It is largely known that the range of an insect diet is mostly determined by oviposition behavior, mainly in species with endophytic larvae such as Zabrotes subfasciatus. However, the proximate factors determining host choice and the subsequent steps leading to the expansion or reduction of the host number and occasional host shifts are largely unknown. We analyzed various factors determining host preference of Z. subfasciatus through the evaluation of: (i) oviposition preference of a wild population of Z subfasciatus on the usual host (bean) and unusual hosts (lentil, chickpea and soy), and the performance of the offspring; (ii) artificial selection for increasing preference for hosts initially less frequently chosen; (iii) comparison of oviposition behavior between two different populations (reared for similar to 30 generations in beans or chickpeas, respectively); (iv) oviposition timing on usual and unusual hosts; and (v) identification of preference hierarchies. We found that when using unusual hosts, there is no correlation between performance and preference and that the preference hierarchy changes only slightly when the population passes through several generations on the less frequently accepted host. We also found a positive response to artificial selection for increasing oviposition on the less preferred host; however, when the host-choice experiment involved two varieties of the usual host, the response was faster than when the choice involved usual and unusual hosts. Finally, beetles reared on an unusual host (chickpea) for 26 generations showed similar good fitness on both usual and unusual hosts, indicating that the use of a new host does not necessarily result in the loss of performance on the original host. Nevertheless, this population showed lower fitness on the usual host than that of the original population, suggesting an underlying partial trade-off phenomenon which may contribute to a broadening of diet of this insect species.

Understanding the mating patterns of populations of tree species is a key component of ex situ genetic conservation. In this study, we analysed the genetic diversity, spatial genetic structure (SGS) and mating system at the hierarchical levels of fruits and individuals as well as pollen dispersal patterns in a continuous population of Theobroma cacao in Para State, Brazil. A total of 156 individuals in a 0.56 ha plot were mapped and genotyped for nine microsatellite loci. For the mating system analyses, 50 seeds were collected from nine seed trees by sampling five fruits per tree (10 seeds per fruit). Among the 156 individuals, 127 had unique multilocus genotypes, and the remaining were clones. The population was spatially aggregated; it demonstrated a significant SGS up to 15m that could be attributed primarily to the presence of clones. However, the short seed dispersal distance also contributed to this pattern. Population matings occurred mainly via outcrossing, but selfing was observed in some seed trees, which indicated the presence of individual variation for self-incompatibility. The matings were also correlated, especially within ((r) over cap (p(m)) = 0.607) rather than among the fruits ((r) over cap (p(m)) = 0.099), which suggested that a small number of pollen donors fertilised each fruit. The paternity analysis suggested a high proportion of pollen migration (61.3%), although within the plot, most of the pollen dispersal encompassed short distances (28m). The determination of these novel parameters provides the fundamental information required to establish long-term ex situ conservation strategies for this important tropical species. Long-term stability of dentin matrix following treatment with various natural collagen cross-linkers

Objectives: Collagen disorganization is one of the main degradation patterns found in unsuccessful adhesive restorations. The hypothesis of this study was that pretreatment using natural collagen cross-linking agents rich in proanthocyanidin (PA) would improve mechanical properties and stability over time of the dentin collagen and, thus, confer a more resistant and lasting substrate for adhesive restorations. Methods: PA-based extracts, from grape seed (GSE), cocoa seed (CSE), cranberry (CRE), cinnamon (CNE) and acai berry (ACE) were applied over the demineralized dentin. The apparent elastic modulus (E) of the treated dentin collagen was analyzed over a 12 month period. Specimens were immersed in the respective solution and E values were obtained by a micro-flexural test at baseline, 10, 30, 60, 120 and 240 min. Samples were stored in artificial saliva and re-tested after 3, 6 and 12 months. Data was analyzed using ANOVA and Tukey test. Results: GSE and CSE extracts showed a time-dependent effect and were able to improve [240 min (MPa): GSE = 108.96 +/- 56.08: CSE = 59.21 +/- 24.87] and stabilize the E of the organic matrix [12 months (MPa): GSE = 40.91 +/- 19.69; CSE = 42.11 +/- 13.46]. CRE and CNE extracts were able to maintain the E of collagen matrices constant over 12 months [CRE = 11.17 +/- 7.22; CNE = 9.96 +/- 6.11; MPa]. ACE (2.64 +/- 1.22 MPa) and control groups immersed in neat distilled water (1.37 +/- 0.69 MPa) and ethanol-water (0.95 +/- 0.33 MPa) showed no effect over dentin organic matrix and enable their degradation and reduction of mechanical properties. Significance: Some PA-based extracts were capable of improving and stabilizing collagen matrices through exogenous cross-links induction. (C) 2011 Elsevier Ltd. All rights reserved.

