Background

Product adherence is a pivotal issue in the development of effective vaginal microbicides to reduce sexual transmission of HIV. To date, the six Phase III studies of vaginal gel products have relied primarily on self-reporting of adherence. Accurate and reliable methods for monitoring user adherence to microbicide-releasing vaginal rings have yet to be established.

Methods

A silicone elastomer vaginal ring prototype containing an embedded, miniature temperature logger has been developed and tested in vitro and in cynomolgus macaques for its potential to continuously monitor environmental temperature and accurately determine episodes of ring insertion and removal.

Results

In vitro studies demonstrated that DST nano-T temperature loggers encapsulated in medical grade silicone elastomer were able to accurately and continuously measure environmental temperature. The devices responded quickly to temperature changes despite being embedded in different thickness of silicone elastomer. Prototype vaginal rings measured higher temperatures compared with a subcutaneously implanted device, showed high sensitivity to diurnal fluctuations in vaginal temperature, and accurately detected periods of ring removal when tested in macaques.

Conclusions

Vaginal rings containing embedded temperature loggers may be useful in the assessment of product adherence in late-stage clinical trials.

OBJECTIVES: Microneedle (MN) arrays could offer a pain-free, minimally invasive approach to monitoring. This is envisaged to be particularly beneficial for younger patients, but parents' views to date are unknown. The aim of this study was to explore parental perceptions of MN-mediated ISF monitoring, as an alternative to the use of conventional blood sampling, and to understand the important factors for technique approval.

METHODS: Semi-structured interviews were conducted with parents with recent experience of a premature birth. Recruitment was through the Northern Ireland premature infant charity, Tinylife. Interviews progressed until data saturation was reached and thematic analysis employed.

KEY FINDINGS: The study included 16 parents. Parental support for MN-mediated monitoring was evident, alongside the unpopularity of traditional blood sampling in neonates. Factors facilitating MN approval included the opportunity for pain reduction, the simplicity of the procedure, the potential for increased parental involvement and the more favourable appearance, owing to the minute size of MNs and similarities with a sticking plaster. Confirmation of correct application, a pain-free patch removal and endorsement from trusted healthcare professionals were important.

CONCLUSION: These findings will inform researchers in the field of MN development and enlighten practitioners regarding parental distress resulting from conventional blood sampling. Further work is necessary to understand MN acceptability among practitioners. This work should assist in the development of an acceptable MN device and facilitate the reduction of parental distress.

Microneedle (MN) arrays could offer an alternative method to traditional drug delivery and blood sampling methods. However, acceptance among key end-users is critical for new technologies to succeed. MNs have been advocated for use in children and so, paediatricians are key potential end-users. However, the opinions of paediatricians on MN use have been previously unexplored. The aim of this study was to investigate the views of UK paediatricians on the use of MN technology within neonatal and paediatric care. An online survey was developed and distributed among UK paediatricians to gain their opinions of MN technology and its use in the neonatal and paediatric care settings, particularly for MN-mediated monitoring. A total of 145 responses were obtained, with a completion response rate of 13.7 %. Respondents believed an alternative monitoring technique to blood sampling in children was required. Furthermore, 83 % of paediatricians believed there was a particular need in premature neonates. Overall, this potential end-user group approved of the MN technology and a MN-mediated monitoring approach. Minimal pain and the perceived ease of use were important elements in gaining favour. Concerns included the need for confirmation of correct application and the potential for skin irritation. The findings of this study provide an initial indication of MN acceptability among a key potential end-user group. Furthermore, the concerns identified present a challenge to those working within the MN field to provide solutions to further improve this technology. The work strengthens the rationale behind MN technology and facilitates the translation of MN technology from lab bench into the clinical setting.

The accumulation of biogenic greenhouse gases (methane, carbon dioxide) in organic sediments is an important factor in the redevelopment and risk management of many brownfield sites. Good practice with brownfield site characterization requires the identification of free-gas phases and pathways that allow its migration and release at the ground surface. Gas pockets trapped in the subsurface have contrasting properties with the surrounding porous media that favor their detection using geophysical methods. We have developed a case study in which pockets of gas were intercepted with multilevel monitoring wells, and their lateral continuity was monitored over time using resistivity. We have developed a novel interpretation procedure based on Archie’s law to evaluate changes in water and gas content with respect to a mean background medium. We have used induced polarization data to account for errors in applying Archie’s law due to the contribution of surface conductivity effects. Mosaics defined by changes in water saturation allowed the recognition of gas migration and groundwater infiltration routes and the association of gas and groundwater fluxes. The inference on flux patterns was analyzed by taking into account pressure measurements in trapped gas reservoirs and by metagenomic analysis of the microbiological content, which was retrieved from suspended sediments in groundwater sampled in multilevel monitoring wells. A conceptual model combining physical and microbiological subsurface processes suggested that biogas trapped at depth may have the ability to quickly travel to the surface.

