The placenta contains several types of feto-maternal interfaces where zygote-derived cells interact with maternal cells or maternal blood for the promotion of fetal growth and viability. The genetic factors regulating the interactions between different cell types within feto-maternal interfaces and the relative contributions of the maternal and zygotic genomes are poorly understood. Genomic imprinting, the epigenetic process responsible for parental origin-dependent functional differences between homologous chromosomes, has been proposed to contribute to these events. Previous studies showed that mouse conceptuses with an absence of imprinted differences between the two copies of chromosome 12 (upon paternal inheritance of both copies) die late in gestation and have a variety of defects, including placentomegaly. Here we examined the role of chromosome 12 imprinting in these placentae in more detail. We show that the spatial interactions between different cell types within feto-maternal interfaces are defective and identify abnormal behaviors in both zygote-derived and maternal cells that are attributed to the genome of the zygote but not the mother. These include compromised invasion of the maternal decidualized endometrium and the central maternal artery situated within it by zygote-derived trophoblast, abnormalities in the wall of the central maternal artery, and defects within the zygote-derived cellular layer of the labyrinth, which is in direct contact with maternal blood. These findings demonstrate multiple roles for chromosome 12 imprinting in the placenta that have not previously been associated with imprinting effects. They provide insights into the function of imprinting in placental development and have evolutionary and clinical implications.

The biological realm has inherited symmetries from the physicochemical realm, but with the increasing complexity at higher phenomenological levels of life, some inherited symmetries are broken while novel symmetries appear. These symmetries are of two types, structural and operational. Biological novelties result from breaking operational symmetries. They are followed by acquisition of regularity and stability, in a recurrent process throughout complexity levels.

The mechanisms that initiate reproductive development after fertilization are not understood. Reproduction in higher plants is unique because it is initiated by two fertilization events in the haploid female gametophyte. One sperm nucleus fertilizes the egg to form the embryo. A second sperm nucleus fertilizes the central cell to form the endosperm, a unique tissue that supports the growth of the embryo. Fertilization also activates maternal tissue differentiation, the ovule integuments form the seed coat, and the ovary forms the fruit. To investigate mechanisms that initiate reproductive development, a female-gametophytic mutation termed fie (fertilization-independent endosperm) has been isolated in Arabidopsis. The fie mutation specifically affects the central cell, allowing for replication of the central cell nucleus and endosperm development without fertilization. The fie mutation does not appear to affect the egg cell, suggesting that the processes that control the initiation of embryogenesis and endosperm development are different. FIE/fie seed coat and fruit undergo fertilization-independent differentiation, which shows that the fie female gametophyte is the source of signals that activates sporophytic fruit and seed coat development. The mutant fie allele is not transmitted by the female gametophyte. Inheritance of the mutant fie allele by the female gametophyte results in embryo abortion, even when the pollen bears the wild-type FIE allele. Thus, FIE carries out a novel, essential function for female reproductive development.

The neurodegeneration and amyloid deposition of sporadic Alzheimer disease (AD) also occur in familial AD and in all trisomy-21 Down syndrome (DS) patients, suggesting a common pathogenetic mechanism. We investigated whether defective processing of damaged DNA might be that mechanism, as postulated for the neurodegeneration in xeroderma pigmentosum, a disease with defective repair not only of UV radiation-induced, but also of some oxygen free radical-induced, DNA lesions. We irradiated AD and DS skin fibroblasts or blood lymphocytes with fluorescent light, which is known to cause free radical-induced DNA damage. The cells were then treated with either beta-cytosine arabinoside (araC) or caffeine, and chromatid breaks were quantified. At least 28 of 31 normal donors and 10 of 11 donors with nonamyloid neurodegenerations gave normal test results. All 12 DS, 11 sporadic AD, and 16 familial AD patients tested had abnormal araC and caffeine tests, as did XP-A cells. In one of our four AD families, an abnormal caffeine test was found in all 10 afflicted individuals (including 3 asymptomatic when their skin biopsies were obtained) and in 8 of 11 offspring at a 50% risk for AD. Our tests could prove useful in predicting inheritance of familial AD and in supporting, or rendering unlikely, the diagnosis of sporadic AD in patients suspected of having the disease.

