Polyhydroxyalkanoates (PHAs) are natural biologically synthesized polymers that have been the subject of much interest in the last decades due to their biodegradability. Thus far, its microbial production is associated with high operational costs, which increases PHA prices and limits its marketability. To address this situation, this thesis’ work proposes the utilization of photosynthetic mixed cultures (PMC) as a new PHA production system that may lead to a reduction in operational costs. In fact, the operational strategies developed in this work led to the selection of PHA accumulating PMCs that, unlike the traditional mixed microbial cultures, do not require aeration, thus permitting savings in this significant operational cost. In particular, the first PHA accumulating PMC tested in this work was selected under non-aerated illuminated conditions in a feast and famine regime, being obtained a consortium of bacteria and algae, where photosynthetic bacteria accumulated PHA during the feast phase and consumed it for growth during the famine phase, using the oxygen produced by algae. In this symbiotic system, a maximum PHA content of 20% cell dry weight (cdw) was reached, proving for the first time, the capacity of a PMC to accumulate PHA. During adaptation to dark/light alternating conditions, the culture decreased its algae content but maintained its viability, achieving a PHA content of 30% cdw. Also, the PMC was found to be able to utilize different volatile fatty acids for PHA production, accumulating up to 20% cdw of a PHA co-polymer composed of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (HV) monomers. Finally, a new selective approach for the enrichment of PMCs in PHA accumulating bacteria was tested. Instead of imposing a feast and famine regime, a permanent feast regime was used, thus selecting a PMC that was capable of simultaneously growing and accumulating PHA, being attained a maximum PHA content of 60% cdw, the highest value reported for a PMC thus far. The results presented in this thesis prospect the utilization of cheap, VFA-rich fermented wastes as substrates for PHA production, which combined with this new photosynthetic technology opens up the possibility for direct sunlight illumination, leading to a more cost-effective and environmentally sustainable PHA production process.

226 methanol and water extracts representing 74 mainly native plant species found in Amazonas State, Brazil, were tested at a standard concentration of 500 μg/mL for lethality towards larvae of the brine shrimp species Artemia franciscana. Several cytotoxic plant species were identified in this work: Aspidosperma marcgravianum, A. nitidum, Croton cajucara, Citrus limetta, Geissospermum argenteum, Minquartia guianensis, Piper aduncum, P. amapense, P. capitarianum, P. tuberculatum and Protium aracouchini. The results were analyzed within the context of the available traditional knowledge and uses for these plants.

Os estudos anatômicos das folhas e caules de Picrolemma sprucei revelaram que as epidermes das folhas são glabras, apresentam células de paredes onduladas e estômatos anomocíticos. O mesofilo é dorsiventral com uma camada de células em paliçada e um parênquima lacunoso. Os feixes vasculares são bicolaterais e apresentam idioblastos escuros de conteúdo tânico. No caule evidencia-se um parênquima cortical desenvolvido, com células de natureza esclerenquimática, isoladas ou reunidas em pequenos grupos, de paredes espessas. Internamente observa-se a região vascular com vasos xilemáticos isolados ou em grupos imersos em tecido fibroso, sendo o parênquima do tipo paratraqueal, vasicêntrico. Na região central encontra-se uma medula desenvolvida. Testes histoquímicos (sudam III e cloreto férrico) realizados na nervura mediana do terço médio do limbo foliar e no pecíolo revelaram respectivamente, a presença de um conteúdo tânico e óleo-resinoso em algumas de suas células.

Ethanol, methanol and water extracts representing mostly native plant species found in the Amazon region were prepared, respectively, by maceration, continuous liquid-solid extraction and infusion, followed by evaporation and freeze-drying. The freeze-dried extracts were tested for lethality toward Aedes aegypti larvae at test concentrations of 500 mg / mL. In general, methanol extracts exhibited the greatest larvicidal activity. The following 7 methanol extracts of (the parts of) the indicated plant species were the most active, resulting in 100% mortality in A. aegypti larvae: Tapura amazonica Poepp. (root), Piper aduncum L. (leaf and root), P. tuberculatum Jacq. (leaf, fruit and branch). and Simaba polyphylla (Cavalcante) W.W. Thomas (branch).

