Annually, ovarian cancer (OC) affects 240,000 women worldwide and is the most lethal gynaecological malignancy. Such mortality is predominantly associated with the development of an intrinsic and acquired resistance to chemotherapy, the lack of targeted therapies and the lack of biomarkers predicting therapeutic response.
Our clinical data demonstrates that increased miR-433 expression in primary high grade serous OC (HGSOCs) is significantly associated with poor PFS (n=46, p=0.024). Interestingly, the IHC analysis of two miR-433 targets: MAD2 [Furlong et al., 2012 PMID:22069160] and HDAC6 shows that low IHC levels of both proteins is also significantly associated with worse outcome (p=0.002 and 0.002 respectively; n=43). Additionally, the analysis of miR 433 in the publicly available TCGA dataset corroborates that high miR-433 is significantly correlated with worse OS for patients presenting with OC (n=558 and p=0.027). In vitro, in a panel of OC cell lines, higher miR-433 and lower MAD2 and HDAC6 levels were associated with resistance to paclitaxel.
To further investigate the role of miR-433 in the cellular response to chemotherapy, we generated an OC cell line stably expressing miR-433, or miR-control. MTT viability assays and Western Blot analyses established that miR-433 cells were more resistant to paclitaxel treatment (50nM) compared to miR-controls. Importantly, we have shown for the first time that miR 433 induced senescence, exemplified by a flattened morphology and down-regulation of phosphorylated Retinoblastoma (p-Rb), a molecular marker of senescence. Surprisingly, miR 433 induced senescence was independent from two well recognised senescent drivers: namely p53/p21 and p16. To explore this further we performed an in silico analysis of seven microRNA platforms which indicated that miR 433 potentially targets Cyclin-dependent kinase CDK6, which promotes sustained phosphorylation of Rb and thus cell cycle progression. In vitro, the overexpression of pre-miR-433 resulted in diminished CDK6 expression demonstrating a novel interaction between miR-433 and CDK6.
In conclusion, this study demonstrates that high miR-433 expression predicts poor outcome in OC patients by putatively rendering OC cells resistant to paclitaxel treatment through the induction of cellular senescence identifying this microRNA as a potential marker of chemoresponse.

Epithelial ovarian carcinoma (EOC) is characterised by late diagnosis and recurrences, both of which contribute to the high morbidity and mortality of this cancer. Unfortunately, EOC has an innate susceptibility to become chemo-resistant. Specifically, up to 30% of patients may not respond to current standard chemotherapy (paclitaxel and platinum in combination) and of those who have an initial response, some patients relapse within a few months. Therefore, in order to improve patient outcome it is crucial to establish what factors influence a patients' individualised response to chemotherapy. We analysed MAD2 protein expression in a patient cohort of 35 ovarian tumours and a panel of 5 ovarian cancer cell lines. We have demonstrated that low nuclear MAD2 expression intensity was significantly associated with chemo-resistant ovarian tumours (p=0.0136). Moreover, in vitro studies of the 5 ovarian cancer cell lines revealed that reduced MAD2 expression was associated with paclitaxel resistance. In silico analysis identified a putative miR-433 binding domain in the MAD2 3′UTR and expression profiling of miR-433 in the ovarian cancer cell lines showed that low MAD2 protein expression was associated with high miR-433 levels. In vitro over-expression of miR-433 attenuated MAD2 protein expression with a concomitant increase in cellular resistance to paclitaxel. Over-expression of a morpholino oligonucleotide that blocks miR-433 binding to MAD2 3′UTR stabilised MAD2 protein expression and protects from miR-433 induced degradation. Furthermore, miR-433 expression analysis in 35 ovarian tumour samples revealed that high miR-433 expression was associated with advanced stage presentations (p=0.0236). In conclusion, ovarian tumours that display low nuclear MAD2 intensity are chemo-resistant and stabilising MAD2 expression by antagonising miR-433 activity is a potential mechanism for restoring chemo-responsiveness in these tumours.