An important determinant of wheat grain quality is the hardness of the grain. The trait is controlled by a major locus, Ha, on the short arm of chromosome 5D. Purified starch granules from soft-grained wheats have associated with them 15-kDa polypeptides called grain softness proteins (GSPs) or "friabilins." Genes that encode one family of closely related GSP polypeptides - GSP-1 genes - were mapped using chromosome substitution lines to the group 5 chromosomes. An F2 population segregating for hard and soft alleles at the Ha locus on a near-isogenic background was used in a single-seed study of the inheritance of grain softness and of GSP-1 alleles. Grain softness versus grain hardness was inherited in a 3:1 ratio. The presence versus absence of GSPs in single seed starch preparations was coinherited with grain softness versus hardness. This showed that grain softness is primarily determined by seed, and not by maternal, genotype. In addition, no recombination was detected in 44 F2 plants between GSP-1 restriction fragment length polymorphisms and Ha alleles. Differences between hard and soft wheat grains in membrane structure and lipid extractability have been described and, of the three characterized proteins that are part of the mixture of 15-kDa polypeptides called GSPs, at least two, and probably all three, are proteins that bind polar lipids. The data are interpreted to suggest that the Ha locus may encode one or more members of a large family of lipid-binding proteins.

DNA and RNA are the polynucleotides known to carry genetic information in life. Chemical variants of DNA and RNA backbones have been used in structure-function and biosynthesis studies in vitro, and in antisense pharmacology, where their properties of nuclease resistance and enhanced cellular uptake are important. This study addressed the question of whether the base(s) attached to artificial backbones encodes genetic information that can be transferred in vivo. Oligonucleotides containing chemical variants of DNA or RNA were used as primers for site-specific mutagenesis of bacteriophage f1. Progeny phage were scored both genetically and physically for the inheritance of information originally encoded by bases attached to the nonstandard backbones. Four artificial backbone chemistries were tested: phosphorothioate DNA, phosphorothioate RNA, 2'-O-methyl RNA and methylphosphonate DNA. All four were found capable of faithful information transfer from their attached bases when one or three artificial positions were flanked by normal DNA. Among oligonucleotides composed entirely of nonstandard backbones, only phosphorothioate DNA supported genetic information transfer in vivo.

Mutations causing mitochondrial defects were induced in a virulent strain of the chestnut blight fungus Cryphonectria parasitica (Murr.) Barr. Virulence on apples and chestnut trees was reduced in four of six extensively characterized mutants. Relative to the virulent progenitor, the attenuated mutants had reduced growth rates, abnormal colony morphologies, and few asexual spores, and they resembled virus-infected strains. The respiratory defects and attenuated virulence phenotypes (hypovirulence) were transmitted from two mutants to a virulent strain by hyphal contact. The infectious transmission of hypovirulence occurred independently of the transfer of nuclei, did not involve a virus, and dynamically reflects fungal diseases caused by mitochondrial mutations. In these mutants, mitochondrial mutations are further implicated in generation of the attenuated state by (i) uniparental (maternal) inheritance of the trait, (ii) presence of high levels of cyanide-insensitive mitochondrial alternative oxidase activity, (iii) cytochrome deficiencies, and (iv) structural abnormalities in the mtDNA. Hence, cytoplasmically transmissible hypovirulence phenotypes found in virus-free strains of C. parasitica from recovering trees may be caused by mutant forms of mtDNA.

Inheritance of specific apolipoprotein E (apoE) alleles determines, in large part, the risk and mean age of onset of late-onset familial and sporadic Alzheimer disease. The mechanism by which the apoE isoforms differentially contribute to disease expression is, however, unknown. Isoform-specific differences have been identified in the binding of apoE to the microtubule-associated protein tau, which forms the paired helical filament and neurofibrillary tangles, and to amyloid beta peptide, a major component of the neuritic plaque. These and other isoform-specific interactions of apoE give rise to testable hypotheses for the mechanism(s) of pathogenesis of Alzheimer disease. An unresolved issue of increasing importance is the relationship between the structural pathological lesions and the cellular pathogenesis responsible for the clinical disease phenotype, progressive dementia. The identification of apoE in the cytoplasm of human neurons and the characterization of isoform-specific binding of apoE to the microtubule-associated proteins tau and MAP-2 present the possibility that apoE may affect microtubule function in the Alzheimer brain.

