The inability to maintain genomic stability and control proliferation are hallmarks of many cancers, which become exacerbated in the presence of unrepaired DNA damage. Such genotoxic stresses trigger the p53 tumor suppressor network to activate transient cell cycle arrest allowing for DNA repair; if the damage is excessive or irreparable, apoptosis or cellular senescence is triggered. One of the major DNA repair pathway that mends DNA double strand breaks is non-homologous end joining (NHEJ). Abrogating the NHEJ pathway leads to an accumulation of DNA damage in the lymphoid system that triggers p53-mediated apoptosis; complete deletion of p53 in this system leads to aggressive lymphomagenesis. Therefore, to study the effect of p53-dependent cell cycle arrest, we utilized a hypomorphic, separation-of-function mutant, p53p/p, which completely abrogates apoptosis yet retains partial cell cycle arrest ability. We crossed DNA ligase IV deficiency, a downstream ligase crucial in mending breaks during NHEJ, into the p53p/p background (Lig4-/-p53p/p). The accumulation of DNA damage activated the p53/p21 axis to trigger cellular senescence in developing lymphoid cells, which absolutely suppressed tumorigenesis. Interestingly, these mice progressively succumb to severe diabetes. Mechanistic analysis revealed that spontaneous DNA damage accumulated in the pancreatic b-cells, a unique subset of endocrine cells solely responsible for insulin production to regulate glucose homeostasis. The genesis of adult b-cells predominantly occurs through self-replication, therefore modulating cellular proliferation is an essential component for renewal. The progressive accumulation of DNA damage, caused by Lig4-/-, activated p53/p21-dependent cellular senescence in mutant pancreatic b-cells that lead to islet involution. Insulin levels subsequently decreased, deregulating glucose homeostasis driving overt diabetes. Our Lig4-/-p53p/p model aptly depicts the dichotomous role of cellular senescence—in the lymphoid system prevents tumorigenesis yet in the endocrine system leads to the decrease of insulin-producing cells causing diabetes. To further delineate the function of NHEJ in pancreatic b-cells, we analyzed mice deficient in another component of the NHEJ pathway, Ku70. Although most notable for its role in DNA damage recognition and repair within the NHEJ pathway, Ku70 has NHEJ-independent functions in telomere maintenance, apoptosis, and transcriptional regulation/repression. To our surprise, Ku70-/-p53p/p mutant mice displayed a stark increase in b-cell proliferation, resulting in islet expansion, heightened insulin levels and hypoglycemia. Augmented b-cell proliferation was accompanied with the stabilization of the canonical Wnt pathway, responsible for this phenotype. Interestingly, the progressive onset of cellular senescence prevented islet tumorigenesis. This study highlights Ku70 as an important modulator in not only maintaining genomic stability through NHEJ-dependent functions, but also reveals a novel NHEJ-independent function through regulation of pancreatic b-cell proliferation. Taken in aggregate, these studies underscore the importance for NHEJ to maintain genomic stability in b-cells as well as introduces a novel regulator for pancreatic b-cell proliferation.

Multiple Endocrine Neoplasia type 1 (MEN1) is a hereditary cancer syndrome characterized by tumors of the endocrine system. Tumors most commonly develop in the parathyroid glands, pituitary gland, and the gastro-entero pancreatic tract. MEN1 is a highly penetrant condition and age of onset is variable. Most patients are diagnosed in early adulthood; however, rare cases of MEN1 present in early childhood. Expert consensus opinion is that predictive genetic testing should be offered at age 5 years, however there are no evidence-based studies that clearly establish that predictive genetic testing at this age would be beneficial since most symptoms do not present until later in life. This study was designed to explore attitudes about the most appropriate age for predictive genetic testing from individuals at risk of having a child with MEN1. Participants who had an MEN1 mutation were invited to complete a survey and were asked to invite their spouses to participate as well. The survey included several validated measures designed to assess participants’ attitudes about predictive testing in minors. Fifty-eight affected participants and twenty-two spouses/partners completed the survey. Most participants felt that MEN1 genetic testing was appropriate in healthy minors. Younger age and increased knowledge of MEN1 genetics and inheritance predicted genetic testing at a younger age. Additionally, participants who saw more positive than negative general outcomes from genetic testing were more likely to favor genetic testing at younger ages. Overall, participants felt genetic testing should be offered at a younger age than most adult onset conditions and most felt the appropriate time for testing was when a child could understand and participate in the testing process. Psychological concerns seemed to be the primary focus of participants who favored later ages for genetic testing, while medical benefits were more commonly cited for younger age. This exploratory study has implications for counseling patients whose children are at risk of developing MEN1 and illustrates issues that are important to patients and their spouses when considering testing in children.