1300 ppm (1.3 g / L), water and ethanol extracts prepared from stems or roots of Picrolemma sprucei Hook. f. were lethal (85-90 % mortality) in vitro to Haemonchus contortus (Barber Pole Worm) larvae, a gastrointestinal nematode parasite found in domestic and wild ruminants. Neosergeolide and isobrucein B were isolated in 0.0083 and 0.0070 % yield from dry, ground P. sprucei stems (0.89 kg). Neosergeolide, isobrucein B and the anthelmintic drug standard levamisole all caused comparable mortality rates (68-77 %) in vitro to H. contortus at similar concentrations (81-86 ppm). The anthelmintic activity of P. sprucei infusions (teas), alcohol extracts, and neosergeolide and isobrucein B, has therefore been demonstrated for the first time.

Maytenus guyanensis Klotzch. is an Amazonian medicinal tree species known in Brazil by the common name chichuá and in Peru and Colombia by the name chuchuhuasi. It is used in traditional medicine as stimulant, tonic, and muscle relaxant, for the relief of arthritis, rheumatism, hemorrhoids, swollen kidney, skin eruptions, and skin cancer prevention, among others. Initially, different extraction solvents and methods were applied to dried, ground bark which made possible the preparation of extracts having both significant lethality to brine shrimp larvae (Artemia franciscana Leach) as well as antioxidant activity in vitro based on tests involving reactions with 2,2,-diphenyl-1-picrylhydrazyl (DPPH). Analysis of fractions from serial extractions with solvents of increasing polarity supports the notion that antioxidant activity is associated with compounds of intermediate polarity and cytotoxicity is associated with compounds of low to intermediate polarity. Variation of extraction time and conditions revealed that hot, continuous ethanol extraction provided good yields of bark extract in several hours. Hot extraction also provided ethanol extracts having greater lethality to brine shrimp and antioxidant activity (compared to the flavonoid rutin in semi-quantitative methods based on DPPH) than extracts obtained from maceration at room temperature. Freeze-dried ethanol extracts were prepared by: 1) maceration at room temperature and 2) hot extraction (eight hours) on several hundred gram scales and the latter extract was shown to have partial screening effects on UVB light. In this work, cytotoxic, antioxidant and potential sun-screening activity are shown for the first time in M. guyanensis.

Tabebuia incana A.H. Gentry (Bignoniaceae) is a tree from the Brazilian Amazon having medicinal uses and is one several Tabebuia spp. known as pau d'arco or palo de arco in this region. Fractionation of the bark ethanolic extract afforded a mixture of 5 and 8-hydroxy-2-(1-hydroxyethyl)naphtho[2,3-b]furan-4,9-diones (1 and 2, respectively) identified on the basis of nuclear magnetic resonance (NMR), infrared (IR) and mass (MS) spectra, whose in vitro antimalarial and antitumor activity have been shown previously. This is the first study on T. incana bark, and 2 are described in this species for the first time. Also, high performance liquid chromatography (HPLC) analysis of T. incana bark tea revealed the presence of the 1 + 2 mixture peak corresponding to a concentration in the range 10-6-10-5 M. The chromatograms of teas prepared from commercial pau d' arco and T. incana bark were also studied and the presence of the 1 + 2 peak has potential for quality control of commercial plant materials.

Quassinoids neosergeolide and isobrucein B, obtained from Picrolemma sprucei, have proven in vitro antitumor, antimalarial, anthelminthic, cytotoxic, insecticide and leishmanicidal activities. There is interest in the in vivo pharmacological study of these natural compounds and their semi-synthetic derivatives, however, the quantities obtained in previous extraction processes have been shown to be a limiting factor for continuation of these studies. Herein, we describe a method for obtaining grams of these quassinoids whose purification relies only on recrystallization.

