Breeding parameters of Great Cormorants (PkaZac/iOCOfiCLX CCUibo dCUtbo) and Double-crested Cormorants (P. CLU/uXuA CMJhLtllb) were examined at two mixed species colonies at Cape Tryon and Durell Point, Prince Edward Island from 1976 to 1978. Differential access to nests at the two colony sites resulted in more complete demographic data for P. CCUibo than for P. CLUJiituA. In 1911j P. CCtfibo was present at both colonies by 21 March, whereas P. auAAJtuA did not return until 1 April and 16 April at Cape Tryon and Durell Point, respectively. Differences in the arrival chronology by individuals of each species and differences in the time of nest site occupation according to age, are suggested as factors influencing the nest site distribution of P. CXUtbo and P. aiVtituA at Cape Tryon. Forty-eight P. dOJtbo chicks banded at the Durell Point colony between 19 74 and 19 76 returned there to nest as two- to four-year olds in 19 77 and 19 78. Unmarked individuals with clutch-starts in April were likely greater than four years old as all marked two to four-year olds (with one possible exception) in 19 77 and 1978 had clutch-starts in May and June. Seasonal variation in the breeding success of P. dOJibo individuals was examined at Durell Point in 1977. Mean clutch-size, hatching success and fledging success exhibited a seasonal decline. Four- and 5-egg clutches represented the majority (75%) of all P. CCUibo clutches at Durell Point in 1977 and had the highest reproductive success (0.48 and 0.43 chicks fledged per egg laid respectively). Smaller clutches produced small broods with significantly higher chick mortality while larger clutches suffered high egg loss prior to clutch completion.

Patterns of intra-clutch egg size variation and intra-clutch hatch intervals in the Ring-billed gull (Larus delawarensis) were documented during the peak nesting period of two consecutive breeding seasons, at a colony near Port Colborne, Ontario. Egg size decreased with laying order; third laid eggs were significantly smaller than first laid eggs. Hatching of the third egg was delayed from that of first and second eggs. Intraclutch egg size differences established initial size disparities among chicks at hatch. Hatch intervals further exaggerated size disparities during the early post brood completion period. Competitive asymmetries among chicks were associated with increased mortality rates among third hatched chicks despite the lack of evidence of a sibling feeding hierarchy. Fledging success in 1987 was greater than in 1988. A "brood reduction strategy" appears to have enabled parents in 1987, to obtain an extra unit of reproductive fitness, while in 1988 parents were often unable to raise the entire brood and third chicks likely represented insurance reproductive value. Experimental broods (1988) were created in which hatch intervals were double those of natural intervals. The size disparities among chicks were significantly greater than in control broods, and the pattern of mortality among chicks suggested that first chicks benefited at a cost to second and third chicks. Parents of peak experimental broods achieved a fledging success rate similar to that of control broods. Characteristics of chick adoptions were also recorded. In each study year, 9 chicks abandoned their natal territories, 6 of which were adopted. Chicks consistently established themselves into broods where they were older than resident chicks. No direct evidence of cost to foster parents, or benefits to adopted chicks was obtained, although fledging success of adopted chicks was high.

The regenerating urodele limb is a useful model system in which to study, in vivo, the controls of cell proliferation and differentiation. Techniques are available which enable one to experimentally manipulate mitogenic influences upon the blastema, as well the morphogenesis of the regenerating 11mb. Although classical regeneration studies have generated a wealth of knowledge concerning tissue interactions, little 1s known about the process at the level of gene expression. The aim of this project was to clone potentially developmentally regulated genes from a newt genomic library for use in future studies of gene expression during limb regeneration. We decided to clone the cytoskeletal actin gene for the following reasons: 1. its expression reflects the proliferative and differentiatlve states of cells in other systems 2. the high copy number of cytoplasmic actin pseudogenes in other vertebrates and the high degree of evolutionary sequence conservation among actin genes increased the chance of cloning one of the newt cytoplasmic actin genes. 3. Preliminary experiments indicated that a newt actin could probably be identified using an available chick ~-actln gene for a molecular probe. Two independent recombinant phage clones, containing actin homologous inserts, were isolated from a newt genomic library by hybridization with the chick actin probe. Restriction mapping identified actin homologous sequences within the newt DNA inserts which were subcloned into the plasmid pTZ19R. The recombinant plasmids were transformed into the Escherichia coli strain, DHsa. Detailed restriction maps were produced of the 5.7Kb and 3.1Kb newt DNA inserts in the plasmids, designated pTNAl and pTNA2. The short «1.3 Kb) length of the actin homologous sequence in pTNA2 indicated that it was possibly a reverse transcript pseudogene. Problems associated with molecular cloning of DNA sequences from N. viridescens are discussed with respect to the large genome size and abundant highly repetitive DNA sequences.

