Adaptation and acclimation to different temperatures of obligate psychrophilic, facultative psychrophilic and mesophilic yeasts. Production of ω-3 and ω-6 polyunsaturated fatty acids by fermentative way. Obligate psychrophilic, facultative psychrophilic and mesophilic yeasts were cultured in a carbon rich medium at different temperatures to investigate if growth parameters, lipid accumulation and fatty acid composition were adaptive and/or acclimatory responses. Acclimation of facultative psychrophiles and mesophiles to lower temperature negatively affected their specific growth rate. Obligate psychrophiles exhibited the highest biomass yield (YX/S), followed by facultative psychrophiles, then by mesophiles. The growth temperature did not influence the YX/S of facultative psychrophiles and mesophiles. Acclimation to lower temperature caused the increase in lipid yield (YL/X) in mesophilic yeasts, but did not affect YL/X in facultative psychrophiles. Similar YL/X were found in both facultative and obligated psychrophiles, suggesting that lipid accumulation is not a distinctive character of adaptation to permanently cold environments. The extent of unsaturation of fatty acids was one major adaptive feature of the yeasts which colonize permanently cold ecosystems. Remarkable amounts of α-linolenic acid were found in obligate psychrophiles at the expenses of linoleic acid, whereas it was generally scarce or absent in all the others strains. Increased unsaturation of fatty acids was also an acclimatory response of mesophiles and facultative psychrophiles to lower temperature. It’s well known that omega-3 polyunsaturated fatty acids (PUFAs) display a variety of beneficial effects on various organ systems and diseases, therefore a process for the microbial production of omega-3 PUFAs would be of great interest. This work sought also to investigate if one of the better psychrophilic yeast, Rhodotorula glacialis DBVPG 4785, stimulated by acclamatory responses, produced omega-3 PUFAs. In fact, the adaptation of psychrophilic yeasts to cold niches is related to the production of higher amounts of lipids and to increased unsaturation degree of fatty acids, presumably to maintain membrane fluidity and functionality at low temperatures. Bioreactor fermentations of Rhodotorula glacialis DBVPG 4785 were carried out at 25, 20, 15, 10, 5, 0, and -3°C in a complex medium with high C:N ratio for 15 days. High biomass production was attained at all the temperatures with a similar biomass/glucose yield (YXS), between 0.40 and 0.45, but the specific growth rate of the strain decreased as the temperature diminished. The coefficients YL/X have been measured between a minimum of 0.50 to a maximum of 0.67, but it was not possible to show a clear effect of temperature. Similarly, the coefficient YL/S ranges from a minimum of 0.22 to a maximum of 0.28: again, it does not appear to be any significant changes due to temperature. Among omega-3 PUFAs, only α-linolenic acid (ALA, 18:3n-3) was found at temperatures below to 0°C, while, it’s remarkable, that the worthy arachidonic acid (C20:4,n-6), stearidonic acid (C20:4,n-3) C22:0 and docosahexaenoic acid (C22:6n-3) were produced only at the late exponential phase and the stationary phase of batch fermentations at 0 and -3°C. The docosahexaenoic acid (DHA) is a beneficial omega-3 PUFA that is usually found in fatty fish and fish oils. The results herein reported improve the knowledge about the responses which enable psychrophilic yeasts to cope with cold and may support exploitation of these strains as a new resource for biotechnological applications.

