The main objective of this study is to determine the activity of Po-210, Pb-210 and gross alpha/beta radioactivity in groundwater collected in Botucatu, municipality of the São Paulo state, Brazil. Samples were collected in wells to perform the radiochemical analysis. Most wells are registered in DAEE, Sabesp or CPRM. The activity of polonium was obtained by the method of alpha spectrometry, held in LABIDRO at UNESP, Rio Claro. All samples showed radioactivity values below the maximum allowed by WHO, that are: gross alpha radioactivity = 0,5 Bq/L, gross beta radioactivity = 1,0 Bq/L, Po210 = 0,2 Bq/L and Pb210 = 0,1 Bq/L. The results of hydrochemical analysis were compared with the 1469 Ordinance of the Health Ministry, and all values are bellow the maximum allowed. The data also allowed obtain the samples classification using the Piper diagram. The samples AAB, Quinta do Manacás, Faz. Quatro Irmãs, Sesi, Sitio São José and ITE are classified as calcic bicarbonate; the samples Staroup and Caio are potassic; the samples Banespa and BTC are bicarbonate and mixed interms of dissolved cations; Sitio das Palmeiras sample is sodium bicarbonate; the sample Faz. São Paulo is chlorinated calcic; the sample Sítio Btu-Pardinho chlorinated and mixed interms of dissolved cations

Nuclear medicine is a medical specialty related to imagery that deals with imaging techniques, diagnosis and therapy, allowing observing the physiological state of tissues noninvasively by marking the molecules participating of these physiological processes with radioactive isotopes, thus creating the called radionuclides. The image of a radionuclide is one of the most important applications of radioactivity in nuclear medicine. The equipment’s of nuclear medicine imaging use the principle of radiation detection, turning it into an electrical signal which, through specific algorithms, allows forming tomographic images that provide information about the functional status of organs. New detection systems have been developed for tomographic acquisitions using solid state detectors. These devices use crystals of cadmium zinc telluride (CdZnTe). Some of the advantages of this detector are a significant improvement of signal to noise ratio, the increased spectral and spatial resolution, which in sum, result in greater clarity of the images obtained, opening new perspectives for imaging protocols previously unattainable. In contrast, all other gamma-cameras equipped with vacuum tubes have remained relatively unchanged for nearly fifty years. In these gamma-cameras, the images are obtained using two steps significantly less efficient: the gamma rays are converted to light through a first device, and then the light is converted into an electrical signal through a second device. One of functions the Medical Physicist is related to the quality control of equipment. This control ensures that the information and images provided are true and thus credible to be used in medical reports. This study aimed to discuss the recent contributions of History of Science to theteaching of chemistry. The theme of the teaching unit was on the accident of cesium-137 in Goiânia ? Goiás, Brazil (1987). This was a qualitative study in which researcherand professor use the methodology of teaching on research. Thus, students should seekinformation, organize the data collected and establish relationships. Students producedposters in an exhibition at the school on the twentieth anniversary of the accident.Results demonstrated the importance of this resource. It was possible to exploit theabundance of detail and illustrations utilized what themes they students considerimportant and which they exploited. A poster exhibition on the theme indicated thestrong interest of students to participate in all activities proposed. The study alsorevealed the importance of using elements of the History of Science in ChemicalEducation and its relationship to the daily life of students.Key words: history of science; radioactivity; cesium-137

Several reefs of the world have undergone changes in community due to sedimentation processes. It has been suggested that Abrolhos reefs (Brazil/South-West Atlantic) are subjected to a steady coastal influence, although there is still little information regarding this assumption. In this work, we have analyzed a set of environmental parameters concerning sedimentation characteristics at the Abrolhos reefs, near 18 degrees S-39 degrees W. The analysis included remote sensing, model and in situ data to provide a three-dimensional quantitative description of the processes that influence sediment apportionment to the reefs. Mineralogy and natural radioactivity of sediment trapped at three reef sites in a transect perpendicular