Thymic atrophy is known to occur during infections; however, there is limited understanding of its causes and of the cross-talk between different pathways. This study investigates mechanisms involved in thymic atrophy during a model of oral infection by Salmonella enterica serovar Typhimurium (S.typhimurium). Significant death of CD4+CD8+ thymocytes, but not of single-positive thymocytes or peripheral lymphocytes, is observed at later stages during infection with live, but not heat-killed, bacteria. The death of CD4+CD8+ thymocytes is Fas-independent as shown by infection studies with lpr mice. However, apoptosis occurs with lowering of mitochondrial potential and higher caspase-3 activity. The amounts of cortisol, a glucocorticoid, and interferon- (IFN-), an inflammatory cytokine, increase upon infection. To investigate the functional roles of these molecules, studies were performed using Ifn/ mice together with RU486, a glucocorticoid receptor antagonist. Treatment of C57BL/6 mice with RU486 does not affect colony-forming units (CFU), amounts of IFN- and mouse survival; however, there is partial rescue in thymocyte death. Upon infection, Ifn/ mice display higher CFU and lower survival but more surviving thymocytes are recovered. However, there is no difference in cortisol amounts in C57BL/6 and Ifn/ mice. Importantly, the number of CD4+CD8+ thymocytes is significantly higher in Ifn/ mice treated with RU486 along with lower caspase-3 activity and mitochondrial damage. Hence, endogenous glucocorticoid and IFN--mediated pathways are parallel but synergize in an additive manner to induce death of CD4+CD8+ thymocytes during S.typhimurium infection. The implications of this study for host responses during infection are discussed.

Many aspects of skeletal muscle biology are remarkably similar between mammals and tiny insects, and experimental models of mice and flies (Drosophila) provide powerful tools to understand factors controlling the growth, maintenance, degeneration (atrophy and necrosis), and regeneration of normal and diseased muscles, with potential applications to the human condition. This review compares the limb muscles of mice and the indirect flight muscles of flies, with respect to the mechanisms of adult myofiber formation, homeostasis, atrophy, hypertrophy, and the response to muscle degeneration, with some comment on myogenic precursor cells and common gene regulatory pathways. There is a striking similarity between the species for events related to muscle atrophy and hypertrophy, without contribution of any myoblast fusion. Since the flight muscles of adult flies lack a population of reserve myogenic cells (equivalent to satellite cells), this indicates that such cells are not required for maintenance of normal muscle function. However, since satellite cells are essential in postnatal mammals for myogenesis and regeneration in response to myofiber necrosis, the extent to which such regeneration might be possible in flight muscles of adult flies remains unclear. Common cellular and molecular pathways for both species are outlined related to neuromuscular disorders and to age-related loss of skeletal muscle mass and function (sarcopenia). The commonality of events related to skeletal muscles in these disparate species (with vast differences in size, growth duration, longevity, and muscle activities) emphasizes the combined value and power of these experimental animal models.

