Binding studies have been performed between amitriptyline and i) native alpha 1-acid glycoprotein (AAG); ii) its desialylated form; iii) its two variants, S-AAG and F-AAG; and iv) a mixture of S-AAG and F-AAG. Scatchard analysis revealed the presence of two classes of binding sites on AAG. For native AAG, the first class (of high affinity) has an association constant (Ka1) of 1.5 x 10(6) L mol-1 and a number of binding sites per mole of protein (n1) of 0.25, while the second class (of low affinity) has a Ka2 of 3.2 x 10(4) L mol-1 and a n2 of 0.94. Similar data were found for desialylated AAG. S-AAG and F-AAG do not differ in their association constants measured with amitriptyline, but in their number of binding sites per mole of protein (n): S-AAG: n1 = 0.56, n2 = 0.52; F-AAG: n1 = 0.17, n2 = 0.71. These results confirm those of a previous study, in which a higher affinity of S-AAG towards various basic drugs in comparison with F-AAG has been found.

Current nuclear medicine techniques for the localization of inflammatory processes are based on injection of 111In labelled autologous granulocytes which need to be isolated and radiolabelled in vitro before reinjection. A new technique is presented here that obviates the need for cell isolation by the direct intravenous injection of a granulocyte specific 123I labelled monoclonal antibody. In this publication the basic parameters of the antibody granulocyte interaction are described. Antibody binding does not inhibit vital functions of the granulocytes, such as chemotaxis and superoxide generation. Scatchard analysis of binding data reveals an apparent affinity of the antibody for granulocytes of 6.8 X 10(9) l/mol and approximately 7.1 X 10(4) binding sites per cell. Due to the high specificity of the antibody, the only expected interference is from CEA producing tumors.

Levels of circulating glucose are tightly regulated. To identify new loci influencing glycemic traits, we performed meta-analyses of 21 genome-wide association studies informative for fasting glucose, fasting insulin and indices of beta-cell function (HOMA-B) and insulin resistance (HOMA-IR) in up to 46,186 nondiabetic participants. Follow-up of 25 loci in up to 76,558 additional subjects identified 16 loci associated with fasting glucose and HOMA-B and two loci associated with fasting insulin and HOMA-IR. These include nine loci newly associated with fasting glucose (in or near ADCY5, MADD, ADRA2A, CRY2, FADS1, GLIS3, SLC2A2, PROX1 and C2CD4B) and one influencing fasting insulin and HOMA-IR (near IGF1). We also demonstrated association of ADCY5, PROX1, GCK, GCKR and DGKB-TMEM195 with type 2 diabetes. Within these loci, likely biological candidate genes influence signal transduction, cell proliferation, development, glucose-sensing and circadian regulation. Our results demonstrate that genetic studies of glycemic traits can identify type 2 diabetes risk loci, as well as loci containing gene variants that are associated with a modest elevation in glucose levels but are not associated with overt diabetes.

BACKGROUND: A device to perform sutureless end-to-side coronary artery anastomosis has been developed by means of stent technology (GraftConnector). The present study assesses the long-term quality of the GraftConnector anastomosis in a sheep model. METHODS: In 8 adult sheep, 40-55 kg in weight, through left anterior thoracotomy, the right internal mammary artery (RIMA) was prepared and connected to the left anterior descending artery (LAD) by means of GraftConnector, on beating heart, without using any stabilizer. Ticlopidine 250 mg/day for anticoagulation for 4 weeks and Aspirin 100 mg/day for 6 months were given. The animals were sacrificed after 6 months and histological examination of anastomoses was carried out after slicing with the connector in situ for morphological analysis. RESULTS: All animals survived at 6 months. All anastomoses were patent and mean luminal width at histology was 1.8 +/- 0.2 mm; mean myotomia hyperplasia thickness was 0.21 +/- 0.1 mm. CONCLUSIONS: Long-term results demonstrate that OPCABGs performed with GraftConnector had 100% patency rate. The mean anastomotic luminal width corresponds to mean LAD's adult sheep diameter. We may speculate that myotomia hyperplasia occurred as a result of local device oversizing.

Purpose: To report on the clinical and electrophysiological findings in a patient with oculo-auricular syndrome due to HMX1 mutation, with a follow-up of 12 years. Background: Oculo-auricular syndrome (MIM: 612109) is a rare developmental recessive condition affecting the eye and external ear that results from a mutation in the HMX1 gene. Previously described ocular abnormalities include bilateral microcornea, posterior synechiae, cataract, chorioretinal colobomas and rod-cone dystrophy. Methods: Retrospective chart review of an affected boy followed over a period of 12 years who had serial complete ophthalmologic examinations, fundus photographs, Goldmann perimetry and full-field electroretinograms (ERG). Results: Initial ERG tracings revealed generalized rod more than cone dysfunction. Thereafter, a rapid deterioration in rod and cone function was detected on follow up ERGs. Conclusion: The retinal degeneration in the recessively inherited oculo-auricular syndrome is a progressive rod-cone dystrophy. Visual prognosis is guarded considering the progressive nature of the retinal dystrophy in early infancy.