to the coastline were used in conjunction with numerical weather prediction model and remote sensing databases. We have observed an increase of around 100% of sediment flux during the summer compared to the winter season. A comparison of regional rainfall regime, sediment plume dynamics and a year-around monitoring of polar fronts trajectories and surface wind showed that the wind-driven resedimentation due to polar front activity is the major contributor to the intensification of sedimentation processes at the offshore area of Abrolhos reefs, despite river runoff from mainland. OBJECTIVE: Glucose intolerance is frequently associated with an altered plasma lipid profile and increased cardiovascular disease risk. Nonetheless, lipid metabolism is scarcely studied in normolipidemic glucose-intolerant patients. The aim of this study was to investigate whether important lipid metabolic parameters, such as the kinetics of LDL free and esterified cholesterol and the transfer of lipids to HDL, are altered in glucose-intolerant patients with normal plasma lipids. METHODS: Fourteen glucose-intolerant patients and 15 control patients were studied; none of the patients had cardiovascular disease manifestations, and they were paired for age, sex, race and co-morbidities. A nanoemulsion resembling a LDL lipid composition (LDE) labeled with C-14-cholesteryl ester and H-3-free cholesterol was intravenously injected, and blood samples were collected over a 24-h period to determine the fractional clearance rate of the labels by compartmental analysis. The transfer of free and esterified cholesterol, triglycerides and phospholipids from the LDE to HDL was measured by the incubation of the LDE with plasma and radioactivity counting of the supernatant after chemical precipitation of non-HDL fractions. RESULTS: The levels of LDL, non-HDL and HDL cholesterol, triglycerides, apo A1 and apo B were equal in both groups. The 14 C-esterified cholesterol fractional clearance rate was not different between glucose-intolerant and control patients, but the H-3-free- cholesterol fractional clearance rate was greater in glucose-intolerant patients than in control patients. The lipid transfer to HDL was equal in both groups. CONCLUSION: In these glucose-intolerant patients with normal plasma lipids, a faster removal of LDE free cholesterol was the only lipid metabolic alteration detected in our study. This finding suggests that the dissociation of free cholesterol from lipoprotein particles occurs in normolipidemic glucose intolerance and may participate in atherogenic signaling.

Background: High-density-lipoprotein (HDL) has several antiatherogenic properties and, although the concentration of HDL-cholesterol negatively correlates with incidence of coronary artery disease (CAD), this is not sufficient to evaluate the overall HDL protective role. The aim was to investigate whether precocious CAD patients show abnormalities in lipid transfers to HDL, a fundamental step in HDL metabolism and function. Methods: Thirty normocholesterolemic CAD patients aged <50 y and 30 controls paired for sex, age and B.M.I. were studied. Fasting blood samples were collected for the in vitro lipid transfer assay and plasma lipid determination. A donor nanoemulsion labeled with radioactive free-cholesterol. cholesteryl esters, phospholipids and triglycerides was incubated with whole plasma and after chemical precipitation of non-HDL fractions, supernatant was counted for radioactivity in HDL. Results: LDL and HDL-cholesterol and triglycerides were equal in both groups. Transfers of free-cholesterol (3.8 +/- 1.2%vs 7.0 +/- 33%,p<0.0001) and triglycerides (3.7 +/- 1.7%vs 4.9 +/- 1.9%, p = 0.0125) were diminished in CAD patients whereas cholesteryl ester transfer increased (6.5 +/- 1.9%vs 4.8 +/- 1.8%, p = 0.0008); phospholipid transfer was equal (17.8 +/- 3.5% vs19.5 +/- 3.9%). Conclusion: Alterations in the transfer of lipids to HDL may constitute a new marker for precocious CAD and relation of this metabolic alteration with HDL antiatherogenic function should be investigated in future studies. Pharmaceuticals are useful tools to prevent and treat human and animal diseases. Following administration, a significant fraction of pharmaceuticals is excreted unaltered into faeces and urine and may enter the aquatic ecosystem and agricultural soil through irrigation with recycled water, constituting a significant source of emerging contaminants into the environment. Understanding major factors influencing their environmental fate is consequently needed to value the risk, reduce contamination, and set up bioremediation technologies. The antiviral drug Tamiflu (oseltamivir carboxylate, OC) has received recent attention due