Ethylene gas is burnt to generate soot which is collected thermophoretically from different locations of the flame. Tribological performance of the collected soot in hexadecane suspension is compared with that of carbon black and diesel soot. The soots are analysed to yield a range of mechanical properties, physical structures and chemistry. The paper correlates these property variations with the corresponding variations in friction and wear when the soot suspended in hexadecane is used to lubricate a steel on steel sliding interaction. The particles are dispersed in hexadecane by a non-ionic surfactant, poly-isobutylene succinimide (PIBS), which is mono-functional with no free amine group. The grafting of the surfactant on the soot particles is found to have a profound effect on the dispersion of the soot, in general, while, between the different soot types, the tribology is differentiated by the physical structure and chemistry.

In the present investigation an attempt has been made to develop a new co-polymeric material for controlled release tablet formulations. The acrylamide grafting was successfully performed on the backbone of sago starch. The modified starch was tested for acute toxicity and drug-excipient compatibility study. The grafted material was used in making of controlled release tablets of lamivudine. The formulations were evaluated for physical characteristics such as hardness, friability, %drug content and weight variations. The in vitro release study showed that the optimized formulation exhibited highest correlation (R) value in case of Higuchi model and the release mechanism of the optimized formulation predominantly exhibited combination of diffusion and erosion process. There was a significant difference in the pharmacokinetic parameters (T-max, C-max, AUC, V-d, T-1/2 and MDT) of the optimized formulation as compared to the marketed conventional tablet Lamivir (R) was observed. The pharmacokinetics parameters were showed controlled pattern and better bioavailability. An in situ carbon-grafted alkaline iron electrode prepared from the active material obtained by decomposing the alpha-FeC2O4 center dot 2H(2)O-polyvinyl alcohol (PVA) composite at 600 degrees C in a vacuum is reported. The active material comprises a mixture of a-Fe and Fe3O4 with the former as the prominent component. A specific discharge capacity in excess of 400 mA h g(-1) at a current density of 100 mA g(-1) is obtained with a faradaic efficiency of 80% for the iron electrode made from carbon-grafted active material (CGAM). The enhanced performance of the alkaline iron electrode is attributed to the increased amount of metallic iron in the active material and its concomitant in situ carbon grafting.

Polyolefin based blends have tremendous commercial importance in view of their exceptional properties. In this study the interface of a biphasic polymer blend of PE (polyethylene) and PEO (polyethylene oxide) has been tailored to reduce the interfacial tension between the phases and to render finer morphology. This was accomplished by employing various strategies like addition of maleated PE (PE grafted maleic anhydride), immobilizing PE chains, ex situ, onto MWNTs by covalent grafting, and in situ grafting of PE chains onto MWNTs during melt processing. Multiwalled nanotubes (MWNTs) with different surface functional groups have been synthesized either a priori or were facilitated during melt mixing at higher temperature. NH2 terminated MWNTs were synthesized by grafting ethylene diamine (EDA) onto carboxyl functionalized carbon nanotubes (COOH(MWNTs) and further, was used to reactively couple with maleated PE to immobilize PE chains on the surface of MWNTs. The covalent coupling of maleated PE with NH2 terminated MWNTs was also realized in situ in the melt extruder at high temperature. Both NH2 terminated MWNTs and the in situ formed PE brush on MWNTs during melt mixing, revealed a significant improvement in the mechanical properties of the blend besides remarkably improving the dispersion of the minor phase (PEO) in the blends. Structural properties of the composites were evaluated and the tensile fractured morphology was assessed using scanning electron microscopy.