Fresh egg-weights and feeding rates to chicks were related to chick survival as one means of quantifying apportionment of parental investment wi thin broods of Caspian Terns (SterDI casRla) at a colony in Georgian Bay. Lake Huron, during 1978 and 1979. Ftrst-laid eggs from 2-egg clutches were Significantly heavier and usually hatched one to three days earlier than second-laid eggs in both years of the study. In both years, first-hatched chicks were larger and generally better fed than second-hatched siblings. The disparity between feedIng rates of first- and second-hatched ehicks was greater in 1979. Brood feeding I rates correlated positively with the percentage of food fed to the least-fed sibUng through the period of B-chick ages zero to 10 days in 1978. I suggest that after this age period, parental control over whlcb cbick was fed diminished. In 1978, 10 of 16 secondhatched chicks were fed more than their older siblings during their first 5 days. 'lb.is is interpreted as a parental response to reduce the competitive advantage of the larger first-hatched chicks. Most chick losses were apparently caused by starvation or preda. tion. In 1979, seeorvl-hatched chick disappearance (due to predation) was -related to low feeding rates, whereas first-hatched chick disappearance was related to low fresh egg-weights.. First-hatched chicks survived better than second-hatched chicks both years, and more pairs fledged two chicks in 1978. Maximum estimated feeding rates at the nest and fledging ages suggested that food was more avatlable in 1978 than in 1979. In 1979, second eggs apparently functioned as "insurance" eggs. When the first-laid egg falled to hatch, or the first-hatched chick died, the second-hatched chick was often successfully fledged. When first-hatched chicks survived, the second-hatched chick usually starved or was preyed upon, reducing the brood to one chick. Parental investment patterns favored first-hatched chicks. Brood reduction, when employed, discouraged total nest failure, however, under appropriate conditions, brood reduction was avoided and full broods (or two chicks) were fledged.

How does fire affect the plant and animal community of the boreal forest? This study attempted to examine the changes in plant composition and productivity, and small mammal demography brought about by fire in the northern boreal environment at Chick Lake, N.W.T. (65053fN, 128°14,W). Two 5*6 ha plots measuring 375m x 150m were selected for study during the summers of 1973 and 197^. One had been unburned for 120 years, the other was part of a fire which burned in the spring of 1969. Grids of 15m x 15m were established in each plot and meter square quadrats taken at each of the 250 grid intersections in order to determine plant composition and density. Aerial primary production was assessed by clipping and drying 80 samples of terminal new production for each species under investigation. Small mammal populations were sampled by placing a Sherman live trap at each grid intersection for ten days in every month. The two plots were similar in plant species composition which suggested that most regrowth in the burned area was from rootstocks which survived the fire. The plant data were submitted to a cluster analysis that revealed nine separate species associations, six of which occured in the burned area and eight of which occured in the control. These were subsequently treated as habitats for purposes of comparison with small mammal distributions. The burned area showed a greater productivity in flowers and fruits although total productivity in the control area was higher due to a large contribution from the non-vascular component. Maximum aerial productivity as dry wieght was measured at 157.1 g/m and 207.8 g/m for the burn and control respectively. Microtus pennsylvanicus and Clethrionomys rutilus were the two most common small mammals encountered; Microtus xanthognathus, Synaptomys borealis, and Phenacomys intermedius also occured in the area. Populations of M. pennsylvanicus and C. rutilus were high during the summer of 1973; however, M. pennsylvanicus was rare on the control but abundant on the burn, while C. rutilus was rare on the burn but abundant in the control. During the summer of 197^ populations declined, with the result that few voles of any species were caught in the burn while equal numbers of the two species were caught in the control. During the summer of 1973 M. pennsylvanicus showed a positive association to the most productive habitat type in the burn which was avoided by C. rutilus. In the control £• rutilus showed a similar positive association to the most productive habitat type which was avoided by M. pennsylvanicus. In all cases for the high population year of 1973# the two species never overlapped in habitat preference. When populations declined in 197^f "both species showed a strong association for the most productive habitat in the control. This would suggest that during a high population year, an abundant species can exclude competitors from a chosen habitat, but that this dominance decreases as population levels decrease. It is possible that M. pennsylvanicus is a more efficient competitor in a recently burned environment, while C. rutilus assumes this role once non-vascular regrowth becomes extensive.

