Objective: Some previous studies have shown that gingipains, trypsin-like proteases produced by Porphyromonas gingivalis, up-regulate human beta defensin-2 (HBD-2) mRNA expression through protease-activated receptor-2 (PAR(2)) in gingival epithelial cells. This study aimed at investigating salivary HBD-2 levels and crevicular PAR(2) mRNA expression in human chronic periodontitis and evaluating whether periodontal treatment affected this process. Methods: Salivary and gingival crevicular fluid (GCF) samples were collected from periodontally healthy (control) and chronic periodontitis patients at baseline and 50 days after nonsurgical periodontal treatment. Salivary HBD-2, and GCF TNF-alpha levels were analysed by ELISA, and PAR(2) mRNA at the GCF was evaluated by RT-PCR. Results: P. gingivalis was significantly (p < 0.05) more prevalent in patients with chronic periodontitis when compared to controls. This prevalence decreased after periodontal therapy (p < 0.0001). The control group showed statistically significant lower levels of HBD-2, TNF-alpha, and PAR(2) expression when compared to the chronic periodontitis group. In addition, periodontal treatment significantly reduced PAR(2) expression and HBD-2 levels in chronic periodontitis patients (p < 0.001). Conclusions: Our results suggest that salivary HBD-2 levels and PAR(2) mRNA expression from GCF are higher in subjects with chronic periodontitis than in healthy subjects, and that periodontal treatment decreases both HBD-2 levels and PAR(2) expression. Objective. The aim of this study was to investigate the effect of CAPE on the insulin signaling and inflammatory pathway in the liver of mice with high fat diet induced obesity. Material/Methods. Swiss mice were fed with standard chow or high-fat diet for 12-week. After the eighth week, animals in the HFD group with serum glucose levels higher than 200 mg/dL were divided into two groups, HFD and HFD receiving 30 mg/kg of CAPE for 4 weeks. After 12 weeks, the blood samples could be collected and liver tissue extracted for hormonal and biochemical measurements, and insulin signaling and inflammatory pathway analyzes. Results. The high-fat diet group exhibited more weight gain, glucose intolerance, and hepatic steatosis compared with standard diet group. The CAPE treatment showed improvement in glucose sensitivity characterized by an area under glucose curve similar to the control group in an oral glucose tolerance test Furthermore, CAPE treatment promoted amelioration in hepatic steatosis compared with the high-fat diet group. The increase in glucose sensitivity was associated with the improvement in insulin-stimulated phosphorylation of the insulin receptor substrate-2, followed by an increase in Akt phosphorylation. In addition, it was observed that CAPE reduced the induction of the inflammatory pathway, c-jun-N- terminal kinase, the nuclear factor kappa B, and cyclooxygenase-2 expression, respectively. Conclusions. Overall, these findings indicate that CAPE exhibited anti-inflammatory activity that partly restores normal metabolism, reduces the molecular changes observed in obesity and insulin resistance, and therefore has a potential as a therapeutic agent in obesity. We investigate how the initial geometry of a heavy-ion collision is transformed into final flow observables by solving event-by-event ideal hydrodynamics with realistic fluctuating initial conditions. We study quantitatively to what extent anisotropic flow (nu(n)) is determined by the initial eccentricity epsilon(n) for a set of realistic simulations, and we discuss which definition of epsilon(n) gives the best estimator of nu(n). We find that the common practice of using an r(2) weight in the definition of epsilon(n) in general results in a poorer predictor of nu(n) than when using r(n) weight, for n > 2. We similarly study the importance of additional properties of the initial state. For example, we show that in order to correctly predict nu(4) and nu(5) for noncentral collisions, one must take into account nonlinear terms proportional to epsilon(2)(2) and epsilon(2)epsilon(3), respectively. We find that it makes no difference whether one calculates the eccentricities over a range of rapidity or in a single slice at z = 0, nor is it important whether one uses an energy or entropy density weight. This knowledge will be important for making a more direct link between experimental observables and hydrodynamic initial conditions, the latter being poorly constrained at present.

The development of an electroanalytical method for simultaneous determination of copper and lead ions in sugar cane spirit (cachaça) using carbon paste electrode modified with ascorbic acid and carbon nanotubes (CPE-AaCNT) is described. Squarewave voltammetry (SWV) with anodic stripping was employed, and this technique was optimized with respect to the following parameters: frequency (50 Hz), amplitude (100 mV) and scan increment (9 mV). The analytical curves were linear in the range from 0.0900 to 7.00 mg L- 1 for lead and copper. The limits of detection were 48.5 and 23.9 µg L- 1 for lead and copper, respectively. The developed method was applied to the simultaneous determination of copper and lead in five commercial samples of sugar cane spirit. The results were in good agreement with those obtained by F AAS/GF AAS (flame atomic absorption spectrometry/graphite furnace atomic absorption spectrometry) and showed that CPE-AaCNT can be successfully employed in the simultaneous determination of these metals in real sugar cane spirit samples.

