Ce texte est un « droit de réponse » par les auteurs de l'article « Vers un naturalisme social. À la croisée des sciences sociales et des sciences cognitives », publié par SociologieS en octobre 2011, au débat qu'il a suscité. Après une brève mise au point sur la forme même du débat, ainsi que sur les dissensions ponctuelles qui opposent les différents protagonistes, l'article répond aux inquiétudes parfaitement légitimes et aux questions de fond que soulève le naturalisme social.

We report a Spanish family with autosomal-dominant non-neuropathic hereditary amyloidosis with a unique hepatic presentation and death from liver failure, usually by the sixth decade. The disease is caused by a previously unreported deletion/insertion mutation in exon 4 of the apolipoprotein AI (apoAI) gene encoding loss of residues 60-71 of normal mature apoAI and insertion at that position of two new residues, ValThr. Affected individuals are heterozygous for this mutation and have both normal apoAI and variant molecules bearing one extra positive charge, as predicted from the DNA sequence. The amyloid fibrils are composed exclusively of NH2-terminal fragments of the variant, ending mainly at positions corresponding to residues 83 and 92 in the mature wild-type sequence. Amyloid fibrils derived from the other three known amyloidogenic apoAI variants are also composed of similar NH2-terminal fragments. All known amyloidogenic apoAI variants carry one extra positive charge in this region, suggesting that it may be responsible for their enhanced amyloidogenicity. In addition to causing a new phenotype, this is the first deletion mutation to be described in association with hereditary amyloidosis and it significantly extends the value of the apoAI model for investigation of molecular mechanisms of amyloid fibrillogenesis.

La monnaie a été étudiée par des économistes hétérodoxes, des sociologues et des historiens qui ont souligné ses rapports à l'ordre collectif, mais elle n'est que rarement analysée sous l'angle de la citoyenneté. Notre thèse propose une réflexion théorique sur quatre types de fonctions (politique, symbolique, socioéconomique et psychoaffective) qui permettent à la monnaie de jouer un rôle de médiation de la citoyenneté. A partir d'une perspective qui combine les apports de l'économie politique internationale et de l'école de la régulation, nous montrons que cette médiation ne mobilise pas seulement des mécanismes sociopolitiques nationaux, mais aussi des mécanismes internationaux qui rétroagissent sur la sphère domestique des États et affectent leur capacité à définir leur régime de citoyenneté. Cette relation est analysée dans le contexte de l'institutionnalisation du système monétaire international de Bretton Woods (1944) et du développement de la globalisation financière depuis les années 1970. Si la monnaie a été mise au service d'un principe de protection des droits sociaux des citoyens contre les pressions financières extérieures après la Seconde guerre mondiale, elle contribue aujourd'hui à l'ouverture de la sphère domestique des Etats aux flux de capitaux transnationaux et à la création d'un ordre politique et juridique favorable aux droits des investisseurs. Cette dynamique est impulsée par l'essor de nouveaux intermédiaires financiers (notamment les agences de notation et les investisseurs institutionnels) et l'émergence concomitante d'une nouvelle forme d'Etat légitimée à partir d'un discours politique néolibéral insistant sur la quête de compétitivité, la réduction de la protection sociale et la responsabilisation individuelle. Elle se traduit par la privatisation des régimes de retraite et le développement des politiques d'éducation financière qui incitent les citoyens à se comporter en « preneurs de risques » actifs et responsables, assurant eux-mêmes leur sécurité économique à travers le placement de leur épargne retraite sur les marchés financiers. Nous soulignons toutefois les difficultés institutionnelles, cognitives et socioéconomiques qui rendent cette transformation de la citoyenneté contradictoire et problématique. Money has been studied by heterodox economists, sociologists and historians who stressed its relationship to collective order. However, it has hardly been analysed from the viewpoint of its relationship to citizenship. We propose a theoretical account of four types of functions (political, symbolic, socioeconomic and psychoaffective) enabling money to operate as a mediation of citizenship. From a perspective that combines the contributions of international political economy and the regulation school, we show that this mediation mobilises not only national sociopolitical mechanisms, but also international mechanisms which feed back on the domestic sphere of states and affect their capacity to define their regime of citizenship. This relationship is analysed in the context of the institutionalisation of the international monetary system of Bretton Woods (1944) and the development of financial globalization since the 1970s. If money has served to protect the social rights of citizens against external financial pressures after the Second World War, today it contributes to the opening of the domestic sphere of states to transnational capital flows and to the creation of a political and legal order favorable to the rights of investors. This dynamic is driven by the rise of new financial intermediaries (in particular rating agencies and institutional investisors) and the simultaneous emergence of a new form of state legitimized from a neoliberal political discourse emphasizing the quest for competitiveness, reduced social protection and individual responsibilization. It results in the privatization of pension systems and the development of policies of financial education that encourage citizens to behave as active and responsible « risk takers », ensuring their own economic security through the investment of their savings retirement on financial markets. The bacterial insertion sequence IS21 contains two genes, istA and istB, which are organized as an operon. IS21 spontaneously forms tandem repeats designated (IS21)2. Plasmids carrying (IS21)2 react efficiently with other replicons, producing cointegrates via a cut-and-paste mechanism. Here we show that transposition of a single IS21 element (simple insertion) and cointegrate formation involving (IS21)2 result from two distinct non-replicative pathways, which are essentially due to two differentiated IstA proteins, transposase and cointegrase. In Escherichia coli, transposase was characterized as the full-length, 46 kDa product of the istA gene, whereas the 45 kDa cointegrase was expressed, in-frame, from a natural internal translation start of istA. The istB gene, which could be experimentally disconnected from istA, provided a helper protein that strongly stimulated the transposase and cointegrase-driven reactions. Site-directed mutagenesis was used to express either cointegrase or transposase from the istA gene. Cointegrase promoted replicon fusion at high frequencies by acting on IS21 ends which were linked by 2, 3, or 4 bp junction sequences in (IS21)2. By contrast, cointegrase poorly catalyzed simple insertion of IS21 elements. Transposase had intermediate, uniform activity in both pathways. The ability of transposase to synapse two widely spaced IS21 ends may reside in the eight N-terminal amino acid residues which are absent from cointegrase. Given the 2 or 3 bp spacing in naturally occurring IS21 tandems and the specialization of cointegrase, the fulminant spread of IS21 via cointegration can now be understood.

