The present study was designed to compare the homeostasis model assessment (HOMA) and quantitative insulin sensitivity check index (QUICKI) with data from forearm metabolic studies of healthy individuals and of subjects in various pathological states. Fifty-five healthy individuals and 112 patients in various pathological states, including type 2 diabetes mellitus, essential hypertension and others, were studied after an overnight fast and for 3 h after ingestion of 75 g of glucose, by HOMA, QUICKI and the forearm technique to estimate muscle uptake of glucose combined with indirect calorimetry (oxidative and non-oxidative glucose metabolism). The patients showed increased HOMA (1.88 ± 0.14 vs 1.13 ± 0.10 pmol/l x mmol/l) and insulin/glucose (I/G) index (1.058.9 ± 340.9 vs 518.6 ± 70.7 pmol/l x (mg/100 ml forearm)-1), and decreased QUICKI (0.36 ± 0.004 vs 0.39 ± 0.006 (µU/ml + mg/dl)-1) compared with the healthy individuals. Analysis of the data for the group as a whole (patients and healthy individuals) showed that the estimate of insulin resistance by HOMA was correlated with data obtained in the forearm metabolic studies (glucose uptake: r = -0.16, P = 0.04; non-oxidative glucose metabolism: r = -0.20. P = 0.01, and I/G index: r = 0.17, P = 0.03). The comparison of QUICKI with data of the forearm metabolic studies showed significant correlation between QUICKI and non-oxidative glucose metabolism (r = 0.17, P = 0.03) or I/G index (r = -0.37, P < 0.0001). The HOMA and QUICKI are good estimates of insulin sensitivity as data derived from forearm metabolic studies involving direct measurements of insulin action on muscle glucose metabolism.

This study aimed to evaluate the use of rosemary (Rosmarinus officinalis) extract (RE), celery (Apium graveolis), and low levels of NO3 and NO2 as natural agents to enhance the quality of colonial salami. Salami was produced according to three treatments: (A) Control: 0.1% curing salt; (B) Rosemary: 0.05% curing salt + 0.5% RE (rosemary extract); and (C) Rosemary+celery: 0.14% Veg 503 + 0.27% Veg 504 (sea salt plus celery) + 0.5% of RE (rosemary extract). There was no effect (P > 0.05) of the treatments on water activity, Na content, and residual NO3 and NO2. Fatty acids C18:2 and C20:4 were reduced (P < 0.05) during the ripening period in the Control treatment indicating possible oxidation. The use of celery resulted in lower pH values (P < 0.05) in the salami. Reduced addition of NO3 and NO2 resulted in salami lighter in color (higher L* values, P < 0.05) at the 12th day of ripening. In conclusion, celery-based products proved to be an effective source of NO2 and NO3 for color development, but the low pH of the product indicates the need for better evaluation of its use in fermented salami. The RE (rosemary extract) reduced fat oxidation in salami, but this needs further evaluation.

La pharmacopée Cris est riche en plantes médicinales et plusieurs d’entre elles sont étudiées par notre laboratoire pour leur potentiel antidiabétique. Certaines espèces ont démontré leur capacité à stimuler la protéine kinase activée par l’AMP (AMPK), une enzyme qui favorise la translocation de transporteurs de glucose à la membrane (effet hypoglycémiant). L’AMPK stimule également d’autres fonctions, telle l’oxydation des graisses, dans le but de rétablir l’énergie cellulaire. Ce projet a comme objectifs d’évaluer, premièrement, le stress métabolique induit par huit des extraits dans des cellules musculaires et des hépatocytes, effet qui serait responsable de l’activation de l’AMPK. Ce stress peut être déterminé en mesurant l’acidification du milieu extracellulaire ainsi que la déplétion du contenu en ATP des cellules suite aux traitements. Le deuxième objectif est de mesurer l’efficacité des extraits à réduire le contenu en gras (oxydation des graisses) et à ainsi normaliser la résistance à l’insuline dans des hépatocytes rendus insulino-résistants. Les hépatocytes sont rendus résistants à l’insuline (condition fortement lié à l’obésité) via traitement avec un acide gras saturé, le palmitate. Les résultats montrent que la majorité des extraits semble induire un stress métabolique de courte durée dans les cellules. Parmi les extraits, seul un a réussi à faire diminuer significativement le taux de triglycérides intracellulaire suite au traitement au palmitate sans toutefois améliorer la sensibilité à l’insuline. En conclusion, le potentiel hypoglycémiant des extraits serait du à leur capacité à affecter la respiration mitochondriale (stress métabolique). Toutefois, leur capacité à améliorer la sensibilité à l’insuline n’a pu être établie.

