Leptospirosis is a public health problem worldwide and its etiology remains unclear. Its pathogenesis involves a complex interaction between host and infecting microorganism. The inflammatory reaction that controls the infection process also underscores many pathophysiological events occurring in leptospirosis. We investigated the presence of tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) in renal tissues by immunohistochemical and histopathological examination in animals experimentally inoculated with Leptospira serovar Canicola. All the tests were carried out 2, 7, 14, 21 or 28 days after inoculation. Although TNF-alpha and IL-6 had been detected in tissues throughout the observation period, these cytokines appeared more intensely during the initial phase of infection. Therefore, both TNF-alpha and IL-6 were associated with the immunopathogenesis of leptospirosis. This profile suggests a high immunocellular response throughout the early infection stages followed by subsequent humoral response.

The action of man has led, over the years, major impacts on the environment, especially in aquatic ecosystems, leading to an impairment of water quality, considered one of the essential factors for the maintenance of vital functions and consequently the life of the planet. Among the activities considered a risk for the environment are linked to pollution from many sources and even entire watersheds, whether by industrial waste, sewage, or for substances used in agriculture, such as pesticides, herbicides and fertilizers. The (2,4-D) 2,4- dichlorophenoxyacetic acid is used worldwide, and the fact that its genotoxicity is proven by several studies and by its long persistence in soil, which enables the leaching and percolation of compounds affecting water bodies, toxicity studies are relevant and justifiable. Thus, this study aimed to evaluate the toxicity of 2,4-D by examining the liver of the fish Oreochromis niloticus exposed to different dilutions. Portions of liver were collected and fixed for histological and histochemical techniques to detect total proteins, polysaccharides and lipids. lipids. Treatment with 2,4-D herbicide apparently did not alter the lipid profiles, the accumulation of polysaccharides, and the presence of total proteins. The 2.5 and 5.0% were lethal to fish. These mortalities are probably of high toxic and cytotoxic potential of 2,4-D herbicide results. Several histopathological changes were found, such as: loss of cytoplasmic integrity, loss of cell limit, nuclear deformation, vacuolated cytoplasm, tissue disorganization and hydropic degeneration. Statistically significant changes were: hydropic degeneration and vacuolated cytoplasm. It is concluded, therefore, that the qualitative morphological analysis is an important method for observing changes in liver toxicology studies. Background: Ventral root avulsion is an experimental model of proximal axonal injury at the central/peripheral nervous system interface that results in paralysis and poor clinical outcome after restorative surgery. Root reimplantation may decrease neuronal degeneration in such cases. We describe the use of a snake venom-derived fibrin sealant during surgical reconnection of avulsed roots at the spinal cord surface. The present work investigates the effects of this fibrin sealant on functional recovery, neuronal survival, synaptic plasticity, and glial reaction in the spinal motoneuron microenvironment after ventral root reimplantation. Methodology/Principal Findings: Female Lewis rats (7 weeks old) were subjected to VRA and root replantation. The animals were divided into two groups: 1) avulsion only and 2) replanted roots with fibrin sealant derived from snake venom. Post-surgical motor performance was evaluated using the CatWalk system twice a week for 12 weeks. The rats were sacrificed 12 weeks after surgery, and their lumbar intumescences were processed for motoneuron counting and immunohistochemistry (GFAP, Iba-1 and synaptophysin antisera). Array based qRT-PCR was used to evaluate gene regulation of several neurotrophic factors and receptors as well as inflammatory related molecules. The results indicated that the root reimplantation with fibrin sealant enhanced motor recovery, preserved the synaptic covering of the motoneurons and improved neuronal survival. The replanted group did not show significant changes in microglial response compared to VRA-only. However, the astroglial reaction was significantly reduced in this group. Conclusions/Significance: In conclusion, the present data suggest that the repair of avulsed roots with snake venom fibrin glue at the exact point of detachment results in neuroprotection and preservation of the synaptic network at the microenvironment of the lesioned motoneurons. Also such procedure reduced the astroglial reaction and increased mRNA levels to neurotrophins and anti-inflammatory cytokines that may in turn, contribute to improving recovery of motor function.

Members of the subfamily Crotalinae are considered to be essentially nocturnal and most of the data about these snakes have been collected from the field. Information on how nutritional status affects the movement rate and activity patterns is a key point to elucidating the ecophysiology of snakes. In this study, we distributed 28 lancehead Bothrops moojeni into three groups under distinct feeding regimens after a month of fasting. Groups were divided as follows: ingestion of meals weighing (A) 40%, (B) 20%, or (C) 10% of the snake body mass. Groups were monitored for five days before and after food intake and the activity periods and movement rates were recorded. Our results show that B. moojeni is prevalently nocturnal, and the activity peak occurs in the first three hours of the scotophase. After feeding, a significant decrease in activity levels in groups A and B was detected. The current results corroborate previous field data that describe B. moojeni as a nocturnal species with low movement rates. The relationship between motion and the amount of food consumed by the snake may be associated with its hunting strategy.

Surprisingly little is known of the toxic arsenal of cnidarian nematocysts compared to other venomous animals. Here we investigate the toxins of nematocysts isolated from the jellyfish Olindias sambaquiensis. A total of 29 unique ms/ms events were annotated as potential toxins homologous to the toxic proteins from diverse animal phyla, including conesnails, snakes, spiders, scorpions, wasp, bee, parasitic worm and other Cnidaria. Biological activities of these potential toxins include cytolysins, neurotoxins, phospholipases and toxic peptidases. The presence of several toxic enzymes is intriguing, such as sphingomyelin phosphodiesterase B (SMase B) that has only been described in certain spider venoms, and a prepro-haystatin P-IIId snake venom metalloproteinase (SVMP) that activates coagulation factor X, which is very rare even in snake venoms. Our annotation reveals sequence orthologs to many representatives of the most important superfamilies of peptide venoms suggesting that their origins in higher organisms arise from deep eumetazoan innovations. Accordingly, cnidarian venoms may possess unique biological properties that might generate new leads in the discovery of novel pharmacologically active drugs.