Ziel dieser Arbeit war es hydrophile Lipopolymere darzustellen, mit denen es möglich sein sollte polymerunterstützte Lipiddoppelschichten auf festen Substratoberflächen zu fixieren. Die Polymere sollten einen Oberflächenanker, eine lipophile Gruppe und ein hydrophiles Polymerrückgrat enthalten. Hierzu wurden Alpha,Omega-funktionalisierte Polymere ausgehend von lipophilen Initiatoren dargestellt. Ausgehend von hydrophoben 2-Brompropionsäureamiden konnte eine kontrollierte radikalische Polymerisation (ATRP) von verschiedenen Acrylamiden durchgeführt werden. So wurden verschiedene Copolymere aus Acrylamid, N-Isopropylacrylamid und N-(3-Dimethylaminopropyl)-acrylamid synthetisiert. Der Einbau des Oberflächenankers als Funktionalität erfolgte indirekt durch Polymerisation eines N-Acryloxysuccinimid Endblocks, welcher in einer anschließenden polymeranalogen Reaktion mit Cysteaminmethyldisulfid umgesetzt wurde.In Ladungsuntersuchungen (PCD) konnte das pH-abhängige Verhalten der Polymere untersucht werden. Der Knäuelkollaps (LCST) der Poly-(N-isopropylacrylamide) wurde mittels Turbidimetrie und DSC charakterisiert. Die Adsorption der Polymere auf Goldoberflächen wurde mit Hilfe der Oberflächenplasmonen Spektroskopie (SPS) aus wässriger Lösung nachgewiesen werden. Dabei bildeten sich ultradünne Filme von 15-20 Å Dicke aus. Kontaktwinkelmessungen wiesen diesen adsorbierten Polymerfilmen ein sehr hydrophiles Verhalten nach. In Lösung adsorbierten die Polymere auf Vesikeloberflächen. Auf ultradünnen Polymerfilmen adsorbierten die Vesikel, wobei mit Hilfe der SPS eine Dickenzunahme um etwa 50 Å nachgewiesen werden konnte. Die ultradünnen Filme der Poly(N-isopropylacrylamide) wiesen eine temperaturabhängige Attraktivität gegenüber Vesikeln auf. Durch gezielte Polystyrol-Entnetzung konnten strukturierte Träger erhalten werden.

Questa relazione finale è stata frutto di un lavoro sperimentale svolto in collaborazione con un’azienda del territorio ed ha avuto come obiettivo principale quello di verificare i parametri qualitativi e compositivi di una nuova tipologia di salume in fase di studio. L’azienda sta studiando una nuova tipologia di “prodotto funzionale” addizionando componenti ad attività nutraceutica al salame ottenuto da sole parti magre di suino. L’intenzione dell’azienda è di arricchire il proprio prodotto con acidi grassi polinsaturi della serie omega-3 (da olio di semi di lino), noti per gli effetti benefici che possono esercitare sulla salute umana. A questo scopo, sul prodotto di nuova formulazione (ed in parallelo sul prodotto a formulazione classica e su un prodotto del tutto simile commercializzato da un’altra azienda) sono state effettuate sia la determinazione della percentuale lipidica, al fine di individuare il quantitativo totale di grasso, che una caratterizzazione gascromatografica del profilo quali-quantitativo in acidi grassi, analsi fondamentale per verificare la conformità con i requisiti indicati dal Reg. 1924/06, e successive modifiche, previsti per le indicazioni da inserire in etichetta (claims). Inoltre, è stato importante controllare che la concentrazione voluta di acido alfa-linolenico si mantenga tale durante tutto il periodo di shelf-life del prodotto (45 gg), perciò per soddisfare quest’ultima finalità, le analisi sono state eseguite sia all’inizio (T0) che alla fine (T1) della vita commerciale del prodotto. E’ apparso poi fondamentale monitorare un possibile decadimento/modifica della qualità organolettica del prodotto di nuova formulazione, a causa del possibile aumento di ossidabilità della frazione lipidica causata da una maggiore presenza di acidi grassi polinsaturi; ed infine, con l’intento di verificare se la nuova formulazione potesse comportare una variazione significativa delle caratteristiche sensoriali del prodotto, sono stati condotti un test descrittivo quantitativo (QDA), in grado di descrivere il