An understanding of the mechanisms underlying the development of resistance to chemotherapy treatment is a gateway to the introduction of novel therapies and improved outcomes for women presenting with ovarian cancer (OC). The desired apoptotic death post-chemotherapy depends on an intact and fully functioning cell cycle machinery.
In this study we demonstrate that stable expression of miR-433 renders OC cells more resistant to paclitaxel treatment. Interestingly, only cells with the highest miR-433 survived paclitaxel suggesting the possible role of miR-433 in cancer recurrence. Importantly, for the first time we demonstrate that miR 433 induces cellular senescence, exemplified by a flattened morphology, the downregulation of phosphorylated Retinoblastoma (p Rb) and increased β galactosidase activity. Surprisingly, miR 433 induced senescence was independent of two well recognised senescent drivers: p21 and p16. Further in silico analysis followed by in vitro experiments identified CKD6 as a novel miR-433 target gene possibly explaining the observed p21 and p16-independent induction of cellular senescence. Another in silico identified miR-433 target gene was CDC27, a protein involved in the regulation of the cell cycle during mitosis. We demonstrate that the overexpression of pre-miR-433 leads to the downregulation of CDC27 in vitro revealing a novel interaction between miR-433 and CDC27, an integral cell cycle regulating protein.
Interestingly, miR-433 expressing cells also demonstrated an ability to impact their tumour microenvironment. We show that miR-433 is present in exosomes released from miR-433 overexpressing and high miR-433 naïve cells. Moreover, growth condition media (GCM) harvested from cells with high miR-433 have higher levels of IL-6 and IL-8, two key cytokines involved in the senescence associated secretory phenotype (SASP). Importantly, GCM from miR-433-enriched cells repressed the growth of co-cultured cells with initial studies showing a GCM-dependent induction of chemoresistance.
In conclusion, data in this study highlights how the aberrant expression miR-433 contributes to chemoresistance in OC cells. We postulate that standard chemotherapy, particularly paclitaxel, used to treat women with OC may have an attenuated ability to kill cells harbouring increased levels of miR-433, allowing for a subsequent chemoresistant phenotype post-therapy.

Higher expression of the miR-433 microRNA (miRNA) is associated with poorer survival outcomes in patients with HGSOC that may be overcome by a greater understanding of the functional role of this miRNA. We previously described miR-433 as a critical cell cycle regulator and mediator of cellular senescence. Downregulation of the mitotic arrest deficiency 2 (MAD2) protein by miR-433 led to increased cellular resistance to paclitaxel in epithelial ovarian cancer cells (EOC). Furthermore immunohistochemical (IHC) analysis of MAD2 expression in patients with HGSOC showed that loss of MAD2 was significantly associated with poorer patient survival. Higher miR-433 expression is also associated with an increased resistance to the platins which is unrelated to loss of MAD2 expression. In silico analysis of the miR-433 target proteins in the TCGA database identified the association between a number of miR-433 targets and poorer patient survival. IHC analysis of the miR-433 target, histone deacetylase 6 (HDAC6), confirmed that its expression was significantly associated with a decrease in patient overall survival. The knock-down of HDAC6 by siRNA in EOC cells did not attenuate apoptotic responses to paclitaxel or platin although lower endogenous HDAC6 expression was associated with more resistant EOC cell lines. In vitro analysis revealed that EOC cells which survived chemotherapeutic kill with high doses of paclitaxel expressed higher miR-433 and concomitant decreased expression of the miR-433 targets. These cells were more chemoresistant compared to the parental cell line and repopulated as 3d organoid cultures in non-adherent stem cell selective conditions; thus indicating that the cells which survive chemotherapy were viable, capable of regrowth and had an increased potential for pluripotency. In conclusion, our data suggests that chemotherapy is not driving the transcriptional upregulation of miR-433 but rather selecting a population of cells with high miR-433 expression that may contribute to chemoresistant disease and tumour recurrence.

Cytokine secretion and degranulation represent key components of CD8(+) T-cell cytotoxicity. While transcriptional blockade of IFN-γ and inhibition of degranulation by TGF-β are well established, we wondered whether TGF-β could also induce immune-regulatory miRNAs in human CD8(+) T cells. We used miRNA microarrays and high-throughput sequencing in combination with qRT-PCR and found that TGF-β promotes expression of the miR-23a cluster in human CD8(+) T cells. Likewise, TGF-β up-regulated expression of the cluster in CD8(+) T cells from wild-type mice, but not in cells from mice with tissue-specific expression of a dominant-negative TGF-β type II receptor. Reporter gene assays including site mutations confirmed that miR-23a specifically targets the 3'UTR of CD107a/LAMP1 mRNA, whereas the further miRNAs expressed in this cluster-namely, miR-27a and -24-target the 3'UTR of IFN-γ mRNA. Upon modulation of the miR-23a cluster by the respective miRNA antagomirs and mimics, we observed significant changes in IFN-γ expression, but only slight effects on CD107a/LAMP1 expression. Still, overexpression of the cluster attenuated the cytotoxic activity of antigen-specific CD8(+) T cells. These functional data thus reveal that the miR-23a cluster not only is induced by TGF-β, but also exerts a suppressive effect on CD8(+) T-cell effector functions, even in the absence of TGF-β signaling.

