O Trypanosoma cruzi, agente etiológico da doença de Chagas, possui um ciclo de vida complexo, deve lidar com diversas condições do ambiente e depende dos hospedeiros para suprir suas necessidades nutricionais. Uma delas é a necessidade de captar a molécula de heme (Fe-protoporfirina IX) que será utilizada como fator de crescimento. Os mecanismos envolvendo o metabolismo de heme são cruciais para a sobrevivência do T. cruzi pois o parasito não possui várias enzimas de biossíntese dessa porfirina e o heme livre pode apresentar citotoxicidade para célula. Na tentativa de perseguir o destino final do heme no parasito, nós estudamos essa via inexplorada no T. cruzi. Nessa tese, nós demonstramos que epimastigotas cultivados com heme, produziram os compostos, α-meso hidroxiheme, verdoheme e biliverdina (identificados por HPLC acoplado á espectrofotômetria). Além disso, nós observamos através de análise dos extratos de epimastigotas no espectrômetro de massas (LQT Orbitrap), espécies iônicas de m/z 583,4 e m/z 619,3. A fragmentação subsequente desses íons originaram espécies filhas típicas das moléculas de biliverdina e verdoheme, respectivamente. Nós observamos também, espécies iônicas de m/z 1397,4 e m/z 1135,4. A fragmentação dessas espécies produziram íons, sendo um deles com a mesma massa molecular de heme (m/z 616,3). Essa espécie iônica por sua vez, gerou fragmentos iônicos idênticos a uma molécula de heme, confirmando que esses intermediários são produtos da modificação da porfirina. Baseado nesses resultados, nós propomos um modelo onde o catabolismo de heme em T. cruzi, envolveria a conjugação da bis(glutationil)spermina, um derivado da tripanotiona presente em tripanossomatídeos, à porfirina (m/z 1137,4), seguido da remoção de dois resíduos de ácidos glutâmicos (m/z 1135,4). Embora o significado bioquímico e fisiológico da adição desse resíduo tiol na molécula de heme ainda é pouco compreendido, alguns trabalhos demonstram a abilidade desses compostos em ligar na porfirina, sem contar também, que esse heme conjugado poderia resultar em uma forma efetiva de prevenção de danos à membrana e a célula ocasionados pelo acúmulo de heme livre. Em conjunto, esses resultados fornecem novas abordagens do metabolismo de heme em T. cruzi, revelando possíveis alvos de intervenção quimioterápica futuros. Nossa proposta está direcionada para uma via ativa de catabolismo de heme que inclui a adição de grupos tiol (derivado da tripanotiona) à heme e a clivagem do anel porfirínico originando a molécula de biliverdina.

No presente trabalho foram desenvolvidos modelos de classificação aplicados à mineração de dados climáticos para a previsão de eventos extremos de precipitação com uma hora de antecedência. Mais especificamente, foram utilizados dados observacionais registrados pela estação meteorológica de superfície localizada no Instituto Politécnico da Universidade do Estado do Rio de Janeiro em Nova Friburgo RJ, durante o período de 2008 a 2012. A partir desses dados foi aplicado o processo de Descoberta de Conhecimento em Banco de Dados (KDD Knowledge Discovery in Databases), composto das etapas de preparação, mineração e pós processamento dos dados. Com base no uso de algoritmos de Redes Neurais Artificiais e Árvores de Decisão para a extração de padrões que indicassem um acúmulo de precipitação maior que 10 mm na hora posterior à medição das variáveis climáticas, pôde-se notar que a utilização da observação meteorológica de micro escala para previsões de curto prazo é suscetível a altas taxas de alarmes falsos (falsos positivos). Para contornar este problema, foram utilizados dados históricos de previsões realizadas pelo Modelo Eta com resolução de 15 km, disponibilizados pelo Centro de Previsão de Tempo e Estudos Climáticos do Instituto Nacional de Pesquisas Espaciais CPTEC/INPE. De posse desses dados, foi possível calcular os índices de instabilidade relacionados à formação de situação convectiva severa na região de Nova Friburgo e então armazená-los de maneira estruturada em um banco de dados, realizando a união entre os registros de micro e meso escala. Os resultados demonstraram que a união entre as bases de dados foi de extrema importância para a redução dos índices de falsos positivos, sendo essa uma importante contribuição aos estudos meteorológicos realizados em estações meteorológicas de superfície. Por fim, o modelo com maior precisão foi utilizado para o desenvolvimento de um sistema de alertas em tempo real, que verifica, para a região estudada, a possibilidade de chuva maior que 10 mm na próxima hora.