ABSTRACT The aim of this thesis is to contribute to the development and use of solar energy as an alternative for producing hot water in the agribusiness sector. Hot water supply is a key issue for a great many agribusinesses and agricultural holdings. Both hot water demand times and required tank water temperature (rTWT) are highly variable, where the demand profile tends to differ from domestic use. Further research is needed on how differences in rTWT influence the performance and feasibility of these systems. The main objective of this thesis is to characterize the performance and test the feasibility of an evacuated tube collector (ETC) solar water heating (SWH) system providing water at a higher temperature than is usual for such systems. The aim is to determine what influence the rTWT has on the system’s energy efficiency, quantify the volume of hot water that the system is capable of supplying at the respective rTWT and establish whether SWH is feasible as a booster supply system for the different analysed rTWTs. To do this, a prototype water heating system has been designed, installed and commissioned and its performance monitored at different rTWTs under real operating conditions. The quantitative results show that a higher rTWT results in a lower energy supply to the tank, where the differences may be greater than 1000 Wh m-2 d-1 from 40 ºC to 80 ºC for insolation values of around 8000 Wh m-2 d-1 (system efficiency ranges from 73% to 56%). The drop in supply is due to lower collector efficiency and greater heat losses from the pipe system. As regards water supplied at the rTWT, the insolation required for the first withdrawal of water to take place is greater at higher rTWTs, where the time between withdrawals increases and the number of withdrawals decreases throughout the day. As rTWT increases, the volume of water supplied at the rTWT decreases due to factors such as lower collector efficiency, pipe system heat losses, energy stored in the water at below the rTWT and more energy being extracted from the system by water heating. For a rTWT of 80 ºC, much of the energy is stored in the tank at below the rTWT at the end of the day. A booster supply system would be required to take advantage of this energy, as overnight tank heat losses may significantly reduce the usable energy available on the following day. It is often not feasible to use the solar system as a single supply system, especially at high rTWTs, as, unlike the supply from the solar heating system which does not produce hot water on many days of the year because insolation is below the required minimum, hot water demand is not seasonal. On the other hand, investment in a solar system as a booster system to meet part of a plant’s heat energy demand is highly recommended. The solar system’s estimated annual usable energy ranges from 1322 kWh m-2 to 1084 kWh m-2. Cost efficiency would be greatest if there were an existing electric boiler, where the payback period would be just a few years —from 5.7 years at 40 ºC to 7.2 years at 80 ºC—. Cost efficiency is also high if there is an existing diesel boiler with payback periods of under 10 years. In a fictitious industry with a demand of 100 kWh day-1 and an existing diesel boiler, the investment in the solar plant would be highly recommended at any rTWT, with a net present value similar to investment costs —12000 € at 80 ºC and 15000 € at 40 ºC— and a payback period of 10 years. The results of this study are potentially very useful for determining the feasibility of using similar systems for meeting the hot water demand of agribusinesses and arable and livestock farms or for other applications demanding water at temperatures not typical of domestic demand (60ºC). Performance and cost efficiency will be determined by the regional insolation, the required water temperature and the demand curve of the specific processes in each case.

In the state of Veracruz, to the south of Mexico, there are located companies dedicated to the production of ethanol from molasses of cane sugar. The smallest, have a average production of 20,000 L ethanol/day. The effluent of production of ethanol include water of condensers, water originated from the cleanliness of tanks of fermentation and vinasses, the above mentioned are more effluent pollutants in the distilleries, for the poor organic matter degradability. The vinasses are generated in high volumes, producing from 12 to 15 L of vinasses per every liter of distilled ethanol. These effluent are characterized by its high temperature, pH acid and a high concentration of DQO as well as high concentration of TS. The determination of the anaerobic degradability of a waste water, it allows to estimate the fraction of DQO that can be transformed potentially into methane and the recalcitrant DQO that stays in the effluent. For the development of degradability test, it is important to consider factors related to the composition of the water to be treated, composition of the sludge and the conditions under which the test is carried out. The anaerobic digestion of industrial wastes water is used commonly in the whole world, it offers significant advantages for the treatment of effluent highly loaded. The anaerobic treatment of industrial wastes water include technologies with biofilms, this anaerobic treatment whit biofilms systems, is a well-established technology for treatment of industrial effluents. The Anaerobic Inverse Fluidized Bed Reactor (IFBR) has been developed to provide biological treatment of high strength organic wastewater for their large specific surface and their low energy requirements for fluidization. In this work, there is carried out the analysis of a process of production of ethanol, identifying the effluent ones that are generated in the process. One determined that the effluent end is composed principally by the vinasses originated from the process of distillation. In the characterization of the vinasses originated from the process of production of ethanol from cane sugar molasses, there were average values of DQO of 193.35 gDQO/L, average values of solid of 109.78 gST/L and pH of 4.64. In this investigation there was carried out a anaerobic degradability test of the vinasses generated in the production of ethanol. In the characterization of the sludge used in the essay, the specific methanogenic activity (SMA) was 0.14 gDQO/gSSV.d. The average removal of DQO of the vinasses was 62.7 %, k equal to 0.031 h-1 was obtained one and a rate of removal substrate of 1.26 gDQO/d. The methane yield was 0.19 LCH4/gDQO removed and the anaerobic biodegradability was a 54.1 %. This study describes the performance of IFBR with Extendospher®, for the treatment of vinasses. The start-up was made in batch, increasing gradually the Organic Load Rate (OLR): 0.5, 1.0, 3.3, 6.8 and 10.4 g COD/L.d. Different Hydraulic Retention Times (HRT) were evaluated: 10, 5 and 1 days. During the operation in batch, the COD removal obtained was of 81 %, and for OLR of 0.5, 1.0, 3.3, 6.8 g COD/L.d the removal obtained was 58, 67, 59 and 50 % respectively. For a maximum OLR of 10.4 g COD/L.d, the COD removal was 38 %, and the system presented instability and decrease of the yield methane. The methane production initiated after 110 days of the start-up of the IFBR, to organic load rate of 1.0 g COD/L.d. The system reached values in the methane yield from 0.15 up to 0.34 LCH4/g CODremoved, for the different organic load rates. During the operation to a constant OLR of 6.4 g COD/L.d, and a HRT of 1 day, the Anaerobic Inverse Fluidized Bed Reactor reached a maximum efficiency of removal of 52 %.
