O objetivo deste trabalho foi o de estimar o consumo total e o diário de matéria seca do pasto, de vacas mestiças Holandês - Zebu, em piquetes de capim-elefante (Pennisetum purpureum Schum.). Três grupos de 24 vacas foram usados em piquetes (4,5 vacas/ha), pastejados por três dias e submetidos a diferentes períodos de descanso durante dois anos. Os tratamentos consistiram de descansos de 30 dias sem concentrado e 30, 37,5 e 45 dias com a suplementação de 2 kg de concentrado (20,6% de proteína bruta). de julho a outubro, as vacas receberam, como suplementação, cana-de-açúcar mais 1% de uréia. O consumo total de matéria seca foi estimado a partir da digestibilidade in vitro da matéria seca da extrusa e da produção fecal obtida com óxido crômico. Independentemente do tratamento, o consumo total foi 2,7; 2,9 e 2.9±0,03%, e o consumo de matéria seca do pasto foi de 1,9; 2,1 e 2,1±0,03% do peso vivo (p<0,05), respectivamente, no primeiro, segundo e terceiro dia de ocupação do piquete. Somente no verão, o consumo do pasto foi semelhante nos três dias de pastejo. A mistura cana-de-açúcar e uréia substituiu o pasto, principalmente no primeiro dia de pastejo, ocasião em que o consumo do pasto era mais baixo.

Determinou-se a prevalência dos genes de virulência expressando fimbrias, produção de hemolisina, colicina e aerobactina em cepas de Escherichia coli obtidas do trato genital de vacas saudáveis que não apresentam sinais clínicos indicativos de infecção. A presença dos genes responsáveis pela expressão de fimbrias (pap, sfa, afa) foi avaliada através de reação em cadeia da polimerase utilizando primers especificos para cada um dos genes, nenhum deles foi detectado em qualquer uma das cepas isoladas. A prevalência dos fatores de virulência foi de 90,4%, 69,8%, 28,5% para colicina, hemolisina e aerobactina, respectivamente. A análise da patogenicidade das cepas do trato genital pode contribuir para o entendimento do comportamento das cepas de E. coli.

Dez vacas multíparas, secas, foram distribuídas aleatoriamente em dois grupos de cinco animais cada. Nos dias 8 a 12 do diestro, o primeiro grupo recebeu 100 ml de anti-soro contra líquido folicular livre de esteróides (anti-LFb) produzido em ovelhas ovariectomizadas. O segundo grupo (controle) recebeu 100 ml de soro de ovelhas não-imunizadas. Seis horas após a aplicação, os dois grupos foram superovulados com FSH (18 NIH-FSH-S1 unidades) e LH (0,29 NIH-LH-S1 unidades) administrados em quantidades decrescentes durante quatro dias. Na manhã do terceiro dia, foi administrada uma dose luteolítica de cloprostenol. Duas inseminações foram realizadas 48 e 60 horas após. Os embriões foram recuperados pelo método cervical 7 dias após a primeira inseminação. Amostras de sangue foram coletadas durante todo o período experimental para determinar, por radioimunoensaio, as concentrações plasmáticas de FSH, LH e progesterona. Todas as vacas do grupo imunizado e 3 do grupo controle apresentaram mais de 2 CL. Não existiu diferença significativa (P>0,05) na taxa de ovulação entre os grupos imunizado e controle (14,4 e 9,9, respectivamente). O número de embriões recuperado não foi significativamente diferente (P>0,05) entre os grupos, embora o grupo imunizado tenha apresentado maior número de embriões transferíveis (3,4 ± 1,0 versus 0,8 ± 0,4, P<0,05). As concentrações de gonadotrofinas plasmáticas não foram correlacionadas com a taxa de ovulação ou com o número de embriões recuperados. As concentrações de progesterona plasmática foram positivamente correlacionadas (r = 0,88, P<0,01) com a taxa de ovulação. Os resultados sugerem que o anti-LFb, aplicado antes da superovulação, não reduz a variabilidade da resposta ovariana.

