Objectives: Combination microbicide vaginal rings may be more effective than single microbicide rings at reducing/preventing sexual transmission of HIV. Here, we report the preclinical development and macaque pharmacokinetics of matrix-type silicone elastomer vaginal rings containing dapivirine and darunavir.

Methods: Macaque rings containing 25 mg dapivirine, 300 mg darunavir and 100 mg dapivirine, and 300 mg darunavir were manufactured and characterised by differential scanning calorimetry. In vitro release was assessed into isopropanol/water and simulated vaginal fluid. Macaque vaginal fluid and blood serum concentrations for both antiretrovirals were measured during 28-day ring use. Tissue levels were measured on day 28. Ex vivo challenge studies were performed on vaginal fluid samples and IC50 values calculated.

Results: Darunavir caused a concentration-dependent reduction in the dapivirine melting temperature in both solid drug mixes and in the combination ring. In vitro release from rings was dependent on drug loading, the number of drugs present, and the release medium. In macaques, serum concentrations of both microbicides were maintained between 101–102 pg/mL. Vaginal fluid levels ranged between 103–104 ng/g and 104–105 ng/g for dapivirine and darunavir, respectively. Tissue concentrations ranges for each drug were: vagina (1.8×103–3.8×103 ng/g) > cervix (9.4×101–3.9×102 ng/g) > uterus (0–108 ng/g) > rectum (0–40 ng/g). Measured IC50 values were > 2 ng/mL for both compounds.

Conclusions: Based on these results, and in light of recent clinical progress of the 25mg dapivirine ring, a combination vaginal ring containing dapivirine and darunavir is a viable second-generation HIV microbicide candidate.

Submerged reefs are important recorders of palaeo-environments and sea-level change, and provide a substrate for modern mesophotic (deep-water, light-dependent) coral communities. Mesophotic reefs are rarely, if ever, described from the fossil record and nothing is known of their long-term record on Great Barrier Reef (GBR). Sedimentological and palaeo-ecological analyses coupled with 67 14C AMS and U–Th radiometric dates from dredged coral, algae and bryozoan specimens, recovered from depths of 45 to 130 m, reveal two distinct generations of fossil mesophotic coral community development on the submerged shelf edge reefs of the GBR. They occurred from 13 to 10 ka and 8 ka to present. We identified eleven sedimentary facies representing both autochthonous (in situ) and allochthonous (detrital) genesis, and their palaeo-environmental settings have been interpreted based on their sedimentological characteristics, biological assemblages, and the distribution of similar modern biota within the dredges. Facies on the shelf edge represent deep sedimentary environments, primarily forereef slope and open platform settings in palaeo-water depths of 45–95 m. Two coral–algal assemblages and one non-coral encruster assemblage were identified: 1) Massive and tabular corals including Porites, Montipora and faviids associated with Lithophylloids and minor Mastophoroids, 2) platy and encrusting corals including Porites, Montipora and Pachyseris associated with melobesioids and Sporolithon, and 3) Melobesiods and Sporolithon with acervulinids (foraminifera) and bryozoans. Based on their modern occurrence on the GBR and Coral Sea and modern specimens collected in dredges, these are interpreted as representing palaeo-water depths of < 60 m, < 80–100 m and > 100 m respectively. The first mesophotic generation developed at modern depths of 85–130 m from 13 to 10.2 ka and exhibit a deepening succession of < 60 to > 100 m palaeo-water depth through time. The second generation developed at depths of 45–70 m on the shelf edge from 7.8 ka to present and exhibit stable environmental conditions through time. The apparent hiatus that interrupted the mesophotic coral communities coincided with the timing of modern reef initiation on the GBR as well as a wide-spread flux of siliciclastic sediments from the shelf to the basin. For the first time we have observed the response of mesophotic reef communities to millennial scale environmental perturbations, within the context of global sea-level rise and environmental changes.

One of the basic aspects of some neural networks is their attempt to approximate as much as possible their biological counterparts. The goal is to achieve a simple and robust network, easy to understand and able of simulating the human brain at a computational level. Recently a third generation of neural networks (NN) [1], called Spiking Neural Networks(SNN) was appeared. This new kind of networks use the time of a electrical pulse, or spike, to encode the information. In the first and second generation of NN analog values are used in the communication between neurons.