to the potential use as a first line defence against H5N1 and H1N1 influenza viruses. Research has shown that OC is not removed during conventional wastewater treatments, thus having the potential to enter surface water bodies. A series of laboratory experiments investigated the fate and the removal of OC in surface water systems in Italy and Japan and in a municipal wastewater treatment plant. A preliminary laboratory study investigated the persistence of the active antiviral drug in water samples from an irrigation canal in northern Italy (Canale Emiliano Romagnolo). After an initial rapid decrease, OC concentration slowly decreased during the remaining incubation period. Approximately 65% of the initial OC amount remained in water at the end of the 36-day incubation period. A negligible amount of OC was lost both from sterilized water and from sterilized water/sediment samples, suggesting a significant role of microbial degradation. Stimulating microbial processes by the addition of sediments resulted in reduced OC persistence. Presence of OC (1.5 μg mL-1) did not significantly affect the metabolic potential of the water microbial population, that was estimated by glyphosate and metolachlor mineralization. In contrast, OC caused an initial transient decrease in the size of the indigenous microbial population of water samples. A second laboratory study focused on basic processes governing the environmental fate of OC in surface water from two contrasting aquatic ecosystems of northern Italy, the River Po and the Venice Lagoon. Results of this study confirmed the potential of OC to persist in surface water. However, the addition of 5% of sediments resulted in rapid OC degradation. The estimated half-life of OC in water/sediment of the River Po was 15 days. After three weeks of incubation at 20 °C, more than 8% of 14C-OC evolved as 14CO2 from water/sediment samples of the River Po and Venice Lagoon. OC was moderately retained onto coarse sediments from the two sites. In water/sediment samples of the River Po and Venice Lagoon treated with 14C-OC, more than 30% of the 14C-residues remained water-extractable after three weeks of incubation. The low affinity of OC to sediments suggests that the presence of sediments would not reduce its bioavailability to microbial degradation. Another series of laboratory experiments investigated the fate and the removal of OC in two surface water ecosystems of Japan and in the municipal wastewater treatment plant of the city of Bologna, in Northern Italy. The persistence of OC in surface water ranged from non-detectable degradation to a half-life of 53 days. After 40 days, less than 3% of radiolabeled OC evolved as 14CO2. The presence of sediments (5%) led to a significant increase of OC degradation and of mineralization rates. A more intense mineralization was observed in samples of the wastewater treatment plant when applying a long incubation period (40 days). More precisely, 76% and 37% of the initial radioactivity applied as 14C-OC was recovered as 14CO2 from samples of the biological tank and effluent water, respectively. Two bacterial strains growing on OC as sole carbon source were isolated and used for its removal from synthetic medium and environmental samples, including surface water and wastewater. Inoculation of water and wastewater samples with the two OC-degrading strains showed that mineralization of OC was significantly higher in both inoculated water and wastewater, than in uninoculated controls. Denaturing gradient gel electrophoresis and quantitative PCR analysis showed that OC would not affect the microbial population of surface water and wastewater. The capacity of the ligninolytic fungus Phanerochaete chrysosporium to degrade a wide variety of environmentally persistent xenobiotics has been largely reported in literature. In a series of laboratory experiments, the efficiency of a formulation using P. chrysosporium was evaluated for the removal of selected pharmaceuticals from wastewater samples. Addition of the fungus to samples of the wastewater treatment plant of Bologna significantly increased (P < 0.05) the removal of OC and three antibiotics, erythromycin, sulfamethoxazole, and ciprofloxacin. Similar effects were also observed in effluent water. OC was the most persistent of the four pharmaceuticals. After 30 days of incubation, approximately two times more OC was removed in bioremediated samples than in controls. The highest removal efficiency of the formulation was observed with the antibiotic ciprofloxacin. The studies included environmental aspects of soil contamination with two emerging veterinary contaminants, such as doramectin and oxibendazole, wich are common parasitic treatments in cattle farms.