Foram estudados os efeitos da temperatura cutânea (Ts) sobre a taxa de termólise por evaporação cutânea (Es) de vacas Holandesas cronicamente expostas ao sol, considerando a pigmentação do pelame. Dezesseis vacas puras de origem foram medidas quanto à evaporação e à temperatura cutâneas às 13 h, após 6 horas de exposição ao sol, no mesmo local (flanco, pescoço e glúteo) e considerando separadamente as malhas negras e as brancas. A evaporação cutânea foi medida por meio de cápsula ventilada. Nas áreas negras a taxa de sudação (138,9 ± 8,5 g.m-2.h-1), a taxa de termólise por evaporação cutânea (93,3 ± 5,7 W.m-2) e a temperatura da superfície cutânea (33,1 ± 0,2°C) foram maiores que nas áreas brancas (109,5 ± 9,7 g.m-2.h-1, 73,6 ± 6,5 W.m-2 e 32,6 ± 0,2°C, respectivamente). Há uma relação exponencial entre evaporação e temperatura cutâneas, que pode ser representada pela equação Es = 31,5+3,67 exp{(Ts-27,9)/2,19115}, com coeficiente de determinação r²=0,68. A taxa de termólise por evaporação cutânea permanece quase constante (cerca de 48 W.m-2) até que a temperatura cutânea atinge aproximadamente 31°C.

Milk serum proteins such as alpha-lactalbumin (ALA) and beta-lactoglobulin (BLG) present biochemical polymorphism which is under the control of codominant autosomal alleles. In the present report, we propose modifications of traditional electrophoretic techniques such as increasing the running gel concentration from 5 to 10% and the addition of 5 M urea to the stacking gel, which permitted the detection of two variants (A and B) at the ALA and BLG loci. About 8 mul of milk serum (6 mg/ml protein) and 10 pl of total fresh milk were applied. Bovine serum albumin (BSA) and immunolactoglobulins (ILG) could also be discriminated. Total fresh milk was as useful as the purified serum milk proteins for the discrimination of ALA and BLG serum milk protein polymorphism by alkaline vertical slab polyacrylamide gel electrophoresis. However, BSA and ILG ran with caseins, which prevented their characterization in this system.

This paper deals with the effects of hair coat characteristics on the sweating rate of Brazilian Braford cows and estimation of heritabilities and genetic correlations of these traits. Data (n=1607) on hair length, coat thickness, hair diameter, number of hairs per unit area, coat reflectance and sweating rate were recorded from heifers and cows of a commercial herd managed on range under extensive system. The data were analyzed considering the following effects on the model for hair coat traits: classes of sires and contemporary groups; linear effects of month and genotype; linear and quadratic effects of age. The effect of sire was important (P<0.05) for all hair coat traits, except for number of hairs; contemporary groups affected (P<0.05) all hair coat traits; the effect of sampling month was important (P<0.05) for hair length and reflectance; genotype affected (P<0.05) hair length, diameter and coat reflectance; the quadratic effect of age was important (P<0.05) only for coat reflectance. Two models were used to analyze the sweating rate. The first model considered the following fixed effects: classes of contemporary groups and sires; linear effect of genotype, coat thickness, hair length, hair diameter, number of hairs, coat reflectance; linear and quadratic effects of time of day, age, air temperature, partial vapour pressure and radiant heat load. The second model used for the sweating rate considered the same fixed effects for the first model, except that the hair coat characteristics were adjusted for important effects used in the models to analyze hair coat traits. All meteorological factors and contemporary groups were important (P<0.05) on variation of sweating rate in both models. The Restricted Maximum. Likelihood (REML) method was used to estimate variance and covariance components under the sire model. Results included heritability estimates in narrow (h(2)) and broad (H) sense for single-trait analyzes: hair thickness (h(2)=0.16; H-2=0.26); hair length (h(2)=0.18; H-2=0.39); number of hairs (h(2)=0.08 +/- 0.07; H-2=0.08 +/- 0.07); hair diameter (h(2)=0.12 +/- 0.07; H-2=0.12 +/- 0.07); coat reflectance (h(2)=0.30; H-2=0.42); and sweating rate (h(2)=0.10 +/- 0.07; H-2=0.10 +/- 0.07). In general, the genetic correlations between the adaptive traits were favorable as for the direction to select for adaptation in tropical environment; however, they presented high standard errors. The results of this study imply that hair coat characteristics and sweating ability are important for the adaptability to heat stress and they must be better studied and further considered for selection for genetic progress of adaptation in tropical environment. Dry matter intake (DMI) of coast-cross grazing by crossbred Holstein-Zebu and Zebu lactating cows was calculated using in vitro dry matter digestibility from extrusa (four esophageal fistulated cows) and fecal output estimate with mordent chromium. Pasture was rotationally grazed with three days grazing period and 27 days testing period, adopting a stocking rate of 1.6 and 3.2 cows/ha, during the dry and rainy season respectively. Voluntary DMI was estimated from degradation characteristics using different equations. Predicted coast-cross DMI varied with models. The prediction of tropical forages dry matter intake from equations based in ruminal degradation parameters needs farther investigation before being employed in practice.