A evolução do veneno, uma das misturas mais complexas da natureza, tem sustentado o sucesso da diversificação de inúmeras linhagens de animais. Serpentes deslizantes ou medusas flutuantes utilizam o veneno, um coquetel de peptídeos farmacologicamente ativos, sais e moléculas orgânicas. Esses animais surpreendentes têm provocado grande fascínio ao longo da história humana. Nesta dissertação propomos um estudo da evolução dos venenos no filo Cnidaria, englobando dados proteômicos e genômicos. Este projeto teve como objetivos: (1) caracterizar e elucidar a evolução da composição do veneno em Cnidaria por meio da comparação de listas de proteínas; (2) testar a hipótese de que a variação na família de toxinas específica de cnidários tem sido o resultado de um regime de seleção positiva; e (3) determinar a extensão em que a duplicação de genes pode ser considerada como a principal razão para a diversificação de toxinas em Cnidaria. O capítulo \"Comparative proteomics reveals common components of a powerful arsenal in the earliest animal venomous lineage, the cnidarians\" propõe o estudo comparado mais completo sobre a composição do veneno de cnidários e uma hipótese sobre a montagem evolutiva do complexo arsenal bioquímico de cnidários e do veneno ancestral desse grupo basal. Vinte e oito famílias de proteínas foram identificadas. Destas, 13 famílias foram registradas pela primeira vez no proteoma de Cnidaria. Pelo menos 15 famílias de toxinas foram recrutadas no proteoma de veneno de cnidários antes da diversificação dos grupos Anthozoa e Medusozoa. Nos capítulos \"Evidence of episodic positive selection in the evolution of jellyfish toxins of the cnidarian venom\" e \"Gene duplications are extensive and contribute significantly to the toxic proteome of nematocysts isolated from Acropora digitifera (Cnidaria: Anthozoa: Scleractinia)\", nossas análises demonstram que as famílias de toxinas nos cnidários se diversificam amplamente mediante a duplicação de genes. Além disso, em contraste com as famílias de toxinas do veneno na maioria das linhagens animais; nós identificamos um padrão diferente na família de toxinas específica de cnidários, em que há uma seleção purificadora por longos períodos seguindo longos tempos de diversificação ou vice-versa

En la provincia de Córdoba existen 35 especies de serpientes de las cuales solamente cinco son consideradas venenosas y de riesgo para la salud humana; se trata de las yararás chica (Bothrops diporus), grande (Bothrops alternatus) y ñata (Bothrops ammodytoides), la cascabel (Crotalus durissus) y la coral (Micrurus pyrrhocryptus). Estas cinco especies, conocidas comúnmente como “víboras”, se distribuyen por el oeste de nuestra provincia. Los accidentes producidos por mordeduras de serpientes venenosas representan en dicha provincia una importante problemática y los grupos afectados son trabajadores rurales, hacheros y carboneros del monte, mineros, personal que trabaja en obras viales, policías, bomberos, turistas y personas que van de campamento (Reati, 2013). Las estadísticas indican que anualmente se producen en Argentina unos 500 casos anuales, de los cuales 70-90 corresponden a la Provincia de Córdoba. Las continuas intervenciones humanas sobre las áreas silvestres por urbanizaciones, realización de obras de infraestructura, el crecimiento de las actividades agrícolas y turísticas, entre otras acciones, aumentan las probabilidades de encuentro entre personas y serpientes, lo que provoca mayor frecuencia de picaduras, como ha ocurrido en muchos países. A su vez, las serpientes –venenosas o no- se enfrentan con serios problemas de conservación debido a las alteraciones ambientales ya mencionadas, el comercio para la utilización del cuero de algunas especies, el mascotismo y fundamentalmente la mortandad en manos del hombre, que las mata indiscriminadamente por temor sin diferenciar si son o no peligrosas. Existe un gran sector de la población que no tiene posibilidad de acceder a la formación en estas problemáticas y la escuela constituye uno de los espacios más importantes para desarrollar esta tarea por su rol en la construcción de conocimientos. Sin embargo, los profesores de nivel medio no disponen de material didáctico desde una perspectiva específicamente regional para desarrollar temas curriculares sobre aspectos biológicos y ecológicos de la fauna de ofidios de la provincia de Córdoba y los problemas de conservación que deben afrontar, como tampoco acerca de la prevención de accidentes ofídicos y los primeros auxilios que se deben realizar en caso de mordeduras de serpientes. Estas circunstancias se contraponen con la ley provincial N° 8113 donde se incluyen, dentro del sistema educativo, aspectos inherentes a conservación, brindando también una serie de lineamientos pedagógicos que establecen la regionalización del currículo; además, asigna especial importancia al medio natural y al contexto sociocultural del alumno. Se presenta aquí como propuesta la producción de material teórico-práctico desde un enfoque transdisciplinar, de manera tal que la temática que aquí se plantea, por su complejidad, pueda ser abordada no solo desde las Ciencias Naturales, sino también desde otras ciencias, ya que abarca no solamente aspectos biológicos y ecológicos de los ofidios, sino también cuestiones relacionadas con la conservación de la biodiversidad y con la salud humana.