We describe the food habits of the Sowerby’s beaked whale (Mesoplodon bidens) from observations of 10 individuals taken as bycatch in the pelagic drift gillnet fishery for Swordfish (Xiphias gladius) in the western North Atlantic and 1 stranded individual from Kennebunk, Maine. The stomachs of 8 bycaught whales were intact and contained prey. The diet of these 8 whales was dominated by meso- and benthopelagic fishes that composed 98.5% of the prey items found in their stomachs and cephalopods that accounted for only 1.5% of the number of prey. Otoliths and jaws representing at least 31 fish taxa from 15 families were present in the stomach contents. Fishes, primarily from the families Moridae (37.9% of prey), Myctophidae (22.9%), Macrouridae (11.2%), and Phycidae (7.2%), were present in all 8 stomachs. Most prey were from 5 fish taxa: Shortbeard Codling (Laemonema barbatulum) accounted for 35.3% of otoliths, Cocco’s Lanternfish (Lobianchia gemellarii) contributed 12.9%, Marlin-spike (Nezumia bairdii) composed 10.8%, lanternfishes (Lampanyctus spp.) accounted for 8.4%; and Longfin Hake (Phycis chesteri) contributed 6.7%. The mean number of otoliths per stomach was 1196 (range: 327–3452). Most of the fish prey found in the stomachs was quite small, ranging in length from 4.0 to 27.7 cm. We conclude that the Sowerby’s beaked whales that we examined in this study fed on large numbers of relatively small meso- and benthopelagic fishes that are abundant along the slope and shelf break of the western North Atlantic.

Psednos rossi new species (Teleostei: Liparidae) is described from two specimens collected in the North Atlantic Ocean off Cape Hatteras, North Carolina, at depths of 500–674 m. Psednos rossi belongs to the P. christinae group, which includes six other species and is characterized by 46–47 vertebrae and the absence of a coronal pore. Psednos rossi differs from those six species by having characters unique within the genus: straight spine, body not humpbacked at the occiput, and a very large mouth with a vertical oral cleft. Other distinguishing characters include a notched pectoral fin with 15–16 rays, eye 17–19% SL, and color in life orange-rose. With P. rossi, the genus Psednos as currently known includes 26 described and five undescribed species of small meso- or bathypelagic liparids from the Atlantic, Pacific, and Indian Oceans.

The Austrian-Ceylonese hydrobiological mission of 1970 investigated and made collections from 36 flowing water systems (brooks, torrents, rivers); of these, 34 water systems were in the mountains regions of south-west and south-east of Sri Lanka. In the crystalline mountain region, the water systems are extremely poor in electrolytes, very soft and slightly acid; these torrential streams have strong falls, high flow velocities and boulder bottoms. The water temperatures increase from the sources and brooks at 2,000 m altitude to the mouths from 15°C to 28°C. The density of animal population (macro and meso-fauna) increases from the river bank regions (and pools) towards the sections with strong current and reaches on the rocks in the cascades a density of 500 to appr. 750 individuals/1/16m².

A Gram-positive bacterium, designated strain CW 7(T), was isolated from forest soil in Anhui Province, south-east China. Cells were strictly aerobic, motile with peritrichous flagella and rod-shaped. The strain grew optimally at 30-37 degrees C and pH 7.0-8.0. The major fatty acids of strain CW 7(T) were anteiso-C-15:0, iso-C-15:0 and anteiso-C-17:0. The predominant menaquinone was MK-7. The cell-wall peptidoglycan contained meso-diaminopimelic acid. The G + C content of the genomic DNA was 42.3 mol%. Phylogenetic analysis indicated that strain CW 7(T) belonged to a monophyletic cluster within the genus Bacillus and showed 16S rRNA gene sequence similarities of less than 96.5% to recognized species of the genus Bacillus. The results of the polyphasic taxonomic study, including phenotypic, chemotaxonomic and phylogenetic analyses, showed that strain CW 7(T) represents a novel species of the genus Bacillus, for which the name Bacillus pallidus sp. nov. is proposed. The type strain is CW 7(T) (=KCTC 13200(T)=CCTCC AB 207188(T)=LMG 24451(T)).