After having covered the international regulation in the labour market and in the control mechanisms fighting the irregular and the concealed labour, the Author concentrates on the Italian system’s answers to the requests coming from the European Union. Starting from the White Book on the labour market in Italy, the problems originated by the Legislative Decree 124/2004 and, moreover, those which have affected the fight against the concealed labour are dealt with. The aim study is to verify that the juridical regulation adopted by the national lawmaker has contributed to solve the problems connected to the labour flexibility and the consequent temporary employment. The analysis of the problems has led to the conclusion that, regardless the lawmaker’s good intentions, the basic principles of the national juridical system do not allow the achievement of a full realization of the objectives. This is mainly because the lack of effective systems to protect the flexible employees often treated as temporary employees, and because of the difficulties to introduce the legal culture on which the whole system should stand.

Entscheidend für die Sauerstoffversorgung im ischämischen Gewebe ist die Bildung von Blutgefäßen. Dieser Vorgang findet im erwachsenen Organismus in Form von Arteriogenese, Angiogenese und Vaskulogenese statt. Die Entdeckung, dass endotheliale Progenitorzellen (EPC) aus dem Knochenmark mobilisiert werden können, um sich im Ischämiegebiet an der Bildung neuer Kapillaren zu beteiligen, eröffnet einen vollkommen neuen therapeutischen Ansatz. In der hier vorliegenden Arbeit konnte in drei unterschiedlichen Tiermodellen, dem Matrigelmodell, dem Hinterlaufischämiemodell und dem Infarktmodell der Nacktmaus gezeigt werden, dass eine Zelltherapie mit EPC die Neovaskularisation steigert und zu einer myokardialen Funktionsverbesserung beiträgt. Der entscheidende Beitrag der Arbeit liegt jedoch in der Erforschung des Zeitraums der Wirkung der Stammzelltherapie. In allen drei Tiermodellen konnte durch ein spezifisches Abtöten der mit der viralen Thymidinkinase (TK) transduzierten EPC der positive Effekt auf die Neovaskularisation gestoppt werden. Im Herzinfarktmodell der Nacktmaus kam es sogar zu einer signifikanten Verschlechterung der Herzfunktion sowie zu einer Vergrößerung des Infarktareals. Dieser Effekt war durch Apoptose der Zellen in der dritten und vierten Woche nach Infarkt und Zellinfusion zu beobachten. Somit besitzen EPC nicht nur eine Rolle in der initialen Freisetzung von Zytokinen, sondern tragen auch langfristig zur Aufrechterhaltung des zelltherapeutischen Effektes bei. Ob hierfür allein der Mechanismus der Differenzierung verantwortlich ist, bleibt in weiteren Untersuchungen abzuklären. Denkbar wäre auch eine Beeinflussung des Remodeling über parakrine Langzeiteffekte. Im zweiten Teil der Doktorarbeit wurde versucht, das eingeschränkte zelltherapeutische Potential von Progenitorzellen von Patienten mit „Koronarer Herzkrankheit“ (KHK) und ischämischer Kardiomyopathie mit Hilfe zweier eNOSTranskriptionsverstärker, „eNOS-enhancer“, zu verbessern. Im Matrigelmodell der Maus konnten wir eine Verbesserung des Neovaskularisationspotentials von Knochenmarkszellen (BMC) von Patienten nach Präinkubation mit dem eNOS-enhancer nachweisen. Auch im Myokardinfarktmodell der Maus konnten eine Verbesserung der Herzfunktion und eine Reduktion der Infarktgröße beobachtet werden. Beim direkten Vergleich der beiden eNOS-enhancer konnte kein Unterschied gefunden werden. Zusammenfassend leistet die hier vorliegende Arbeit einen wichtigen Beitrag zum Verständnis für die Bedeutung von Progenitorzellen im Rahmen der Stammzelltherapie nach Myokardinfarkt. Ferner wurde die Möglichkeit aufgezeigt, durch gezielte Beeinflussung der Progenitorzellen ihr therapeutisches Potential signifikant zu steigern.

pegylated liposomal doxorubicin (PLD) and bevacizumab are active agents in the treatment of metastatic breast cancer (MBC). We carried out a multicenter, single-arm phase II trial to evaluate the toxicity and efficacy of PLD and bevacizumab as first-line treatment in MBC patients.