L’augmentation de la dépense énergétique (DE) par une augmentation de l'activité physique (AP) participe au maintien et à l’amélioration de la santé. La mesure à grande échelle de la DE totale (DET) en général et AP en particulier se heurte à des difficultés pratiques de recueil de données, de validité et aux coûts. Bien que dans la littérature de nombreux accéléromètres permettent d’estimer la DET, il y a encore des limites quant à la mesure de l’intensité de l’AP, élément qui influence l’état de la balance énergétique et le bénéfice pour la santé de l’AP. De plus, peu de comparaisons entre les différents accéléromètres sont disponibles. La présente étude avait pour but d’évaluer la capacité de deux accéléromètres (Actical et Sensewear Armband) pour estimer la DET en comparaison avec la technique de l’eau doublement marquée (EDM) ; d’évaluer la concordance entre les accéléromètres dans la mesure de la DE au repos (DER) en comparaison avec la technique de la calorimétrie indirecte (CI) et d’évaluer la DE liée à l’AP en comparaison avec la technique de la CI. Les résultats montrent qu’il y a une très bonne corrélation entre la CI et les accéléromètres dans la mesure de la DER(r > 0.80, p<0.001) et une bonne corrélation entre les accéléromètres et l’EDM dans la mesure de la DET(r>0.75, p<0.001). Pour la mesure de l’intensité de l’AP, l’ACT est plus précis (r=0.80, p<0.001)) que le SWA à la marche, le SWA est meilleur(r=0.80, p<0.001) au vélo. Cette étude permet d’affirmer que le SWA et l’ACT sont des alternatives valables pour mesurer la DE.

Antioxidants are substances that when present at low concentrations compared to that of an oxidisable substrate significantly delays or inhibits oxidation of that substrate in food products or in living systems. Antioxidants are either endogenous to the body or derived from the diet. Several types of synthetic antioxidants like BHT, BHA, TBHQ etc. are also used in the food industry. However, findings and subsequent publicity has fostered significant consumer resistance to the use of synthetic food additives as antioxidants, colourants etc. and therefore food industry is in search of potential natural antioxidants from edible sources.The major dietary sources of antioxidant phytochemicals are cereals, legumes, fruits, vegetables, oilseeds, beverages, spices and herbs. In the present study, we have focused on rice bran and its byproducts. Rice is one of the oldest of food crops and has been a staple food in India from very ancient times. It is also the staple food for about 60% of the world's population. Rice bran is a byproduct of the rice milling industry and is a potential commercial source of a healthy edible oil viz. rice bran oil and a variety of bio-active phytochemicals.Defatted rice bran (DRB), a byproduct of rice bran oil extraction, is also a good source of insoluble dietary fiber, protein, phytic acid, inosito I, vitamin B and a variety of other phytochemicals. Though the antioxidant potential of DRB has been demonstrated, it still remained a relatively unexplored source material, which demanded further investigation especially with regard to its detailed phytochemical profile leading to practical application. The focus of the present investigation therefore has been on DRB primarily to establish its phytochemical status and feasibility of using it as a source of bio-active phytochemicals and natural antioxidants leading to value addition of DRB otherwise used as cattle feed. To gain a better understanding of the value of rice bran as a source of phytochemicals, five popular rice varieties of the region viz. PTB 50, PTB 39, PTB 38, JA Y A, and MO 10 and a wild variety (oryza nivara) that is mainly used for medicinal applications in traditional ayurvedic system were characterized along with commercial samples of rice bran. The present study also explains the feasibility of a process for the extraction, enrichment, and isolation of antioxidant compounds from DRB. The antioxidant potential of the extracts were evaluated both in bulk oils and in food relevant model emulsions, using standard in vitro models. Radical scavenging effects, indicative of possible biological effects, were also evaluated.