We report the exploration of some unique metabolic pathways in Perkinsus olseni a marine protist parasite, responsible to significant mortalities in mollusks, especially in bivalves all around the world. In Algarve, south of Portugal carpet shell clam Ruditapes decussatus mortalities can reach up to 70%, causing social and economic losses. The objective of studying those unique pathways, is finding new therapeutic strategies capable of controlling/eliminating P. olseni proliferation in clams. In that sense metabolic pathways, were explored, and drugs affecting these cycles were tested for activity. The first step involved the identification of the genes behind those pathways, the reconstitution of the main steps, and molecular characterization of those genes and later on, the identification of possible targets within the genes studied. Metabolic cycles were screened due to the fact of not being present in host or differ in a critical way, such as the following pathways: shikimate, MEP-­‐ isoprenoids, Leloir cycle for chitin production, purine biosynthesis (unique among protists), the de novo synthesis of folates (absent in metazoa) and some unique genes like, the alternative oxidase (a branch of respiratory chain) and the hypoxia sensor HPH. All those pathways were covered and possible chemical inhibition using therapeutic drugs was tested with positive results. The relation between the common host Ruditapes decussatus and P. olseni was also explored in a dimension not possible some years ago. With the accessibility to second generation sequencers and microarray analysis platforms, genes involved in host defense or parasite virulence and resistance to the host were deciphered, allowing aiming to new targets (mechanisms and pathways), offering new possibilities for the control of Perkinsus in close environments. The thousands of genes, generated by this work, sequenced and analyzed from this commercial valuable clam and for Perkinsus olseni will be an important and value tool for the scientific community, allowing a better understanding of host-­‐parasite interactions, promoting the usage of P. olseni as an emerging model for alveolata parasites.

Against Beck’s claims that conventional sociological concepts and categories are zombie categories, this paper argues that Durkheim’s theoretical framework in which suicide is a symptom of an anomic state of society can help us understand the diversity of trajectories that transnational migrants follow and that shape their suicide rates within a cosmopolitan society. Drawing on ethnographic data collected on eight suicides and three attempted suicide cases of second-generation male Alevi Kurdish migrants living in London, this article explains the impact of segmented assimilation/adaptation trajectories on the incidence of suicide and how their membership of a ‘new rainbow underclass’, as a manifestation of cosmopolitan society, is itself an anomic social position with a lack of integration and regulation.

Contemporaneamente o Homem depara-se com um dos grandes desafios que é o de efetivar a transição para um futuro sustentável. Assim, o setor da energia tem um papel fundamental neste processo de transição, com principal enfoque no setor dos automóveis, sendo este um setor que contribui com elevadas quantidades de gases de efeito estufa libertados para a atmosfera. Também a escassez dos recursos petrolíferos constitui um ponto fundamental no tema apresentado. Com a necessidade de combater esses problemas é que se tem vindo a tentar desenvolver combustíveis renováveis e neutros quanto às emissões. A primeira geração de biocombustíveis obtidos através de culturas agrícolas terrestres preenche em parte esses requisitos, porém, não atinge os valores da procura e ainda competem com a produção de alimentos. Daí o interesse na aposta de uma segunda geração de biocombustíveis produzidos de fontes que não pertencem à cadeia alimentar e são residuais mas, que mesmo assim não permitem satisfazer as necessidades de matériaprima. A terceira geração de biocombustíveis vem justamente responder a estas questões pois assenta em matérias-primas que não competem pela utilização do solo agrícola nem são usadas para fins alimentares, tendo produtividades areais substancialmente superiores às que as culturas convencionais ou biomassas residuais conseguem assegurar. A matéria prima de terceira geração são portanto as microalgas, cujas produtividades em biomassa são extremamente elevadas, para além de produtividades muito superiores em lípidos, hidratos de carbono e/ou outros produtos de valor elevado. No entanto, este tipo de produção de biocombustível ainda enfrenta alguns problemas técnicos que o tornam num processo dispendioso para competir economicamente com outros tipos de produção de biodiesel. Na linha do que foi dito anteriormente, este trabalho apresenta um estudo de viabilidade económica e energética do biodiesel produzido através da Chlorella vulgaris, apresentando as técnicas e resultados de cultivo da Chlorella vulgaris e posteriormente de produção do biodiesel através dos lípidos obtidos através da mesma. Para melhorar a colheita das microalgas, que é uma das fases mais dispendiosas, testou-se o aumento de pH e a adição de um floculante (Pax XL-10), sendo que o primeiro não permitiu obter resultados satisfatórios, enquanto o segundo permitiu obter resultados de rendimento na ordem dos 90%. Mesmo com a melhoria da etapa da colheita, o preço mínimo do biodiesel produzido a partir do óleo de Chlorella vulgaris, com as condições ótimas de cultivo e produtividades máximas encontradas na literatura, foi de 8,76 €/L, pois, na análise económica, o Pax XL-10 revelou-se extremamente caro para utilizar na floculação de microalgas para obtenção de um produto de baixo valor, como é o biodiesel. A não utilização da floculação reduz o preço do biodiesel para 7,85 €/L. O que se pode concluir deste trabalho é que face às técnicas utilizadas, a produção de biodiesel Chlorella vulgaris apenas, não é economicamente viável, pelo que para viabilizar a sustentabilidade do processo seria ainda necessário desenvolver mais esforços no sentido de otimizar a produção de biodiesel, eventualmente associando-a à produção de um outro biocombustível produzido a partir da biomassa extraída residual e/ou da recuperação de outros produtos de maior valor.