In the present study, different aerial parts from twelve Amazonian plant species found in the National Institute for Amazon Research's (INPA's) Adolpho Ducke Forest Reserve (in Manaus, Amazonas, Brazil) were collected. Separate portions of dried, ground plant materials were extracted with water (by infusion), methanol and chloroform (by continuous liquid-solid extraction) and solvents were removed first by rotary evaporation, and finally by freeze-drying which yielded a total of seventy-one freeze-dried extracts for evaluation. These extracts were evaluated initially at concentrations of 500 and 100 µg/mL for in vitro hemolytic activity and in vitro inhibition of platelet aggregation in human blood, respectively. Sixteen extracts (23 % of all extracts tested, 42 % of all plant species), representing the following plants: Chaunochiton kappleri (Olacaceae), Diclinanona calycina (Annonaceae), Paypayrola grandiflora (Violaceae), Pleurisanthes parviflora (Icacinaceae), Sarcaulus brasiliensis (Sapotaceae), exhibited significant inhibitory activity towards human platelet aggregation. A group of extracts with antiplatelet aggregation activity having no in vitro hemolytic activity has therefore been identified. Three extracts (4 %), all derived from Elaeoluma nuda (Sapotaceae), exhibited hemolytic activity. None of the plant species in this study has known use in traditional medicine. So, these data serve as a baseline or minimum of antiplatelet and hemolytic activities (and potential usefulness) of non-medicinal plants from the Amazon forest. Finally, in general, these are the first data on hemolytic and inhibitory activity on platelet aggregation for the genera which these plant species represent.

Objective: The aim of this study is to improve the understanding of self-changes after an intervention for depression focused on implicative dilemmas, a type of cognitive conflict related to identity. As recent research has highlighted the relevance of identity-related dilemmas in clients with depression, we sought to assess the way in which clients resolve such inner conflicts after a tailored dilemma-focused intervention and how this is reflected in the clients’ self-narratives. Method: We used three instruments to observe differences between good (n = 5) and poor (n = 5) outcome cases: (i) the Repertory Grid Technique to track the resolution of dilemmas, (ii) the Change Interview to compile clients’ accounts of changes at posttreatment, and (iii) the Innovative Moments Coding System to examine the emergence of clients’ novelties at the Change Interview. Results: Groups did not differ in terms of the number and relevance of client-identified significantly helpful events. However, between-group differences were found for the resolution of dilemmas and for the proportion of high-level innovative moment (IM) types. Furthermore, a greater self-narrative reconstruction was associated with higher levels of symptom improvement. Conclusions: Good outcome cases seem to be associated with the resolution of conflicts and high-level IMs.

El extenso uso de pavimentos impermeables trae consigo, sobre todo en áreas de un importante desarrollo urbano, considerables problemas en la evacuación de las aguas lluvias. El uso indiscriminado de estas estructuras en áreas urbanas, disminuye notoriamente la capacidad de recarga natural de agua en los terrenos, e incrementa de forma considerable el volumen y el caudal del escurrimiento superficial, aumentando el riesgo de provocar inundaciones. Además, cuando el agua de las lluvias escurre, arrastra consigo materiales sólidos y contaminantes depositados en calles y estacionamientos. Una de las alternativas posibles para la remediación de esta problemática es el uso de pavimentos porosos. Lamentablemente, en nuestro país no se ha extendido la aplicación de estas tecnologías que conllevan importantes beneficios económicos y ambientales, entre los cuales pueden citarse la mejora en la calidad de los efluentes pluviales, la flora arbórea urbana más longeva y mejor desarrollada, la reducción del efecto de “isla térmica” en las grandes ciudades, la reducción notable de la contaminación acústica, el aumento de la seguridad vial, y reducción de costos, considerando integralmente el sistema vial y el sistema de drenaje pluvial. Los beneficios ambientales descriptos justifican plenamente su amplio uso en los países desarrollados. Como objetivo principal se plantea caracterizar experimentalmente el comportamiento hidrológico-ambiental de pavimentos porosos que puedan ser fabricados con materiales y tecnología local, e inferir los posibles beneficios que su uso pudiera acarrear en el medio ambiente. Como metodología se propone el estudio teórico-experimental, desde la tecnología de los materiales, en el diseño y conocimiento de las propiedades en estado fresco y endurecido y en la durabilidad de hormigones porosos elaborados con materiales locales. Para la caracterización hidrológica se fabricarán parcelas de pavimento poroso, las cuales serán ensayadas mediante el uso de un microsimulador de lluvia, midiendo la intensidad de lluvia generada y el escurrimiento superficial resultante. Estos experimentos podrán repetirse para distintas intensidades de lluvia generada, la cual puede graduarse en el rango de 65 a 120 mm/h. A partir de los datos recopilados, podrán ajustarse parámetros hidrológicos que permitan describir el comportamiento de estas superficies, en comparación con la situación natural y la actual (pavimentos impermeables). Se ajustarán modelos de infiltración que permitan extrapolar en forma hipotética la situación a escalas mayores, y cuantificar preliminarmente los beneficios hidrológico-ambientales que produciría este tipo de tratamiento superficial en el ambiente urbano. Como resultados de este proyecto se espera obtener: una verificación de las posibilidades de los materiales locales, brindando bases evolucionadas para su empleo en la construcción; una metodología para caracterizar en forma preliminar la capacidad de infiltración de muestras reducidas de pavimento poroso; un sensor electrónico para automatizar las mediciones del microsimulador de lluvia; una metodología para caracterizar experimentalmente la capacidad de infiltración de los pavimentos porosos propuestos, basada en el uso del simulador de lluvia disponible en el Laboratorio de Hidráulica; un conjunto de resultados experimentales que posibiliten analizar el comportamiento hidrológico-ambiental de estas superficies; un conjunto de parámetros que permitan cuantificar la respuesta hidrológica de estos pavimentos y una estimación preliminar de los hipotéticos beneficios asociados al uso de estas tecnologías. Se considera que el Proyecto puede servir como puntapié inicial para el análisis de viabilidad, a nivel local, del uso de tecnologías de pavimentación novedosas en el país, pero con varias décadas de experiencia a nivel internacional. Los beneficios medioambientales asociados a los pavimentos porosos, justifican plenamente su investigación a nivel local.

