本学位论文报道了西藏产三种藏族传统植物药材的化学成分研究。论文由四章组成,前三章是实验部分,分别报道了尼泊尔黄堇(Corydalis hendersonii Hemsl.)、藏波罗花(Incarvillea younghusbandii Sprague)和全缘叶绿绒蒿(Meconopsis interifolia Franch.)的化学成分研究结果。从这三种青藏高原药用植物中共分离鉴定出33 个化合物,其中1 个是新化合物。第四章概述了罂粟科紫堇属植物的化学和药理研究进展。 第一章为尼泊尔黄堇的化学成分研究。通过正、反相硅胶柱色谱等分离方法从药用植物尼泊尔黄堇的地上部分共分离纯化得到12 个化合物。运用MS、1H-NMR、13C-NMR、DEPT、HMBC、NOESY 等现代波谱学方法将它们的结构鉴定为:刺罂粟碱(1) , 普托品(2) , 新那亭(3) , 斯可任(4) , tetrahydrothalifendine (5) ,9-methyl-decumbenine C (6),tetrahydroberberrubine (7),隐品碱(8),α-别隐品碱(9),6,7-methylenedioxy-1(2H)-oxoisoquinolinone (10),6-丙酮基-5,6 -二氢血根碱(11)和β-谷甾醇(12)。其中化合物6 为新化合物,为首次发现的分子骨架上C-9 位连有甲基的苯肽异喹啉类型生物碱。另外,除化合物1 和2 外,其它9 个生物碱(3~11)均为首次从该种植物中分离得到。同时,我们还对对尼泊尔黄堇中的总生物碱进行了串联质谱分析。 第二章为藏波罗花的化学成分研究。从该药用植物的地上部分共分离得到16个化合物,通过理化常数和波谱数据鉴定为:异佛手柑内酯(1),6-甲氧基当归素(2),欧前胡素(3),花椒毒内酯(4),珊瑚菜素(5),水合氧化前胡素(6),rivulobirin A (7),齐墩果酸甲酯(8),咖啡酸甲酯(9),银桦酸(10),(D)-boschniakinic acid (11),对羟基苯甲酸(12) , tert-O-β-D-glucopyranosyl-(R)-heraclenol (13) , 5-methoxy-8-O-β-D-glucopyranosyloxypsoralen (14),前胡苷V(15)和苯乙醇-O-β-D-吡喃葡萄糖-(1→2)-O-β-D-吡喃葡萄糖苷(16)。所有以上化合物均为首次从该种植物中分离得到。另外我们还首次对藏波罗花挥发油的化学成分进行了气相色谱-质谱(GC-MS)联用分析,共鉴定出39 个挥发性成分。 第三章为全缘叶绿绒蒿化学成分的分离鉴定。从传统藏药材全缘叶绿绒蒿地上部分共分离纯化出8 个化合物。通过理化常数和波谱数据将他们的结构分别鉴定为:去甲血根碱(1),β-谷甾醇(2),3-羟基-齐墩果烷-12(13)-烯-30-酸(3),6-丙酮基-5,6-二氢血根碱(4),木犀草素(5),胡萝卜苷(6),quercetin 3-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside (7)和普托品(8)。其中化合物1,4 和7 为首次从该种药用植物中分离得到。 第四章为综述,总结和归纳了近年来罂粟科紫堇属植物的化学和药理研究进展。 This dissertation consists of four parts. The first, second and third parts report the studies on the chemical constituents from the medicinal plants of Corydalis hendersonii, Incarvillea younghusbandii and Meconopsis interifolia. The forth part reviews the progress of the studies on Corydalis species. The first chapter is about the isolations and identifications of alkalids from the aerial parts of C. hendersonii which is a traditional Tibetan medicine to treat febrifuge, high blood pressure and hepatitis. A new isoquinoline alkaloid, 9-methyl-decumbenine C (6), together with ten known alkaloids, stylopine (1), protopine (2), canadine (3), scoulerine (4), tetrahydrothalifendine (5), tetrahydroberberrubine (7), cryptopine (8), α-allocryptopine (9), 6,7-methylenedioxy-1(2H)-isoquinolinone (10) and 6-acetonyl-5,6-dihydrosanguinarine (11), and β-sitosterol (12) were isolated. Their structures were elucidated by spectroscopic methods. Furthermore, the total alkaloids were analyzed by ESI-MSn. The second chapter is about the isolations and identifications of chemical constituents from the aerial parts of I. younghusbandii. Sixteen compounds were isolated and purified by normal and reversed phase silica gel column chromatography. By spectral analysis, there structures were identified as isobergapten (1), sphondin (2), imperatorin (3), xanthotoxin (4), phellopterin (5), heraclenol (6), rivulobirin A (7), methyl oleanolate (8), methyl caffeate (9), grevillic acid (10), (D)-boschniakinic acid (11), 4-hydroxybenzoic acid (12), tert-O-β-D-glucopyranosyl-(R)-heraclenol (13), 5-methoxy-8-O-β-D-glucopyranosyloxypsoralen (14), decuroside Ⅴ(15), and phenylethyl-O-β-Dglucopyranosyl-(1→2)-β-D-glucopyranoside (16). All of these compounds were isolated from this plant for the first time.By the way, the chemical components of the essential oil from I. younghusbandii were analyzed by GC-MS for the first time. The third chapter is about the the isolations and identifications of the chemical constituents of M. interifolia. Eight compounds were isolated and identified as norsanguinarine (1), β-sitosterol (2), 3-hydroxyolean-12(13)-en-30-oic acid (3), 6-acetonyl-5,6-dihydrosanguinarine (4), luteolin (5), daucosterol (6), quercetin 3-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside (7) and protopine (8). The compounds 1, 4 and 7 were isolated from this herb for the first time. The last chapter is a review of the research progress of the studies on Corydalis species, which includes the chemical constituents in this genus and their pharmacology.