Influence of different tropical fruits on biological and behavioral aspects of the Mediterranean fruit fly Ceratitis capitata (Wiedemann) (Diptera, Tephritidae). Studies on Ceratitis capitata, a world fruit pest, can aid the implementation of control programs by determining the plants with higher vulnerability to attacks and plants able to sustain their population in areas of fly distribution. The objective of the present study was to evaluate the influence of eight tropical fruits on the following biological and behavioral parameters of C. capitata: emergence percentage, life cycle duration, adult size, egg production, longevity, fecundity, egg viability, and oviposition acceptance. The fruits tested were: acerola (Malpighia glabra L.), cashew (Anacardium occidentale L.), star fruit (Averrhoa carambola L.), guava (Psidium guajava L.), soursop (Annona muricata L.), yellow mombin (Spondias mombin L.), Malay apple (Syzygium malaccense L.), and umbu (Spondias tuberosa L.). The biological parameters were obtained by rearing the recently hatched larvae on each of the fruit kinds. Acceptance of fruits for oviposition experiment was assessed using no-choice tests, as couples were exposed to two pieces of the same fruit. The best performances were obtained with guava, soursop, and star fruit. Larvae reared on cashew and acerola fruits had regular performances. No adults emerged from yellow mombin, Malay apple, or umbu. Fruit species did not affect adult longevity, female fecundity, or egg viability. Guava, soursop, and acerola were preferred for oviposition, followed by star fruit, Malay apple, cashew, and yellow mombin. Oviposition did not occur on umbu. In general, fruits with better larval development were also more accepted for oviposition.

Los volúmenes de plancton variaron entre 0,09 y 3,15 mL.m-3, promedio 0,76 mL.m-3. El fitoplancton de red fue predominante sólo en 32% de las muestras dentro de 30 mn. A 10 m de profundidad las concentraciones fluctuaron entre 26 x 103 cel.L-1 en Pisco y 4 930 x 103 cel.L-1 en Callao; la diversidad varió entre 0,54 bits.cel-1 en Chicama y 3,03 bits.cel-1 frente a Pisco. El microplancton presentó las mayores densidades seguido del nanoplancton. Protoperidinium obtusum, indicador de ACF estuvo costero desde Punta Falsa hasta San Juan; en Chicama, Pisco y entre Punta Mendieta-Punta Caballas se distribuyó hasta 40 mn. Ceratium praelongum, indicador de ASS presentó una distribución variada entre Paita y Matarani, alcanzó máximo acercamiento a la costa en Chala.

BACKGROUND: Within the frame of a twinning programme with Nicaragua, The La Mascota project, we evaluated in our study the contribution of cytogenetic characterization of acute lymphoblastic leukemia (ALL) as prognostic factor compared to clinical, morphological, and immunohistochemical parameters. METHODS: All patients with ALL treated at the only cancer pediatric hospital in Nicaragua during 2006 were studied prospectively. Diagnostic immunophenotyping was performed locally and bone marrow or blood samples were sent to the cytogenetic laboratory of Zurich for fluorescence in situ hybridization (FISH) analysis and G-banding. RESULTS: Sixty-six patients with ALL were evaluated. Their mean age at diagnosis was 7.3 years, 31.8% were >or=10 years. Thirty-four patients (51.5%) presented with hyperleucocytosis >or=50 x 10(9)/L, 45 (68.2%) had hepatosplenomegaly. Immunophenotypically 63/66 patients (95%) had a B-precursor, 2 (3%) a T- and 1 (1.5%) a B-mature ALL. FISH analysis demonstrated a TEL/AML1 fusion in 9/66 (14%), BCR/ABL fusion in 1 (1.5%), MLL rearrangement in 2 (3.1%), iAMP21 in 2 (3.1%), MYC rearrangement in 1 (1.5%), and high-hyperdiploidy in 16 (24%). All patients but two with TEL/AML1 fusion and high-hyperdiploidy were clinically and hematologically in the standard risk group whereas those with poor cytogenetic factors had clinical high-risk features and were treated intensively. CONCLUSIONS: Compared to Europe, the ALL population in Nicaragua is older, has a higher proportion of poor prognostic clinical and hematological features and receives more intensive treatment, while patients with TEL/AML1 translocations and high-hyperdiploidy are clinically in the standard risk group. Cytogenetics did not contribute as an additional prognostic factor in this setting.