O glyphosate é o principal herbicida utilizado no manejo de plantas daninhas na agricultura, aplicado em alguns sistemas de forma repetitiva ao longo de cada ano. Esta prática selecionou biótipos resistentes de espécies de plantas daninhas, sendo o capim-amargoso (Digitaria insularis) selecionado no Brasil. Portanto, se tornam necessários estudos para entender, manejar e reduzir a infestação do capim-amargoso resistente ao glyphosate. Dessa forma, esta pesquisa foi desenvolvida com os objetivos de: (i) mapear áreas do Brasil com possíveis infestações de capim-amargoso resistente ao glyphosate; (ii) avaliar alternativas químicas de seu manejo; (iii) elucidar os mecanismos de resistência ao glyphosate e; (iv) avaliar a herança genética dos genes que conferem resistência ao glyphosate. Para o desenvolvimento dos experimentos foram coletadas sementes de biótipos potencialmente resistentes de diversas regiões do Brasil onde ocorreram falhas de controle de D. insularis após a aplicação de glyphosate. Na primeira etapa da pesquisa foram realizados experimentos para determinação de uma dose discriminatória de triagementre as populações resistentes e suscetíveis ao glyphosate, através de curvas de dose-resposta, para identificar a resistência ao Glyphosate, sendo que estes dados foram utilizados para mapear a ocorrência de biótipos resistentes em algumas regiões do país. Na segunda etapa foi conduzido um experimento em casa-de-vegetação visando encontrar herbicidas alternativos ao Glyphosate para controle do capim-amargoso, utilizando herbicidas recomendados para as culturas do milho e algodão, tanto em condições de aplicação de pré como em pós-emergência da planta daninha. Na terceira etapa foram realizados ensaios para determinar a existência de absorção e translocação diferencial do glyphosate em biótipos suscetíveis e resistentes, juntamente com a análise molecular para comparar a região 106 do gene que codifica a EPSPs nestes biótipos. Por fim um estudo de polinização cruzada foi conduzido para avaliar se genes de resistência ao glyphosate são transferidos para a geração seguinte após inflorescências de biótipos suscetíveis serem acondicionadas com as de biótipos resistentes, submetendo a geração seguinte a experimentos de curva de dose-resposta com o glyphosate. Através do modelo de curva dose-resposta do programa estatístico R, determinou-se a dose de 960 g e.a ha-1, como a dose utilizada para triagem dos biótipos oriundos de diferentes regiões do Brasil. Com isto foram gerados mapas indicando a presença ou ausência de resistência ao herbicida, sendo que as região oeste do Paraná e sul do Mato Grosso do Sul apresentam maior número de localidades com a presença de biótipos resistentes. As alternativas de controle viáveis como pós-emergentes no estádio de um a dois perfilhos, foram os herbicidas Nicosulfuron, Imazapic + Imazapyr, Atrazine, Haloxifop-methyl e Tepraloxydim. Na pré-emergência do capim-amargoso os herbicidas Atrazine, Isoxaflutole, S-metolachlor, Clomazone, Diuron e Flumioxazin se apresentaram como eficazes para o controle desta espécie. Os resultados do experimento de absorção, translocação e comparação da região 106 não mostraram diferenças entre os biótipos resistente e suscetível. O experimento sobre cruzamento entre biótipos resistente e suscetível determinou a espécie D. insularis como autógama e sem transferência de genes que causam a resistência ao glyphosate.

Esta pesquisa aponta alguns dos efeitos subjetivos e estratégias singulares de resistência frente à desigualdade racial no nosso país, abordando as vicissitudes de inscrição no laço social de mulheres negras e pobres. É fruto de uma intervenção clínico-política com um grupo de adolescentes em uma Escola Municipal de Ensino Fundamental de São Paulo na qual foi se evidenciando, para nós, a necessidade de cada um desses adolescentes de defender intransigentemente a honra e o valor de suas mães frente aos outros membros do grupo. Tanto pelo seu excesso como pela sua repetição, essa situação nos sugeria um mal-estar e um não dito referido às configurações familiares e à posição destas mulheres nesta comunidade escolar, que nos levou a escutá-las. Tomando a indicação freudiana de que a psicologia individual seria também psicologia social e a formulação lacaniana de que podemos considerar o Inconsciente como sendo a Política, acreditamos ser indispensável escutar o sujeito levando em consideração o Outro, entendido tanto do ponto de vista sócio-histórico, como libidinal. Isso significa que não poderíamos escutar estas mulheres sem considerar o campo de desigualdades sociais e raciais no qual estavam inscritas discursivamente, o que nos exigiu uma interlocução fundamental tanto com pesquisas da antropologia social e da sociologia, como da história. A fala destas mulheres foi nos revelando que, além de outras identificações contingentes, o fato de serem reconhecidas e se reconhecerem como mulheres negras era um elemento fundamental nas suas vivências cotidianas. Uma vez que nosso passado escravista não teria sido suficientemente lembrado e admitido, alguns traços se fariam presentes através de uma transmissão simbólica, pelos subterrâneos da cultura, de uma posição de servidão a elas atribuída. Permaneceria de uma forma atualizada e insidiosa uma divisão racializada da nossa sociedade, ancorada na herança de uma cisão entre a mulher mundana cujo corpo seria visto como um corpo de gozo, mas sem valor social, a mucama, e a que seria valorizada socialmente à custa de um corpo assexuado, casta e educada, esposa do senhor de escravos. Apesar de tantos avanços, as conquistas femininas das últimas décadas não seriam totalmente estendidas a essas mulheres, negras e pobres, que seguiriam, frequentemente, apresentando no imaginário social um corpo ao qual se atribuiria a capacidade de satisfazer os desejos mais inconfessáveis de um homem à custa de ser visto como propriedade e domínio deste. A atitude racista se faria presente em relação a elas, entendida como o ato de segregação do gozo inadmitido de um sujeito no corpo de um outro, ou ainda, como Lacan apontou, impondo a um outro, seu modo de gozo. Mais do que uma identidade das mulheres negras, consideramos fundamental conceber a particularidade de um laço que se estabeleceria na relação com elas, na medida em que seu corpo seria capaz de despertar e revelar a relação do sujeito com o mais íntimo e insuportável de si mesmo: ela seria a estrangeira frente a um homem, por ser mulher; e seria estrangeira frente a uma mulher ou homem branco, por ser negra. A sua condição de estrangeira a deixaria assim como figura paradigmática de um Outro sexo, um sexo Outro, um gozo Outro, recaindo sobre ela as reações mais violentas de extirpação desse gozo. As estratégias de como manter o que seria próprio do gozo feminino não balizado pelo gozo fálico, posto que seria suplementar a ele frente a essa injunção de segregação e depreciação, seriam sempre singulares. Apresentamos um caso clínico, Silvana, apontando suas estratégias de resistência frente a um discurso social que a desqualificaria tentando lhe impor um estreitamento de sua vida erótica e sua redução a um modo único de gozo