The parental behaviour of male and female Common Terns (Sterna hirundo) was documented throughout two breeding seasons at a colony near Port Colborne , Ontario. Thirteen and fourteen pairs of terns were chosen for intensive study in 1982 and 1983, respectively. The delivery of fish by males to their mates ("courtship feeding") occurred prior-to, during, and following the egg-laying period. Following the laying of the second egg, courtship feeding rates declined significantly. There was a significant, positive correlation bebween courtship feeding rates and subsequent chick feeding rates by males. The incubation rates of females were significantly higher than those of males, especially during the first ten days of incubation. Territorial attendance rates during the incubation stage were similar for males and females. During the chick stage, territorial attendance rates of females were significantly higher than those of males. The size of fish fed to chicks by males increased as the chicks grew older and chick feeding rates of males were approximately three times higher than female rates. Based on these quantitative differences in parental care activities, the cumulative parental time investment by the two sexes was very similar. However, the energetic investment by males was likely greater than that by females, since male parental contributions (e.g. courtship feeding and chick feeding) often entailed extensive foraging behaviour.

Several factors influencing reproductive success were investigated at a Common Tern colony at Port Colborne, Ontario in 1976. In general three egg clutches hatched better than two egg clutches and early started clutches hatched eggs and fledged chicks better than late clutches; the fledging success of two and three egg clutches was similar. Early clutches took longer to hatch and hatched more synchronously than did late clutches. While hatching success differed with nesting substrate used fledging success' did not* No relationship was found between either incubation attentiveness and reproductive success or between incubation attentiveness and clutch size* At no time did food availability appear to be a factor limiting the successful upbringing of two chick broods. While fCf chicks (i.e. chicks hatching from the last laid eggs of three egg clutches) generally survived and grew poorly relative to their brood mates they grew best when they originated from clutches that hatched relatively asynchronously.

Aspects of the breeding biology of two Lake Erie Herri ng Gull colonies were studied in 1975 and 1976. In 1976 the incubation attention given 2-egg and 3-egg clutches initiated early and late in the season was measured. Brood size at one colony was artificially increased or decreased by addition of chicks shortly after hatching. Hatching success was not consistently re~ated to clutch size but early nesters were more successful than late nes'ters. Differences in hatching success between 2-egg and 3-egg clutches were a function of the time of clutch initiation with the clutch size having the greater proportion of its nests initiated early in the season being more successful. The incubation attentiveness of parents of 2-egg and 3-ev,g , and early and late clutches was similar. Most nests were incubated greater than 95% of the time although t heir hatching success was similar ' to those incubated less than 75% of the time. Fledging success, chick growth and weight at fledging were similar among broods of one, two and three chicks and artificially increased broods of four and five chicks. Fledging success was highest for o.e chick broods reduced from two and three chick broods.