A novel model-based principal component analysis (PCA) method is proposed in this paper for wide-area power system monitoring, aiming to tackle one of the critical drawbacks of the conventional PCA, i.e. the incapability to handle non-Gaussian distributed variables. It is a significant extension of the original PCA method which has already shown to outperform traditional methods like rate-of-change-of-frequency (ROCOF). The ROCOF method is quick for processing local information, but its threshold is difficult to determine and nuisance tripping may easily occur. The proposed model-based PCA method uses a radial basis function neural network (RBFNN) model to handle the nonlinearity in the data set to solve the no-Gaussian issue, before the PCA method is used for islanding detection. To build an effective RBFNN model, this paper first uses a fast input selection method to remove insignificant neural inputs. Next, a heuristic optimization technique namely Teaching-Learning-Based-Optimization (TLBO) is adopted to tune the nonlinear parameters in the RBF neurons to build the optimized model. The novel RBFNN based PCA monitoring scheme is then employed for wide-area monitoring using the residuals between the model outputs and the real PMU measurements. Experimental results confirm the efficiency and effectiveness of the proposed method in monitoring a suite of process variables with different distribution characteristics, showing that the proposed RBFNN PCA method is a reliable scheme as an effective extension to the linear PCA method.

There is an increasing interest in the biomedical field to create implantable medical devices to provide a temporary mechanical function for use inside the human body. In many of these applications bioresorbable polymer composites using PLLA with β-TCP , are increasingly being used due to their biocompatability, biodegradability and mechanical strength.1,3 These medical devices can be manufactured using conventional plastics processing methods such as injection moulding and extrusion, however there is great need to understand and control the process due to a lack of knowledge on the influence of processing on material properties. With the addition of biocompatible additives there is also a requirement to be able to predict the quality and level of dispersion within the polymer matrix. On-line UV-Vis spectroscopy has been shown to monitor the quality of fillers in polymers. This can eliminate time consuming and costly post-process evaluation of additive dispersion. The aim of this work was to identify process and performance relationships of PLLA/β-TCP composites with respect to melt-extrusion conditions. This is part of a wider study into on-line process monitoring of bioresorbable polymers as used in the medical industry.
These results show that final properties of the PLLA/ β-TCP composite are highly influenced by the particle size and loading. UV-Vis spectroscopy can be used on-line to monitor the final product and this can be utilised as a valuable tool for quality control in an application where consistent performance is of paramount importance.

Landslides and debris flows, commonly triggered by rainfall, pose a geotechnical risk causing disruption to transport routes and incur significant financial expenditure. With infrastructure maintenance budgets becoming ever more constrained, this paper provides an overview of some of the developing methods being implemented by Queen’s University, Belfast in collaboration with the Department for Regional Development to monitor the stability of two distinctly different infrastructure slopes in Northern Ireland. In addition to the traditional, intrusive ground investigative and laboratory testing methods, aerial LiDAR, terrestrial LiDAR, geophysical techniques and differential Global Positioning Systems have been used to monitor slope stability. Finally, a comparison between terrestrial LiDAR, pore water pressure and soil moisture deficit (SMD) is presented to outline the processes for a more informed management regime and to highlight the season relationship between landslide activity and the aforementioned parameters.

Those living with an acquired brain injury often have issues with fatigue due to factors resulting from the injury. Cognitive impairments such as lack of memory, concentration and planning have a great impact on an individual’s ability to carry out general everyday tasks, which subsequently has the effect of inducing cognitive fatigue. Moreover, there is difficulty in assessing cognitive fatigue, as there are no real biological markers that can be measured. Rather, it is a very subjective effect that can only be diagnosed by the individual. Consequently, the traditional way of assessing cognitive fatigue is to use a self-assessment questionnaire that is able to determine contributing factors. State of the art methods to evaluate cognitive! fa tigue employ cognitive tests in order to analyse performance on predefined tasks. However, one primary issue with such tests is that they are typically carried out in a clinical environment, therefore do not have the ability to be utilized in situ within everyday life. This paper presents a smartphone application for the evaluation of fatigue, which can be used daily to track cognitive performance in order to assess the influence of fatigue.