Diabetes mellitus umfasst eine heterogene Gruppe von Stoffwechselfunktionsstörungen, die durch hohe Blut-Glukose-Werte gekennzeichnet sind. Zwei Haupttypen von Diabetes mellitus wurden definiert: Typ 1- und Typ 2-Diabetes. Repaglinid ist ein neuer, schnell wirksamer, bei Typ 2-Diabetikern eingesetzter prandialer Glukose-Regulator mit einer kurzen Plasmahalbwertszeit (<1 Stunde) und der erste Vertreter der Carbamoylmethylbenzoesäure Familie, der in klinischen Studien getestet wurde. Die 18F- und 11C-markierten Repaglinid-Derivate (S)-2-(2-[18F]Fluorethoxy)-4-((3-methyl-1-(2-piperidin-1-yl-phenyl)-butylcarbamoyl)-methyl)-benzoesäure ([18F]Fluorethoxy-desethoxy-Repaglinid) und (S)-2-([11C]Methoxy)-4-([3-methyl-1-(2-piperidin-1-yl-phenyl)-butyl-carba-moyl]-benzoesäure ([11C]Methoxy-desethoxy-Repaglinid) wurden als potentielle Tracer für die nicht-invasive Quantifizierung des Sulfonylharnstoffrezeptor-Typ1-Status (SUR-1) der Insulin-sezernierenden -Zellen mittels Positronen-Emissions-Tomographie (PET) synthetisiert. [18F]Fluorethoxy-desethoxy-Repaglinide konnte in einer radiochemischen Ausbeute (RCA) von 20% nach 135 Minuten mit einer radiochemischen Reinheit >98% unter Verwendung des sekundären Markierungsvorläufers 2-[18F]Fluorethyltosylat erhalten werden. Die spezifische Aktivität lag im Bereich von 50-60 GBq/µmol. Für die radioaktive Synthese des [11C]Methoxy-desethoxy-Repaglinids wurde der sekundäre Markierungsvorläufer [11C]Methyliodid verwendet. Der 11C-Radiotracer wurde in einer RCA von 35% (bezogen auf [11C]CO2) mit einer spezifischen Aktivität von 40-70 GBq/µmol erhalten. Um die Eigenschaften des fluorierten sowie des methoxylierten Repaglinids zu charakterisieren, wurde die Affinität beider Verbindungen zum humanen SUR-1 evaluiert. [19F]Fluorethoxy-desethoxy-Repaglinid und Methoxy-desethoxy-Repaglinid induzierten Verdrängungskurven mit Hill-Koeffizienten nahe 1 und ergaben Dissotiationskonstanten (KD) von 142 nM beziehungsweise 83 nM - vergleichsweise geringe Verluste relativ zu Original-Repaglinid. Die biologische Aktivität wurde mittels Insulin-Sekretionstests an isolierten Ratten-Inselzellen gezeigt und war ebenfalls mit der des Repaglinids vergleichbar. Schließlich wurde die Biodistribution des [18F]Fluorethoxy-desethoxy-Repaglinids in gesunden Sprague-Dawley-Ratten durch Messung der Konzentration der Verbindung in verschiedenen Organen nach intravenöser Injektion untersucht. Das pankreatische Gewebe zeigte im Zeitintervall zwischen 10 und 30 Minuten nach Injektion eine stabile Akkumulation von etwa 0.12% der injizierten Dosis. 50% dieser Tracer-Akkulmulation konnten durch zusätzliche Injektion von nicht-radioaktiv-markiertem Repaglinid verdrängt werden, was auf eine mögliche Eignung des [18F]Fluorethoxy-desethoxy-Repaglinids für in vivo-Untersuchungen mittels PET schließen lässt. Eine erste humane PET-Studie zeigte zwar ebenfalls eine stabile, allerdings nur geringere Akkumulation von [18F]Fluorethoxy-desethoxy-Repaglinid im Pankreas und eine überproportional hohe Aktivitätsanreicherung in der Leber. Die Radioaktivitäts-akkumulation im Blut fiel nach wenigen Minuten unter die des Pankreas.