The relationships among chick feeding, size and type of prey item, and foraging time away from the brood have not been well studied in seabirds. This study investigated spatial and temporal patterns of foraging and chick-provisioning among 23 radio-tagged male common terns nesting at Hamilton Harbour, Lake Ontario during 1991 and 1992. Telemetry data were collected concurrently with behavioural observations from an elevated blind. Terns fitted with transmitters did not differ from controls with respect to either brood attendance, patterns of chick mortality, species and size distributions of prey delivered to offspring, or chick-provisioning rates. There was a clear separation of parental roles: males were primarily responsible for feeding chicks while females allocated more time to brood attendance. The prey species most commonly delivered to chicks by adults were rainbow smelt (Osmerus mordax) and alewife (A/osa pseudoharengus), followed in importance by larval fish, emerald shiner (Notropis antherinoides), salmonids, and fathead minnows (Pimepha/es prome/as). The relative proportions of various fish speCies delivered to chicks by males differed over the course of each breeding season, and there was also much variability in species composition of prey between years. Sizes of prey delivered to chicks also differed between sampling periods. The modal size of fish brought to chicks during Peak 1991 was 1.5 bill lengths, while the majority of prey in Late 1991 were small larval fish. The reverse trend occurred in 1992 when small fish were delivered to chicks predominantly during the Peak nesting period. During periods when predominantly small fish were delivered to chicks, the foraging activity of radio-tagged males was concentrated within a two kilometer radius of the colony. The observed variation in prey composition and foraging locations during the study likely reflects temporal variation in the availability of prey in the vicinity of the colony. Males delivered fish to chicks at a constant rate, while females 4 increased their feeding frequency over the first six to ten brood days. The mean length of fish delivered to chicks by adults increased significantly with increasing chick age. As a group, within each nesting period, transmittered males either foraged predominantly in the same directional bearing (north during Peak 1991, south during Late 1992), or concentrated foraging activity in the immediate vicinity of the colony (Late 1991, Peak 1992). However, individual radio-tagged males exhibited unique and predictable foraging patterns, often favouring specific locations within these areas and differing in their secondary foraging patterns. Overall, the Lake Ontario shoreline between NCB Bay" (3.5 km south of colony) and the lift bridge canal (4 km north of colony) was the foraging area used most frequently by radiotagged males during the chick-rearing period. Foraging patterns of transmittered males at Windermere Basin are similar to patterns of peak-nesting common terns, but differ from those of late-nesters, at a nearby colony (Port Colborne, Lake Erie). Differences between the foraging patterns of late-nesting terns at these colonies likely reflect differences in annual patterns of fish availability between the two locations. No relationship was found between foraging proficiency of adults and survival of offspring. Stochastic factors, such as predation by black-crowned nightherons (Nycticorax nycticorax) and adverse weather conditions during the early stages of chick rearing, may be more important determinants of common tern breeding success than parental quality or fish availability.

Une cascade de facteurs de transcription composée de SIM1, ARNT2, OTP, BRN2 et SIM2 est requise pour la différenciation des cinq types cellulaires qui peuplent le noyau paraventriculaire (PVN) de l’hypothalamus, un régulateur critique de plusieurs processus physiologiques essentiels à la survie. De plus, l’haploinsuffisance de Sim1 est aussi une cause d’hyperphagie isolée chez la souris et chez l’homme. Nous désirons disséquer le programme développemental du PVN, via une approche intégrative, afin d’identifier de nouveaux gènes qui ont le potentiel de réguler l’homéostasie chez l’individu adulte. Premièrement, nous avons utilisé une approche incluant l’analyse du transcriptome du PVN à différents stades du développement de la souris pour identifier de tels gènes. Nous avons comparé les transcriptomes de l’hypothalamus antérieur chez des embryons de souris Sim1+/+ et Sim1-/- à E12.5 issus de la même portée. De cette manière, nous avons identifié 56 gènes agissant en aval de Sim1 dont 5 facteurs de transcription - Irx3, Sax1, Rxrg, Ror et Neurod6. Nous avons également proposé un modèle de développement à deux couches de l’hypothalamus antérieur. Selon ce modèle, les gènes qui occupent un domaine médial dans la zone du manteau caractérisent des cellules qui peupleront le PVN alors que les gènes qui ont une expression latérale identifient des cellules qui donneront plus tard naissance aux structures ventrolatérales de l’hypothalamus. Nous avons aussi démontré que Sim1 est impliqué à la fois dans la différenciation, la migration et la prolifération des neurones qui peuplent le PVN tout comme Otp. Nous avons également isolé par microdissection au laser le PVN et l’hypothalamus médiobasal chez des souris de type sauvage à E14.5 pour en comparer les transcriptomes. Ceci nous a permis d’identifier 34 facteurs de transcription spécifiques au PVN et 76 facteurs spécifiques à l’hypothalamus médiobasal. Ces gènes représentent des régulateurs potentiels du développement hypothalamique. Deuxièmement, nous avons identifié 3 blocs de séquences au sein de la région 5’ d’Otp qui sont conservés chez l’homme, la souris et le poisson. Nous avons construit un transgène qui est composé d’un fragment de 7 kb contenant ces blocs de séquences et d’un gène rapporteur. L’analyse de 4 lignées de souris a montré que ce transgène est uniquement exprimé dans le PVN en développement. Nous avons généré un deuxième transgène dans lequel le fragment de 7 kb est inséré en amont de l’ADNc de Brn2 ou Sim1 et de Gfp. Nous avons obtenu quatre lignées de souris dans lesquels le profil d’expression de Brn2 et de Gfp reproduit celui d’Otp. Nous étudierons le développement du PVN et la prise alimentaire chez ces souris. En parallèle, nous croisons ces lignées avec les souris déficientes en Sim1 pour déterminer si l’expression de Brn2 permet le développement des cellules du PVN en absence de Sim1. En résumé, nous avons généré le premier transgène qui est exprimé spécifiquement dans le PVN. Ce transgène constitue un outil critique pour la dissection du programme développemental de l’hypothalamus. Troisièmement, nous avons caractérisé le développement de l’hypothalamus antérieur chez l’embryon de poulet qui représente un modèle intéressant pour réaliser des études de perte et de gain de fonction au cours du développement de cette structure. Il faut souligner que le modèle de développement à deux couches de l’hypothalamus antérieur semble être conservé chez l’embryon de poulet où il est aussi possible de classer les gènes selon leur profil d’expression médio-latéral et le devenir des régions qu’ils définissent. Finalement, nous croyons que cette approche intégrative nous permettra d’identifier et de caractériser des régulateurs du développement du PVN qui pourront potentiellement être associés à des pathologies chez l’adulte telles que l’obésité ou l’hypertension.