There is no complete overview or discussion of the literature of the economics of federalism and fiscal decentralization, even though scholarly interest in the topic has been increasing significantly over recent years. This paper provides a general, brief but comprehensive overview of the main insights from the literature on fiscal federalism and decentralization. In doing so, literature on fiscal federalism and decentralization is grouped into two main approaches: “first generation of theories” and “second generation of theories”.

This contribution aims at exploring the significance of the new generation of UNESCO conventions for the recognition of higher education qualifications. It discusses three possible scenarios and links them to the empirical findings of a study that compares the enabling conditions of the first generation of recognition conventions established in the 1970s and 1980s with the ones establishing the second generation today. Taking an interdisciplinary approach, the paper argues that the changes illustrate a more general shift in the architecture of the global order and highlights a new role of UNESCO.

To assess the preferred methods to quit smoking among current smokers. Cross-sectional, population-based study conducted in Lausanne between 2003 and 2006 including 988 current smokers. Preference was assessed by questionnaire. Evidence-based (EB) methods were nicotine replacement, bupropion, physician or group consultations; non-EB-based methods were acupuncture, hypnosis and autogenic training. EB methods were frequently (physician consultation: 48%, 95% confidence interval (45-51); nicotine replacement therapy: 35% (32-38)) or rarely (bupropion and group consultations: 13% (11-15)) preferred by the participants. Non-EB methods were preferred by a third (acupuncture: 33% (30-36)), a quarter (hypnosis: 26% (23-29)) or a seventh (autogenic training: 13% (11-15)) of responders. On multivariate analysis, women preferred both EB and non-EB methods more frequently than men (odds ratio and 95% confidence interval: 1.46 (1.10-1.93) and 2.26 (1.72-2.96) for any EB and non-EB method, respectively). Preference for non-EB methods was higher among highly educated participants, while no such relationship was found for EB methods. Many smokers are unaware of the full variety of methods to quit smoking. Better information regarding these methods is necessary.

The Rankin Inlet area, on the west shore of Hudson Bay in the Northwest Territories, is in the Churchill Structural Province. Metamorphosed volcanic and sedimentary rocks, previously mapped as Archean and part of the Kaminak Group, underlie most of the area. The Rankin Inlet Group consists of greywacke, with minor conglomeratic greywacke, quartzite and dolomite, overlain by massive and pillowed basaltic flows. Gabbro sills intrude the sediments near the base of the volcanic sequence and three serpentinite sills outcrop at the base of the volcanic sequence. The sediments are in fault-contact with quartz monzonite to the south and were intruded by granitic rocks to the northwest. Two periods of folding were defined by the mapping. The first generation folds are recumbent isoclinal folds, with northwest-trending and northeast-dipping axial planes, formed through gravitational sliding. The second generation folds are symmetrically disposed about the axis of the granitic intrusion and have east-southeast trending and nearly vertical axial planes. Whole-rock analysis of 64 rock samples indicates that metasomatic alteration accompanied the intrusion of both the granitic rocks and the serpentinite. The volcanic rocks, gabbro and serpentinite were derived from a magma of oceanic tholeiitic affinities. The stratigraphic sequence and chemistry of the volcanic rocks of the Rankin Inlet Group indicate that this assemblage is correlative with the Hurwitz Group rather than the Kaminak Group and is therefore Aphebian in age.