Dispersion of nanoparticles in polymer nanocomposite films determines the application potential of these systems as novel materials with unique physical properties. Grafting polymers to, mostly inorganic, nanoparticles has been suggested as an effective strategy to enhance dispersion and hence the efficacy of materials. In this review, we discuss the various parameters which control dispersion of polymer grafted nanoparticles in polymer nanocomposite films. We discuss how surface x-ray scattering and microscopy can provide complementary and unique information in thin polymer nanocomposite films to unravel the subtle interplay of entropic and surface interactions, mediated by confinement, that leads to enhanced dispersion of the nanoparticles in these films. Herein, we present six new lipopolymers based on low molecular weight, branched polyethylenimine (BPEI 800 Da) which are hydrophobically modified using ferrocene terminated alkyl tails of variable lengths. The effects of degree of grafting, spacer length and the redox state of ferrocene in the lipopolymers on the self assembly properties were investigated in detail by TEM, AFM, DLS and zeta potential measurements. The assemblies displayed an oxidation induced increase in the size of the aggregates. The co-liposomes comprising the lipopolymer and a helper lipid, 1,2-dioleoyl phosphatidyl ethanolamine (DOPE), showed excellent gene (pDNA) delivery capability in a serum containing environment in two cancer cell lines (HeLa and U251 cells). Optimized formulations showed remarkably higher transfection activity than BPEI (25 kDa) and were also significantly better than a commercial transfection reagent, Lipofectamine 2000 as evidenced from both the luciferase activity and GFP expression analysis. Oxidation of ferrocene in the lipopolymers led to drastically reduced levels of gene transfection which was substantiated by reduced cellular internalization of fluorescently labelled pDNA as detected using confocal microscopy and flow cytometry. Moreover, the transfection inactive oxidized lipopolyplexes could be turned transfection active by exposure to ascorbic acid (AA) in cell culture medium during transfection. Endocytosis inhibition experiments showed that gene expression mediated by reduced formulations involved both clathrin and caveolae mediated pathways while the oxidized formulations were routed via the caveolae. Cytotoxicity assays revealed no obvious toxicity for the lipopolyplexes in the range of optimized transfection levels in both the cell lines studied. Overall, we have exploited the redox activity of ferrocene in branched PEI-based efficient polymeric gene carriers whose differential transfection activities could be harnessed for spatial or temporal cellular transfections.

We present the results of combined experimental and theoretical (molecular dynamics simulations and integral equation theory) studies of the structure and effective interactions of suspensions of polymer grafted nanoparticles (PGNPs) in the presence of linear polymers. Due to the absence of systematic experimental and theoretical studies of PGNPs, it is widely believed that the structure and effective interactions in such binary mixtures would be very similar to those of an analogous soft colloidal material-star polymers. In our study, polystyrene-grafted gold nanoparticles with functionality f = 70 were mixed with linear polystyrene (PS) of two different molecular weights for obtaining two PGNP: PS size ratios, xi = 0.14 and 2.76 (where, xi = M-g/M-m, M-g and M-m being the molecular weights of grafting and matrix polymers, respectively). The experimental structure factor of PGNPs could be modeled with an effective potential (Model-X), which has been found to be widely applicable for star polymers. Similarly, the structure factor of the blends with xi = 0.14 could be modeled reasonably well, while the structure of blends with xi = 2.76 could not be captured, especially for high density of added polymers. A model (Model-Y) for effective interactions between PGNPs in a melt of matrix polymers also failed to provide good agreement with the experimental data for samples with xi = 2.76 and high density of added polymers. We tentatively attribute this anomaly in modeling the structure factor of blends with xi = 2.76 to the questionable assumption of Model-X in describing the added polymers as star polymers with functionality 2, which gets manifested in both polymer-polymer and polymer-PGNP interactions especially at higher fractions of added polymers. The failure of Model-Y may be due to the neglect of possible many-body interactions among PGNPs mediated by matrix polymers when the fraction of added polymers is high. These observations point to the need for a new framework to understand not only the structural behavior of PGNPs but also possibly their dynamics and thermo-mechanical properties as well. The use of copolymer and polymer blends widened the possibility of creating materials with multilayered architectures. Hierarchical polymer systems with a wide array of micro and nanostructures are generated by thermally induced phase separation (TIPS) in partially miscible polymer blends. Various parameters like the interaction between the polymers, concentration, solvent/non-solvent ratio, and quenching temperature have to be optimized to obtain these