Objectives To characterize the properties of dentin matrix treated with two proanthocyanidin rich cross-linking agents and their effect on dentin bonded interfaces. Methods Sound human molars were cut into 0.5mm thick dentin slabs, demineralized and either treated with one of two cross-linking agents (grape seedGSE and cocoa seedCOE extracts) or left untreated. The modulus of elasticity of demineralized dentin was assessed after 10 or 60min and the swelling ratio after 60min treatment. Bacterial collagenase was also used to assess resistance to enzymatic degradation of samples subjected to ultimate tensile strength. The effect of GSE or COE on the resindentin bond strength was evaluated after 10 or 60min of exposure time. Data were statistically analyzed at a 95% confidence interval. Results Both cross-linkers increased the elastic modulus of demineralized dentin as exposure time increased. Swelling ratio was lower for treated samples when compared to control groups. No statistically significant changes to the UTS indicate that collagenase had no effect on dentin matrix treated with either GSE or COE. Resindentin bonds significantly increased following treatment with GSE regardless of the application time or adhesive system used. Significance Increased mechanical properties and stability of dentin matrix can be achieved by the use of PA-rich collagen cross-linkers most likely due to the formation of a PAcollagen complex. Objectives: The effects of interactions between cross-linking proanthocyanidins (PA) in polar solvents and type-I collagen of demineralized dentin were investigated. Methods: Three PA-rich extracts, two from grape seed (GSEP and GSES) and one from cocoa (COE), were dissolved (water, ethanol:water and acetone:water) and analyzed for their ability to increase the modulus of elasticity of demineralized dentin. Sound dentin beams (0.5 mm x 1.7 mm x 7 mm) were fully demineralized and divided into 12 groups according to the type of cross-linking agent and solvents used. Specimens were immersed in the respective solutions and tested at baseline, 10, 30, 60, 120 and 240 min. Results: The elastic modulus (EM) of dentin was significantly increased by the PA treatment regardless of time (p < 0.05 for all times). The extracts showed different solubility in different solvents. GSEP showed the highest increase in EM when diluted in distilled water and acetone at all exposure times. Both GSEs showed superior results when diluted in distilled water and after 4 h of treatment, while COE produced strongest enhancement when dissolved in ethanol:water. Conclusions: The results indicates that herbal extraction process and other pharmacognostic parameters have an important influence on extract solubility as well as constitution and, consequently, on the PA-dentin matrix interaction. This study evaluated the effect of the C-factor and dentin preparation method (DPM) in the bond strength (BS) of a mild self-etch adhesive; the study also observed the SEM superficial aspects of the corresponding smear layer. For purposes of this study, 25 molars (n=5) were used in a bond strength test. The molars were divided into two parts (buccal and lingual): one part received a Class V cavity (C-factor=3) and the other received a flat surface (C-factor=0) with the same bur type (coarse diamond or carbide bur and fine diamond or carbide bur), both within the same dentin depth. Five teeth were prepared with wet 60-grit and 600-grit SiC papers. After restoration with Clearfil SE Bond, microtensile beans (0.8 mm(2)) were prepared and tested after 24 hours in a universal testing machine (0.5 mm/minute). An additional two teeth for each DPM were prepared for SEM evaluation of the smear layer superficial aspects. The BS values were submitted to one-way ANOVA, considering only the DPM (flat surfaces) and two-way ANOVA (C-Factor x DPM, considering only burs) with p=0.05. Although the DPM in the flat surfaces was not significant, the standard deviations of carbide bur-prepared specimens were markedly lower. The BS was significantly lower in cavities. The fine carbide bur presented the most favorable smear layer aspect. It was concluded that different dentin preparation methods could not prevent the adverse effect in bond strength of a high C-factor. A coarse cut carbide bur should be avoided prior to a mild self-etch adhesive, because it adversely affected bond strength. In contrast, a fine cut carbide bur provided the best combination: high bond strength with low variability, which suggests a more reliable bond strength performance.