Dentro do recorte temporal dos anos 1930-1940, esta tese pretende investigar de que forma as características e especificidades da corrente nacionalista e do Nacionalismo musical se espelham na criação artística e musical brasileira em especial no corpus da obra para piano solo para concerto daquele momento. A investigação consiste em dois momentos distintos de pesquisa, sendo o primeiro compreendido de revisão bibliográfica sobre a corrente nacionalista, Nacionalismo e Nacionalismo musical, além de uma contextualização histórica acerca dos anos 1930 no Brasil, Europa e Estados Unidos da América e também biografia de compositores nacionalistas brasileiros do período; seu segundo momento compreende elaboração de dados acerca de programas de concerto de piano relativos ao Nacionalismo musical, análise de peças além de discussões acerca de notícias de periódicos daquele momento. Discussões em torno de problemáticas correlatas ao tema são aqui também proporcionadas.

As células estaminais hematopoiéticas residem na medula óssea e possuem capacidade para se auto-renovar e dar origem a todos os tipos de células sanguíneas. O endotélio da medula óssea é constituído por células endoteliais de medula óssea (BMEC) e compreende dois nichos com funções distintas: o nicho osteoblástico e o nicho vascular. O nicho osteoblásctico proporciona condições para a quiescência de células estaminais hematopoiéticas, enquanto no nicho vascular ocorre proliferação e diferenciação das mesmas. Quando ocorre um desequilíbrio na expressão de genes que codificam para proteínas envolvidas na mobilização de células do nicho osteoblástico para o nicho vascular – factores angiócrinos – ocorre uma desestabilização do microambiente medular, que se pode traduzir num processo tumoral. Os microRNAs (miRNAs) são uma classe de RNAs não codificantes, de cadeia simples, que regula a expressão génica. Os miRNAs são sequências endógenas de RNA que possuem entre 19 e 25 nucleótidos de tamanho. Os miRNAs são reguladores da expressão genica, induzindo o silenciamento a nível da pós-transcrição, através da sua ligação com uma sequência específica para a qual possuem afinidade, na região 3’ não traduzida (3’ UTR) dos seus mRNA alvo, conduzindo à inibição da tradução ou à sua degradação. Os miRNAs estão envolvidos na regulação de genes de diversas vias afectando processos fundamentais como hematopoiese, apoptose, proliferação celular e tumorigénese. Os níveis de expressão dos miRNAs estão alterados no cancro, podendo actuar directamente como supressores de tumor ou como oncogenes, sendo neste caso denominados de oncomirs. Os perfis dos níveis de expressão de vários miRNAs foram estudados, tendo-se verificado que se alteram durante o processo de carcinogénese, podendo actuar directamente como supressores de tumor ou como oncogenes, sendo neste caso denominados de oncomirs. Apesar do miR-363* estar envolvido na regulação da expressão de genes que regulam propriedades das células endoteliais e medula óssea, os genes sobre os quais exerce a sua função ainda não foram identificados.O objectivo do presente estudo é a identificação dos genes directamente regulados pelo miR-363* (genes alvo) e a sua relevância para a disfunção medular e a sua caracterização nos síndromes mielodisplásicos. A estratégia usada baseou-se na redução ou aumento forçados dos níveis de miR-363* em células endoteliais e subsequente análise da expressão génica através de microarrays de cDNA do genoma humano. A redução do miR-363* vai implicar o aumento da expressão dos seus genes alvo, assim como o aumento dos níveis do miR-363* vai induzir a degradação e consequente redução dos seus genes alvos. A intersecção dos dados gerados através do estudo da expressão com bases de dados que possuem algoritmos para previsão de genes alvo directos dos miRNAs (miRBase e MicroCosm Targets) permitiu restringir os genes a analisar a sete genes, nomeadamente BST1, ESAM, FCER1G, IKBKG, SELE, THBS3 e TIMP1. A interacção directa destes candidatos a alvos directos do miR-363* foi posteriormente validada. Para tal, as 3’UTR dos genes foram clonadas num vector que contém o gene da luciferase. Uma vez as clonagens realizadas, efectuaram-se ensaios funcionais em células endoteliais, nomeadamente HUVEC, nas quais se co-transfectaram os vectores gerados, anti-miRs ou pre-miRs (para diminuir ou aumentar o nível de miRNA) e o plasmídeo controlo da Renilla para normalização dos ensaios de luciferase. A variação da luminescência obtida em presença do aumento ou redução do miR-363* deu uma forte indicação da regulação directa do miR-363* nesses alvos. No entanto, a confirmação desta interacção directa foi efectuada através de ensaios de mutagénese, nos quais de induziram mutações na 3’UTR nos locais de ligação do miRNA, seguidos dos ensaios funcionais como acima descritos. Esta estratégia sugere que o TIMP1, inibidor da metaloprotease-9 (MMP-9), é regulado directamente pelo miR-363*. Adicionalmente, os níveis de expressão dos alvos directos do miR-363* foram estudados em 17 amostras de aspirados de medula óssea de doentes com síndromes mielodisplásicos. Os síndromes mielodisplásicos são caracterizados como um grupo heterogéneo de condições, que apresentam citopenias (produção deficiente de eritrócitos, leucócitos e/ou megacariócitos) e medula óssea displástica e hipercelular. A escalonagem dos doentes foi feita de acordo com o sistema de prognóstico IPSS elaborado pela Organização Mundial de Saúde, e que consiste numa tabela de risco de progressão de síndromes mielodisplásicos para leucemia mielóide aguda (LMA) e que agrupa os doentes em baixo risco – que compreende os níveis baixo e intermédio 1 – e em alto risco – que compreende os níveis intermédio 2 e alto. Dos genes regulados pelo miR-363*, o destacam-se o TIMP1, estando aumentando em doentes com mau prognóstico, e o THBS3 que apresenta um aumento nos doentes com prognóstico intermédio. Em suma, os estudos realizados permitiram a identificação de genes regulados pelo miR-363* e contribuiram para o conhecimento de como o miR-363* contribui para a disfunção medular, particularmente em síndromes mielodisplásicos, pela desregulação das propriedades endoteliais.

