Binary probes are oligonucleotide probe pairs that hybridize adjacently to a complementary target nucleic acid. In order to detect this hybridization, the two probes can be modified with, for example, fluorescent molecules, chemically reactive groups or nucleic acid enzymes. The benefit of this kind of binary probe based approach is that the hybridization elicits a detectable signal which is distinguishable from background noise even though unbound probes are not removed by washing before measurement. In addition, the requirement of two simultaneous binding events increases specificity. Similarly to binary oligonucleotide probes, also certain enzymes and fluorescent proteins can be divided into two parts and used in separation-free assays. Split enzyme and fluorescent protein reporters have practical applications among others as tools to investigate protein-protein interactions within living cells. In this study, a novel label technology, switchable lanthanide luminescence, was introduced and used successfully in model assays for nucleic acid and protein detection. This label technology is based on a luminescent lanthanide chelate divided into two inherently non-luminescent moieties, an ion carrier chelate and a light harvesting antenna ligand. These form a highly luminescent complex when brought into close proximity; i.e., the label moieties switch from a dark state to a luminescent state. This kind of mixed lanthanide complex has the same beneficial photophysical properties as the more typical lanthanide chelates and cryptates - sharp emission peaks, long emission lifetime enabling time-resolved measurement, and large Stokes’ shift, which minimize the background signal. Furthermore, the switchable lanthanide luminescence technique enables a homogeneous assay set-up. Here, switchable lanthanide luminescence label technology was first applied to sensitive, homogeneous, single-target nucleic acid and protein assays with picomolar detection limits and high signal to background ratios. Thereafter, a homogeneous four-plex nucleic acid array-based assay was developed. Finally, the label technology was shown to be effective in discrimination of single nucleotide mismatched targets from fully matched targets and the luminescent complex formation was analyzed more thoroughly. In conclusion, this study demonstrates that the switchable lanthanide luminescencebased label technology can be used in various homogeneous bioanalytical assays.

This thesis is about development broadband feed for two-mirror antenna system that match following requirements: beamwidth from 45 to 90 degrees at -3 dB level, circular polarization, absence of radiation to the lower hemisphere area. Literature review was done in the areas of the UWB antennas creation. During the work attempts were made to create a feed in a form of the quad ridged horn and "eleven" antennas. The latter is introduced as the most effective feed among all antennas discussed in thesis. Radiation patterns and other results for "eleven" antenna were obtained. Results were saved as far field sources and placed slightly below focal point into the two-mirror antenna system, because phase center of the “eleven” antenna is predominantly shifted upwards. Directivity patterns for the two-mirror system were obtained and the conclusions about the work results have been made

Différentes méthodes ayant pour objectif une utilisation optimale d'antennes radio-fréquences spécialisées en imagerie par résonance magnétique sont développées et validées. Dans un premier temps, il est démontré qu'une méthode alternative de combinaison des signaux provenant des différents canaux de réception d'un réseau d'antennes mène à une réduction significative du biais causé par la présence de bruit dans des images de diffusion, en comparaison avec la méthode de la somme-des-carrés généralement utilisée. Cette réduction du biais engendré par le bruit permet une amélioration de l'exactitude de l'estimation de différents paramètres de diffusion et de diffusion tensorielle. De plus, il est démontré que cette méthode peut être utilisée conjointement avec une acquisition régulière sans accélération, mais également en présence d'imagerie parallèle. Dans une seconde perspective, les bénéfices engendrés par l'utilisation d'une antenne d'imagerie intravasculaire sont étudiés. Suite à une étude sur fantôme, il est démontré que l'imagerie par résonance magnétique intravasculaire offre le potentiel d'améliorer significativement l'exactitude géométrique lors de mesures morphologiques vasculaires, en comparaison avec les résultats obtenus avec des antennes de surface classiques. Il est illustré qu'une exactitude géométrique comparable à celle obtenue grâce à une sonde ultrasonique intravasculaire peut être atteinte. De plus, plusieurs protocoles basés sur une acquisition de type balanced steady-state free-precession sont comparés dans le but de mettre en évidence différentes relations entre les paramètres utilisés et l'exactitude géométrique obtenue. En particulier, des dépendances entre la taille du vaisseau, le rapport signal-sur-bruit à la paroi vasculaire, la résolution spatiale et l'exactitude géométrique atteinte sont mises en évidence. Dans une même optique, il est illustré que l'utilisation d'une antenne intravasculaire permet une amélioration notable de la visualisation de la lumière d'une endoprothèse vasculaire. Lorsque utilisée conjointement avec une séquence de type balanced steady-state free-precession utilisant un angle de basculement spécialement sélectionné, l'imagerie par résonance magnétique intravasculaire permet d'éliminer complètement les limitations normalement engendrées par l'effet de blindage radio-fréquence de l'endoprothèse.

