The three-dimensional structure of Aspergillus niger pectin lyase B (PLB) has been determined by crystallographic techniques at a resolution of 1.7 Å. The model, with all 359 amino acids and 339 water molecules, refines to a final crystallographic R factor of 16.5%. The polypeptide backbone folds into a large right-handed cylinder, termed a parallel β helix. Loops of various sizes and conformations protrude from the central helix and probably confer function. The largest loop of 53 residues folds into a small domain consisting of three antiparallel β strands, one turn of an α helix, and one turn of a 310 helix. By comparison with the structure of Erwinia chrysanthemi pectate lyase C (PelC), the primary sequence alignment between the pectate and pectin lyase subfamilies has been corrected and the active site region for the pectin lyases deduced. The substrate-binding site in PLB is considerably less hydrophilic than the comparable PelC region and consists of an extensive network of highly conserved Trp and His residues. The PLB structure provides an atomic explanation for the lack of a catalytic requirement for Ca2+ in the pectin lyase family, in contrast to that found in the pectate lyase enzymes. Surprisingly, however, the PLB site analogous to the Ca2+ site in PelC is filled with a positive charge provided by a conserved Arg in the pectin lyases. The significance of the finding with regard to the enzymatic mechanism is discussed.

Mammalian hematopoietic stem cell (HSC) commitment and differentiation into lymphoid lineage cells proceed through a series of developmentally restricted progenitor compartments. A complete understanding of this process, and how it differs from HSC commitment and differentiation into cells of the myeloid/erythroid lineages, requires the development of model systems that support HSC commitment to the lymphoid lineages. We now describe a human bone marrow stromal cell culture that preferentially supports commitment and differentiation of human HSC to CD19+ B-lineage cells. Fluorescence activated cell sorterpurified CD34++/lineage-cells were isolated from fetal bone marrow and cultured on human fetal bone marrow stromal cells in serum-free conditions containing no exogenous cytokines. Over a period of 3 weeks, CD34++/lineage- cells underwent commitment, differentiation, and expansion into the B lineage. Progressive changes included: loss of CD34, acquisition of and graded increases in the level of cell surface CD19, and appearance of immature B cells expressing mu/kappa or mu/lambda cell surface Ig receptors. The tempo and phenotype of B-cell development was not influenced by the addition of IL-7 (10 ng/ml), or by the addition of goat anti-IL-7 neutralizing antibody. These results indicate a profound difference between mouse and human in the requirement for IL-7 in normal B-cell development, and provide an experimental system to identify and characterize human bone marrow stromal cell-derived molecules crucial for human B lymphopoiesis.

Induction of cytochrome P4501A1 (CYP1A1) in the hepatoma Hepa1c1c7 cell line results in an elevation in the excretion rate of 8-oxoguanine (oxo8Gua), a biomarker of oxidative DNA damage and the major repair product of 8-oxo-2'-deoxyguanosine (oxo8dG) residues in DNA. Treatment of this cell line with 2,3,7,8-tetrachloro-p-dibenzodioxin (TCDD), a nonmetabolized environmental contaminant, and indolo(3,2-b)carbazole (ICZ), a metabolite of a natural pesticide found in cruciferous vegetables, is shown to both induce CYP1A1 activity and elevate the excretion rate of oxo8Gua; 7,8-benzoflavone (7,8-BF or alpha-naphthoflavone), an inhibitor of CYP1A1 activity and an antagonist of the aryl hydrocarbon (Ah) receptor, reduced the excretion rate of oxo8Gua. The essential role of Ah-receptor, which mediates the induction of CYP1A1, is shown by the inability of TCDD to induce CYP1A1 and to increase excretion of oxo8Gua in Ah receptor-defective c4 mutant cells. While there was a significant 7.0-fold increase over 2 days in the excretion rate of oxo8Gua into the growth medium of TCDD-treated Hepa1c1c7 cells compared to control, no significant increase was detected in the steady-state level of oxo8dG in the DNA presumably due to efficient DNA repair. Thus, the induction of CYP1A1 appears to lead to a leak of oxygen radicals and consequent oxidative DNA damage that could lead to mutation and cancer.

Cytokines are important regulators of hematopoesis. Mutations in gamma c, which is a subunit shared by the receptors for interleukin (IL) 2, IL-4, and IL-7, have been causally associated with human X chromosome-linked severe combined immunodeficiency disease. This finding indicates a mandatory role for cytokine receptor signaling at one or more stages of lymphocyte development. To evaluate the cellular level at which gamma c is critical for lymphopoiesis, the effect of monoclonal antibodies to gamma c on the capacity of syngeneic bone marrow cells to reconstitute the hematopoietic compartment of lethally irradiated recipient mice was examined. We show that monoclonal antibody to gamma c blocked lymphocyte development at or before the appearance of pro-B cells and prior to or at the seeding of the thymus by precursor cells while erythromyeloid cell development was normal. These results suggest that one level of lymphocyte development that requires gamma c is a point in hematopoietic cell differentiation near the divergence of lymphopoiesis and erythromyelopoesis.

O sistema imunológico preserva a integridade do organismo perante o ambiente que ele está inserido. As reações imunológicas são essenciais para controle e eliminação da infecção, no entanto se os mecanismos contra regulatórios da resposta imunológica forem superados, a homeostasia pode falhar levando a um desequilíbrio no processo de reparo do organismo, podendo causar danos, insuficiência de órgão e até a morte. Muitos estudos têm demonstrado a interação entre o sistema imunológico e os aminoácidos. Visto que a glutamina é utilizada como substrato energético para enterócitos, além de fornecer nitrogênio para síntese de purinas e pirimidinas para proliferação celular e a taurina participa da hemostasia, estabilização de membranas, mobilização de cálcio, além de ser importante agente antioxidante, nos propusemos nesse trabalho investigar os efeitos da glutamina e taurina sobre aspectos relacionados a resposta imunológica de células da linhagem Raw 264.7. Para tanto foram avaliadas: viabilidade celular; a capacidade proliferativa e o ciclo celular; a capacidade de síntese de citocinas: IL-1 α, IL-1β, IL-6, IL-10 e TNF-α e a expressão do fator de transcrição NFκB, bem como de seu inibidor IκBα. Foi possível observar aumento da viabilidade e proliferação celular para células tratadas com glutamina, entretanto não foi observado o mesmo efeito para taurina. Quando ambos aminoácidos foram associados, houve prevalência dos efeitos da glutamina. Observamos neste trabalho, que houve uma tendência da diminuição de expressão da relação de p-NFκB/NFκB quando se aumentou a concentração de glutamina. Paralelamente, o mesmo foi encontrado para relação p-IκB/IκB. Esses resultados corroboram com os resultados encontrados na produção das citocinas pró-inflamatórias IL-1α, Il-1β e TNF-α, uma vez que ao aumentarmos a concentração de glutamina bem como glutamina e taurina, observamos menor produção das mesmas. Complementarmente, encontramos resultados opostos para a citocina anti-inflamatória IL-10, a qual teve maior síntese em resposta ao aumento da concentração dos aminoácidos. Portanto concluímos que tanto a glutamina quanto a taurina possuem capacidade de modular aspectos da resposta imunológica de células Raw 264.7.
