Gas permeability coefficients of a series of aromatic polyetherimides prepared from 1,4-bis(3,4-dicarboxyphenoxy) benzene dianhydride (HQDPA) and four (methylene dianiline)s with a methyl side group to H-2, CO2, O-2, N-2, and CH4 were measured under 7 atm and within a temperature range from 30 to 150 degrees C. The gas permeabilities and permselectivities of these polymers were compared with those of the HQDPA-based polyetherimides from methylene dianiline (MDA) and isopropylidene dianiline (IPDA). The number and position of the methyl side groups on the benzene rings of the diamine residues strongly affect the gas permeabilities and permselectivities of the HQDPA-based polyetherimides. The gas permeability of the polyetherimide progressively increases with an increase in the number of the methyl side groups. Both the gas permeability and permselectivity of the polyetherimides with methyl side groups are higher than those of HQDPA-MDA. The polyetherimide prepared from 3,3'-dimethyl 4,4'-methylene dianiline (DMMDA1) possesses both higher permeability and permselectivity than the polyetherimides prepared from 2,2'-dimethyl 4,4'-methylene dianiline (DMMDA2). However, two of the polyetherimides prepared 2,2',3,3'-tetramethyl 4,4'-methylene dianiline (TMMDA1) or 2,2', 5,5'-tetramethyl 4,4'-methylene dianiline (TMMDA2) possess almost the same gas permeability and permselectivity.

New poly(azomethine sulfones) with linear structures containing sulfonyl bis(4-phenoxyphenylene) and oxo bis(benzylideneaniline) or methylene bis(benzylideneaniline) units were prepared in the conventional literature manner by condensing the dialdehyde sulfone monomer (V) with diamines such as 4,4'-oxydianiline (IIIa) and 4,4'-methylenedianiline (IIIb), or by condensing an azomethine biphenol (IX) with 4,4'-sulfonyldichlorobenzene (II). Three model compounds which reproduced the above structures were also synthesized. The resulting polymers were confirmed by IR, H-1-NMP, and elemental analysis, and were characterized by inherent viscosities, thermogravimetric analysis (TGA), and x-ray diffraction. The thermotropic liquid crystalline (TLC) behavior was studied using polarization light microscopy (PLM), thermooptical analysis (TOA), and DSC. A nematic texture was observed only for 4,4'-oxydianiline-units-based polymers. The reaction of polymer VIIIb containing -CH2- links between the mesogens with the model compound IX led to polymer X which exhibited TLC behavior.

The catalytic activity of heteropoly compounds in the oxidation of benzyl alcohol and cyclohexa nol under phase transfer conditions has been studied. The catalytic activity of six kinds of heteropoly acids with Keggin structure will drop by the order of GeMo12 (H4GeMo12O40). PW12, PMo12, SiMo12, GeW12 and SiW12. When the three protons of H3PW12O40 Were replaced by Na+ step by step, the catalytic activity will raise gradually with the drop of acidity. The addition of base and trace amount of sulfuric acid to the reaction system resulted in an increase of catalytic activity. It was found that catalytic activity of mono-lacunary heteropoly compounds is higher than that of the primary heteropoly acids (or salts). The catalytic oxidation system of HPA-H2O2-PTC is very active in the oxidation of benzyl alcohol ana cyclohexanol, but it has little activity in the oxidation of inactive compounds such as n(or iso)-proplalcohol. n-butyl alcohol and n-hexanol. Solvent has great effect on reaction, when polar compounds such as water were used as solvent, the catalytic activity is better than that when non-polar compounds were used as solvent.

A series of polyimides with different structures have been synthesized and studied by dynamic mechanical analysis. The results obtained indicate that the beta relaxation in polyimides is related to the rotation of rigid segment(s) of p-phenylene and imide groups around 'hinges' such as -O-, -CH2- and so on in diamines. It is noticed that two kinds of polyimides both with [GRAPHICS] imide groups have verv weak beta relaxation below the glass transition temperature. This phenomenon is due to the fact that the configuration of chains with the above imide groups hinders the rotation of the rigid segments in the chains.