micro/nanophase structures. Alternatively, the addition of nanoparticles is another strategy to design materials with desired hetero-phase structures. The dynamics of the polymer nanocomposite depends on the statistical ordering of polymers around the nanoparticle, which is dependent on the shape of the nanoparticle. The entropic loss due to deformation of polymer chains, like the repulsive interactions due to coiling and the attractive interactions in the case of swelling has been highlighted in this perspective article. The dissipative particle dynamics has been discussed and is correlated with the molecular dynamics simulation in the case of polymer blends. The Cahn Hillard Cook model on variedly shaped immobile fillers has shown difference in the propagation of the composition wave. The nanoparticle shape has a contributing effect on the polymer particle interaction, which can change the miscibility window in the case of these phase separating polymer blends. Quantitative information on the effect of spherical particles on the demixing temperature is well established and further modified to explain the percolation of rod shaped particles in the polymer blends. These models correlate well with the experimental observations in context to the dynamics induced by the nanoparticle in the demixing behavior of the polymer blend. The miscibility of the LCST polymer blend depends on the enthalpic factors like the specific interaction between the components, and the solubility product and the entropic losses occurring due to the formation of any favorable interactions. Hence, it is essential to assess the entropic and enthalpic interactions induced by the nanoparticles independently. The addition of nanoparticles creates heterogeneity in the polymer phase it is localized. This can be observed as an alteration in the relaxation behavior of the polymer. This changes the demixing behavior and the interaction parameter between the polymers. The compositional changes induced due to the incorporation of nanoparticles are also attributed as a reason for the altered demixing temperature. The particle shape anisotropy causes a direction dependent depletion, which changes the phase behavior of the blend. The polymer-grafted nanoparticles with varying grafting density show tremendous variation in the miscibility of the blend. The stretching of the polymer chains grafted on the nanoparticles causes an entropy penalty in the polymer blend. A comparative study on the different shaped particles is not available up to date for understanding these aspects. Hence, we have juxtaposed the various computational studies on nanoparticle dynamics, the shape effect of NPs on homopolymers and also the cases of various polymer blends without nanoparticles to sketch a complete picture on the effect of various particles on the miscibility of LCST blends.

Polyolefinic membranes have attracted a great deal of interest owing to their ease of processing and chemical inertness. In this study, porous polyolefin membranes were derived by selectively etching PEO from PE/PEO (polyethylene/poly(ethylene oxide)) blends. The hydrophobic polyolefin (low density polyethylene) was treated with UV-ozone followed by dip coating in chitosan acetate solution to obtain a hydrophilic-antibacterial surface. The chitosan immobilized PE membranes were further characterized by Fourier transform infrared spectroscope (FTIR) and X-ray photoelectron spectroscope (XPS). It was found that surface grafting of chitosan onto PE membranes enhanced the surface roughness and the concentration of nitrogen (or amine) scaled with increasing concentration of chitosan (0.25 to 2% wt/vol), as inferred from Kjeldahl nitrogen analysis. The pure water flux was almost similar for chitosan immobilized PE membranes as compared to membranes without chitosan. The bacterial population, substantially reduced for membranes with higher concentration of chitosan. For instance, 90 and 94% reduction in Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) colony forming unit respectively was observed with 2% wt/vol of chitosan. This study opens new avenues in designing polyolefinic based antibacterial membranes for water purification.

Lehenengoa CdSe nanopartikulen sintesia, eta ondoren hauen funtzionalizazioa blokezko kopolimero bati gehitzean blokeetako batean hauen dispertsio egokia lortzeko. Horrela CdSe nanopartikulen kasuan ondoren aurkezten diren propietateetan tamainak duen garrantzia kontutan izanik, sintesi parametro ugari aztertu dira (egonkortzaile mota, pH eta Cd:Se erlazioa) nanopartikulen sintesia optimizatzeko,. honez gain, nanopartikularen gainazalean ondorengo eraldaketetarako egokia den talde funtzionala lortuaz. Behin nanopartikulen sintesia optimizatuta, hauen gainazala eraldatzeko teknika ezberdinak aztertu dira, ondoren blokezko kopolimeroei gehitzean nanokonpositeetan dispertsio onak lortzeko. Funtzionalizaziorako teknika ezberdinak erabili dira, hala nola amonio gatzekin eraldaketa edota grafting teknika ezberdinak (grafting from eta grafting through). Azken honen bitartez nanopartikulen dispertsio on bat lortu delarik blokezko kopolimero baten poliestireno fasean.Nanokonposite egokiak lorturik, hauen eta hauek osatzen dituzten nanopartikula eraldatuen propietate elektrostatikoak aztertu dira indar elektrostatikozko mikroskopia neurketa bidez.