El inadecuado aporte nutricional en los pacientes con enfermedades criticas, ha llevado al desarrollo de complicaciones que incrementan la mortalidad y los costos de la atención en salud. Muchos factores están involucrados en el consumo de los nutrientes por el organismo, como: los traslados, las intervenciones quirúrgicas, el uso de vasopresores, la ventilación mecánica, entre otros. Si se presenta imprecisión en la de-terminación del gasto energético, puede conllevar a un sobre aporte alimenticio en el paciente el cual puede afectar la evolución y pronosti-co del individuo. Es de conocimiento universal que los costos en salud cada día se in-crementan, en especial cuando se presenta requerimiento del manejo de un paciente en la unidad de cuidados intensivos. Para predecir el gasto energético basal de los paciente en las UCI se cuentan con herramientas de evaluación sencillas, de fácil uso y económicas, como la ecuación de Harris-Benedict, o herramientas complejas y de difícil manejo como la Calorimetría Indirecta. El incremento en la demanda de servicios con mayor tecnología en el tratamiento de los pacientes, enfrenta al personal de salud para ser más crítico en el uso de la nueva tecnología, por tal motivo, se evaluó la presencia de correlación entre las ecuaciones de Harris-Benedict y Calorimetría Indirecta, encontrando que se presenta una buena correlación entre las ecuaciones, con un valor de Pearson de 0,700 y una p = 0.002. Por lo que se puede concluir que las ecuaciones pueden ser utilizadas para estimar el gasto energético basal de los pacientes en la UCI.

Avaliaram-se 20 cordeiros machos ¾ mestiços Suffolk submetidos a dietas isoprotéicas (21% proteína bruta) e isoenergéticas (2,9 Mcal EM/kg MS), nas formas farelada e peletizada, e abatidos com 26 e 28 kg de peso vivo. Os cordeiros tiveram acesso ao creep feeding, permanecendo com as ovelhas até atingirem o peso de abate. A ração peletizada promoveu maior rendimento de carcaça quente (54,18% para ração peletizada e 52,04% para ração farelada) e maior rendimento verdadeiro (58,37% peletizada e 56,66% farelada). O peso vivo final não interferiu nos pesos das carcaças quente e fria (12,79 e 13,05 kg para 26 e 28 kg de peso vivo final, respectivamente). Não houve interferência da forma física da ração e do peso vivo final dos animais na área do olho de lombo e no comprimento do lombo. A largura do lombo e a máxima espessura de gordura foram maiores nas carcaças dos animais que receberam ração peletizada e a mínima espessura foi inferior nos animais abatidos com 26 kg de peso vivo (1,48 e 1,77 cm para os cordeiros abatidos com 26 e 28 kg, respectivamente). Concluiu-se que o uso da ração peletizada proporcionou aos cordeiros melhoria dos índices de rendimento de carcaça quente, rendimento biológico, profundidade do lombo e índice de compacidade da carcaça. O peso vivo de 26 kg apresentou-se como o mais satisfatório para o abate de cordeiros, por apresentar características de carcaça semelhantes àqueles abatidos com 28 kg.