profilo sensoriale del prodotto ed un test discriminante qualitativo (metodo triangolare) capace di valutare l’eventuale comparsa di cambiamenti in seguito alle modifiche apportate al prodotto; per ultimo è stato poi eseguito un test affettivo quantitativo (test di preferenza, condotto su scala di laboratorio) in grado fornire informazioni sul gradimento e di evidenziare i principali vettori che guidano le scelte dei consumatori. I risultati delle analisi chimiche e sensoriali, eseguite per soddisfare tali richieste, hanno evidenziato come il salume presenti un quantitativo di tali acidi grassi polinsaturi in linea con il claim “Alimento fonte di acidi grassi omega-3”, (requisiti indicati dal Reg. 1924/06 e successive modifiche), risultato confermato sia all’inizio che al termine della vita commerciale del prodotto (pari a circa 45 giorni). Inoltre, esso risulta essere piuttosto interessante dal punto di vista nutrizionale, poiché caratterizzato da un contenuto di grasso per la categoria dei salumi, relativamente limitato, pari a circa il 15%, con percentuali più favorevoli in acidi grassi insaturi. L’analisi del profilo sensoriale condotta ad inizio shelf-life ha invece evidenziato come questa tipologia di prodotto sia caratterizzata sia al gusto che all’olfatto da note più sapide e speziate , oltre ad una maggiore consistenza della fetta, rispetto agli altri campioni analizzati, mentre i risultati relativi al termine della shelf-life hanno evidenziato come esso tenda ad essere soggetto ad alterazioni sensoriali rilevabili proprio al termine della fase di conservazione dovute nello specifico alla formazione di composti responsabili dell’odore di rancido. I test discriminanti, condotti con giudici non allenati, non hanno invece fatto registrare differenze sensoriali significative tra il prodotto con formulazione classica e quello di nuova formulazione, confrontati ad inizio e a fine shelf-life. Riassumendo, dai risultati ottenuti da questo lavoro sperimentale, il prodotto di nuova formulazione, arricchito in acidi grassi polinsaturi della serie omega-3, risulta essere in linea con il claim “Alimento fonte di acidi grassi omega-3”, presenta in generale un contenuto di grasso totale inferiore a quello di altre tipologie di salumi ed un rapporto più favorevole tra acidi grassi insaturi e saturi, per questo può essere considerato un prodotto interessante dal punto di vista della salute e della nutrizione. Proprio per la sua formulazione più ricca in acidi grassi polinsaturi, tende però a fine shelf-life a presentare una leggera nota di rancido, riconducibile ad una maggiore ossidabilità della frazione lipidica. Tale variazione sensoriale è risultata comunque percepita solo da un panel allenato, mentre un test di tipo discriminante, condotto con giudici non allenati, non ha messo in luce differenze significative tra il salume di nuova formulazione e quello con formulazione classica né all’inizio, né al termine della vita commerciale del prodotto.

In the course of this work the effect of metal substitution on the structural and magnetic properties of the double perovskites Sr2MM’O6 (M = Fe, substituted by Cr, Zn and Ga; M’ = Re, substituted by Sb) was explored by means of X-ray diffraction, magnetic measurements, band structure calculations, Mößbauer spectroscopy and conductivity measurements. The focus of this study was the determination of (i) the kind and structural boundary conditions of the magnetic interaction between the M and M’ cations and (ii) the conditions for the principal application of double perovskites as spintronic materials by means of the band model approach. Strong correlations between the electronic, structural and magnetic properties have been found during the study of the double perovskites Sr2Fe1-xMxReO6 (0 < x < 1, M = Zn, Cr). The interplay between van Hove-singularity and Fermi level plays a crucial role for the