Pan-viral DNA array (PVDA) and high-throughput sequencing (HTS) are useful tools to identify novel viruses of emerging diseases. However, both techniques have difficulties to identify viruses in clinical samples because of the host genomic nucleic acid content (hg/cont). Both propidium monoazide (PMA) and ethidium bromide monoazide (EMA) have the capacity to bind free DNA/RNA, but are cell membrane-impermeable. Thus, both are unable to bind protected nucleic acid such as viral genomes within intact virions. However, EMA/PMA modified genetic material cannot be amplified by enzymes. In order to assess the potential of EMA/PMA to lower the presence of amplifiable hg/cont in samples and improve virus detection, serum and lung tissue homogenates were spiked with porcine reproductive and respiratory virus (PRRSV) and were processed with EMA/PMA. In addition, PRRSV RT-qPCR positive clinical samples were also tested. EMA/PMA treatments significantly decreased amplifiable hg/cont and significantly increased the number of PVDA positive probes and their signal intensity compared to untreated spiked lung samples. EMA/PMA treatments also increased the sensitivity of HTS by increasing the number of specific PRRSV reads and the PRRSV percentage of coverage. Interestingly, EMA/PMA treatments significantly increased the sensitivity of PVDA and HTS in two out of three clinical tissue samples. Thus, EMA/PMA treatments offer a new approach to lower the amplifiable hg/cont in clinical samples and increase the success of PVDA and HTS to identify viruses.

A dual-port microstrip antenna with a crescent shaped patch with excellent isolation betwecn the ports has been reportcd [I]. Since circular-sided geometries are inore compact than rectangular oncs, thcy find morc applications in microstrip arrays. The crcscent shaped antenna geometry [ I ] provides greater area rcductioii compared to other circular sided patches for broadband operation [2]. In this Lctter, formulac for calculating thc TM, I and TMZI mode resonant frequencies of this microstrip antenna, obtained by modifying the equations of a standard circular patch [3] are presentcd. Thcorctical results are compared with experimental observations aid the validity of the computation is established.

The arrow shaped microstrip antenna, which produces dual frequency dual polarisation operation with considera-ble size reduction compared to conventional patches has been reported [I]. These antennas provide greater area reduction and improved gain compared to drum shaped patches [2]. Prediction of the resonance frequency of drum shaped patches [3] and circular patches for broadband operation [4] are available in the literature. In this Letter, we propose empirical formulas for calculating the resonance frequencies of the arrow shaped microstrip antenna. These antennas can be employed for obtaining dual frequency with the same polarisation, bandwidth enhancement, circular polarisation etc. by varying its different parameters or by introducing slots. The proposed design equations provide an easier and simple way of predicting the resonant frequencies of these patches.

A new microstrip antenna geometry with considerable reduction in size, with similar radiation characteristics to those of an equivalent rectangular patch antenna is proposed. A relationship has been suggested for fmding out the resonant frequency of the new geometry, and its validity has been established by the experimental results. Without increasing the aperture area, this geometry also offers a facility for considerably reducing the resonant frequency compared to conventional patches.

A circular miqrostrip antenna with a modified structure is presented. By adjusting the feed location along the circumference of the patch it is possible to match the antenna with a C microstrip line of any impedance. The impedance bandwidth and radiation characteristics are unaffected by this structural V modification.

The main objective of carrying out this investigation is to develop suitable transducer array systems so that underwater pipeline inspection could be carried out in a much better way, a focused beam and electronic steering can reduce inspection time as well. Better results are obtained by optimizing the array parameters. The spacing between the elements is assumed to be half the wavelength so that the interelement interaction is minimum. For NDT applications these arrays are operated at MHz range. The wavelengths become very small in these frequency ranges. Then the size of the array elements becomes very small, requiring hybrid construction techniques for their fabrication. Transducer elements have been fabricated using PVDF as the active, mild steel as the backing and conducting silver preparation as the bonding materials. The transducer is operated in the (3,3) mode. The construction of a high frequency array is comparatively complicated. The interelement spacing between the transducer elements becomes considerably small when high frequencies are considered. It becomes very difficult to construct the transducer manually. The electrode connections to the elements can produce significant loading effect. The array has to be fabricated using hybrid construction techniques. The active materials has to be deposited on a proper substrate and etching techniques are required to fabricate the array. The annular ring, annular cylindrical or other similar structural forms of arrays may also find applications in the near future in treatments were curved contours of the human body are affected.

A new dual port microstrip antenna geometry for dual frequency operation is presented. The structure consists of the intersection of two circles of the same radius with their centres displaced by a small fraction of the wavelength . This antenna provides wide impedance bandwidth and excellent isolation between its ports. The gain of the antenna is comparable to that of a standard circular microstrip antenna operating at the same resonant frequency. A theoretical analysis for calculating the resonant frequencies of the two ports is also presented

A novel technique for obtaining dual-hand circular polarization (CP) radiation of a single-feed circular microstrip antenna in proposed and demonstrated. By embedding two pain of arc shaped slots of proper lengths close to the boundary of a circular patch, and protruding one of the arc-shaped slots with a narrow slit, the circular microstrip antenna can perform dual-hand CP radiation using a single probe feed. A single-feed rectangular-ring microstrip antenna is proposed for indoor communication under the Bluetooth protocol. The dimensions of the antenna together with the location of the feed point are optimized through field simulations in order to cover the Bluetooth bandwidth and to avoid linear polarization. The performance and the efficiency of the antenna are illustrated in a real indoor environment