OBJETIVOS: Realizou-se um estudo transversal para avaliar a taxa de metabolismo de repouso (TMR) e condições socioeconômicas em 15 crianças escolares do sexo feminino; eutróficas (EU= estatura/idade 3 95% e peso/idade entre 90-110%) e 15 com desnutrição pregressa (DP= estatura/idade < 95% e peso/estatura entre 90-110%) moradoras em favelas no município de São Paulo. MÉTODOS: Avaliou-se a TMR por calorimetria indireta, e a situação socioeconômica por entrevista domiciliar. RESULTADOS: O grupo DP apresentou TMR mais alta quando expressa por unidade de peso corpóreo (EU= 40,5 Kcal/kg/dia; DP=44,4 Kcal/kg/dia, p<0,05) e por quilograma de massa magra (EU= 49,2 Kcal/kg/dia; DP=52,5 Kcal/kg/dia, p<0,05); e diferenças significantes para renda per capita, analfabetismo materno, número de parasitas por criança, número de ordem entre os filhos e número de irmãos. em análise multivariada as variáveis associadas à desnutrição foram renda per capita e analfabetismo materno. CONCLUSÕES: Embora os dois grupos tenham peso/estatura normais, a presença de baixa estatura leve foi acompanhada por alterações metabólicas e socioeconômicas típicas de um quadro de desnutrição.

The pyrH-encoded uridine 5′-monophosphate kinase (UMPK) is involved in both de novo and salvage synthesis of DNA and RNA precursors. Here we describe Mycobacterium tuberculosis UMPK (MtUMPK) cloning and expression in Escherichia coli. N-terminal amino acid sequencing and electrospray ionization mass spectrometry analyses confirmed the identity of homogeneous MtUMPK. MtUMPK catalyzed the phosphorylation of UMP to UDP, using ATP-Mg 2+ as phosphate donor. Size exclusion chromatography showed that the protein is a homotetramer. Kinetic studies revealed that MtUMPK exhibits cooperative kinetics towards ATP and undergoes allosteric regulation. GTP and UTP are, respectively, positive and negative effectors, maintaining the balance of purine versus pyrimidine synthesis. Initial velocity studies and substrate(s) binding measured by isothermal titration calorimetry suggested that catalysis proceeds by a sequential ordered mechanism, in which ATP binds first followed by UMP binding, and release of products is random. As MtUMPK does not resemble its eukaryotic counterparts, specific inhibitors could be designed to be tested as antitubercular agents. © 2010 Elsevier Inc. All rights reserved.

Tuberculosis remains as one of the main cause of mortality worldwide due to a single infectious agent, Mycobacterium tuberculosis. The aroK-encoded M. tuberculosis Shikimate Kinase (MtSK), shown to be essential for survival of bacilli, catalyzes the phosphoryl transfer from ATP to the carbon-3 hydroxyl group of shikimate (SKH), yielding shikimate-3-phosphate and ADP. Here we present purification to homogeneity, and oligomeric state determination of recombinant MtSK. Biochemical and biophysical data suggest that the chemical reaction catalyzed by monomeric MtSK follows a rapid-equilibrium random order of substrate binding, and ordered product release. Isothermal titration calorimetry (ITC) for binding of ligands to MtSK provided thermodynamic signatures of non-covalent interactions to each process. A comparison of steady-state kinetics parameters and equilibrium dissociation constant value determined by ITC showed that ATP binding does not increase the affinity of MtSK for SKH. We suggest that MtSK would more appropriately be described as an aroL-encoded type II shikimate kinase. Our manuscript also gives thermodynamic description of SKH binding to MtSK and data for the number of protons exchanged during this bimolecular interaction. The negative value for the change in constant pressure heat capacity (ΔCp) and molecular homology model building suggest a pronounced contribution of desolvation of non-polar groups upon binary complex formation. Thermodynamic parameters were deconvoluted into hydrophobic and vibrational contributions upon MtSK:SKH binary complex formation. Data for the number of protons exchanged during this bimolecular interaction are interpreted in light of a structural model to try to propose the likely amino acid side chains that are the proton donors to bulk solvent following MtSK:SKH complex formation. © 2013 Rosado et al.