Experiments were performed to determine the mechanism by which recombinant bovine interferon-alpha(I)1 (rbIFN-alpha) causes an acute reduction in plasma concentrations of progesterone. In experiment 1, administration of a prostaglandin synthesis inhibitor blocked rbIFN-alpha-induced hyperthermia but did not prevent the decline in plasma concentrations of progesterone. The decline in progesterone concentrations caused by rbIFN-alpha was, therefore, not a direct consequence of the associated hyperthermia or of pathways mediated through prostaglandin synthesis. It is also unlikely that rbIFN-alpha acts to increase the clearance of progesterone since injection of rbIFN-alpha did not decrease plasma concentrations of progesterone in ovariectomized cows given an intravaginal implant of progesterone (experiment 2). In experiment 3, rbIFN-alpha did not affect basal and LH-induced release of progesterone from cultured luteal slices, indicating that rbIFN-alpha is unlikely to affect luteal function directly. Injection of rbIFN-alpha did, however, cause a decrease in plasma concentrations of LH in ovariectomized cows (experiment 4) that coincided temporally with the decrease in progesterone concentrations seen in cows having a functional corpus luteum. The present results strongly suggest that rbIFN-alpha acts to reduce secretion of progesterone by interfering with pituitary support for luteal synthesis of progesterone. The finding that rbIFN-alpha can inhibit LH secretion implies that interferon-alpha molecules should be considered among the cytokines that can regulate hypothalamic or pituitary function.

The objectives of the current study were to assess the feasibility of using stayability traits to improve fertility of Nellore cows and to examine the genetic relationship among the stayabilities at different ages. Stayability was defined as whether a cow calved every year up to the age of 5 (Stay5), 6 (Stay6), or 7 (Stay7) yr of age or more, given that she was provided the opportunity to breed. Data were analyzed based on a maximum a posteriori probit threshold model to predict breeding values on the liability scale, whereas the Gibbs sampler was used to estimate variance components. The EBV were obtained using all animals included in the pedigree or bulls with at least 10 daughters with stayability observations, and average genetic trends were obtained in the liability and transformed to the probability scale. Additional analyses were performed to study the genetic relationship among stayability traits, which were compared by contrasting results in terms of EBV and the average genetic superiority as a function of the selected proportion of sires. Heritability estimates and SD were 0.25 +/- 0.02, 0.22 +/- 0.03, and 0.28 +/- 0.03 for Stay5, Stay6, and Stay7, respectively. Average genetic trends, by year, were 0.51 +/- 0.34, and 0.38% for Stay5, Stay6, and Stay7, respectively. Estimates of EBV SD, in the probability scale, for all animals included in the pedigree and for bulls with at least 10 daughters with stayability observations were 7.98 and 12.95, 6.93 and 11.38, and 8.24 and 14.30% for Stay5, Stay6, and Stay7, respectively. A reduction in the average genetic superiorities in Stay7 would be expected if the selection were based on Stay5 or Stay6. Nonetheless, the reduction in EPD, depending on selection intensity, is on average 0.74 and 1.55%, respectively. Regressions of the sires' EBV for Stay5 and Stay6 on the sires' EBV for Stay7 confirmed these results. The heritability and genetic trend estimates for all stayability traits indicate that it is possible to improve fertility with selection based on a threshold analysis of stayability. The SD of EBV for stayability traits show that there is adequate genetic variability among animals to justify inclusion of stayability as a selection criterion. The potential linear relationship among stayability traits indicates that selection for improved female traits would be more effective by having predictions on the Stay5 trait.

Thirteen cows, Bos indicus, of the Nellore breed were superovulated with 22 mg of follicle stimulating hormone (FSH) administered by intramuscular route during four consecutive days (D10, D11, D12 and D13), starting on the 10th day of the estrous cycle (day 0 = estrus). Prostaglandin (PGF2alpha, 1.0 mg, im) was administered on D12, 48 h after the first FSH injection, for the induction of estrus on D14, when artificial insemination was performed. Seven days later (D21 of the cycle), embryos were collected, and evaluated, and the ovarian response was estimated on the basis of number of corpora lutea determined by rectal palpation. Blood samples were obtained for the determination of plasma 17-beta estradiol on D10, D11, D12, D13, D14 and D21 and plasma progesterone on D14 by RIA. The donors were divided into two groups according to progesterone levels on D14, the day of the induced estrus (GI: P4 less-than-or-equal-to 1.00 ng/ml, N = 5 and GII: P4 > 1.00 ng/ml, N = 8). A linear positive correlation was observed between plasma 17-beta estradiol concentration on the day of estrus and viable embryo number. We conclude that plasma 17-beta estradiol and progesterone concentrations on the day of estrus can be used to predict the viability of embryos recovered from Nellore cows superovulated with FSH.