Estrogen signalling is critical for ovarian differentiation in reptiles with temperature-dependent sex determination (TSD). To elucidate the involvement of estrogen in this process, adrenal-kidney-gonadal (AKG) expression of estrogen receptor (ER alpha) was studied at female-producing temperature (FPT) in the developing embryos of the lizard, Calotes versicolor which exhibits a distinct pattern of TSD. The eggs of this lizard were incubated at 31.5 +/- 0.5 degrees C (100% FPT). The torso of embryos containing adrenal-kidney-gonadal complex (AKG) was collected during different stages of development and subjected to Western blotting and immunohistochemistry analysis. The ER alpha, antibody recognized two protein bands with apparent molecular weight similar to 55 and similar to 45 kDa in the total protein extracts of embryonic AKG complex of C. versicolor. The observed results suggest the occurrence of isoforms of ER alpha. The differential expression of two different protein isoforms may reveal their distinct role in cell proliferation during gonadal differentiation. This is the first report to reveal two isoforms of the ER alpha in a reptile during development. Immunohistochemical studies reveal a weak, but specific, cytoplasmic ER alpha immunostaining exclusively in the AKG during late thermo-sensitive period suggesting the responsiveness of AKG to estrogens before gonadal differentiation at FPT. Further, cytoplasmic as well as nuclear expression of ER alpha in the medulla and in oogonia of the cortex (faint activity) at gonadal differentiation stage suggests that the onset of gonadal estrogen activity coincides with sexual differentiation of gonad. Intensity and pattern of the immunoreactions of ER alpha in the medullary region at FPT suggest endogenous production of estrogen which may act in a paracrine fashion to induce neighboring cells into ovarian differentiation pathway. Astrocytes are fundamental for brain homeostasis and the progression and outcome of many neuropathologies including Alzheimer's disease (AD). In the triple transgenic mouse model of AD (3xTg-AD) generalised hippocampal astroglia atrophy precedes a restricted and specific beta-amyloid (A beta) plaque-related astrogliosis. Astrocytes are critical for CNS glutamatergic transmission being the principal elements of glutamate homeostasis through maintaining its synthesis, uptake and turnover via glutamate-glutamine shuttle. Glutamine synthetase (GS), which is specifically expressed in astrocytes, forms glutamine by an ATP-dependent amination of glutamate. Here, we report changes in GS astrocytic expression in two major cognitive areas of the hippocampus (the dentate gyrus, DG and the CA1) in 3xTg-AD animals aged between 9 and 18 months. We found a significant reduction in Nv (number of cell/mm(3)) of GS immunoreactive (GS-IR) astrocytes starting from 12 months (28.59%) of age in the DG, and sustained at 18 months (31.65%). CA1 decrease of GS-positive astrocytes Nv (33.26%) occurs at 18 months. This Nv reduction of GSIR astrocytes is paralleled by a decrease in overall GS expression (determined by its optical density) that becomes significant at 18 months (21.61% and 19.68% in DG and CA1, respectively). GS-IR Nv changes are directly associated with the presence of A beta deposits showing a decrease of 47.92% as opposed to 23.47% in areas free of A beta. These changes in GS containing astrocytes and GS-immunoreactivity indicate AD-related impairments of glutamate homeostatic system, at the advanced and late stages of the disease, which may affect the efficacy of glutamatergic transmission in the diseased brain that may contribute to the cognitive deficiency.

In the kelp forests of Carmel Bay there are six common rockfishes (Sebastes). Three are pelagic (S. serranoides, S. mystinus, and S. melanops) and two are demersal (S. chrysomelas and S. carnatus). The sixth (S. atrovirens) is generally found a few meters above the sea floor. The pelagic rockfishes which are spatially overlapping have different feeding habits. All rockfishes except S. mystinus utilize juvenile rockfishes as their primary food source during the upwelling season. Throughout the non-upwelling season, most species consume invertebrate prey. The pelagic rockfishes have shorter maxillary bones and longer gill rakers than their demersal congeners, both specializations for taking smaller prey. They also have longer intestines, enabling them to utilize less digestable foods. S. mystinus, which has the longest intestine, may be able to use algae as a food source. Fat reserves are accumulated from July through October, when prey is most abundant. Fat is depleted throughout the rest of the year as food becomes scarce and development of sexual organs takes place. Gonad development occurs from November through February for all species except S. atrovirens.

ENGLISH: Three hundred and twenty-six collections of anchoveta (Cetengraulis mysticetus), an important tuna bait species, taken between April 1951 and April 1960 from seven major baiting areas in the Eastern Tropical Pacific Ocean (Almejas Bay, Guaymas, Ahome Point, Banderas Bay, Gulf of Fonseca, coast of Colombia and Ecuador-Peru) are the basis of this study of age, growth, sexual maturity and spawning. The study of the temporal progression of modal size groups from plots of monthly length-frequency distributions provided estimates of age and rate of growth. The study of sexual maturity and time of spawning was based on gross examination of ovaries, and application of the gonad index.