Die Nutzung des Like-Buttons wurde bisher nur oberflächlich und vor allem auf den US-Raum begrenzt erforscht. Dabei lag der Fokus ausschließlich auf wirtschaftlich orientierten Unternehmen und dem monetären Wert, den ein Fan für dieses hat. In der vorliegenden Untersuchung wird eine erweiterte Perspektive gewählt. Aus Sicht deutschsprachiger Nutzer wird erforscht, welche Motive und Bedürfnisse eine Person dazu veranlassen, sich freiwillig per Gefällt-mir zu verknüpfen. Hierzu wird der Uses and Gratifications-Ansatz herangezogen. Es stehen nicht Unternehmen, sondern generell alle Formen von Organisationen im Fokus. Gleichzeitig wird untersucht, wie diese Beziehungen verlaufen: Welche Erwartungen stellen Fans? Welche Handlungen nehmen sie wie häufig vor und welchen Effekt hat eine Fanbeziehung auf die Einstellung eines Nutzers gegenüber einer Organisation? Die Motive hinter der Nutzung des Gefällt mir-Buttons sind umfangreicher, als es bisherige Forschung vermuten ließ. Fans wollen informiert werden, sie wollen ihr Gefallen schnell und einfach bekunden und sich durch die Verknüpfung selbst darstellen. Weniger wichtig, aber dennoch von Relevanz sind die Motive der Partizipation und Unterstützung, der Empfehlung, der Vorteilssuche, der Unterhaltung sowie der Netzwerkerweiterung und Gruppenzugehörigkeit. Die Verwendung des Like-Buttons dient somit der Befriedigung einer ganzen Reihe von Nutzungsbedürfnissen. Der Verlauf der Beziehungen zwischen Organisationen und Usern ist eher von Passivität geprägt. Fans wollen zwar informiert werden und stellen konkrete Ansprüche an die Art und Form von Organisationsmeldungen, sie wollen aber nur selten wirklich direkt mit einem Unternehmen, einem Verein o.ä. interagieren. Langweilen die Meldungen einer Organisation, werden zu viele oder nicht relevante Informationen an die Fans weitergeben, zögern diese nicht, eine Verknüpfung zu beenden. Ein Effekt der Fanschaft auf die Einstellung von Nutzern gegenüber der jeweiligen Organisation konnte nur bedingt festgestellt werden. Letztere zieht einen Gewinn nicht aus einer erhöhten Ausgabebereitschaft seiner Fans, sondern aus deren stärkerer Aufmerksamkeit gegenüber Organisationsmeldungen.