The present thesis outlines our latest findings on the reactivity of the Burgess reagent with oxiranes. Structural, mechanistic, and computational studies are presented. Included is the development of a (-)-menthyl version of the Burgess reagent and its application to the synthesis of enantiomerically pure ~-amino alcohols. This methodology has been exploited in the formal enantiodivergent synthesis of the (+)- and (-)-isomers of balanol. Also described is a second generation approach to both balanol enantiomers; each commencmg with the chemoenzymatic dihydroxylation of bromobenzene. This study also describes the steric and functional limitations of the toluene dioxygenase-mediated oxidation of benzoate esters. The metabolite derived from ethyl benzoate was employed in a formal synthesis of oseltamivir. Finally, several synthetic approaches to oseltamivir and its analogs are presented, each proceeding through a different vinyl aziridine derived from bromobenzene and ethyl benzoate.

ABSTRACT Photosystem II (PSII) of oxygenic photosynthesis has the unique ability to photochemically oxidize water, extracting electrons from water to result in the evolution of oxygen gas while depositing these electrons to the rest of the photosynthetic machinery which in turn reduces CO2 to carbohydrate molecules acting as fuel for the cell. Unfortunately, native PSII is unstable and not suitable to be used in industrial applications. Consequently, there is a need to reverse-engineer the water oxidation photochemical reactions of PSII using solution-stable proteins. But what does it take to reverse-engineer PSII’s reactions? PSII has the pigment with the highest oxidation potential in nature known as P680. The high oxidation of P680 is in fact the driving force for water oxidation. P680 is made up of a chlorophyll a dimer embedded inside the relatively hydrophobic transmembrane environment of PSII. In this thesis, the electrostatic factors contributing to the high oxidation potential of P680 are described. PSII oxidizes water in a specialized metal cluster known as the Oxygen Evolving Complex (OEC). The pathways that water can take to enter the relatively hydrophobic region of PSII are described as well. A previous attempt to reverse engineer PSII’s reactions using the protein scaffold of E. coli’s Bacterioferritin (BFR) existed. The oxidation potential of the pigment used for the BFR ‘reaction centre’ was measured and the protein effects calculated in a similar fashion to how P680 potentials were calculated in PSII. The BFR-RC’s pigment oxidation potential was found to be 0.57 V, too low to oxidize water or tyrosine like PSII. We suggest that the observed tyrosine oxidation in BRF-RC could be driven by the ZnCe6 di-cation. In order to increase the efficiency of iii tyrosine oxidation, and ultimately oxidize water, the first potential of ZnCe6 would have to attain a value in excess of 0.8 V. The results were used to develop a second generation of BFR-RC using a high oxidation pigment. The hypervalent phosphorous porphyrin forms a radical pair that can be observed using Transient Electron Paramagnetic Resonance (TR-EPR). Finally, the results from this thesis are discussed in light of the development of solar fuel producing systems.