magnetic properties. Substitution of Fe by Cr in Sr2FeReO6 leads to a non-monotonic behaviour of the saturation magnetization (MS) and an enhancement for substitution levels up to 10 %. The Curie temperatures (TC) monotonically increase from 401 to 616 K. In contrast, Zn substitution leads to a continuous decrease of MS and TC. The diamagnetic dilution of the Fe-sublattice by Zn leads to a transition from an itinerant ferrimagnetic to a localized ferromagnetic material. Thus, Zn substitution inhibits the long-range ferromagnetic interaction within the Fe-sublattice and preserves the long-range ferromagnetic interaction within the Re-sublattice. Superimposed on the electronic effects is the structural influence which can be explained by size effects modelled by the tolerance factor t. In the case of Cr substitution, a tetragonal – cubic transformation for x > 0.4 is observed. For Zn substituted samples the tetragonal distortion linearly increases with increasing Zn content. In order to elucidate the nature of the magnetic interaction between the M and M’ cations, Fe and Re were substituted by the valence invariant main group metals Ga and Sb, respectively. X-ray diffraction reveals Sr2FeRe1-xSbxO6 (0 < x < 0.9) to crystallize without antisite disorder in the tetragonal distorted perovskite structure (space group I4/mmm). The ferrimagnetic behaviour of the parent compound Sr2FeReO6 changes to antiferromagnetic upon Sb substitution as determined by magnetic susceptibility measurements. Samples up to a doping level of 0.3 are ferrimagnetic, while Sb contents higher than 0.6 result in an overall antiferromagnetic behaviour. 57Fe Mößbauer results show a coexistence of ferri- and antiferromagnetic clusters within the same perovskite-type crystal structure in the Sb substitution range 0.3 < x < 0.8, whereas Sr2FeReO6 and Sr2FeRe0.9Sb0.1O6 are “purely” ferrimagnetic and Sr2FeRe0.1Sb0.9O6 contains antiferromagnetically ordered Fe sites only. Consequently, a replacement of the Re atoms by a nonmagnetic main group element such as Sb blocks the double exchange pathways Fe–O–Re(Sb)–O–Fe along the crystallographic axis of the perovskite unit cell and destroys the itinerant magnetism of the parent compound. The structural and magnetic characterization of Sr2Fe1-xGaxReO6 (0 < x < 0.7) exhibit a Ga/Re antisite disorder which is unexpected because the parent compound Sr2FeReO6 shows no Fe/Re antisite disorder. This antisite disorder strongly depends on the Ga content of the sample. Although the X-ray data do not hint at a phase separation, sample inhomogeneities caused by a demixing are observed by a combination of magnetic characterization and Mößbauer spectroscopy. The 57Fe Mößbauer data suggest the formation of two types of clusters, ferrimagnetic Fe- and paramagnetic Ga-based ones. Below 20 % Ga content, Ga statistically dilutes the Fe–O–Re–O–Fe double exchange pathways. Cluster formation begins at x = 0.2, for 0.2 < x < 0.4 the paramagnetic Ga-based clusters do not contain any Fe. Fe containing Ga-based clusters which can be detected by Mößbauer spectroscopy firstly appear for x = 0.4.

Questa relazione finale è incentrata sul lavoro sperimentale richiesto dalla collaborazione con l’azienda Natural Salumi ed ha come obiettivo principale quello di verifica dei parametri qualitativi e compositivi di una nuova tipologia di salume in fase di studio. L’azienda ha infatti messo a punto un nuovo “prodotto funzionale” addizionando componenti aventi attività nutraceutica al salame ottenuto da sole parti magre di suino (tipo NaturalFetta) e si propone di arricchire il proprio prodotto con acidi grassi polinsaturi della serie omega-3 (da olio di semi di lino), noti per gli effetti benefici esercitati sulla salute umana. Sul prodotto di nuova formulazione si effettueranno sia la determinazione della percentuale lipidica mediante metodo ufficiale Soxhlet ed automatizzato