The Antrim Coast Road stretching from the seaport of Larne in the East of Northern Ireland has a well-deserved reputation for being one of the most spectacular roads in Europe (Day, 2006). However the problematic geology; Jurassic Lias Clay and Triassic Mudstone overlain by Cretaceous Limestone and Tertiary Basalt, and environmental variables result in frequent instances of slope instability manifested in both shallow debris flows and occasional massive rotational movements, creating a geotechnical risk to this highway. This paper describes how a variety of techniques are being used to both assess instability and monitor movement of these active slopes near one site at Straidkilly Point, Glenarm. An in-depth understanding of the geology was obtained via boreholes, resistivity surveys and laboratory testing. Environmental variables recorded by an on-site weather station were correlated with measured pore water pressure and soil moisture infiltration data. Terrestrial LiDAR (TLS), with surveys carried out on a bi-monthly basis allowed for the generation of Digital Elevation Models (DEMs) of difference, highlighting areas of recent movement, accumulation and depletion. Morphology parameters were generated from the DEMs and include slope, curvature and multiple measures of roughness. Changes in the structure of the slope coupled with morphological parameters were characterised and linked to progressive failures from the temporal monitoring. In addition to TLS monitoring, Aerial LiDAR datasets were used for the spatio-morphological characterisation of the slope on a macro scale. A Differential Global Positioning System (dGPS) was also deployed on site to provide a real-time warning system for gross movements, which were also correlated with environmental conditions. Frequent electrical resistivity tomography (ERT) surveys were also implemented to provide a better understanding of long-term changes in soil moisture and help to define the complex geology. The paper describes how the data obtained via a diverse range of methods has been combined to facilitate a more informed management regime of geotechnical risk by the Northern Ireland Roads Service.

The coast of the Bulgarian Black Sea is a popular summer holiday destination. The Dam of Iskar is the largest artificial dam in Bulgaria, with a capacity of 675 million m3. It is the main source of tap water for the capital Sofia and for irrigating the surrounding valley. There is a close relationship between the quality of aquatic ecosystems and human health as many infections are waterborne. Rapid molecular methods for the analysis of highly pathogenic bacteria have been developed for monitoring quality. Mycobacterial species can be isolated from waste, surface, recreational, ground and tap waters and human pathogenicity of nontuberculose mycobacteria (NTM) is well recognized. The objective of our study was to perform molecular analysis for key-pathogens, with a focus on mycobacteria, in water samples collected from the Black Sea and the Dam of Iskar. In a two year period, 38 water samples were collected-24 from the Dam of Iskar and 14 from the Black Sea coastal zone. Fifty liter water samples were concentrated by ultrafiltration. Molecular analysis for 15 pathogens, including all species of genus Mycobacterium was performed. Our results showed presence of Vibrio spp. in the Black Sea. Rotavirus A was also identified in four samples from the Dam of Iskar. Toxigenic Escherichia coli was present in both locations, based on markers for stx1 and stx2 genes. No detectable amounts of Cryptosporidium were detected in either location using immunomagnetic separation and fluorescence microscopy. Furthermore, mass spectrometry analyses did not detect key cyanobacterial toxins. On the basis of the results obtained we can conclude that for the period 2012-2014 no Mycobacterium species were present in the water samples. During the study period no cases of waterborne infections were reported.

In recent years, Structural Health Monitoring (SHM) systems have been developed to monitor bridge deterioration, assess real load levels and hence extend bridge life and safety. A road bridge is only safe if the stresses caused by the passing vehicles are less than the capacity of the bridge to resist them. Conventional SHM systems can be used to improve knowledge of the bridges capacity to resist stresses but generally give no information on the causes of any increase in stresses (based on measuring strain). The concept of in Bridge Weigh-in-Motion (B-WIM) is to establish axle loads, without interruption to traffic flow, by using strain sensors at a bridge soffit and subsequently converting the data to real time axle loads or stresses. Recent studies have shown it would be most beneficial to develop a portable system which can be easily attached to existing and new bridge structures for a specified monitoring period. The sensors could then be left in place while the data acquisition can be moved for various other sites. Therefore it is necessary to find accurate sensors capable of capturing peak strains under dynamic load and suitable methods for attaching these strain sensors to existing and new bridge structures. Additionally, it is important to ensure accurate strain transfer between concrete and steel, the adhesives layer and the strain sensor. This paper describes research investigating the suitably of using various sensors for the monitoring of concrete structures under dynamic vehicle load. Electrical resistance strain (ERS) gauges, vibrating wire (VW) gauges and fibre optic sensors (FOS) are commonly used for SHM. A comparative study will be carried out to select a suitable sensor for a bridge Weigh in Motion System. This study will look at fixing methods, durability, scanning rate and accuracy range. Finite element modeling is used to predict the strains which are then validated in laboratory trials.

