A infecção pelo HTLV-1/2 já foi investigada em diversas populações, mostrando prevalência variável conforme a área geográfica, grupos étnicos e subpopulações analisadas. Em estudos brasileiros foi observada uma maior prevalência nos Estados da Bahia e do Pará. O presente estudo teve como objetivo avaliar a prevalência do HTLV em mulheres grávidas na cidade de Belém, Pará, por meio de técnicas sorológicas e moleculares. Foram coletadas 1.027 amostras de sangue e alíquotas de plasmas foram testadas para a presença de anticorpos anti-HTLV-1/2, utilizando o método imunoenzimático do tipo ELISA. A confirmação da infecção e diferenciação dos tipos e subtipos virais foi realizada por meio da Reação em Cadeia mediada pela Polimerase em duas etapas (Nested PCR) através da amplificação gênica das regiões pX, env e 5’LTR seguido da análise de RFLP e seqüenciamento. Foram detectados seis (0,58%) casos de sororreatividade para o HTLV que posteriormente foram confirmados pela amplificação de um fragmento de 159 pb da região pX. A análise de RFLP com a enzima TaqI mostrou que quatro (66,7%) amostras eram positivas para HTLV-1 e duas (33,3%) para HTLV-2. Uma das amostras HTLV-2 teve o fragmento de 630 pb do gene env amplificado. A análise com a endonuclease XhoI mostrou a presença de um perfil de RFLP característico dos subtipos HTLV-2a/2c. Duas amostras HTLV-1 e uma HTLV-2 tiveram a região 5’LTR amplificada e seqüenciada. Por meio da análise filogenética foi possível classificar as amostras HTLV-1 como Cosmopolita Transcontinental e a HTLV-2 como pertencente ao subtipo HTLV-2c. Esse estudo mostra a relevância de se introduzir o teste para HTLV na triagem pré-natal, a fim de reduzir transmissão vertical e horizontal em nosso estado.

In the title compound, C12H15N3S, the molecule deviates slightly from planarity, with a maximum deviation from the mean plane of the non-H atoms of 0.2756 (6) Å for the S atom and a torsion angle for the N-N-C-N fragment of -7.04 (16)°. In the crystal, molecules are linked by N-H...S hydrogen-bond interactions, forming centrosymmetric dimers. Additionally, one weak intramolecular N-H...N hydrogen-bond interaction is observed. The crystal packing shows a herringbone arrangement viewed along the c axis.

In the search for new therapeutic tools against tuberculosis and to further address the therapeutic potential of pyridine-2-thiol 1-oxide (Hmpo) metal complexes, two new octahedral [M(III)(mpo)3] complexes, with M = Ga or Bi, were synthesized and characterized in the solid state and in solution. Attempts to crystallize [Ga(III)(mpo)3] in CH2Cl2 led to single crystals of the reaction product [GaCl(mpo)2], where the gallium(III) ion is in a square basis pyramidal environment, trans-coordinated at the basis to two pyridine-2-thiolato 1-oxide anions acting as bidentate ligands through their oxygen and sulfur atoms. The biological activity of the new [M(III)(mpo)3] complexes together with that of the previously reported Fe(III) analogous compound and the pyridine-2-thiol 1-oxide sodium salt (Na mpo) was evaluated on Mycobacterium tuberculosis. The compounds showed excellent activity, both in the standard strain H37Rv ATCC 27294 (pan-susceptible) and in five clinical isolates that are resistant to the standard first-line anti-tuberculosis drugs isoniazid and rifampicin. These pyridine-2-thiol 1-oxide derivatives are promising compounds for the treatment of resistant tuberculosis.

The ( Z)-4,4,4-trifluoro-3-(2-hydroxyethylamino)-1-(2-hydroxyphenyl)-2-buten-1-one (C12H12F3NO3) compound was thoroughly studied by IR, Raman, UV-visible, and C-13 and F-19 NMR spectroscopies. The solid-state molecular structure was determined by X-ray diffraction methods. It crystallizes in the P2(1)/c space group with a = 12.1420(4) angstrom, b = 7.8210(3) angstrom, c := 13.8970(5) angstrom, beta = 116.162(2)degrees, and Z = 4 molecules per unit cell. The molecule shows a nearly planar molecular skeleton, favored by intramolecular OH center dot center dot center dot 0 and NH center dot center dot center dot 0 bonds, which are arranged in the lattice as an OH center dot center dot center dot 0 bonded polymer coiled around crystallographic 2-fold screw-axes. The three postulated tautomers were evaluated using quantum chemical calculations. The lowest energy tautomer (I) calculated with density functional theory methods agrees with the observed crystal structure. The structural and conformational properties are discussed considering the effect of the intra- and intermolecular hydrogen bond interactions.