Signal relay by guidance receptors at the axonal growth cone is a process essential for the assembly of a functional nervous system. We investigated the in vivo function of Src family kinases (SFKs) as growth cone guidance signaling intermediates in the context of spinal lateral motor column (LMC) motor axon projection toward the ventral or dorsal limb mesenchyme. Using in situ mRNA detection we determined that Src and Fyn are expressed in LMC motor neurons of chick and mouse embryos at the time of limb trajectory selection. Inhibition of SFK activity by C-terminal Src kinase (Csk) overexpression in chickLMCaxons using in ovo electroporation resulted inLMC axons selecting the inappropriate dorsoventral trajectory within the limb mesenchyme, with medial LMC axon projecting into the dorsal and ventral limb nerve with apparently random incidence. We also detected LMC axon trajectory choice errors in Src mutant mice demonstrating a nonredundant role for Src in motor axon guidance in agreement with gain and loss of Src function in chickLMCneurons which led to the redirection ofLMCaxons. Finally, Csk-mediated SFK inhibition attenuated the retargeting ofLMCaxons caused by EphA or EphB over-expression, implying the participation of SFKs in Eph-mediated LMC motor axon guidance. In summary, our findings demonstrate that SFKs are essential for motor axon guidance and suggest that they play an important role in relaying ephrin:Eph signals that mediate the selection of motor axon trajectory in the limb.