Sediment particle size analysis (PSA) is routinely used to support benthic macrofaunal community distribution data in habitat mapping and Ecological Status (ES) assessment. No optimal PSA Method to explain variability in multivariate macrofaunal distribution has been identified nor have the effects of changing sampling strategy been examined. Here, we use benthic macrofaunal and PSA grabs from two embayments in the south of Ireland. Four frequently used PSA Methods and two common sampling strategies are applied. A combination of laser particle sizing and wet/dry sieving without peroxide pre-treatment to remove organics was identified as the optimal Method for explaining macrofaunal distributions. ES classifications and EUNIS sediment classification were robust to changes in PSA Method. Fauna and PSA samples returned from the same grab sample significantly decreased macrofaunal variance explained by PSA and caused ES to be classified as lower. Employing the optimal PSA Method and sampling strategy will improve benthic monitoring. © 2012 Elsevier Ltd.

Harmful algal blooms (HABs) are a natural global phenomena emerging in severity and extent. Incidents have many economic, ecological and human health impacts. Monitoring and providing early warning of toxic HABs are critical for protecting public health. Current monitoring programmes include measuring the number of toxic phytoplankton cells in the water and biotoxin levels in shellfish tissue. As these efforts are demanding and labour intensive, methods which improve the efficiency are essential. This study compares the utilisation of a multitoxin surface plasmon resonance (multitoxin SPR) biosensor with enzyme-linked immunosorbent assay (ELISA) and analytical methods such as high performance liquid chromatography with fluorescence detection (HPLC-FLD) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) for toxic HAB monitoring efforts in Europe. Seawater samples (n = 256) from European waters, collected 2009-2011, were analysed for biotoxins: saxitoxin and analogues, okadaic acid and dinophysistoxins 1/2 (DTX1/DTX2) and domoic acid responsible for paralytic shellfish poisoning (PSP), diarrheic shellfish poisoning (DSP) and amnesic shellfish poisoning (ASP), respectively. Biotoxins were detected mainly in samples from Spain and Ireland. France and Norway appeared to have the lowest number of toxic samples. Both the multitoxin SPR biosensor and the RNA microarray were more sensitive at detecting toxic HABs than standard light microscopy phytoplankton monitoring. Correlations between each of the detection methods were performed with the overall agreement, based on statistical 2 × 2 comparison tables, between each testing platform ranging between 32% and 74% for all three toxin families illustrating that one individual testing method may not be an ideal solution. An efficient early warning monitoring system for the detection of toxic HABs could therefore be achieved by combining both the multitoxin SPR biosensor and RNA microarray.