Context: Neolignans are usually dimers formed by oxidative coupling of allyl and propenyl phenols, and the neolignan analogue, 2-phenoxy-1-phenylethanone (LS-2) is a promising antimycobacterial compound showing very weak cytotoxicity in mammalian cells and lack of acute toxicity in murine models. Objectives: To investigate the mechanism of action of LS-2 in rat hepatocytes by evaluating the activity levels of enzymes related to oxidation status and drug-metabolizing activity. Materials and methods: Hepatocytes were treated with LS-2 from 0.05 up to 1 mM, for 24 and 48 h, and reduced glutathione (GSH), lipid peroxidation and cytochrome P450 enzyme (CYP450) activity were assayed. A homologous series of phenoxazone ethers were used as substrates to measure the enzymatic profile. The biotransformation of LS-2 was studied in hepatocytes by gas chromatography-mass spectrometry (GC-MS) for detection and analysis of possible metabolites. Results: Hepatocytes treated with LS-2 up to 1 mM for 24 or 48 h did not induce the formation of GSH and lipid peroxidation. O-Dealkylation activities of the isoenzymes CYP4501A1, CYP4501A2, CYP4502B1 and CYP4502B2 were also not detected in the hepatocytes treated with LS-2 for 24 or 48 h. Discussion and conclusion: The results indicate that LS-2 or its two detected metabolites, 2-phenoxy-1-phenylethanol and 2,4-(2-hydroxy-2-phenylethoxy) phenol, are not cytotoxic to rat hepatocytes. These compounds maintain a balance between the production of pro-oxidant agents and their respective antioxidant systems. The data show that enzymes related to oxidation status and drug-metabolizing activities are not involved in the mechanism of action of LS-2.

Conventional treatment of tuberculosis (TB) demands a long course therapy (6 months), known to originate multiple drug resistant strains (MDR-TB), which emphasizes the urgent need for new antituberculous drugs. The purpose of this study was to investigate a novel treatment for TB meant to improve patient compliance by reducing drug dosage frequency. Polymeric microparticles containing the synthetic analogue of neolignan, 1-phenyl-2-phenoxiethanone (LS-2), were obtained by a method of emulsification and solvent evaporation and chemically characterized. Only representative LS-2-loaded microparticles were considered for further studies involving experimental murine TB induced by Mycobacterium tuberculosis H37Rv ATCC 27294. The LS-2-loaded microparticles were spherical in shape, had a smooth wall and showed an encapsulation efficiency of 93% in addition to displaying sustained release. Chemotherapeutic potential of LS-2 entrapped in microparticles was comparable to control groups. These findings are encouraging and indicate that LS-2-loaded microparticles are a potential alternative to conventional chemotherapy of TB.

The electrochromic behavior of iron complexes derived from tetra-2-pyridyl-1,4-pyrazine (TPPZ) and a hexacyanoferrate species in polyelectrolytic multilayer adsorbed films is described for the first time. This complex macromolecule was deposited onto indium-tin oxide (ITO) substrates via self-assembly, and the morphology of the modified electrodes was studied using atomic force microscopy (AFM), which indicated that the hybrid film containing the polyelectrolyte multilayer and the iron complex was highly homogeneous and was approximately 50 nm thick. The modified electrodes exhibited excellent electrochromic behavior with both intense and persistent coloration as well as a chromatic contrast of approximately 70%. In addition, this system achieved high electrochromic efficiency (over 70 cm(2) C-1 at 630 nm) and a response time that could be measured in milliseconds. The electrode was cycled more than 10(3) times, indicating excellent stability.

PURPOSE: To compare gene expression of the chemokines RANTES and eotaxin-2, its receptor, CCR-3, adhesion molecule ICAM-1 and its receptor LFA-1 in eosinophilic polyps and in control normal nasal mucosa. METHODS: Gene expression was quantified by Real Time PCR in polyps (n=35) and in healthy nasal mucosa (n=15). RESULTS: Eosinophilic polyps showed a higher expression of eotaxin-2 and RANTES, but not of CCR-3, ICAM-1 or LFA-1 compared to control nasal mucosa. CONCLUSION: Eosinophilic polyps present greater expression of eotaxin-2 and RANTES, but not of CCR-3, ICAM-1 or LFA-1 compared to control nasal mucosa.