Soxtec, al fine di individuare il quantitativo totale della parte grassa, che una caratterizzazione del profilo quali-quantitativo in acidi grassi, fondamentale per verificare la conformità con i requisiti indicati dal Reg. 1924/06 (e successivi come il Reg. UE 432/2012) previsti per le indicazioni da inserire in etichetta (claims) per i prodotti arricchiti con acidi grassi polinsaturi (PUFA). La determinazione del profilo in acidi grassi e, nello specifico, del contenuto in acido -linolenico, sarà realizzata mediante gascromatografia previa estrazione della frazione lipidica dei salumi con metodo di Folch modificato, che prevede un'estrazione solido-liquido. La concentrazione di acido alfa-linolenico sarà inoltre monitorata durante tutto il periodo di shelf-life del prodotto (45 gg) al fine di valutare eventuali variazioni durante la conservazione. Per soddisfare quest’ultima finalità, le analisi saranno eseguite sia all’inizio (T0) che alla fine (T1) della vita commerciale del prodotto. Le stesse analisi verranno inoltre condotte sia sul prodotto a formulazione classica che su un prodotto del tutto simile, commercializzato da un’altra azienda e considerato come leader di mercato. In relazione ad un possibile aumento di ossidabilità della frazione lipidica, sarà realizzato un controllo dei parametri chimici, qualitativi e compositivi, con particolare riferimento ai prodotti di ossidazione del colesterolo (date le loro implicazioni in ambito biomedico). La maggiore presenza in formulazione di acidi grassi polinsaturi potrebbe infatti favorire un incremento dei COPs (Cholesterol Oxidation Products), che saranno separati, identificati e quantificati mediante la procedura SPE/GC-MS (estrazione e purificazione in fase solida accoppiata ad analisi gascromatografica con rivelazione mediante spettrometria di massa).

This thesis is concerned with calculations in manifestly Lorentz-invariant baryon chiral perturbation theory beyond order D=4. We investigate two different methods. The first approach consists of the inclusion of additional particles besides pions and nucleons as explicit degrees of freedom. This results in the resummation of an infinite number of higher-order terms which contribute to higher-order low-energy constants in the standard formulation. In this thesis the nucleon axial, induced pseudoscalar, and pion-nucleon form factors are investigated. They are first calculated in the standard approach up to order D=4. Next, the inclusion of the axial-vector meson a_1(1260) is considered. We find three diagrams with an axial-vector meson which are relevant to the form factors. Due to the applied renormalization scheme, however, the contributions of the two loop diagrams vanish and only a tree diagram contributes explicitly. The appearing coupling constant is fitted to experimental data of the axial form factor. The inclusion of the axial-vector meson results in an improved description of the axial form factor for higher values of momentum transfer. The contributions to the induced pseudoscalar form factor, however, are negligible for the considered momentum transfer, and the axial-vector meson does not contribute to the pion-nucleon form factor. The second method consists in the explicit calculation of higher-order diagrams. This thesis describes the applied renormalization scheme and shows that all symmetries and the power counting are preserved. As an application we determine the nucleon mass up to order D=6 which includes the evaluation of two-loop diagrams. This is the first complete calculation in manifestly Lorentz-invariant baryon chiral perturbation theory at the two-loop level. The numerical contributions of the terms of order D=5 and D=6 are estimated, and we investigate their pion-mass dependence. Furthermore, the higher-order terms of the nucleon sigma term are determined with the help of the Feynman-Hellmann theorem.