The prevalence of hypertension in African Americans (AAs) is higher than in other US groups; yet, few have performed genome-wide association studies (GWASs) in AA. Among people of European descent, GWASs have identified genetic variants at 13 loci that are associated with blood pressure. It is unknown if these variants confer susceptibility in people of African ancestry. Here, we examined genome-wide and candidate gene associations with systolic blood pressure (SBP) and diastolic blood pressure (DBP) using the Candidate Gene Association Resource (CARe) consortium consisting of 8591 AAs. Genotypes included genome-wide single-nucleotide polymorphism (SNP) data utilizing the Affymetrix 6.0 array with imputation to 2.5 million HapMap SNPs and candidate gene SNP data utilizing a 50K cardiovascular gene-centric array (ITMAT-Broad-CARe [IBC] array). For Affymetrix data, the strongest signal for DBP was rs10474346 (P= 3.6 × 10(-8)) located near GPR98 and ARRDC3. For SBP, the strongest signal was rs2258119 in C21orf91 (P= 4.7 × 10(-8)). The top IBC association for SBP was rs2012318 (P= 6.4 × 10(-6)) near SLC25A42 and for DBP was rs2523586 (P= 1.3 × 10(-6)) near HLA-B. None of the top variants replicated in additional AA (n = 11 882) or European-American (n = 69 899) cohorts. We replicated previously reported European-American blood pressure SNPs in our AA samples (SH2B3, P= 0.009; TBX3-TBX5, P= 0.03; and CSK-ULK3, P= 0.0004). These genetic loci represent the best evidence of genetic influences on SBP and DBP in AAs to date. More broadly, this work supports that notion that blood pressure among AAs is a trait with genetic underpinnings but also with significant complexity.

Whole-grain foods are touted for multiple health benefits, including enhancing insulin sensitivity and reducing type 2 diabetes risk. Recent genome-wide association studies (GWAS) have identified several single nucleotide polymorphisms (SNPs) associated with fasting glucose and insulin concentrations in individuals free of diabetes. We tested the hypothesis that whole-grain food intake and genetic variation interact to influence concentrations of fasting glucose and insulin. Via meta-analysis of data from 14 cohorts comprising ∼ 48,000 participants of European descent, we studied interactions of whole-grain intake with loci previously associated in GWAS with fasting glucose (16 loci) and/or insulin (2 loci) concentrations. For tests of interaction, we considered a P value <0.0028 (0.05 of 18 tests) as statistically significant. Greater whole-grain food intake was associated with lower fasting glucose and insulin concentrations independent of demographics, other dietary and lifestyle factors, and BMI (β [95% CI] per 1-serving-greater whole-grain intake: -0.009 mmol/l glucose [-0.013 to -0.005], P < 0.0001 and -0.011 pmol/l [ln] insulin [-0.015 to -0.007], P = 0.0003). No interactions met our multiple testing-adjusted statistical significance threshold. The strongest SNP interaction with whole-grain intake was rs780094 (GCKR) for fasting insulin (P = 0.006), where greater whole-grain intake was associated with a smaller reduction in fasting insulin concentrations in those with the insulin-raising allele. Our results support the favorable association of whole-grain intake with fasting glucose and insulin and suggest a potential interaction between variation in GCKR and whole-grain intake in influencing fasting insulin concentrations.

Foi realizado um experimento em casa de vegetação para avaliar os efeitos da inoculação de fungos micorrízicos arbusculares (FMAs) sobre o crescimento e nutrição de mudas de gravioleira (Annona muricata L.) em Latossolo Amarelo álico, fumigado e não fumigado. A inoculação foi feita durante o transplante; após cinco meses foram avaliados o crescimento e a absorção de nutrientes das mudas. A inoculação aumentou significativamente o crescimento da gravioleira no solo fumigado;o maior incremento foi observado nos tratamentos com Scutellospora heterogama e Gigaspora margarita. A eficiência da inoculação no solo fumigado variou de 594% a 1.348%. Embora o efeito da inoculação tenha sido reduzido no solo não fumigado, G. margarita, Entrophospora colombiana e Gigaspora sp. aumentaram significativamente o crescimento das mudas, e a eficiência da inoculação de 61% foi obtida com G. margarita; S. heterogama não se mostrou eficiente. No solo fumigado, G. margarita e Gigaspora sp. promoveram maior porcentagem de colonização radicular; no entanto, a colonização radicular não diferiu entre tratamentos com inoculação no solo não fumigado. Os teores e conteúdos de nutrientes variaram entre os tratamentos. A gravioleira é uma planta responsiva a FMAs, e a inoculação da espécie G. margarita beneficia o desenvolvimento das mudas de gravioleira nos solos fumigado e não fumigado.