Os compostos de tributilestanho (TBT) foram utilizados como biocidas em tintas antivegetativas (AV) e amplamente aplicados, durante décadas, de forma a evitar a bioincrustação em superfícies submersas, principalmente em cascos de embarcações. Os seus efeitos deletérios em organismos não-alvo tornaram-se evidentes após o aparecimento de gastrópodes prosobrâquios com imposex – sobreposição de caracteres sexuais masculinos sobre o tracto reprodutivo das fêmeas. Desde então, a expressão do imposex em prosobrânquios tem sido amplamente utilizada como biomarcador da poluição por TBT. Um dos objectivos da presente tese é avaliar se as mais recentes restrições legais na utilização das tintas AV com compostos organoestânicos (OTs) resultaram numa redução da poluição pelos mesmos na costa continental Portuguesa. Para tal, foi levada a cabo a análise da variação temporal do imposex como biomarcador dos níveis ambientais de TBT e a validação de procedimentos, de modo a seguir de forma precisa a evolução da intensidade deste fenómeno ao longo do tempo. O trabalho de investigação teve início no momento em que a ineficácia da legislação anterior (Directiva 89/677/CEE) na redução da poluição por TBT no litoral Português era reportada na literatura e quando estavam já agendados instrumentos decisivos para a diminuição definitiva deste tipo de poluição: o Regulamento (CE) N.º 782/2003 do Parlamento e do Conselho da União Europeia (UE) que bania as tintas AV baseadas em TBT na sua frota a partir de 1 de Julho de 2003; a "Convenção Internacional sobre o Controlo de Sistemas Antivegetativos Nocivos em Navios” (Convenção AFS), adoptada em 2001 pela Organização Marítima Internacional (OMI), que procurava erradicar os OTs da frota mundial até 2008. Os níveis de imposex e as concentrações de OTs nos tecidos de fêmeas de Nucella lapillus foram medidos em 17 locais de amostragem ao longo da costa Portuguesa em 2003, a fim de se avaliarem os impactos do TBT nas populações desta espécie e de se criar uma base de dados para seguir a sua evolução futura. O índice da sequência do vaso deferente (VDSI), o índice do tamanho relativo do pénis (RPSI) e a percentagem de fêmeas afectadas por imposex (%I) foram utilizados na medição da intensidade deste fenómeno em cada local de amostragem e os seus valores variaram entre 0,20-4,04, 0,0- 42,2% e 16,7-100,0%, respectivamente. Foram encontradas fêmeas estéreis em 3 locais de amostragem, com percentagens a variar entre 4,0 e 6,2%. As concentrações de TBT e dibutilestanho (DBT) nas fêmeas variaram respectivamente entre 23-138 e <10-62 ng Sn.g-1 de peso seco, e o conteúdo em TBT nos tecidos revelou-se significativamente correlacionado com o imposex (nomeadamente com o RPSI e o VDSI). Os níveis de expressão do fenómeno e o conteúdo em OTs nos tecidos, foram superiores na proximidade de portos, confirmando as conclusões obtidas anteriormente por outros autores de que os navios e a actividade dos estaleiros são as principais fontes destes compostos no litoral Português. As infra-estruturas associadas ás principais fontes de OTs – portos, estaleiros e marinas – estão geralmente localizadas no interior de estuários, motivo pelo qual estas áreas têm vindo a ser descritas como as mais afectadas por estes poluentes. Por esta razão, foi levado a cabo o estudo pormenorizado da poluição por TBT na Ria de Aveiro, como caso de estudo representativo da poluição por estes compostos num sistema estuarino em Portugal continental. N. lapillus foi usada como bioindicador para avaliar a tendência temporal da poluição por TBT nesta área entre 1997 e 2007. Foi registada uma diminuição da intensidade do imposex após 2003, embora as melhorias mais evidentes tenham sido observadas entre 2005 e 2007, provavelmente devido à implementação do Regulamento (CE) N.º 782/2003 que proibiu a aplicação de tintas AV com TBT em navios com a bandeira da UE. Apesar desses progressos, as análises ao conteúdo em OTs nos tecidos de fêmeas de N. lapillus e em amostras de água colhidas em 2006 indicaram contaminação recente por TBT na área de estudo, evidenciando assim a permanência de fontes de poluição. A utilização de N. lapillus como bioindicador da poluição por TBT na Ria de Aveiro apresenta algumas limitações uma vez que a espécie não ocorre nas áreas mais interiores da Ria e não vive em contacto com sedimentos. Assim, a informação obtida a partir da sua utilização como bioindicador é fundamentalmente relativa aos níveis de TBT na coluna de água. Foi então necessário recorrer a um bioindicador complementar – Hydrobia ulvae – para melhor avaliar a evolução temporal da poluição por TBT no interior deste sistema estuarino e estudar a persistência de TBT nos sedimentos. Não foi registada uma diminuição dos níveis de imposex em H. ulvae na Ria de Aveiro entre 1998 e 2007, apesar da aplicação do Regulamento (CE) N.º 782/2003. Pelo contrário, houve um aumento global significativo da percentagem de fêmeas afectadas por imposex e um ligeiro aumento do VDSI, contrastando com o que tem sido descrito para outros bioindicadores na Ria de Aveiro no mesmo período. Estes resultados mostram que a diferente biologia/ecologia das espécies indicadoras determina vias distintas de acumulação de TBT, apontando a importância da escolha do bioindicador dependendo do compartimento a ser monitorizado (sedimento versus água). A ingestão de sedimento como hábito alimentar em H. ulvae foi discutida como sendo a razão para a escolha da espécie como indicadora da contaminação dos sedimentos por TBT. Foram também estudados os métodos mais fiáveis para reduzir a influência de variáveis críticas na medição dos níveis de imposex em H. ulvae. As comparações de parâmetros do imposex baseados em medições do pénis devem ser sempre realizadas sob condições de narcotização bem standardizadas uma vez que este procedimento provoca um aumento significativo do comprimento do pénis (PL) em ambos os sexos. O VDSI, a % e o PL em ambos os sexos revelaram ser fortemente influenciados pelo tamanho dos espécimes: a utilização de fêmeas mais pequenas conduz à subestimação do VDSI, da %I e do PL, enquanto que diferenças no tamanho dos machos provocam variações no índice do comprimento relativo do pénis (RPLI), independentemente dos níveis de poluição