The electrochemical behaviour of N-benzylaniline polymerization is determined by the nature of the electrolyte. The voltammograms for a poly-N-benzylaniline modified Pt electrode prepared in 1 M HCl (abbreviated to PBAn(HCl)), and 1 M H2SO4 (PBAn(H2SO4)) tested in 1 M hydrochloric, sulfuric, and perchloric acid were almost superimposable. The polymer film electrode prepared in 1 M HClO4 (abbreviated to PBAn(HClO4)) is electroinactive, and exhibits only charging behaviour in 1 M HClO4 solution and can be activated in hydrochloric or other acid electrolytes with a smaller anion. These interesting phenomena are explained in terms of the anions catalyzing the loss of benzyl groups.

Seven chiral phase-transfer catalysts, among which three have not been reported so far, have been prepared and applied to the asymmetric alkylation of alpha-isopropyl benzyl cyanide and alpha-isopropyl-p-chlorobenzyl cyanide. The result showed that short reaction time, low temperature, high catalyst concentration and non-polar solvent would improve the optical yield. The influence of structure of the catalyst on the asymmetric reaction was preliminarily studied. The optical purity of the products were evaluated by gas chromatography with a chiral column.

The properties of miscible phenolphthalein poly(ether ether ketone)/phenoxy (PEK-C/phenoxy) blends have been measured by dynamic mechanical analysis and tensile testing. The blends were found to have single glass transition temperatures (T(g)) that vary continuously with composition. The tensile moduli exhibit positive deviations from simple additivity. Marked positive deviations were also observed for tensile strength. The tensile strengths of the 90/10 and 75/25 PEK-C/phenoxy blends are higher than those of both the pure components. Embrittlement, or transition from the brittle to the ductile mode of failure, occurs in the composition range of 50-25 wt% PEK-C. These observations suggest that mixing on the segmental level has occurred and that there is enough interaction between the components to decrease its internal mobility significantly. PEK-C was also found to be miscible with the epoxy monomer, diglycidyl ether of bisphenol A (DGEBA), as shown by the existence of a single glass transition temperature (T(g)) within the whole composition range. Miscibility between PEK-C and DGEBA could be considered to be due mainly to entropy. However, PEK-C was judged to be immiscible with the diaminodiphenylmethane-curved epoxy resin (DDM-cured ER). It was observed that the PEK-C/ER blends have two T(g), which remain invariant with composition and are almost the same as those of the pure components, respectively. Scanning electron microscopy showed that the PEK-C/ER blends have a two-phase structure. The different miscibility with PEK-C between DGEBA and the DDM-cured ER is considered to be due to the dramatic change in the chemical and physical nature of ER after curing.

Among marine bacteria isolated from the cytotoxic sponge Hymeniacidon perleve, one strain NJ6-3-1 classified as Pseudomonas sp. showed both cytotoxic and antimicrobial activities. Fatty acid analysis indicated that the bacterial strain consists mainly of C16:1, C16:0, C18:1, C18:0, C15:0, C14:0. One unusual 9,10-cyclopropane-C17:0 fatty acid and C26:0 also constitute major components, as well as the existence of squalene, the precursor of triterpenoids. The major metabolites in the culture broth were identified as alkaloids, including diketopiperazines and indole compounds, namely 3,6-diisopropylpiperazine-2,5-dione, 3-benzyl-3-isopropylpiperazine-2,5-dione, 3,6-bis-(2-methylpropyl)-piperazine-2,5-dione, indole-3-carboxaldehyde, indole-3-carboxylic acid methyl ester, indole-3-ethanol, and quinazoline-2,4-dione.