While polymers with different functional groups along the backbone have intensively been investigated, there is still a challenge in orthogonal functionalization of the end groups. Such well-defined systems are interesting for the preparation of multiblock (co) polymers or polymer networks, for bio-conjugation or as model systems for examining the end group separation of isolated polymer chains. rnHere, Reversible Addition Fragmentation Chain Transfer (RAFT) polymerization was employed as method to investigate improved techniques for an a, w end group functionalization. RAFT produces polymers terminated in an R group and a dithioester-Z group, where R and Z stem from a suitable chain transfer agent (CTA). rnFor alpha end group functionalization, a CTA with an activated pentafluorophenyl (PFP) ester R group was designed and used for the polymerization of various methacrylate monomers, N-isopropylacrylamide and styrene yielding polymers with a PFP ester as a end group. This allowed the introduction of inert propyl amides, of light responsive diazo compounds, of the dyes NBD, Texas Red, or Oregon Green, of the hormone thyroxin and allowed the formation of multiblocks or peptide conjugates. rnFor w end group functionalization, problems of other techniques were overcome through an aminolysis of the dithioester in the presence of a functional methane thiosulfonate (MTS), yielding functional disulfides. These disulfides were stable under ambient conditions and could be cleaved on demand. Using MTS chemistry, terminal methyl disulfides (enabling self-assembly on planar gold surfaces and ligand substitution on gold and semiconductor nanoparticles), butynyl disulfide end groups (allowing the “clicking” of the polymers onto azide functionalized surfaces and the selective removal through reduction), the bio-target biotin, and the fluorescent dye Texas Red were introduced into polymers. rnThe alpha PFP amidation could be performed under mild conditions, without substantial loss of DTE. This way, a step-wise synthesis produced polymers with two functional end groups in very high yields. rnAs examples, polymers with an anchor group for both gold nanoparticles (AuNP) and CdSe / ZnS semi-conductor nanoparticles (QD) and with a fluorescent dye end group were synthesized. They allowed a NP decoration and enabled an energy transfer from QD to dye or from dye to AuNP. Water-soluble polymers were prepared with two different bio-target end groups, each capable of selectively recognizing and binding a certain protein. The immobilization of protein-polymer-protein layers on planar gold surfaces was monitored by surface plasmon resonance.Introducing two different fluorescent dye end groups enabled an energy transfer between the end groups of isolated polymer chains and created the possibility to monitor the behavior of single polymer chains during a chain collapse. rnThe versatility of the synthetic technique is very promising for applications beyond this work.

In this thesis we consider systems of finitely many particles moving on paths given by a strong Markov process and undergoing branching and reproduction at random times. The branching rate of a particle, its number of offspring and their spatial distribution are allowed to depend on the particle's position and possibly on the configuration of coexisting particles. In addition there is immigration of new particles, with the rate of immigration and the distribution of immigrants possibly depending on the configuration of pre-existing particles as well. In the first two chapters of this work, we concentrate on the case that the joint motion of particles is governed by a diffusion with interacting components. The resulting process of particle configurations was studied by E. Löcherbach (2002, 2004) and is known as a branching diffusion with immigration (BDI). Chapter 1 contains a detailed introduction of the basic model assumptions, in particular an assumption of ergodicity which guarantees that the BDI process is positive Harris recurrent with finite invariant measure on the configuration space. This object and a closely related quantity, namely the invariant occupation measure on the single-particle space, are investigated in Chapter 2 where we study the problem of the existence of Lebesgue-densities with nice regularity properties. For example, it turns out that the existence of a continuous density for the invariant measure depends on the mechanism by which newborn particles are distributed in space, namely whether branching particles reproduce at their death position or their offspring are distributed according to an absolutely continuous transition kernel. In Chapter 3, we assume that the quantities defining the model depend only on the spatial position but not on the configuration of coexisting particles. In this framework (which was considered by Höpfner and Löcherbach (2005) in the special case that branching particles reproduce at their death position), the particle motions are independent, and we can allow for more general Markov processes instead of diffusions. The resulting configuration process is a branching Markov process in the sense introduced by Ikeda, Nagasawa and Watanabe (1968), complemented by an immigration mechanism. Generalizing results obtained by Höpfner and Löcherbach (2005), we give sufficient conditions for ergodicity in the sense of positive recurrence of the configuration process and finiteness of the invariant occupation measure in the case of general particle motions and offspring distributions.