por TBT. Existe, portanto, a necessidade de controlar algumas variáveis envolvidas na análise do imposex que mostraram afectar a fiabilidade dos resultados. Uma vez que N. lapillus é o principal bioindicador dos efeitos biológicos específicos do TBT para a área da OSPAR, foi também estudada a influência de algumas variáveis na avaliação dos níveis de imposex nesta espécie, especificamente as relacionadas com o ciclo reprodutor e o tamanho dos espécimes. O estudo do ciclo reprodutor e a variação sazonal/espacial do comprimento do pénis do macho (MPL) incidiu num único local no litoral Português (Areão – região de Aveiro) de forma a avaliar se o RPSI varia sazonal e espacialmente na mesma estação de amostragem e se tais variações influenciam os resultados obtidos em programas de monitorização do imposex. Nos meses de Dezembro de 2005 a Junho de 2007, foram encontrados espécimes de N. lapillus sexualmente maturos e potencialmente aptos para a reprodução. Contudo, foi também evidente um padrão sazonal do ciclo reprodutor – o estado de desenvolvimento da gametogénese nas fêmeas variou sazonalmente e ocorreu uma diminuição do volume da glândula da cápsula e do factor de condição no final do Verão / início do Outono. Contrariamente, a gametogénese nos machos não apresentou variação sazonal significativa, embora os valores mais baixos do factor de condição, do comprimento do pénis e dos volumes de esperma e da próstata tenham também sido registados no final do Verão / início do Outono. Além disso, o MPL mostrou variar, no mesmo local de amostragem, inversamente com a distância aos ninhos de cápsulas ovígeras; um aumento dos valores do MPL foi também observado em espécimes de maior tamanho. Todas estas variações no MPL introduzem desvios nos resultados da avaliação do imposex quando é usado o RPSI. A magnitude do erro envolvido foi quantificada e revelou-se superior em locais com níveis mais elevados de poluição por TBT. Apesar do RPSI ser um índice que fornece informação importante sobre os níveis de poluição por TBT, a sua interpretação deve ser cuidadosa e realizada complementarmente com os outros índices, destacando-se o VDSI como índice mais fiável e com significado biológico mais expressivo. Novas campanhas de monitorização do imposex em N. lapillus foram realizadas ao longo da costa Portuguesa em 2006 e 2008, e os resultados subsequentemente comparados com a base de dados criada em 2003,de forma a avaliar a evolução da poluição por TBT no litoral Português naquele período. Nestes estudos foram aplicados novos procedimentos na monitorização e tratamento dos dados, de forma a minimizar a variação nos índices de avaliação do imposex induzida pelos factores acima descritos, para seguir com maior consistência a tendência da poluição por TBT entre 2003 e 2008. Foi observado um declínio global significativo na intensidade de imposex na área de estudo durante o referido período e a qualidade ecológica da costa Portuguesa, segundo os termos definidos pela Comissão OSPAR, revelou melhorias notáveis após 2003 confirmando a eficácia do Regulamento (CE) N.º 782/2003. Não obstante, as populações de N. lapillus revelaram-se ainda amplamente afectadas por imposex, tendo sido detectadas emissões de TBT na água do mar ao longo da costa em 2006, apesar da restrição anteriormente referida. Estes inputs foram atribuídos principalmente aos navios que à data ainda circulavam com tintas AV com TBT aplicadas antes de 2003, uma vez que a sua utilização nas embarcações foi apenas proibida em 2008. Considerando que o Regulamento (CE) N.º 782/2003 constitui uma antecipação da proibição global da OMI que entrou em vigor em Setembro de 2008, prevê-se, por analogia, que haja uma rápida diminuição da poluição por TBT à escala mundial num futuro próximo. Na sequência desta previsão, é apresentada uma discussão teórica preliminar relativamente ás possíveis estratégias usadas por N. lapillus na recolonização de locais onde, no passado, as populações terão aparentemente sido extintas devido a níveis extremamente elevados de TBT. Estes locais são tipicamente zonas abrigadas junto de infra-estruturas portuárias, cuja recolonização por esta espécie será provavelmente muito lenta dada a mobilidade reduzida dos adultos e o ciclo de vida não apresentar fase larvar pelágica. Foram então equacionadas duas hipotéticas vias de recolonização de zonas abrigadas por espécimes provenientes de populações de costa aberta/exposta: a migração de adultos e/ou a dispersão de juvenis. No entanto, em ambos os casos, estaria implicada a transposição de um problema amplamente descrito na bibliografia: as populações de N. lapillus de costa aberta podem apresentar um fenótipo muito diferente das de zonas abrigadas, podendo inclusivamente variar no número de cromossomas. A recolonização pode portanto não ter sucesso pelo simples facto dos novos recrutas não estarem bem adaptados aos locais a recolonizar. Para analisar este problema, foram estudados tanto a forma da concha de espécimes de N. lapillus ao longo da costa Portuguesa como o respectivo número de cromossomas. Embora a forma da concha tenha revelado diferenças, de acordo com o grau de hidrodinamismo entre as populações de N. lapillus avaliadas, o cariótipo 2n = 26, típico de zonas expostas, foi registado em todos os locais amostrados. Por outro lado, foi também testada em laboratório a possível mortalidade de juvenis em dispersão no interior menos salino dos estuários. Foi então verificada a ocorrência de mortalidade elevada de juvenis expostos a salinidades baixas (=100% após 1 hora a salinidades ≤9 psu), o que também pode comprometer a recolonização de zonas estuarinas menos salinas por esta via. Mesmo assim, os juvenis mostraram um comportamento de flutuação à superfície da água em condições laboratoriais, que pode ser considerado um benefício específico na colonização de áreas mais internas dos estuários, se tal facto vier a ser confirmado em estudos de campo. As conclusões deste estudo contribuem certamente para a descrição do final da “história do TBT” dado que, uma vez controlados alguns factores determinantes no uso do imposex como biomarcador, a avaliação do declínio da poluição por estes compostos, agora esperado à escala global, se torna mais rigorosa.