Eine häufige Art der Chemotherapie ist die Behandlung von Tumoren mit alkylierenden oder chloralkylierenden Zytostatika, die eine Alkylierung von Guanin in der DNA verursachen. Daraus resultieren eine Blockierung der DNA-Synthese und ein Rückgang im Tumorwachstum. Das Enzym O6-Methylguanin-DNA-methyltransferase (MGMT) ist in der Lage, solche Schäden zu reparieren. Da MGMT auch in verschiedenen Tumorarten exprimiert wird, eine Tatsache, die therapeutische Effekte verringern könnte, wird zur Zeit die Gabe von Inhibitoren der MGMT, wie O6-Benzylguanin, vor der eigentlichen Chemotherapie untersucht. Um möglicher Weise die Selektivität dieser Verbindungen für Tumor- vs. gesundem Gewebe und auch die in vivo-Eigenschaften zu verbessern, wurden glycosylierte Inhibitoren vorgeschlagen. Für eine Entwicklung neuer MGMT-Inhibitoren wäre es hilfreich, die in vivo Bioverteilung in Tier und Mensch durch eine Markierung mit geeigneten Isotopen verfolgen zu können. Im Moment existiert keine Möglichkeit, den MGMT-Status eines Tumors nicht-invasiv zu visualisieren. Diese Information kann sehr wichtig für die Planung einer Chemotherapie mit alkylierenden oder chloralkylierenden Zytostatika sein. Mit Methoden wie der Positronen-Emissions-Tomographie (PET) oder der Einzel-Photonen-Emissions-Tomographie (SPECT) ist eine nicht-invasive Quantifizierung von biochemischen Prozessen prinzipiell möglich. Hierfür wurden verschiedenen MGMT-Inhibitoren bereits mit Isotopen wie Fluor-18, Kohlenstoff-11 un Iod-131 markiert, aber sie waren aus unterschiedlichen Gründen nicht geeignet. Das Ziel dieser Arbeit war die Entwicklung von neuen O6-derivatisierten Guaninen, die über einen C8-Spacer an der N9-Position des Guanins mit einer Glucose-Einheit konjugiert werden sollten, geeigneten Markierungsvorläufern und Radioiodierungs-Methoden. Durch Wahl eines geeigneten Radioiodisotops für die Markierung des Restes an der O6-Position des Guanins kann die ex vivo-Bioverteilung dieser Verbindungen in tumortragenden Nacktmäusen (Iod-131) und die Untersuchung der in vivo-Verteilung (Iod-123) durchgeführt werden. Daher wurden O6-(5-Iodothenyl)- (ITG) und O6-(3-Iodbenzyl)guanin-Derivate (IBG) sowie ihre Glucose-Konjugate ITGG und IBGG synthetisiert. Von diesen inaktiven Standard-Verbindungen wurden die IC50-Werte zur MGMT bestimmt. Da sie alle im nM-Bereich lagen, schienen die Verbindungen für weitere Untersuchungen geeignet zu sein. Die Radiomarkierung der Inhibitoren mit Iod-131 bzw. Iod-123 wurde durch Umsetzung der Trialkyl-stannylierten Markierungsvorläufer mit der Chloramin T-Methode in mittleren (Iod-123) bis hohen (Iod-131) radiochemischen Ausbeuten und mit hohen radiochemischen Reinheiten durchgeführt. Mit den 131I-iodierten Verbindungen wurde die spezifische Bindung zur MGMT nachgewiesen, eine Eigenschaft, die essentiell für eine weitere Verwendung dieser Derivate ist. Sie wurden auch zur Bestimmung der ex vivo-Tumor- und Organverteilung in tumortragenden Nacktmäusen (MEX(+), MEX(-), Glioblastom) verwendet. In allen Fällen war die Tumoraufnahme der nicht-konjugierten Guanin-Derivate höher als die der entsprechenden Glucose-Konjugate. Das Tumor-Blut-Verhältnis, das sehr wichtig für einen potentiellen Einsatz der Verbindungen als Tracer des MGMT-Status eines Tumors ist, variierte abhängig von der Kinetik. Zu allen Zeitpunkten war die in vivo-Deiodierung der Glucose-Konjugate deutlich geringer als die von ITG oder IBG. Unter Verwendung von [131I]IBG und [131I]IBGG wurde die