Este estudio es una síntesis de las últimas aportaciones acerca de las vías en al-Andalus (ss. VIII-XV), cuya estructura inicial es herencia de la época romana. Dadas las limitaciones que imponen los escasos datos que hallamos en las fuentes árabes y los pocos restos camineros conocidos, se hace necesario emplear una nueva metodología de investigación: Se aborda el objeto como un estudio global, no sólo de los caminos, sino de todos los elementos interrelacionados con ellos, denominados como "espacios viales". Luego se presenta la indisoluble relación de los "espacios viales" con los castillos y torres ("espacios de control y defensa") en al-Andalus, como los vigilantes de los caminos, y se enuncia la toponimia árabe relacionada tanto con los "espacios viales" como con los "espacios de control y defensa". Finalmente, se recogen los datos que las diversas crónicas árabes nos proporcionan acerca de la política viaria en al-Andalus a lo largo del tiempo, así como sobre quién era el responsable de velar por el buen estado de los tramos viales.

Inheritance of each chromosome depends upon its centromere. A histone H3 variant, centromere protein A (CENP-A), is essential for epigenetically marking centromere location. We find that CENP-A is quantitatively retained at the centromere upon which it is initially assembled. CENP-C binds to CENP-A nucleosomes and is a prime candidate to stabilize centromeric chromatin. Using purified components, we find that CENP-C reshapes the octameric histone core of CENP-A nucleosomes, rigidifies both surface and internal nucleosome structure, and modulates terminal DNA to match the loose wrap that is found on native CENP-A nucleosomes at functional human centromeres. Thus, CENP-C affects nucleosome shape and dynamics in a manner analogous to allosteric regulation of enzymes. CENP-C depletion leads to rapid removal of CENP-A from centromeres, indicating their collaboration in maintaining centromere identity.

1) Our study addresses the role of non-genetic and genetic inheritance in shaping the adaptive potential of populations under a warming ocean scenario. We used a combined experimental approach (transgenerational plasticity and quantitative genetics) to partition the relative contribution of maternal vs. paternal (additive genetic) effects to offspring body size (a key component of fitness), and investigated a potential physiological mechanism (mitochondrial respiration capacities) underlying whole organism growth/size responses. 2) In very early stages of growth (up to 30 days), offspring body size of marine sticklebacks benefited from maternal transgenerational plasticity (TGP): offspring of mothers acclimated to17°C were larger when reared at 17°C, and offspring of mothers acclimated to 21°C were larger when reared at 21°C. The benefits of maternal TGP on body size were stronger and persisted longer (up to 60 days) for offspring reared in the warmer (21°C) environment, suggesting that maternal effects will be highly relevant for climate change scenarios in this system. 3) Mitochondrial respiration capacities measured on mature offspring (F1 adults) matched the pattern of TGP for juvenile body size, providing an intuitive mechanistic basis for the maternal acclimation persisting into adulthood. Size differences between temperatures seen at early growth stages remained in the F1 adults, linking offspring body size to maternal inheritance of mitochondria. 4) Lower maternal variance components in the warmer environment were mostly driven by mothers acclimated to ambient (colder) conditions, further supporting our tenet that maternal effects were stronger at elevated temperature. Importantly, all parent-offspring temperature combination groups showed genotype x environment (GxE) interactions, suggesting that reaction norms have the potential to evolve. 5) To summarise, transgenerational plasticity and genotype x environment interactions work in concert to mediate impacts of ocean warming on metabolic capacity and early growth of marine sticklebacks. TGP can buffer short-term detrimental effects of climate warming and may buy time for genetic adaptation to catch up, therefore markedly contributing to the evolutionary potential and persistence of populations under climate change.
