear conservation laws (for example, Buckley-Leverett, shallow water and Euler equations). The ADBQUICKEST scheme is then used to solve six types of fluid flow problems of increasing complexity: namely, 2D aerosol filtration by fibrous filters; axisymmetric flow in a tubular membrane; 2D two-phase flow in a fluidized bed; 2D compressible Orszag-Tang MHD vortex; axisymmetric jet onto a flat surface at low Reynolds number and full 3D incompressible flows involving moving free surfaces. The numerical simulations indicate that this convective upwinding scheme is a good generic alternative for solving complex fluid dynamics problems. © 2012.

In the northwestern region of the state of São Paulo, Brazil, the freshwater crab Dilocarcinus pagei Stimpson, 1861 is usually sold as bait for several fish species for sport fishing. Thus, knowledge of the reproductive biology of this species is essential to provide information for maintaining natural stocks and implementing future farming initiatives. The objective of this study was to investigate the population dynamics of the red freshwater crab D. pagei, focusing on reproductive and juvenile recruitment periods, sex ratio, and individual growth. Sampling was performed monthly from October 2005 to September 2007 in the Marimbondo-Fumas Hydroelectric Plant reservoir, Rio Grande, municipality of Icém in northwestern São Paulo state. The individuals were sexed, carapace widths were measured, and the gonads were examined macroscopically. In total, 1002 individuals were analyzed, consisting of 568 males and 434 females, of which 4 were ovigerous and 35 carried early juveniles in the incubatory chamber. The largest numbers of reproductive individuals of both sexes were recorded during the winter and spring months before spawning, which occurred in the spring. Intense juvenile recruitment and the highest rainfall levels were observed during the Southern Hemisphere summer. The results indicate that spawning can be considered seasonal, and the simultaneous emergence of juveniles with high rainfall levels is probably the result of the higher availability of food and potential sites for protection against predators. The skewed sex ratio (♂ 1:0.76 ♀) may be associated with growth rates, mortality, food restriction, behavioral differences, migration, and differential habitat use between the sexes. The similar maximum size (CW∞) and growth constant (k) for males (k = 1.78 y-1, CW = 61.43 mm) and females (k = 1.67 y-1, CW = 60.66 mm) may be associated with different energy investment strategies between the sexes. The mandibles of bees contain two types of tegumental glands whose function is not clear, despite the hypotheses put forward by several researchers. Although these glands have been found in all the bee species studied so far, observations have been confined mostly to workers of eusocial species in the forager phase. The work reported here involved a study of the morphology of the glands of newly emerged, nurse and forager workers, virgin and fecundated queens, and newly emerged and sexually mature males of Scaptotrigona postica, seeking to identify changes that may be linked to the bees life phase. Our findings indicate that the two types of glands are present in the species but not in all life phases or individual classes. The glands consisting of class I cells, the epithelial glands are present only in forager workers and fecundated queens. Glands of type III cells were studied in detail, and gland size was estimated from histological sections. The degree of development of the glands varies according to individual classes and life phases, suggesting different functions during the individuals life and from one individual to another. © Koninklijke Brill NV, Leiden, 2012.

This study presents the ovarian morphology and the dynamics of the vitellogenesis process in the oocytes of the diplopod Leptodesmus dentellus. The oocytes are arranged in clusters called ovisacs which are distributed in pairs along the midline of the body forming the ovary. Regions similar to the germarium appear paired in the anterior region of the ovary; however, the development of the oocytes of this species does not follow a regionalisation in the reproductive organ. Cells in three developmental stages are found throughout the length of the ovary. Calcium, proteins, lipids and neutral polysaccharides were detected in the oocytes of L. dentellus. The polysaccharides and the proteins found in the oocytes have a double origin: endogenous, with the participation of the germinative vesicle, and exogenous, from follicular epithelium. The origin of the lipids is exogenous, i.e., they are incorporated into the oocytes, probably derived from the perivisceral fat body, which are highly developed in this region. The deposition of calcium is pre-vitellogenic and probably functions as a reserve during the juvenile stages. © 2012 Koninklijke Brill NV, Leiden.

Apresentamos um novo método de inversão linear bidimensional de dados gravimétricos produzidos por bacias sedimentares com relevo do embasamento descontínuo. O método desenvolvido utiliza um modelo interpretativo formado por um conjunto de fitas horizontais bidimensionais justapostas cujas espessuras são os parâmetros a serem estimados. O contraste de densidade entre o embasamento e os sedimentos é presumido constante e conhecido. As estimativas das espessuras foram estabilizadas com o funcional da Variação Total (VT) que permite soluções apresentando descontinuidades locais no relevo do embasamento. As estimativas do relevo são obtidas através da resolução de um sistema de equações lineares, resolvido na norma L1. Como métodos lineares subestimam as estimativas de profundidade do embasamento de bacias maiores que cerca de 500 m, amplificamos as estimativas de profundidade através da modificação da matriz associada ao modelo interpretativo de fitas. As estimativas obtidas através deste procedimento são em geral ligeiramente superestimadas. Desse modo, elas são corrigidas através de uma correção definida pela expressão da placa Bouguer. Testes em dados sintéticos e reais produziram resultados comparáveis aos produzidos pelo método não linear, mas exigiram menor tempo computacional. A razão R entre os tempos exigidos pelo método não linear e o método proposto cresce com o número de observações e parâmetros. Por exemplo, para 60 observações e 60 parâmetros, R é igual a 4, enquanto para 2500 observações e 2500 parâmetros R cresce para 16,8. O método proposto e o método de inversão não linear foram aplicados também em dados reais do Steptoe Valley, Nevada, Estados Unidos, e da ponte do POEMA, no Campus do Guamá em Belém, produzindo soluções similares às obtidas com o método não linear exigindo menor tempo computacional.