ENGLISH: Catch and effort statistics from the Japanese longline fishery operating in the eastern Pacific Ocean east of 130°W, from 1964 through 1966, were examined to study the geographic distribution, trends in apparent abundance, sexual maturity, and size composition of the tunas and billfishes. Yellowfin and bigeye tuna are generally most abundant in the equatorial regions of the high seas between about 10°N and 20°S, but west of 95°W. The marlins are more coastal in distribution, usually occurring to the east, and to the north and south of the heavy concentration of tropical tunas. Sailfish tend to be associated with coastal areas also, whereas shortbill spearfish are more frequently captured on the high seas. Swordfish are found most abundantly in the coastal regions off northern Mexico, and off northern Peru and southern Ecuador. The albacore, a temperate-water species of tuna, is most abundant in the high-seas area of the southeastern Pacific, Trends in apparent abundance were measured by the hook-rate (i.e. catch per 100 hooks). Hook-rates for bigeye tuna have decreased from about 3.5 fish per 100 hooks in 1958 to about 1.1 fish per 100 hooks in 1966. During the same period, effort was increased substantially and total catch has decreased since 1963. It does not appear that increased effort will result in sustained increased catches of bigeye. Hook-rates for yellowfin tuna in recent years have decreased to about one third of their initial levels. The surface fishery for yellowfin in the eastern Pacific apparently affects recruitment to the longline fishery. Assuming that present conditions in the surface fishery do not change appreciably, increased effort in the longline fishery probably would not produce sustained increased catches, but might in fact result in reduced catch rates. Unlike the situation for the other tunas of the eastern Pacific, it appears that the albacore fishery east of 130°W is not having a marked effect on their abundance. Although a high degree of variability was observed in the hookrates for striped marlin, no obvious trends are evident. Catches have decreased slightly from 13,500 tons in 1964 to about 11,000 tons in 1966. Heavy fishing for sailfish began in 1964 with a hook-rate of 10.6 fish per 100 hooks; by 1966 it had dropped to 5.8. Catches of this species in the area of major concentration dropped from 329,900 fish in 1965 to 173,600 fish in 1966. This fishery has operated for too short a period of time to enable one to determine its effect on the sustainable yield. Length-frequency measurements and gonad samples from yellowfin and bigeye tunas collected in the eastern Pacific were analyzed to determine sexual maturity and growth characteristics. The results corroborate the findings of earlier investigators. (PDF contains 144 pages.)

A short review of the actual assessments on the northeast atlantic groundfish stocks of cod, saithe, haddock, redfish and Greenland halibut provided by the ICES „Arctic Fisheries Working Group“is given. All these stocks are presently considered outside safe biological limits. For the first time since 5 years the cod stock is in an endangered situation, because the assessment was to optimistic and the TAC therefore set to high. According to the recent assessments necessary changes in the commercial fishery of the year 1998 off the Nowegian coast and in the Barent-Sea area are discussed. Information on distribution and fishery of cod, haddock, saithe and redfish is given. Biological investigations were carried out onboard the German factory trawler FMS „KIEL“ in January/March and September/ Occtober 1998 in order to collect data of the German fishery in that area. Aspects of length and age distributions, and stomach- and gonad investigation are represented.

During this years’ International Bottom Trawl Survey (IBTS) of ICES, a total of 407 half hour tows were made in the North Sea including Skagerrak in January/ February, 1998. Results indicate except for sprat no outstanding incoming yearclasses for cod, haddock, saithe, Norway pout, whiting, herring, and mackerel. Most of the adult cod and saithe investigated did not show normal gonad development for this time of the year. The abundance of skates and sharks was still low. Results of 68 hydrographical stations of R.V. “Walter Herwig III” showed in contrast to four preceeding IBTS-surveys a warming of the northern North Sea of approx. 1 K.