Six novel dibenzyl bromophenols (1-6) with different dimerization patterns and two propyl bromophenol derivatives (7 and 8), together with 11 known bromophenol derivatives, were isolated from the ethanolic extract of the brown alga Leathesia nana. On the basis of spectroscopic methods the structures of the new compounds were determined as 5,6'-diethyloxymethyl-3,4,2'-tribromo-2,3',4'-trihydroxydiphenyl ether (1), 2-(2,3-dibromo-4,5-dihydroxybenzyl)-3,5-dihydroxy-4-methoxybenzyl alcohol (2), 6-(2,3-dibromo-4,5dihydroxybenzyl)-2,3-dibromo-4,5-dihydroxy benzyl methyl ether (3), 9,10-dihydro-9,10-dimethoxy-3,4,7,8-tetrabromo-1,2,5,6-tetrahydroxyanthracene (4), (+)-3-(2,3-dibromo-4,5-dihydroxyphenyl)-4-bromo-5,6-dihydroxy-1,3-dihydroisobenzofuran (5), rel-(4aS*,10aR*)-(+/-)-6,7-dibromo-4a-hydroxy-3,8-dihydroxymethyl-10a-methoxy- 1,4,4a, 10a-tetrahydrodibenzo[b,e][1,4]dioxin-1-one (6), (E)-2-methyl-3-(2,3-dibromo-4,5-dihydroxyphenyl)propenal (7), and 2-methyl-3-(2,3-dibromo-4,5-dihydroxyphenyl)-1-propanol (8). Some compounds including 3 showed in vitro selective cytotoxicity against several human cancer cell lines. This is the first brown alga to be reported containing bromophenols.

To investigate the antitumor effect of bromophenol derivatives in vitro and Leathesia nana extract in vivo, six bromophenol derivatives 6-(2,3-dibromo-4,5-dihydroxybenzyl)-2,3-dibromo-4,5-dihydroxy benzyl methyl ether (1), (+)-3-(2,3-dibromo-4,5-dihydroxyphenyl)-4-bromo-5,6-dihydroxy-1,3-dihydroisobenzofuran (2), 3-bromo-4-(2,3-dibromo-4,5-dihydroxybenzyl)-5-methoxymethyl-pyrocatechol (3), 2,2',3,3'-tetrabromo-4,4',5,5'-tetrahydroxy-diphenylmethane (4), bis(2,3-dibromo-4,5-dihydroxybenzyl) ether (5), 2,2',3-tribromo-3',4,4',5-tetrahydroxy-6'-ethyloxymethyldiphenylmethane (6) were isolated from brown alga Leathesia nana, and their cytotoxicity were tested by MTT assays in human cancer cell lines A549, BGC-823, MCF-7, B16-BL6, HT-1080, A2780, Bel7402 and HCT-8. Their inhibitory activity against protein tyrosine kinase (PTK) with over-expression of c-kit was analyzed also by ELISA. The antitumor activity of ethanolic extraction of Leathesia nana (EELN) was evaluated on S-180-bearing mice. All compounds showed very potent cytotoxicity against all of the eight cancer cell lines with IC50 below 10 mu g/mL. In PTK inhibition study, all bromophenol derivatives showed moderate inhibitory activity and compounds 2, 5 and 6 showed significant bioactivity with the inhibition ratio of 77.5%, 80.1% and 71.4%, respectively. Pharmacological studies reveal that EELN could inhibit the growth of Sarcoma 180 tumor and increase the indices of thymus and spleen to improve the immune system remarkably in vivo. Results indicated that the bromophenol derivatives and EELN can be used as potent antitumor agents for PTK over-expression of c-kit and considered in a new therapeutic strategy for treatment of cancer.

The focus of this thesis is the preparation of enantiopure sulfoxides by means of copper-catalysed asymmetric sulfoxidation, with particular emphasis on the synthesis of aryl benzyl and aryl alkyl sulfoxides. Chapter 1 contains a review of the methods employed for the asymmetric synthesis of sulfoxides, compounds with many applications in stereoselective synthesis and in some cases with pharmaceutical application. Chapter 1 describes asymmetric oxidation, including metal-catalysed, non metal-catalysed and enzyme-catalysed, in addition to synthetic approaches via nucleophilic substitution of appropriately substituted precursors. Kinetic resolution in oxidation of sulfoxides to the analogous sulfones is also discussed; in certain cases, access to enantioenriched sulfoxides can be achieved via a combination of asymmetric sulfoxidation and complementary kinetic resolution. The design and synthesis of a series of sulfides to enable exploration of the substituent effects of the copper-mediated oxidation was undertaken, and oxidation to the racemic sulfoxides and sulfones to provide reference samples was conducted. Oxidation of the sulfides using copper-Schiff base catalysis was undertaken leading to enantioenriched sulfoxides. The procedure employed is clean, inexpensive, not air-sensitive and utilises aqueous hydrogen peroxide as oxidant. Extensive investigation of the influence of the reaction conditions such as solvent, temperature, copper salt and ligand was undertaken to lead to the optimised conditions. While the direct attachment of one aryl substituent to the sulfide is essential for efficient enantiocontrol, in the case of the second substituent the enantiocontol is dependent on the steric rather than electronic features of the substituent. Significantly, use of naphthyl-substituted sulfides results in excellent enantiocontrol; notably 97% ee, obtained in the oxidation of 2-naphthyl benzyl sulfide, represents the highest enantioselectivity reported to date for a copper-mediated sulfur oxidation. Some insight into the mechanistic features of the copper-mediated sulfur oxidation has been developed based on this work, although further investigation is required to establish the precise nature of the catalytic species responsible for asymmetric sulfur oxidation. Full experimental details, describing the synthesis and structural characterisation, and determination of enantiopurity are included in chapter 3.