Die vorliegende Arbeit beschäftigt sich mit der Modellierung niederenergetischer elektromagnetischer und hadronischer Prozesse im Rahmen einer manifest lorentzinvarianten, chiralen effektiven Feldtheorie unter expliziter, dynamischer Berücksichtigung resonanter, das heißt vektormesonischer Freiheitsgrade. Diese effektive Theorie kann daher als Approximation der grundlegenden Quantenchromodynamik bei kleinen Energien verstanden werden. Besonderes Augenmerk wird dabei auf das verwendete Zähl- sowie Renormierungschema gelegt, wodurch eine konsistente Beschreibung mesonischer Prozesse bis zu Energien von etwa 1GeV ermöglicht wird. Das verwendete Zählschema beruht dabei im Wesentlichen auf einem Argument für großes N_c (Anzahl der Farbfreiheitsgrade) und lässt eine äquivalente Behandlung von Goldstonebosonen (Pionen) und Resonanzen (Rho- und Omegamesonen) zu. Als Renormierungsschema wird das für (bezüglich der starken Wechselwirkung) instabile Teilchen besonders geeignete complex-mass scheme als Erweiterung des extended on-mass-shell scheme verwendet, welches in Kombination mit dem BPHZ-Renormierungsverfahren (benannt nach Bogoliubov, Parasiuk, Hepp und Zimmermann) ein leistungsfähiges Konzept zur Berechnung von Quantenkorrekturen in dieser chiralen effektiven Feldtheorie darstellt. Sämtliche vorgenommenen Rechnungen schließen Terme der chiralen Ordnung vier sowie einfache Schleifen in Feynman-Diagrammen ein. Betrachtet werden unter anderem der Vektorformfaktor des Pions im zeitartigen Bereich, die reelle Compton-Streuung (beziehungsweise Photonenfusion) im neutralen und geladenen Kanal sowie die virtuelle Compton-Streuung, eingebettet in die Elektron-Positron-Annihilation. Zur Extraktion der Niederenergiekopplungskonstanten der Theorie wird letztendlich eine Reihe experimenteller Datensätze verschiedenartiger Observablen verwendet. Die hier entwickelten Methoden und Prozeduren - und insbesondere deren technische Implementierung - sind sehr allgemeiner Natur und können daher auch an weitere Problemstellungen aus diesem Gebiet der niederenergetischen Quantenchromodynamik angepasst werden.

Produktionsmechanismen für Teilchenproduktion im mittleren Energiebereich wurden in Proton-Proton Kollisionen innerhalb der COMPASS-Kollaboration mit Hilfe des COMPASS-Spektrometers am SPS Beschleuniger am CERN untersucht. Die verschiedenen Produktionsmechanismen werden mittels Produktion der Vektormesonen omega und phi studiert und können die diffraktive Anregung des Strahlteilchens mit anschliessendem Zerfall der Resonanz, zentrale Produktion und den damit verwandten “Shake-off” Mechanismus enthalten. Die für diese Arbeit verwendeten Daten wurden in den Jahren 2008 und 2009 mit 190 GeV/c-Protonen aufgenommen, die auf ein Flüssigwasserstofftarget trafen. Das Target war von einem Rückstoßprotonendetektor umgeben, der ein integraler Bestandteil des neuentwickelten Hadrontriggersystems ist. Für dieses System wurden außerdem einige neue Detektoren gebaut. Die Leistungsfähigkeit des Rückstoßprotonendetektors und des Triggersystems wird untersucht und Effizienzen extrahiert. Außerdem wird sowohl eine Methode zur Rekonstruktion von Rückstoßprotonen als auch eine Methode zur Kalibration des Rückstoßprotonendetektors entwickelt und beschrieben. Die Produktion von omega-Mesonen wurde in der Reaktion pp -> p omega p, omega -> pi+pi-pi0 und die Produktion von phi-Mesonen in der Reaktion pp -> p phi p, phi -> K+K- bei einem Impulsübertrag zwischen 0.1 (GeV/c)^2 und 1 (GeV/c)^2 gemessen. Das Produktionsverhältnis s(pp -> p phi p)/s(pp -> p omega p) wird als Funktion des longitudinalen Impulsanteils xF bestimmt und mit der Vorhersage durch die Zweigregel verglichen. Es ergibt sich eine signifikante Verletzung der Zweigregel, die abhängig von xF ist. Die Verletzung wird in Verbindung zu resonanten Strukturen im pomega-Massenspektrum diskutiert. Die xF-Abhängigkeit verschwindet, wenn man die Region niedriger pomega- und pphi-Masse entfernt, die solche resonanten Strukturen aufweist. Zusätzlich wird die Spinausrichtung bzw. das Spindichtematrixelement rho00 für omega- und phi-Mesonen untersucht. Die Spinausrichtung wird im Helizitätssystemrnanalysiert, welches für eine Abgrenzung von resonanten, diffraktiven Anregungen geeignet ist. Außerdem wird die Spinausrichtung in einem Referenzsystem mit Bezug auf die Richtung des Impulsübertrags untersucht, mit dessen Hilfe zentrale Prozesse wie zentrale Produktion oder “shake-off” abgegrenzt werden. Auch hier wird eine Abhängigkeit von xF und der invarianten Masse des pomega-Systems beobachtet. Diese Abhängigkeit kann wieder auf die resonanten Strukturen in der Produktion von omega-Mesonen zurückgeführt werden. Die Ergebnisse werden abschließend im Hinblick auf die verschiedenen Produktionsmechanismen diskutiert.