Environmental transport of pollutants comprises distinct processes such as volatilization, leaching and surface runoff. Sorption is one of the most important phenomena that affects leaching, and thus the fate of hydrophobic organic pollutants in soils and also control their distribution in the soil/water environment. The work developed focuses the optimization of analytical techniques for monitoring the sorption behaviour of organic pollutants, 17α- ethinylestradiol (EE2) and atrazine, and their fate in aqueous environment. Initially, the development of several analytical techniques, such as micellar electrokinetic chromatography, spectral deconvolution, using UV-Vis and fluorescence spectroscopy, and also enzyme linked immunosorbent assay was performed. Optimization, method performance and recovery tests are described and results discussed. Moreover, in order to evaluate the applicability of the previously optimized method, atrazine and EE2 sorption to soil samples was performed. The work developed provide several options, in terms of methodology to follow sorption of atrazine onto soils, however the choice depends on the laboratory conditions and on the analyst preferences. The advantages and disadvantages of each methodology should be evaluated first. The second part of this work consisted in the sorption behaviour study of those two different hydrophobic organic pollutants onto different soil samples. Soil organic matter chemical characterization, being essential to understand the binding mechanism responsible for the interactions, was made. The results of atrazine binding to organic matter pointed out that carboxyl units and aromaticrich organic matter are the most efficient binding agents for atrazine. EE2 adsorbs strongly to soil organic matter and is mainly stabilized by hydrophobic interactions, through aromatic nuclei face to face with surface and/or another EE2 molecule association. Farmyard manure soil contains higher aromatic and carboxyl units, indicating that this type of manure can be effectively used to minimize the residual toxicity of EE2 and atrazine present in soils, increasing the sorption and reducing leaching onto water resources. Since the final destination of organic pollutants can be ground, surface and/or waste water, atrazine and 17α-ethinylestradiol were quantified in several water samples.