The bacterial insertion sequence IS21 when repeated in tandem efficiently promotes non-replicative cointegrate formation in Escherichia coli. An IS21-IS21 junction region which had been engineered to contain unique SalI and BglII sites close to the IS21 termini was not affected in the ability to form cointegrates with target plasmids. Based on this finding, a novel procedure of random linker insertion mutagenesis was devised. Suicide plasmids containing the engineered junction region (pME5 and pME6) formed cointegrates with target plasmids in an E.coli host strain expressing the IS21 transposition proteins in trans. Cointegrates were resolved in vitro by restriction with SalI or BglII and ligation; thus, insertions of four or 11 codons, respectively, were created in the target DNA, practically at random. The cloned Pseudomonas aeruginosa arcB gene encoding catabolic ornithine carbamoyltransferase was used as a target. Of 20 different four-codon insertions in arcB, 11 inactivated the enzyme. Among the remaining nine insertion mutants which retained enzyme activity, three enzyme variants had reduced affinity for the substrate ornithine and one had lost recognition of the allosteric activator AMP. The linker insertions obtained illustrate the usefulness of the method in the analysis of structure-function relationships of proteins.

This PhD dissertation deals with the question of evaluating social welfare and public policy making through the works of William Stanley Jevons (1835-1882) and Léon Walras (1834-1910), two economists who lived in the 19th century. These authors, well-known for their analyses on pure economics, were also deeply interested in the social problems of their time and proposed solutions to remedy them. In accordance with utilitarianism, Jevons was convinced that the reforms implemented by public authorities should improve social welfare (viewed as dependant on individual utilities). As for Walras, he defined a justice criterion based on a particular definition of natural law, and the reforms he proposed had the objective of restoring the rights following from this definition. We have drawn two principal conclusions from our work. First, the analyses of Jevons and Walras, who are often considered marginalise, are different not only in pure economics (as suggested by Jaffé in the seventies), but also from the point of view of welfare economics (defined as a science dealing with the evaluation of different social states). Secondly, these authors propose two different ways of justifying social reforms which have similarities with modern theory even though neither Jevons nor Walras are considered pioneers of welfare economics or public economics. Based on these two conclusions, we claim that studying these authors' theories might be of interest for the evaluation of public projects by the economists of today. Not only were their problems similar to ours, but also the original ideas present in their analyses may lead to the refinement of modern methods.Résumé en françaisCette these de doctorat porte sur les manières dont deux économistes du XIXe siècle, William Stanley Jevons (1835-1882) et Léon Walras (1834-1910), abordent la question du bien- etre collectif et la prise de décision publique. Connus principalement pour leurs travaux en économie pure, ces auteurs s'intéressent également à la question sociale et proposent des solutions pour y remédier. Jevons, conformément à son adhésion à la tradition utilitariste, estime que les interventions des autorités publiques doivent avoir pour objectif l'amélioration du bien- être collectif. Quant à Walras, il adopte un critère de justice qui relève du « droit naturel » et les propositions sociales qu'il préconise sont justifiées sur la base de leur conformité avec ce dernier. A l'issue du travail effectué dans le cadre de cette thèse, nous avons abouti à deux résultats principaux : (1) Les analyses de Jevons et Walras, deux économistes qui sont souvent considérés comme des « marginalistes » diffèrent fondamentalement non seulement dans leur analyse de l'économie pure, comme Jaffé le soulignait (1976), mais également en termes d'économie du bien-être (comprise comme une science ayant pour objectif l'évaluation des différents états sociaux). (2) Il existe deux voies originales pour justifier les réformes sociales dans les oeuvres de Jevons et Walras qui partagent des similarités importantes avec les théories modernes bien que ces auteurs ne fassent pas partie des jalons généralement retenus de l'histoire de l'économie du bien-être et de l'économie publique. Ces deux résultats nous conduisent à avancer que l'étude des approches de Jevons et de Walras peut contribuer à la formulation de nouvelles solutions à des problèmes rencontrés dans l'évaluation des différentes politiques publiques, car non seulement les préoccupations de ces auteurs était similaires aux nôtres, mais vu les éléments originaux que leurs propositions comportent, elles peuvent permettre de nuancer certaines aspects des méthodes modernes.