Não tem sido frequentes no Brasil estudos de avaliação da qualidade dos serviços de saúde. Tem sido adotado entendimento de qualidade como o grau em que processo de assistência aumenta a probabilidade de resultados favoráveis e diminui a probabilidade de resultados desfavoráveis, dado o estado do conhecimento médico. Indicadores de resultados de efeitos adversos do processo de assistência costumam ser empregados e, entre eles, para aquelas condições e procedimentos onde óbitos ocorrem com frequência, estão as taxas de mortalidade hospitalar. Entre esses procedimentos inclui-se a cirurgia de revascularizaçäo do miocárdio. Apesar de frequentes na literatura, particularmente norte-americana, não há estudos de escala realizados no Brasil. Para estudos deste tipo bases de dados administrativas tem sido empregadas. No Brasil recentemente tem sido exploradas as potencialidades dos bancos de dados do Sistema de Informações Hospitalares do Sistema Único de Saúde do Ministério da Saúde (SIH-SUS) em diversos estudos. Como há registro de óbitos hospitalares no sistema é possível utilizá-lo para a obtenção de dados sobre mortalidade hospitalar. Os bancos de dados do SIH-SUS de 1996 a 1998 foram integrados e as variáveis disponíveis no banco obtido examinadas quanto a possibilidade de inclusão do estudo descritivo de características da cirurgia coronária no país. Foram identificadas aquelas variáveis que poderiam ser utilizadas para proceder algum grau de ajuste de risco para os casos atendidos pelos diferentes hospitais. Para que se obtivesse uma comparação do comportamento do ajuste obtido com essas variáveis com modelos mais completos que incorporassern mais variáveis, inclusive variáveis clínicas, foram estudadas para o mesmo período, as internações realizadas no Hospital Universitário Pedro Ernesto da Universidade do Estado do Rio de Janeiro, utilizando dados de banco específico do Serviço de Cirurgia Cardíaca. Além do estudo descritivo foram desenvolvidos para os casos deste hospital modelos de regressão logística incorporando variáveis pré-operatórias e com as variáveis disponíveis no SIH-SUS para avaliar as diferentes capacidades de ajuste de risco. Após a escolha de um modelo de risco com maior capacidade de ajuste, foram calculadas as taxas de mortalidade hospitalar e obtidos os valores de taxas esperadas após o ajuste de risco. Os hospitais forma ordenados de acordo com as razões entre as taxas observadas e esperadas e identificados aqueles hospitais que apresentavam razões estatisticamente significativas superiores e inferiores a média nacional. Estudou-se também o efeito do volume de casos sobre a mortalidade hospitalar. Foram obtidas informações de 41.989 cirurgias codificadas como cirurgia coronária com circulação extracorpórea realizadas em 131 hospitais brasileiros, em 22 unidades da federaçâo. A taxa anual por 100000 habitantes foi de 8,7 para o Brasil, com São Paulo apresentando taxa de 16,6. Para efeitos de comparação a taxa em anos em torno de 1997 foi de 144,5 nos EUA, 54,4 no Canadá, 90,0 na Austrália e 31,5 em Portugal. A taxa de mortalidade no período foi de 7,2 % (EUA, 2,8%; Canadá, 2,5%: França, 3,2%). A maioria de pacientes operados foi do sexo masculino (67,5%) e a idade média foi de 59,9 anos.

A medula óssea adulta possui duas populações de células-tronco importantes no tratamento de diversas doenças hepáticas: células-tronco hematopoiéticas (CTHs) e células-tronco mesenquimais. A regeneração do fígado após a hepatectomia é um processo complexo que requer a proliferação de todas as células hepáticas. Fatores de crescimento, citocinas e componentes da matriz extracelular são elementos-chave nesse processo. As lamininas são uma família de proteínas de matriz extracelular, com funções adesivas e quimiotáticas pelo recrutamento de integrinas e outros receptores de superfície celular. No fígado normal, a laminina é expressa nas veias porta e centrolobular. O objetivo desse estudo foi investigar a expressão de laminina durante a regeneração hepática induzida por hepatectomia parcial e após o transplante de células mononucleares de medula óssea. As células mononucleares de medula óssea foram obtidas dos fêmures e tíbias de ratos, isoladas, marcadas com DAPI e injetadas pela veia porta em ratos recém-hepatectomizados. Os fígados foram coletados 15 minutos, 1 dia e 3 dias após a hepatectomia e o transplante de células de medula óssea e congelados. Os cortes foram imunomarcados com anticorpos primários anti-CD34 e anti-laminina de rato e observados em microscópio confocal de varredura a laser. Os resultados mostraram que 15 minutos após a hepatectomia parcial, as células-tronco hematopoiéticas CD34+ transplantadas foram encontradas em contato com a laminina localizada nas veias porta e centrolobular, indicando que a laminina poderia participar na adesão inicial das células-tronco a esses vasos logo após o seu transplante. Além disso, 1 e 3 dias após a hepatectomia, as células mononucleares de medula óssea transplantadas foram observadas nos sinusóides hepáticos expressando laminina. Esses resultados sugerem que a laminina pode ser um componente da matriz extracelular importante para a adesão e enxerto de células de medula óssea no fígado após uma lesão. Nós também analisamos a expressão de osteopontina (OPN) em células de medula óssea e CTHs. Os resultados por microscopia confocal demonstraram que a maioria das células mononucleares de medula óssea recém-isoladas expressa quantidades variáveis de OPN. Além disso, algumas CTHs CD34+ também expressam OPN. Após 1 e 4 dias de cultura, observamos uma diminuição de células expressando CD34, e um aumento na expressão de OPN pelas células mononucleares de medula óssea.