Genistein is an isoflavenoid that is abundant in soy beans. Genistein has been reported to have a wide range of biological activities and to play a role in the diminished incidence of breast cancer in populations that consume a soy-rich diet. Genistein was originally identified as an inhibitor of tyrosine kinases; however, it also inhibits topoisomerase II by stabilizing the covalent DNA cleavage complex, an event predicted to cause DNA damage. The topoisomerase II inhibitor etoposide acts in a similar manner. Here we show that genistein induces the up-regulation of p53 protein, phosphorylation of p53 at serine 15, activation of the sequence-specific DNA binding properties of p53, and phosphorylation of the hCds1/Chk2 protein kinase at threonine 68. Phosphorylation and activation of p53 and phosphorylation of Chk2 were not observed in ATM-deficient cells. In contrast, the topoisomerase II inhibitor etoposide induced phosphorylation of p53 and Chk2 in ATM-positive and ATM-deficient cells. In addition, genistein-treated ATM-deficient cells were significantly more susceptible to genistein-induced killing than were ATM-positive cells. Together our data suggest that ATM is required for activation of a DNA damage-induced pathway that activates p53 and Chk2 in response to genistein.

Backhousia citriodora is typical of the many commercially valuable woody Australian Myrtaceae species that are recalcitrant in forming adventitious roots from cuttings after maturation. A series of experiments were conducted to identify an endogenous rooting inhibitor in line with established criteria. Endogenous levels of citral were correlated with the rooting capacities of juvenile versus mature, and easy- versus difficult-to-root genotypes of B. citriodora, in both winter and summer. The biological activity of citral was confirmed in bioassays on mung beans and easy-to-root B. citriodora seedlings. Evidence of a common mechanism of root inhibition with other species in the Myrtaceae and the role of action of citral are discussed.

Two varieties of adzuki grown in Australia, Bloodwood and Erimo, were stored for up to 6 months at three temperatures (10, 20 and 30 degreesC), and two relative humidities (RH; 40 and 65%). The amount of cell wall material increased with time under all storage conditions. This increase was greatest at 30 degreesC and 40% RH. Storage time and conditions did not affect the total pectin levels in the cell wall. Erimo constantly exhibited a higher total pectin level than Bloodwood. The Bloodwood soluble pectin, Ca++ and Mg++ and Erimo Ca++ in the cell wall remained stable during storage, while the Erimo soluble pectin and Mg++ exhibited a slight decrease at 20 and 30 degreesC after 3 months of storage. Three different particular geometrical shapes of parallelepiped, cylinder and sphere were taken from cut green beans (length:diameter = 1:1, 2:1 and 3:1) and potatoes (aspect ratio = 1:1, 2:1 and 3:1) and peas, respectively. Their drying behaviour in a fluidised bed was studied at three different drying temperatures of 30, 40 and 50 degreesC (RH = 15%). Drying curves were constructed using non-dimensional moisture ratio (MR) and time and their behaviour was modelled using exponential (MR = exp(-kt)) and Page (MR = exp(-kt(n))) models. The effective diffusion coefficient of moisture transfer was determined by Fickian method using uni- and three-dimensional moisture movements. The diffusion coefficient was least affected by the size when the moisture movement was considered three-dimensional, whereas the drying temperature had a significative effect on diffusivity as expected. The drying constant and diffusivity coefficients were on the descending order for potato, beans and peas. The Arrhenius activation energy for the peas was also highest, indicating a strong barrier to moisture movement in peas as compared to beans and skinless cut potato pieces. O objetivo deste trabalho foi avaliar os efeitos do óleo de mamona sobre a broca-grande do tomateiro, Helicoverpa zea, e sobre o parasitoide de ovos Trichogramma pretiosum. Foi determinada a mortalidade larval da broca, que foi submetida ao óleo de mamona por ingestão e contato às concentrações 0,5, 1,0, 1,5, 2,0, 2,5 e 3,0% (v v-1). A ação do óleo de mamona sobre T. pretiosum foi determinada mediante testes de seletividade e suscetibilidade. As maiores mortalidades de larvas foram observadas no tratamento por contato com as menores concentrações (0,5 e 1,0%), enquanto com as concentrações intermediárias (1,5; 2,0 e 2,5%), não houve diferença entre as vias de aplicação. À maior concentração testada (3,0%), o tratamento por ingestão proporcionou mortalidade superior àquela obtida por contato. O número de ovos parasitados por T. pretiosum, no teste de seletividade, foi afetado negativamente pelo óleo de mamona, contudo, os demais parâmetros avaliados quanto à seletividade e à suscetibilidade não foram afetados. O óleo de mamona reduz a sobrevivência de larvas de H. zea tanto por ingestão quanto por contato. Além disso, não prejudica o desenvolvimento de T. pretiosum, desde que as pulverizações sejam realizadas após as liberações do parasitoide.