Cette thèse porte sur l’amélioration des techniques d’imagerie à haut-contraste permettant la détection directe de compagnons à de faibles séparations de leur étoile hôte. Plus précisément, elle s’inscrit dans le développement du Gemini Planet Imager (GPI) qui est un instrument de deuxième génération pour les télescopes Gemini. Cette caméra utilisera un spectromètre à champ intégral (SCI) pour caractériser les compagnons détectés et pour réduire le bruit de tavelure limitant leur détection et corrigera la turbulence atmosphérique à un niveau encore jamais atteint en utilisant deux miroirs déformables dans son système d’optique adaptative (OA) : le woofer et le tweeter. Le woofer corrigera les aberrations de basses fréquences spatiales et de grandes amplitudes alors que le tweeter compensera les aberrations de plus hautes fréquences ayant une plus faible amplitude. Dans un premier temps, les performances pouvant être atteintes à l’aide des SCIs présentement en fonction sur les télescopes de 8-10 m sont investiguées en observant le compagnon de l’étoile GQ Lup à l’aide du SCI NIFS et du système OA ALTAIR installés sur le télescope Gemini Nord. La technique de l’imagerie différentielle angulaire (IDA) est utilisée pour atténuer le bruit de tavelure d’un facteur 2 à 6. Les spectres obtenus en bandes JHK ont été utilisés pour contraindre la masse du compagnon par comparaison avec les prédictions des modèles atmosphériques et évolutifs à 8−60 MJup, où MJup représente la masse de Jupiter. Ainsi, il est déterminé qu’il s’agit plus probablement d’une naine brune que d’une planète. Comme les SCIs présentement en fonction sont des caméras polyvalentes pouvant être utilisées pour plusieurs domaines de l’astrophysique, leur conception n’a pas été optimisée pour l’imagerie à haut-contraste. Ainsi, la deuxième étape de cette thèse a consisté à concevoir et tester en laboratoire un prototype de SCI optimisé pour cette tâche. Quatre algorithmes de suppression du bruit de tavelure ont été testés sur les données obtenues : la simple différence, la double différence, la déconvolution spectrale ainsi qu’un nouvel algorithme développé au sein de cette thèse baptisé l’algorithme des spectres jumeaux. Nous trouvons que l’algorithme des spectres jumeaux est le plus performant pour les deux types de compagnons testés : les compagnons méthaniques et non-méthaniques. Le rapport signal-sur-bruit de la détection a été amélioré d’un facteur allant jusqu’à 14 pour un compagnon méthanique et d’un facteur 2 pour un compagnon non-méthanique. Dernièrement, nous nous intéressons à certains problèmes liés à la séparation de la commande entre deux miroirs déformables dans le système OA de GPI. Nous présentons tout d’abord une méthode utilisant des calculs analytiques et des simulations Monte Carlo pour déterminer les paramètres clés du woofer tels que son diamètre, son nombre d’éléments actifs et leur course qui ont ensuite eu des répercussions sur le design général de l’instrument. Ensuite, le système étudié utilisant un reconstructeur de Fourier, nous proposons de séparer la commande entre les deux miroirs dans l’espace de Fourier et de limiter les modes transférés au woofer à ceux qu’il peut précisément reproduire. Dans le contexte de GPI, ceci permet de remplacer deux matrices de 1600×69 éléments nécessaires pour une séparation “classique” de la commande par une seule de 45×69 composantes et ainsi d’utiliser un processeur prêt à être utilisé plutôt qu’une architecture informatique plus complexe.

Les acides biliaires sont des composés naturels existants dans le corps humain. Leur biocompatibilité, leur caractère amphiphile et la rigidité de leur noyau stéroïdien, ainsi que l’excellent contrôle de leurs modifications chimiques, en font de remarquables candidats pour la préparation de matériaux biodégradables pour le relargage de médicaments et l'ingénierie tissulaire. Nous avons préparé une variété de polymères à base d’acides biliaires ayant de hautes masses molaires. Des monomères macrocycliques ont été synthétisés à partir de diènes composés de chaînes alkyles flexibles attachées à un noyau d'acide biliaire via des liens esters ou amides. Ces synthèses ont été réalisées par la fermeture de cycle par métathèse, utilisant le catalyseur de Grubbs de première génération. Les macrocycles obtenus ont ensuite été polymérisés par ouverture de cycle, entropiquement induite le catalyseur de Grubbs de seconde génération. Des copolymères ont également été préparés à partir de monolactones d'acide ricinoléique et de monomères cycliques de triester d’acide cholique via la même méthode. Les propriétés thermiques et mécaniques et la dégradabilité de ces polymères ont été étudiées. Elles peuvent être modulées en modifiant les différents groupes fonctionnels décorant l’acide biliaire et en ayant recours à la copolymérisation. La variation des caractéristiques physiques de ces polymères biocompatibles permet de moduler d’autres propriétés utiles, tel que l’effet de mémoire de forme qui est important pour des applications biomédicales.