脂肪酸是生物体内普遍存在、具重要生理功能的物质,亦是重要的化工原料。研究脂肪酸生物合成及其调控,既是揭示生命活动基本规律的需要,又具巨大的经济价值。多形汉逊氏酵母(Hansenula polymorpha)是一种甲基嗜热酵母,能合成多聚不饱和脂肪酸,是研究脂肪酸生物合成的理想材料之一。为阐明多形汉逊氏酵母细胞中脂肪酸生物合成途径、关键步骤、调节机理,并利用此系统生产有用脂肪酸,我们开展了不饱和脂肪酸生物合成关键酶基因--△9-脂肪酸去饱和酶基因研究。 以P. angusta IFO 1475的P-OLE1基因为探针,Southern杂交分析,发现在亲缘关系很近的不同种类的甲基嗜热酵母如H. pofymorpha、Pichia angusta、P. pastoris、P. methanolica和Candida boMinii中Δ9-脂肪酸去饱和酶基因的结构多形性。 构建了H. polymorpha CBS 1976染色体Δ9-脂肪酸去饱和酶基因座位的限制性酶切图谱,进而分离了3.4 kb BamHI-XhoI基因片段并进行全序列分析,结果表明这个片段含1个与已克隆的酵母Δ59-脂肪酸去饱和酶基因高度同源的、由1353 bp组成的ORF。推导的H-OLE1多肽具脂肪酸去饱和酶的一些基本特征,如含2个结构域:1个位于N一端、含3个保守的组氨酸簇、具催化功能,另1个位于C-端、参与脱饱和反应中电子传递、类似细胞色素b5。将这个序列申报DDBJ,获得Accession number为:AB024576,推导的蛋白的氨基酸序列的Accession number为:BAA75902。 为验证H-OLE1基因的功能,建立了多形汉逊氏酵母DNA电穿孔实验系统,进行了遗传互补测验。发现完整的H-OLE1基因可互补缺乏Δ59-脂肪酸去饱和酶活性的多形汉逊氏酵母营养缺陷型fadl突变体,却不能互补相应的酿酒酵母olel突变体,而由酿酒酵母GAP表达框架和H-OLE1 ORF组成的嵌合基因可互补上述olel突变体。说明H-OLE1基因编码Δ9-脂肪酸去饱和酶,多形汉逊氏酵母的Δ9-脂肪酸去饱和酶和酿酒酵母的脂肪酸脱饱和系统相亲和,而H-OLE1基因的启动子在异源细胞中没有活性。 为研究H-OLE1基因的转录及其调节规律,通过一系列实验,首次找到了可在研究多形汉逊氏酵母基因表达时用作内标的GAP基因。Northern杂交发现,H-OLE1基因在细胞中以较低水平表达,产生1.5 kb的转录子;基因表达略受不饱和脂肪酸的抑制;在多形汉逊氏酵母HOLE1基因的转录调节中,Choi等在酿酒酵母OLE1基因中发现的脂肪酸调节元件FAR可能不是关键的。 利用基因敲除技术,通过转化H-OLE1∷S-LEU2线性DNA到多形汉逊氏酵母二倍体细胞(fadl/FADl)中,首次构建了多形汉逊氏酵母H-OLE1基因的破坏株。遗传学和分子生物学研究表明,破坏株细胞中线性DNA定位串联多拷贝整合到染色体中并置换了fadl突变部位。利用气相色谱分析了ΔH-OLE1破坏株、fadl-2突变株、野生型菌株及含H-OLE1基因转化子的细胞总脂肪酸,发现多形汉逊氏酵母细胞中除18:0→18:1(Δ9)→18:2(Δ9,12)→18:3(Δ9,12,15)这个脂肪酸去饱和主路外,还可能存在其它几个脱饱和反应与延长反应,如16:1(Δ9)→16:2(Δ9,12)→18:2(Δ11,14);16:1(Δ9)→18:1(Δ11)→18:2(Δ11,15)等。 近年维管组织分化研究进展迅速,取得大量可喜结果,也存在许多不足,如细胞分化调节机理,特别是激素诱导的分子机理研究比较薄弱。为建立研究维管组织分化的理想系统,研究嫁接体发育的激素调节机制,在Parkinson和Yeoman发明的离体茎段嫁接系统的基础上,研究了激素对嫁接体发育特别是维管组织分化的影响。 采用不同的嫁接方法,用试管苗对黄瓜离体茎段自体嫁接、亲和性的黄瓜/黑籽南瓜与不亲和性的黄瓜/绿豆离体茎段嫁接组合进行研究,建立了嫁接过程简单、污染率低的试管苗离体茎段嫁接系统。利用往培养基中添加或不加植物激素研究嫁接体发育,发现通过改变培养基中的植物激素,可使亲和的嫁接体难以形成贯通砧木和接穗的维管束桥,也可诱导非亲和性的嫁接体产生维管束桥。初步研究证明利用植物激素可以克服嫁接不亲和性,这一结果是嫁接基础理论研究的一个重要进展,对揭示嫁接亲和性机制具重要意义。由于黄瓜绿豆嫁接组合中,砧木绿豆是可以固氮的豆科植物,研究结果具有潜在的应用前景。 详细地研究了外源IAA和玉米素(ZT)对黄瓜自体嫁接系统中维管束桥形成时间和数目特别是贯通砧木和接穗的管状分子数的影响。