A luminosidade desponta como um dos fatores abióticos de maior importância no estabelecimento das plantas, podendo ocasionar alterações a nível fisiológico e anatômico, interferindo diretamente no desenvolvimento das mesmas e consequentemente, sua produtividade. O cacaueiro (Theobroma cacao – Malvaceae) possui grande interesse econômico devido à utilização de suas sementes para produção de manteiga de cacau e chocolate. Neste sentido, o objetivo deste trabalho foi comparar características fisiológicas e anatômicas de dois genótipos de cacaueiro (PH 16 e IPIRANGA 01), submetidos a diferentes condições de radiação solar de modo a inferir a respeito das condições de luminosidade mais favoráveis ao desenvolvimento dos mesmos. Foram realizadas análises de crescimento, teor de pigmentos, trocas gasosas e anatômicas caulinares e foliares. O delineamento experimental foi em Blocos casualizados (DBC), com 4 repetições em arranjo fatorial 2 x 5, constituído de dois genótipos e 5 níveis de luminosidade (0% - pleno sol -, 18%, 30%, 50% e 80% de sombreamento), totalizando 40 parcelas com 10 plantas cada. Os dados obtidos foram submetidos à análise de variância e regressão. Os resultados de crescimento indicam maior adaptação, de ambos os genótipos, em condições de irradiância alta a moderada. O índice de qualidade de Dickson demonstrou maior capacidade de sobrevivência em condições de campo a níveis altos de irradiância para o genótipo PH 16 e a níveis de irradiância moderada para IPIRANGA 01. Os teores de pigmentos fotossintetizantes de IPIRANGA 01 mostram-se mais elevados na condição de 30% de sombreamento, enquanto que, para PH 16 os maiores teores foram observados em 50% de sombreamento. Ambos os genótipos demonstraram altas taxas de A, Ci, E, A/E, A/Gs e A/Ci, sob elevadas irradiâncias, assim como adaptações anatômicas caulinares e foliares a maiores luminosidades, tais como, maior espessamento do limbo foliar, parênquima paliçádico, esponjoso, limbo foliar, epiderme adaxial e densidade estomática, além de maior densidade e frequência de elementos de vaso e espessura do xilema secundário. Os genótipos PH 16 e IPIRANGA 01 apresentaram uma grande plasticidade em relação aos diferentes níveis de irradiância, no entanto, constatou-se que PH 16 apresentou melhor desempenho sob condições de alta irradiância, como as obtidas nos tratamentos a pleno sol e 18% de sombreamento, enquanto que, IPIRANGA 01 mostrou-se mais adaptado sob sombreamento moderado, a 30% de sombra.

O Estado do Espírito Santo é o maior produtor de grãos de café conilon do Brasil. Devido aos trabalhos na seleção de plantas produtivas, os avanços na produtividade desta cultura são mais pronunciados com a utilização da técnica da irrigação. A utilização da irrigação na cafeicultura possibilita a adoção de novas tecnologias de plantio, expansão da área produtiva, uniformização da produção diminuindo os grãos chochos e mal granados, além de mitigar os problemas da deficiência hídrica nos períodos críticos da cultura. Diante do exposto, objetivou-se com este estudo, avaliar o crescimento, a produção e o rendimento dos treze clones da variedade de café ‘Conilon Vitória’, em condições de déficit hídrico e irrigado. O experimento foi instalado no ano de 2010 no Instituto Federal do Espírito Santo, Campus Alegre, em esquema de parcelas subdivididas 3 x 13, sendo nas parcelas o manejo de irrigação em três níveis (irrigado, irrigado com déficit hídrico de 50% e sem irrigação) e nas subparcelas os clones da variedade ‘Conilon Vitória’ em treze níveis, em um delineamento em blocos casualizados, com três repetições. Cada parcela experimental foi constituída por cinco plantas úteis. O manejo da irrigação adotado foi via clima, onde a ETo foi estimada por meio da equação de Hargreaves e Samani (1985). Os dados foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey para comparações entre o fator manejo e Scott-Knott para comparações entre o fator clone. O crescimento dos ramos das plantas foi diferenciado em relação ao manejo de irrigação aplicado. Plantas sem irrigação apresentaram maiores picos de crescimento. Plantas irrigadas apresentaram menores picos de crescimento dos ramos, porém, com maior período de crescimento. A produtividade média de plantas irrigadas foi cerca de 474,5% superior a plantas sem irrigação. A produtividade média de plantas irrigadas com déficit hídrico de 50% foi cerca de 329,6 % superior a plantas sem irrigação. O rendimento entre plantas irrigadas e irrigadas com déficit hídrico de 50% não diferiu. Já plantas sem irrigação apresentaram menor rendimento em todos os clones. A irrigação influenciou no tamanho dos grãos, plantas irrigadas apresentaram maiores percentagens de grãos retidos em peneiras 13 e superiores. Já plantas sem irrigação apresentaram maiores percentagens de grãos retidos na fundagem.