Les animaux font face à des changements environnementaux brutaux dus aux modifications de milieux liés à l'activité humaine et aux changements climatiques, et doivent s'ajuster rapidement à leur nouvel environnement. Certains processus cognitifs comme l'innovation et l'apprentissage permettent aux animaux d'intégrer de nouveaux comportements à leur répertoire comportemental (flexibilité comportementale), leur donnant l'opportunité d'intégrer un comportement plus optimal pour s'ajuster. Les performances cognitives varient entre espèces et les individus d'une même population et bien que des études récentes se soient intéressées aux causes des variations interindividuelles des performances cognitives, les conséquences restent peu explorées. Dans cette thèse, les questions des pressions de sélection s'exerçant sur les capacités cognitives sont abordées afin de mieux comprendre l'évolution de ces traits au sein d'une population naturelle de mésange charbonnière Parus major. Un nouveau test de résolution de problème a tout d'abord été présenté à des couples reproducteurs directement en milieu naturel. Les résultats ont montré que les couples les plus performants à résoudre la tâche surpassaient les couples les moins performants sur plusieurs mesures de succès reproducteur. Afin de vérifier que la motivation à nourrir les poussins ne biaisait pas cette relation, la taille de nichée a ensuite été manipulée, ce qui n'a pas affecté la performance subséquente des parents. Les couples innovateurs démontraient un meilleur succès reproducteur quel que soit le changement de la taille de nichée subit, ce qui suggère que cette performance influence bien le succès de reproduction, et non l'inverse. De plus, les couples innovateurs approvisionnaient leurs poussins plus souvent que les couples non innovateurs, suggérant que les innovateurs pourraient exploiter leur habitat de façon plus optimale. Dans un troisième temps, plusieurs caractéristiques morphologiques, dont la coloration des plumes, ont été reliées aux performances de résolution de problème et d'apprentissage. Ces liens, bien que complexes et condition-dépendants, pourraient indiquer un rôle de ces performances lors de la sélection sexuelle. Enfin, afin de tester l'effet du parasite sanguin du paludisme sur les traits comportementaux, un médicament contre le paludisme a été injecté à des femelles reproductrices. Cette injection n'a pas modifié leurs performances cognitives mais a augmenté leur niveau d'activité et d'exploration du nichoir en réponse à la tâche de résolution de problème. Ce parasite sanguin, très présent chez les populations depassereaux, pourrait donc expliquer les variations interindividuelles et interpopulationnelles de certains traits comportementaux en milieu naturel, au même titre que dans nombreux autres systèmes hôte-parasites étudiés. Les travaux présentés dans cette thèse ont permis de détailler pour la première fois la relation entre une performance cognitive et le succès reproducteur chez une population aviaire naturelle, une relation robuste et non influencée par la motivation à nourrir la couvée. Cette performance cognitive est reliée à plusieurs traits morphologiques, mais non à la charge parasitaire. Une meilleure exploitation de l'habitat et habileté à s'occuper des poussins pourrait expliquer cette relation.

Objective: Our research program has focused on the development of promising, soft alkylating N-phenyl-N’-(2-chloroethyl)urea (CEU) compounds which acylate the glutamic acid-198 of β-tubulin, near the binding site of colchicum alkaloids. CEUs inhibit the motility of cancerous cells in vitro and, interestingly, exhibit antiangiogenic and anticancer activity in vivo. Mitotic arrest induced by microtubule-interfering agents such as CEUs remains the major mechanism of their anticancer activity, leading to apoptosis. However, we recently demonstrated that microtubule disruption by CEUs and other common antimicrotubule agents greatly alters the integrity and organization of microtubule-associated structures, the focal adhesion contact, thereby initiating anoikis, an apoptosis-like cell death mechanism caused by the loss of cell contact with the extracellular matrix. Methods: To ascertain the activated signaling pathway profile of CEUs, flow cytometry, Western blot, immunohistochemistry and transfection experiments were performed. Wound-healing and chick embryo assays were carried out to evaluate the antiangiogenic potency of CEUs. Results: CEU-induced apoptosis involved early cell cycle arrest in G2/M and increased level of CDK1/cycline B proteins. These signaling events were followed by the specific activation of the intrinsic apoptosis pathway, involving loss of mitochondrial membrane potential (Δψm) and ROS production, cytochrome c release from mitochondria, caspase activation, AIF nuclear translocation, PARP cleavage and nuclear fragmentation. CEUs maintained their efficacy on cells plated on pro-survival extracellular matrices or exhibiting overexpression of P-glycoprotein or the anti-apoptotic protein Bcl-2. Conclusion: Our results suggest that CEUs represent a promising new class of antimicrotubule, antiangiogenic and pro-anoikis agents.