Biodistribution nach Inhibition der Natrium-abhängigen Glucose-Transporter, die zumindests teilweise für die Aufnahme der MGMT-Inhibitoren in Zellen verantwortlich sind, durch Phloretin untersucht. Einen Unterschied in der Tumoraufnahme zwischen den mit Phloretin behandelten und den unbehandelten Mäusen konnte nicht beobachtet werden, wahrscheinlich weil die Akkumulation im Tumor generell niedrig war. Mit den 123I-iodierten Verbindungen [123I]IBG und [123I]IBGG wurden in vivo-Scans an tumortragenden Nacktmäusen (MEX(+), MEX(-)) mit einer Kleintier-SPECT-Kamera durchgeführt. In beiden Fällen wurde eine geringe Akkumulation in den Tumoren im Vergleich zu anderen Organen beobachtet, was die ex vivo-Biodistributionsdaten bestätigte.

For the safety assessment of radioactive waste, the possibility of radionuclide migration has to be considered. Since Np (and also Th due to the long-lived 232-Th) will be responsible for the greatest amount of radioactivity one million years after discharge from the reactor, its (im)-mobilization in the geosphere is of great importance. Furthermore, the chemistry of Np(V) is quite similar (but not identical) to the chemistry of Pu(V). Three species of neptunium may be found in the near field of the waste disposal, but pentavalent neptunium is the most abundant species under a wide range of natural conditions. Within this work, the interaction of Np(V) with the clay mineral montmorillonite and melanodins (as model substances for humic acids) was studied. The sorption of neptunium onto gibbsite, a model clay for montmorillonite, was also investigated. The sorption of neptunium onto γ-alumina and montmorillonite was studied in a parallel doctoral work by S. Dierking. Neptunium is only found in ultra trace amounts in the environment. Therefore, sensitive and specific methods are needed for its determination. The sorption was determined by γ spectroscopy and LSC for the whole concentration range studied. In addition the combination of these techniques with ultrafiltration allowed the study of Np(V) complexation with melanoidins. Regrettably, the available speciation methods (e.g. CE-ICP-MS and EXAFS) are not capable to detect the environmentally relevant neptunium concentrations. Therefore, a combination of batch experiments and speciation analyses was performed. Further, the preparation of hybrid clay-based materials (HCM) montmorillonitemelanoidins for sorption studies was achieved. The formation of hybrid materials begins in the interlayers of the montmorillonite, and then the organic material spreads over the surface of the mineral. The sorption of Np onto HCM was studied at the environmentally relevant concentrations and the results obtained were compared with those predicted by the linear additive model by Samadfam. The sorption of neptunium onto gibbsite was studied in batch experiments and the sorption maximum determined at pH~8.5. The sorption isotherm pointed to the presence of strong and weak sorption sites in gibbsite. The Np speciation was studied by using EXAFS, which showed that the sorbed species was Np(V). The influence of M42 type melanodins on the sorption of Np(V) onto montmorillonite was also investigated at pH 7. The sorption of the melanoidins was affected by the order in which the components were added and by ionic strength. The sorption of Np was affected by ionic strength, pointing to outer sphere sorption, whereas the presence of increasing amounts of melanoidins had little influence on Np sorption.