A expansão da fruticultura no Nordeste brasileiro pode sofrer limitações, já que algumas áreas com elevado potencial de cultivo são abastecidas por água de qualidade insatisfatória. Considerando a potencialidade de cultivo da gravioleira (Annona muricata L.) na região e o fato de não existir dados sobre a produção de mudas enxertadas dessa espécie, sob estresse salino, conduziu-se este trabalho com o objetivo de avaliar os efeitos da salinidade da água de irrigação sobre o tipo 'Crioula' tendo 'Morada' como porta-enxerto. A água de irrigação de condutividade elétrica entre 0,5 e 5,5 dS m-1 foi preparada mantendo-se proporção equivalente a 7:2:1, entre Na:Ca:Mg, respectivamente; o delineamento foi em blocos casualizados, com quatro repetições, 54 plantas por parcela. Foram avaliados: germinação, formação do porta-enxerto (número de folhas, altura de plantas, diâmetro do caule, matéria seca de parte aérea e de raiz, área foliar e número de plantas aptas à enxertia) e número de enxertos vivos. A porcentagem de germinação da gravioleira não foi afetada pelo estresse salino, porém a velocidade de emergência e o acúmulo de fitomassa foram reduzidos com aumento da salinidade da água de irrigação. Pode-se usar água com até 1,5 dS m-1 de condutividade elétrica na formação de mudas de graviola.

O objetivo deste trabalho foi avaliar a diversidade genética em uma população de 148 híbridos de tangor 'Murcott' (Citrus reticulata Blanco x C. sinensis L. Osbeck) e laranja 'Pêra' (C. sinensis L. Osbeck) obtidos por polinização controlada, pelo uso de marcadores fAFLP e RAPD. Marcadores polimórficos (416 marcadores fAFLP e 33 RAPD) foram utilizados para avaliar a similaridade genética entre os híbridos, calculada com o coeficiente Jaccard pelo método UPGMA. A consistência de cada agrupamento foi determinada pelo programa BOOD. Houve alta similaridade genética entre os parentais. A laranja 'Pêra' apresentou maior número (132) de loci em heterozigose em relação ao tangor 'Murcott' (105), corroborando a teoria de origem híbrida para a laranja-doce. Observaram-se dois grupos distintos de plantas, e um deles abrangeu 80% dos híbridos com maior similaridade com a laranja 'Pêra'. A análise bootstrap não revelou consistência estatística entre esses grupos. Marcadores fAFLP são mais eficientes na avaliação do polimorfismo, sendo indicados para seleção de indivíduos híbridos mais próximos a um dos parentais.

Este trabalho teve por objetivo avaliar mudanças físicas, químicas e bioquímicas relacionadas ao amaciamento de graviola (Annona muricata L.) Crioula durante a maturação. Os frutos foram colhidos no estádio de maturidade fisiológica, em plantas cultivadas na Estação Experimental da Embrapa Agroindústria Tropical, em Pacajus, Ceará, armazenados a 26,3±0,6 ºC e 88±12% de umidade relativa, durante 1, 2, 3, 4 e 5 dias. Utilizou-se o delineamento experimental inteiramente casualizado, com quatro repetições. As variáveis analisadas foram: perda de massa, amido, pectina total e solúvel, e atividades das enzimas amilase, pectinametilesterase, poligalacturonase, alfa-galactosidases e beta-galactosidases. Não houve variação significativa no conteúdo de pectina solúvel. Após cinco dias, a perda de massa atingiu 5%, mas não promoveu murcha dos frutos. As reduções nos conteúdos de amido e de pectina total coincidiram com período de expressiva atividade, respectivamente, das enzimas amilase e da poligalacturonase e beta-galactosidase de parede celular. As modificações mais acentuadas no conteúdo de amido, de pectina total, na solubilização de pectinas e na atividade das enzimas amilase, pectinametilesterase, poligalacturonase e beta-galactosidase de parede celular ocorreram no período do segundo ao quarto dia após a colheita.

O objetivo deste trabalho foi comparar formas de análise de medidas repetidas para o melhoramento da produção de frutos de pinha (Annona squamosa). Vinte progênies de meias-irmãs foram avaliadas por três anos (2003, 2004 e 2005) em delineamento de blocos ao acaso, com cinco repetições, com cada parcela constituída de quatro plantas. A característica avaliada foi o número de frutos por indivíduo. Os modelos de simetria composta, de simetria composta com variâncias heterogêneas, autorregressivo com variâncias heterogêneas, e antedependência estruturada, foram analisados com o programa ASReml. A estimação dos componentes de variância e a predição dos valores genéticos foram feitas com o procedimento REML/BLUP. A comparação dos modelos foi realizada pelo teste de razão de verossimilhança e pelo critério de Akaike. O modelo antedependência estruturada, para os fatores progênie e parcela, e o modelo multivariado, para o fator resíduo, são as melhores abordagens para a análise dos dados, pois propiciam eficiência e parcimônia em relação ao modelo multivariado completo. Com o modelo antedependência estruturada, é possível a identificação de famílias superiores, em cada colheita, e também de famílias com maior número total de frutos.