The preliminary results obtained from the study of stomach contents of specimens of Gymnarchus niloticus of a size ranging from 56 cm (66 grams) to 152 cm (12.0 kg.) from August to December, 1982 indicate for these sizes an exclusively piscivorous diet. The predominant prey species in Lake Chad are Tilapia/Sarotherodon , and Clarias . The prey is often cut in two parts before it is swallowed. Small prey may be swallowed whole. The implication of this prey capture method on prey-predator relationship and therefore the impact of the predator is discussed. The breeding season of Gymnarchus niloticus in Lake Chad has been deduced from observation of changes in gonad maturity stages and the results indicate that breeding takes place between August and November. Data are also presented on the fecundity, size of ripe eggs and probable size at maturity

Twenty four matured samples of Bagrus bayad macropterus from the wild (Shiroro Lake, Nigeria) and under captivity, size ranging from 412.69-3300.00 g total body weight, were analysed for sexual maturity, fecundity and egg size. The average fecundity obtained were 53352.59 and 21028.32 eggs for the wild and cultured fish respectively.Positive relationship was observed between fecundity, body size and gonad weight. Fecundity increased as body size increased. A more positive and linear relationship was observed between fecundity and gonad weight than fecundity and total body weight. Egg diameter, length and weight were determined from the egg samples. The mean size range of eggs for cultured fish was 0.74-1.05 mm of diameter; 1.01-1.20 mm of length and 0.25-0.40 mg of weight. Wild samples had mean size range of 0.68-l.09 mm of diameter, 0.85-1.38 mm of length and egg mean weight range was 0.15- 0.40 mg. Sexual maturity is dependent on size (1 kg and above). The egg diameter, egg length and weight bear no relationship with each other. Gonad development study indicated that gonad development was faster under captivity than in wild

Fish samples were collected using seine and trawl nets in Nyanza Gulf, Lake Victoria (Kenya) in order to study the reproductive and gonadal patterns of Oreochromis niloticus; samples were collected from June 1998 to February 1999. The variation in relative condition factor and monthly variation in gonad weight showed spawning took place between June and July. Most ripe fish were recovered between November and February, while a slight drop in GSI for males was noted between November and January. External factors triggering reproductive strategy are now being addressed

The development of the vulva of the nematode Caenorhabditis elegans is induced by a signal from the anchor cell of the somatic gonad. Activity of the gene lin-3 is required for the Vulval Precursor Cells (VPCs) to assume vulval fates. It is shown here that lin-3 encodes the vulval-inducing signal.
lin-3 was molecularly cloned by transposon-tagging and shown to encode a nematode member ofthe Epidermal Growth Factor (EGF) family. Genetic epistasis experiments indicate that lin-3 acts upstream of let-23, which encodes a homologue of the EGF-Receptor.
lin-3 transgenes that contain multiple copies of wild-type lin-3 genomic DNA clones confer a dominant multivulva phenotype in which up to all six of the VPCs assume vulval fates. The properties of these trans genes suggest that lin-3 can act in the anchor cell to induce vulval fates. Ablation of the gonadal precursors, which prevents the development of the AC, strongly reduces the ability of lin-3 transgenes to stimulate vulval development. A lin-3 recorder transgene that retains the ability to stimulate vulval development is expressed specifically in the anchor cell at the time of vulval induction.
Expression of an obligate secreted form of the EGF domain of Lin-S from a heterologous promoter is sufficient to induce vulval fates in the absence of the normal source of the inductive signal. This result suggests that Lin-S may act as a secreted factor, and that Lin-S may be the sole vulval-inducing signal made by the anchor cell.
lin-3 transgenes can cause adjacent VPCs to assume the 1° vulval fate and thus can override the action of the lateral signal mediated by lin-12 that normally prevents adjacent 1° fates. This indicates that the production of Lin-3 by the anchor cell must be limited to allow the VPCs to assume the proper pattern of fates of so 3° 3° 2° 1° 2° 3°.