Die heutige Verfügbarkeit der molekularen Bildgebung ermöglicht einen signifikanten Einfluss auf die Diagnostik und die Therapiekontrolle von neurodegenerativen Erkrankungen, die unter anderem durch Fehlsteuerungen im GABAergen System auftreten können. Die Visualisierung und Quantifizierung des GABAA-alpha5-Subtyps durch PET könnte dabei zu einem besseren Verständnis von Erkrankungen wie Alzheimer und traumatischen Neurosen (emotionales Langzeitgedächtnis) beitragen. Ferner eröffnen GABAA/alpha5-subtypselektive Liganden die Möglichkeit, wesentliche Grundlagen der elementaren Vorgänge von Lernen und Erinnern zu untersuchen. 7,8,9,10-Tetrahydro-(7,10-ethan)-1,2,4-triazol[3,4-alpha]phthalazine stellen sich als vielverspre-chende Leitstrukturen zur Entwicklung neuer 18F-markierter alpha5-subtypselektiver GABAA-Rezeptorliganden für die PET dar. Um diese neuartigen Substanzen hinsichtlich ihrer Potenz als GABAA-alpha5-subtypselektive Radioliganden zu verifizieren, wurden zunächst die entsprechenden 19F-Derivate TC07-TC12 synthetisiert. Diese Referenzverbindungen wurden in Rezeptor-bindungsassays und in Autoradiographien mit [3H]Ro 15-4513 als zu verdrängender Radioligand evaluiert. In beiden Experimenten als auch in in vivo-Verdrängungsexperimenten an Ratten konnte eine hohe Affinität im nanomolaren Bereich als auch eine hohe Selektivität bezüglich der GABAA/alpha5-Untereinheit für einige der dargestellten Referenzverbindungen nachgewiesen werden. Gemäß diesen vielversprechenden Ergebnissen wurden verschiedene Markie-rungsvorläufer für eine 18F-Direktmarkierung der relevantesten Substanz TC07 in einer mehrstufigen organischen Synthese dargestellt. Die anschließende 18F-Markierung erfolgte über eine nukleophile Substitution mit [18F]Fluorid. Die Reaktionsparameter wurden hinsichtlich Reaktionstemperatur und dauer, Markierungsvorläuferkonzentration, Basenabhängigkeit und verschiedenen Markierungsmethoden optimiert. Daraus resultierend konnte [18F]TC07 mit bis zu 45 % radiochemischer Ausbeute erhalten werden. Die zerfallskorrigierte, gesamtradiochemische Ausbeute von nca [18F]TC07 in isotonischer NaCl-Lösung betrug 15 %. Basierend auf den bisher erhaltenen Ergebnissen wurde der Radioligand in in vitro-, ex vivo- und in vivo µPET-Experimenten evaluiert. Die zunächst durchgeführten in vitro-Experimente deuteten auf eine homogene Verteilung der Aktivität hin und zeigten keine spezifische Anreicherung. Diese Ergebnisse wurden sowohl in ex vivo- als auch in in vivo-µPET-Studien bestätigt. Auch hier konnte nur eine niedrige Aktivitätsanreicherung, eine homogene Verteilung im gesamten Gehirn und keine Übereinstimmung mit der bekannten GABAA/alpha5-Subtypverteilung gefunden werden. Eine im Anschluss durchgeführte Metabolismusstudie zeigte eine langsame Metabolisierungsrate des [18F]TC07 und auch eine Organverteilungsstudie zeigte keine außergewöhnlichen Anreicherungen. Aus den erhaltenen Ergebnissen kann geschlossen werden, dass der Radioligand [18F]TC07 kein geeigneter Tracer zur in vivo-Visualisierung der alpha5-Untereinheit des GABAA-Rezeptors ist.