Entre los tumores frecuentes en los caninos, se encuentra el denominado Tumor Venéreo Transmisible (TVT). El origen celular exacto del TVT no está claro, aunque estudios inmunohistoquímicos muestran las características de compatibilidad con células histiocíticas. En nuestra región se diagnostican con frecuencia aunque no se tienen datos precisos de su incidencia. El diagnóstico precoz es la mejor herramienta utilizada tanto en medicina veterinaria como humana para el control de tumores. El tumor puede ser muy agresivo localmente, rara vez metastiza. El manejo terapéutico no ha sido sencillo. Se han probado distintos tratamientos como cirugía, radioterapia, bioterapia y quimioterapia- La quimioterapia ha demostrado ser el tratamiento mas efectivo y la droga mas utilizada es el sulfato de vincristina, pero el tiempo del tratamiento hasta la remisión completa varía ampliamente entre individuos. Si bien existen estadificaciones clínicas y diagnósticos citológicos e histológicos que permiten establecer el grado de malignidad, los mismos tienen componentes subjetivos por su naturaleza cualitativa. Los marcadores tumorales se han utilizado especialmente en el establecimiento de un pronóstico acertado. Entre estos están las Regiones Organizadoras Nucleolares (NORs), que pueden colorearse con tinciones argénticas (AgNOR). dichas regiones corresponden a segmentos de cromosomas que contienen genes ribosomales. Por otro lado, existen marcadores de proliferación celular como el anticuerpo monoclonal Ki 67, el cual detecta un antígeno nuclear que se expresa en las células que entran al ciclo celular (fases G1, S, G2 y mitosis), proporcionando una medida directa de la fracción de crecimiento del tejido. El uso de estos marcadores fue eficiente en patología humana, mientras en veterinaria existen escasos reportes La mayoría de los marcadores se han usado sobre muestras histológicas, existiendo escasa información de su uso en muestras citológicas. Conocer el diagnóstico y el pronóstico del tumor relacionándolo con el tratamiento instaurado permitiría predecir las expectativas y calidad de vida del paciente. El contar en Medicina Veterinaria con una herramienta cuantitativa como son los marcadores tumorales que soporte las observaciones subjetivas que aporta el diagnóstico citomorfológico, podría contribuir a establecer de manera confiable el pronóstico del paciente con TVT, así como también definir el modelo terapéutico. Por esto sería importante la búsqueda de técnicas rápidas y seguras que permitan establecer pronósticos cada vez más precisos.