Avalanches, debris flows, and landslides are geophysical hazards, which involve rapid mass movement of granular solids, water and air as a single-phase system. The dynamics of a granular flow involve at least three distinct scales: the micro-scale, meso-scale, and the macro-scale. This study aims to understand the ability of continuum models to capture the micro-mechanics of dry granular collapse. Material Point Method (MPM), a hybrid Lagrangian and Eulerian approach, with Mohr-Coulomb failure criterion is used to describe the continuum behaviour of granular column collapse, while the micromechanics is captured using Discrete Element Method (DEM) with tangential contact force model. The run-out profile predicted by the continuum simulations matches with DEM simulations for columns with small aspect ratios ('h/r' < 2), however MPM predicts larger run-out distances for columns with higher aspect ratios ('h/r' > 2). Energy evolution studies in DEM simulations reveal higher collisional dissipation in the initial free-fall regime for tall columns. The lack of a collisional energy dissipation mechanism in MPM simulations results in larger run-out distances. Micro-structural effects, such as shear band formations, were observed both in DEM and MPM simulations. A sliding flow regime is observed above the distinct passive zone at the core of the column. Velocity profiles obtained from both the scales are compared to understand the reason for a slow flow run-out mobilization in MPM simulations. © 2013 AIP Publishing LLC.

The mechanics of failure for elastic-brittle lattice materials is reviewed. Closed-form expressions are summarized for fracture toughness as a function of relative density for a wide range of periodic lattices. A variety of theoretical and numerical approaches has been developed in the literature and in the main the predictions coincide for any given topology. However, there are discrepancies and the underlying reasons for these are highlighted. The role of imperfections at the cell wall level can be accounted for by Weibull analysis. Nevertheless, defects can also arise on the meso-scale in the form of misplaced joints, wavy cell walls and randomly distributed missing cell walls. These degrade the macroscopic fracture toughness of the lattice. © 2010 Springer Science+Business Media B.V.

Two types of SiO2 with different mesopore size and HZSM-5 zeolite were used to prepare hybrid supported cobalt-based catalysts. The textual and structural properties of the catalysts were studied using N-2 physisorption, X-ray diffraction (XRD), and H-2 temperature-programmed reduction (TPR) techniques. Fischer-Tropsch synthesis (FTS) performances of the catalysts were carried out in a fixed-bed reactor. The combination effects of the meso- and micropores of the supports as well as the interaction between supports and cobalt particles on FTS activity are discussed. The results indicate that the catalyst supported on the tailor-made SiO2 and HZSM-5 hybrid maintained both meso- and micropore pores during the preparation process without HZSM-5 particles agglomerating. The mesopores provided quick mass transfer channels, while the micropores contributed to high metal dispersion and accelerated hydrocracking/hydroisomerization reaction rate. High CO conversion of 83.9% and selectivity to gasoline-range hydrocarbons (C-5-C-12) of 55%, including more than 10% isoparaffins, were achieved simultaneously on this type of catalyst.

聚丙交酯又称聚乳酸(PLA),是目前最重要的合成生物降解高分子之一。丙交酯有L,L-丙交酯(LLA)、D,D-丙交酯(DLA)和内消旋丙交酯(meso-LA)三种立体异构体。PLA包括全同立构、间同立构、无规立构、不均匀有规立构和嵌段立构等构型。 PLA的微观链结构在很大程度上决定了PLA的理化性质。无规立构和不均匀有规立构聚丙交酯是非晶聚合物,它们可以由meso-LA或者rac-LA(LLA和DLA的等物质的量混合物)的聚合得到;而全同立构、间同立构和嵌段立构的聚丙交酯都是可以结晶的;全同立构的聚丙交酯可由纯的DLA或者LLA聚合而成,高分子量的全同立构PLA(PLLA或者PDLA)的熔点约180ºC;但是,有趣的是,等量PDLA和PLLA形成的外消旋混合物的熔点约230ºC,这与无规的poly(rac-LA)(聚外消旋丙交酯)形成强烈对照;间同和嵌段立构聚丙交酯只是最近几年才被合成出来,目前,只能通过丙交酯的立体选择性聚合才能得到,它们的结晶性和熔点随催化剂的选择性不同变化范围很大;而就降解性能而言,非晶聚丙交酯的降解速率要高于结晶性的聚丙交酯。因为序列结构对聚丙交酯的性质有很大的影响,而聚丙交酯的序列结构可以通过丙交酯的立体选择性聚合来控制,所以,近年来,丙交酯的立体选择性聚合催化剂的开发成为了一个研究热点。 我们设计合成了一系列非手性烯醇式席夫碱-铝/锌化合物,详细的表征了它们的结构,阐明了配体取代基团与催化剂构型之间的关系;将这一系列化合物用于丙交酯的开环聚合,系统的研究了不同结构的催化剂与丙交酯的聚合动力学、立体规整度之间的关系。