当砧木和接穗培养基中都没有添加植物激素时,嫁接接合部难以产生维管束桥,也难以产生贯通的管状分子。当培养基中添加植物激素时,维管束桥数和贯通的管状分子数随激素浓度和种类的不同而不同。本实验的最佳激素条件是:在接穗培养基中加IAA 1.0 mg/L和ZT 0.25 mg/L,在砧木培养基中加ZT 0.25 mg/L。研究表明在试管苗离体茎段自体嫁接系统中,外源激素是嫁接成功的必要条件。试管苗离体茎段嫁接系统是一个理想的研究植物维管组织分化的新系统。 通过对嫁接体发育期接合部及嫁接体各部分IAA、玉米素及玉米素核苷(Z+ZR)的ELISA分析,发现嫁接接合部维管束的再生受IAA和Z+ZR含量的共同调节;连接接穗和砧木维管束桥的分化比维管束的网联要求更高的IAA水平及LAA(Z+ZR)比率。 上述结果为利用嫁接系统研究维管组织分化机理奠定了基础,使进一步研究嫁接体发育的激素调节机理成为可能。

BACKGROUND: When fresh morselized graft is compacted, as in impaction bone-grafting for revision hip surgery, fat and marrow fluid is either exuded or trapped in the voids between particles. We hypothesized that the presence of incompressible fluid damps and resists compressive forces during impaction and prevents the graft particles from moving into a closer formation, thus reducing the graft strength. In addition, viscous fluid such as fat may act as an interparticle lubricant, thus reducing the interlocking of the particles. METHODS: We performed mechanical shear testing in the laboratory with use of fresh-frozen human femoral-head allografts that had been passed through different orthopaedic bone mills to produce graft of differing particle-size distributions (grading). RESULTS: After compaction of fresh graft, fat and marrow fluid continued to escape on application of normal loads. Washed graft, however, had little lubricating fluid and better contact between the particles, increasing the shear resistance. On mechanical testing, washed graft was significantly (p < 0.001) more resistant to shearing forces than fresh graft was. This feature was consistent for different bone mills that produced graft of different particle-size distributions and shear strengths. CONCLUSIONS: Removal of fat and marrow fluid from milled human allograft by washing the graft allows the production of stronger compacted graft that is more resistant to shear, which is the usual mode of failure. Further research into the optimum grading of particle sizes from bone mills is required.