The ability to manipulate the coordination chemistry of metal ions has significant ramifications for the study and treatment of metal-related health concerns, including iron overload, UV skin damage, and microbial infection among many other conditions. To address this concern, chelating agents that change their metal binding characteristics in response to external stimuli have been synthesized and characterized by several spectroscopic and chromatographic analytical methods. The primary stimuli of interest for this work are light and hydrogen peroxide.

Herein we report the previously unrecognized photochemistry of aroylhydrazone metal chelator ((E)-N′-[1-(2-hydroxyphenyl)ethyliden]isonicotinoylhydrazide) (HAPI) and its relation to HAPI metal binding properties. Based on promising initial results, a series of HAPI analogues was prepared to probe the structure-function relationships of aroylhydrazone photochemistry. These efforts elucidate the tunable nature of several aroylhydrazone photoswitching properties.

Ongoing efforts in this laboratory seek to develop compounds called prochelators that exhibit a switch from low to high metal binding affinity upon activation by a stimulus of interest. In this context, we present new strategies to install multiple desired functions into a single structure. The prochelator 2-((E)-1-(2-isonicotinoylhydrazono)ethyl)phenyl (E)-3-(2,4-dihydroxyphenyl)acrylate (PC-HAPI) is masked with a photolabile trans-cinnamic acid protecting group that releases umbelliferone, a UV-absorbing, antioxidant coumarin along with a chelating agent upon UV irradiation. In addition to the antioxidant effects of the coumarin, the released chelator (HAPI) inhibits metal-catalyzed production of damaging reactive oxygen species. Finally a peroxide-sensitive prochelator quinolin-8-yl (Z)-3-(4-hydroxy-2-((4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)benzyl)oxy)phenyl)acrylate (BCQ) has been prepared using a novel synthetic route for functionalized cis-cinnamate esters. BCQ uses a novel masking strategy to trigger a 90-fold increase in fluorescence emission, along with the release of a desired chelator, in the presence of hydrogen peroxide.

The purpose of the present study was to use attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) and target factor analysis (TFA) to investigate the permeation of model drugs and formulation components through Carbosil® membrane and human skin. Diffusion studies of saturated solutions in 50:50 water/ethanol of methyl paraben (MP), ibuprofen (IBU) and caffeine (CF) were performed on Carbosil® membrane. The spectroscopic data were analysed by target factor analysis, and evolution profiles of the signal for each component (i.e. the drug, water, ethanol and membrane) over time were obtained. Results showed that the data were successfully deconvoluted as correlations between factors from the data and reference spectra of the components, were above 0.8 in all cases. Good reproducibility over three runs for the evolution profiles was obtained. From the evolution profiles it was observed that water diffused better through the Carbosil® membrane than ethanol, confirming the hydrophilic properties of the Carbosil® membrane used. IBU diffused slower compared with MP and CF. The evolution profile of CF was very similar to that of water, probably because of the high solubility of CF in water, indicating that both compounds are diffusing concurrently. The second part of the work involved a study of the evolution profiles of the components of a commercial topical gel containing 5% (w/w) of ibuprofen as it permeated through human skin. Although the system was much more complex, data were still successfully deconvoluted and the different components of the formulation identified except for benzyl alcohol which might be attributed to the low concentrations of benzyl alcohol used in topical formulations.