Le problème grandissant de l’antibiorésistance remet en question plusieurs pratiques reliées à l’utilisation des antibiotiques, dont l’usage à grande échelle pour des fins de promotion de la croissance chez les animaux de consommation. Depuis 2006, le retrait des antibiotiques dans les élevages de poulets de chair en Europe a été associé à la résurgence d’entérite nécrotique, une maladie intestinale causée par la bactérie Clostridium perfringens. Alors que la pathogénie de la maladie semblait bien comprise, le retrait des antibiotiques a montré que peu de solutions de remplacement sont disponibles afin de prévenir cette maladie. Très peu d’études se sont intéressées à mieux caractériser la dynamique des populations de C. perfringens dans les élevages avicoles et l’effet que pouvait avoir le retrait des antibiotiques sur l’évolution de ces populations. La présente étude a évalué l’impact du remplacement des antibiotiques promoteurs de croissance et des anticoccidiens pendant une période de 14 mois, dans huit élevages commerciaux de poulets de chair au Québec. Un protocole d’élevage alternatif, combinant des stratégies telles que des produits à base d’huiles essentielles, des acides organiques et inorganique, une vaccination contre la coccidiose ainsi qu’une amélioration des conditions de démarrage des poussins, a été utilisé en remplacement des antimicrobiens. Les performances zootechniques, la santé digestive ainsi que l’occurrence d’entérite nécrotique pour les lots de poulets soumis au protocole d’élevage alternatif ont été comparées avec celles de lots de poulets de chair recevant une ration conventionnelle. Des analyses moléculaires basées sur la PCR et le PFGE ont été utilisées afin de documenter l’impact de la mise en place du protocole d’élevage alternatif sur les populations de C. perfringens. Les résultats obtenus montrent qu’aucune différence entre les groupes soumis aux deux types de protocoles n'est observée en ce qui a trait à la viabilité en élevage, à l'âge d'abattage et aux taux de condamnations à l'abattoir. Toutefois, les lots soumis au traitement alternatif ont eu des performances moindres pour le poids moyen à l'abattage, le gain moyen quotidien et la conversion alimentaire. Un peu plus de 27% des lots soumis au protocole alternatif ont connu un épisode d’entérite nécrotique clinique. Les analyses ont aussi montré que l’un ou l’autre des protocoles ne semble pas exercer d’influence particulière sur la dynamique temporelle des populations de C. perfringens. Pour les lots soumis au protocole d’élevage alternatif, une forte diversité génétique pour C. perfringens a été liée à un risque augmenté de vivre un épisode d’entérite nécrotique. À l’opposé, les lots alternatifs ayant vécu des épisodes récurrents de la maladie ont montré une diminution significative de la diversité génétique, ainsi qu’une augmentation marquée du nombre de souches de C. perfringens transportant plusieurs gènes de virulence. La présente étude a permis de mieux documenter, d’un point de vue économique et scientifique, la production de poulets de chair élevés sans antibiotiques ni anticoccidiens au Québec. Cette étude est la seule du genre s’étant intéressée à la caractérisation et à la comparaison dans le temps des flores de C. perfringens retrouvées dans les deux types d’élevages. Elle a révélé que la production à grande échelle de poulets de chair élevés sans antibiotiques ni anticoccidiens est possible au Québec, mais que celle-ci présente des impacts sur les performances zootechniques des oiseaux, sur leur santé digestive et sur la dynamique des populations de C. perfringens. Vibrio sp. V26 isolated from mangrove sediment showed 98 % similarity to 16S rRNA gene of Vibrio cholerae, V. mimicus, V. albensis and uncultured clones of Vibrio. Phenotypically also it resembled both V. cholerae and V. mimicus.Serogrouping, virulence associated gene profiling, hydrophobicity, and adherence pattern clearly pointed towards the non—toxigenic nature of Vibrio sp. V26. Purification and characterization of the enzyme revealed that it was moderately thermoactive, nonhemagglutinating alkaline metalloprotease with a molecular mass of 32 kDa. The application of alkaline protease from Vibrio sp. V26 (APV26) in sub culturing cell lines (HEp-2, HeLa and RTG-2) and dissociation of animal tissue (chick embryo) for primary cell culture were investigated. The time required for dissociation of cells as well as the viable cell yield obtained by while administeringAPV26 and trypsin were compared. Investigations revealed that the alkaline protease of Vibrio sp. V26 has the potential to be used in animal cell culture for subculturing cell lines and dissociation of animal tissue for the development of primary cell cultures, which has not been reported earlier among metalloproteases of Vibrios.