In order to understand the influence of alkyl side chains on the gas-phase reactivity of 1,4-naphthoquinone derivatives, some 2-hydroxy-1,4-naphthoquinone derivatives have been prepared and studied by electrospray ionization tandem mass spectrometry in combination with computational quantum chemistry calculations. Protonation and deprotonation sites were suggested on the basis of gas-phase basicity, proton affinity, gas-phase acidity (?Gacid), atomic charges and frontier orbital analyses. The nature of the intramolecular interaction as well as of the hydrogen bond in the systems was investigated by the atoms-in-molecules theory and the natural bond orbital analysis. The results were compared with data published for lapachol (2-hydroxy-3-(3-methyl-2-butenyl)-1,4-naphthoquinone). For the protonated molecules, water elimination was verified to occur at lower proportion when compared with side chain elimination, as evidenced in earlier studies on lapachol. The side chain at position C(3) was found to play important roles in the fragmentation mechanisms of these compounds. Copyright (c) 2012 John Wiley & Sons, Ltd.

Die vorliegende Arbeit befasst sich mit der Bedeutung des § 2 Abs. 1 BetrVG für die Reichweite und die Grenzen der Mitbestimmung des Betriebsrates nach § 87 BetrVG.. Ebenso wie im BetrVG 1952 bildet auch im BetrVG 1972 und im BetrVG 2001 der vierte Teil mit den Regelungen über die Mitwirkung und Mitbestimmung der Arbeitnehmer das eigentliche Kernstück des Gesetzes. Zentralvorschrift für die Mitbestimmung des Betriebsrats in sozialen Angelegenheiten ist dabei § 87 BetrVG. Diese Regelung umfasst alle Arbeitsbedingungen, die nicht schon gesetzlich oder tariflich geregelt sind, anderseits aber oft nur einheitlich für alle Arbeitnehmer im Betrieb geregelt werden können. Soweit die Tatbestandsvoraussetzungen der einzelnen Gegenstände in § 87 Abs. 1 Nr. 1 bis Nr. 13 BetrVG erfüllt sind, hat der Betriebsrat eine umfassende funktionelle Zuständigkeit zur Mitbestimmung in sämtlichen sozialen Angelegenheiten. Aufgabe der vorliegenden Arbeit soll es daher sein, Rechtsnatur, Struktur und Anwendungsbereich des Gebots der vertrauensvollen Zusammenarbeit und der Mitbestimmungsrechte gem. § 87 BetrVG zu klären. Hier ist insbesondere zu erörtern, ob Mitbestimmungsrechte des Betriebsrats gem. § 87 Abs. 1 BetrVG außerhalb des Katalogs der gesetzlichen Vorschriften aus dem Grundsatz der vertrauensvollen Zusammenarbeit herzuleiten sind. Ein weiterer Schwerpunkt liegt in der Herausarbeitung der Korrektur- und Schrankenfunktion des § 2 Abs. 1 BetrVG im Bereich der Mitbestimmung nach § 87 Abs. 1 BetrVG.

2-Phenoxyethanol (ethylene glycol monophenyl ether) is used as solvent for cellulose acetate, dyes, inks, and resins; it is a synthetic intermediate in the production of plasticizers, pharmaceuticals, and fragrances. Phenoxyethanol is obtained industrially by reaction of phenol with ethylene oxide, in the presence of an homogeneous alkaline catalyst, typically sodium hydroxide. The yield is not higher than 95-96%, because of the formation of polyethoxylated compounds. However, the product obtained may not be acceptable for use in cosmetic preparations and fragrance formulations, due to presence of a pungent “metallic” odor which masks the pleasant odor of the ether, deriving from residual traces of the metallic catalyst. Here we report a study aimed at using ethylene carbonate in place of ethylene oxide as the reactant for phenoxyethanol synthesis; the use of carbonates as green nucleophilic reactants is an important issue in the context of a modern and sustainable chemical industry. Moreover, in the aim of developing a process which might adhere the principles of Green Chemistry, we avoided the use of solvents, and used heterogeneous basic catalysts. We carried out the reaction by using various molar ratios between phenol and ethylene carbonate, at temperatures ranging between 180 and 240°C, with a Na-mordenite catalyst. Under specific conditions, it was possible to obtain total phenol conversion with >99% yield to phenoxyethanol in few hours reaction time, using a moderate excess of ethylene carbonate. Similar results, but with longer reaction times, were obtained using a stoichiometric feed ratio of reactants. One important issue of the research was finding conditions under which the leaching of Na was avoided, and the catalyst could be separated and reused for several reaction batches.