Craniosynostosis consists of a premature fusion of the sutures in an infant skull that restricts skull and brain growth. During the last decades, there has been a rapid increase of fundamentally diverse surgical treatment methods. At present, the surgical outcome has been assessed using global variables such as cephalic index, head circumference, and intracranial volume. However, these variables have failed in describing the local deformations and morphological changes that may have a role in the neurologic disorders observed in the patients. This report describes a rigid image registration-based method to evaluate outcomes of craniosynostosis surgical treatments, local quantification of head growth, and indirect intracranial volume change measurements. The developed semiautomatic analysis method was applied to computed tomography data sets of a 5-month-old boy with sagittal craniosynostosis who underwent expansion of the posterior skull with cranioplasty. Quantification of the local changes between pre- and postoperative images was quantified by mapping the minimum distance of individual points from the preoperative to the postoperative surface meshes, and indirect intracranial volume changes were estimated. The proposed methodology can provide the surgeon a tool for the quantitative evaluation of surgical procedures and detection of abnormalities of the infant skull and its development.

We present theory and experiments on the dynamics of reaction fronts in two-dimensional, vortex-dominated flows, for both time-independent and periodically driven cases. We find that the front propagation process is controlled by one-sided barriers that are either fixed in the laboratory frame (time-independent flows) or oscillate periodically (periodically driven flows). We call these barriers burning invariant manifolds (BIMs), since their role in front propagation is analogous to that of invariant manifolds in the transport and mixing of passive impurities under advection. Theoretically, the BIMs emerge from a dynamical systems approach when the advection-reaction-diffusion dynamics is recast as an ODE for front element dynamics. Experimentally, we measure the location of BIMs for several laboratory flows and confirm their role as barriers to front propagation.

Human invariant natural killer T (NKT) cell TCRs bind to CD1d via an "invariant" Vα24-Jα18 chain (iNKTα) paired to semi-invariant Vβ11 chains (iNKTβ). Single-amino acid variations at position 93 (p93) of iNKTα, immediately upstream of the "invariant" CDR3α region, have been reported in a substantial proportion of human iNKT-cell clones (4-30%). Although p93, a serine in most human iNKT-cell TCRs, makes no contact with CD1d, it could affect CD1d binding by altering the conformation of the crucial CDR3α loop. By generating recombinant refolded iNKT-cell TCRs, we show that natural single-nucleotide variations in iNKTα, translating to serine, threonine, asparagine or isoleucine at p93, exert a powerful effect on CD1d binding, with up to 28-fold differences in affinity between these variants. This effect was observed with CD1d loaded with either the artificial α-galactosylceramide antigens KRN7000 or OCH, or the endogenous glycolipid β-galactosylceramide, and its importance for autoreactive recognition of endogenous lipids was demonstrated by the binding of variant iNKT-cell TCR tetramers to cell surface expressed CD1d. The serine-containing variant showed the strongest CD1d binding, offering an explanation for its predominance in vivo. Complementary molecular dynamics modeling studies were consistent with an impact of p93 on the conformation of the CDR3α loop.