The effects of high-pressure processing (HPP) in conjunction with an essential oil-based active packaging on the surface of ready-to-eat (RTE) chicken breast were investigated as post-processing listericidal treatment. Three different treatments were used, and all samples were vacuum packed: (i) HPP at 500. MPa for 1. min (control), (ii) active packaging based on coriander essential oil, and (iii) active packaging and HPP. When applied individually, active packaging and pressurisation delayed the growth of Listeria monocytogenes. The combination of HPP and active packaging resulted in a synergistic effect reducing the counts of the pathogen below the detection limit throughout 60. days storage at 4. °C. However, when these samples were stored at 8. °C, growth did occur, but again a delay in growth was observed. The effects on colour and lipid oxidation were also studied during storage and were not significantly affected by the treatments. Active packaging followed by in-package pressure treatment could be a useful approach to reduce the risk of L. monocytogenes in cooked chicken without impairing its quality. Industrial relevance: Ready-to-eat products are of great economic importance to the industry. However, they have been implicated in several outbreaks of listeriosis. Therefore, effective ways to reduce the risk from this pathogenic microorganism can be very attractive for manufacturers. This study showed that the use of active packaging followed by HPP can enhance the listericidal efficiency of the treatment while using lower pressure levels, and thus having limited effects on colour and lipid oxidation of RTE chicken breast.

Esta tese teve como objectivo estudar estratégias de conservação de pescado fresco, recorrendo ao uso de extractos e óleos essenciais de plantas e do processamento por alta pressão (HPP), usando filetes de robalo como um caso de estudo modelo. Relativamente aos extractos e óleos essenciais, avaliaram-se as suas propriedades antibacterianas e antioxidantes. Os extractos aquosos quente de poejo e de orégão e o óleo essencial de cravinho apresentaram a maior actividade antioxidante. Os óleos essenciais foram mais eficientes do que os extractos para inibir o crescimento das estirpes bacterianas testadas, tendo-se observado os menores valores de concentração mínima inibitória nos óleos essenciais de orégão, citronela, alho e orégão Espanhol. De seguida, estudou-se o efeito dos óleos essenciais de orégão Espanhol e de limão na conservação de filetes de robalo fresco tendo em conta critérios microbiológicos, químicos, físicos e sensoriais. A aplicação do óleo essencial de orégão Espanhol aumentou o tempo de vida útil dos filetes sob o ponto de vista bacteriano, mas não em termos sensoriais. A combinação dos óleos essenciais de orégão Espanhol e de limão melhorou o efeito antioxidante e reduziu a intensidade do odor e a sua eficácia em relação às Enterobacteriaceae, comparando com o tratamento com óleo essencial de orégão Espanhol per se. No sentido de reduzir o odor conferido pelos óleos essenciais realizou-se um estudo de conservação para avaliar o efeito de películas com óleos essenciais (citronela, alho e tomilho) em filetes de robalo, recorrendo a uma teste de desafio bacteriano. As películas sem óleos essenciais aumentaram o tempo de vida útil sob o ponto de vista bacteriano, mas este efeito não foi observado com a incorporação dos óleos essenciais nas películas. Em relação ao HPP, testaram-se diversas condições (nível de pressão, tempo de pressurização e taxa de pressurização) e avaliaram-se os efeitos na actividade enzimática, na qualidade global e na conservação de filetes de robalo fresco. Em geral, o aumento do nível de pressão e do tempo de pressurização diminuiu a actividade da fosfatase ácida e das enzimas proteolíticas, a carga bacteriana e a capacidade de retenção de água, enquanto que os filetes ficaram mais brancos. O HPP revelou potencial para o desenvolvimento de novos produtos: mais brancos, não translúcidos, mais firmes e com maior tempo de frescura e estabilidade microbiológica. Em conclusão, os óleos essenciais e o HPP têm potencial para conservar pescado fresco, devido aos seus efeitos na qualidade bacteriana. Ainda assim, mais esforços devem ser feitos no sentido de reduzir a transferência de odor dos óleos essenciais para o pescado e os efeitos do HPP no aspecto do pescado e na oxidação lipídica.

This study aimed to analyse the Brazilian savanna forest from a Legal Reserve (LR) area from a perspective of conservation, reservoir of organic carbon and medicinal biomass for a prospective use of native medicinal plants. An ethnobotanical and ethnopharmacological survey was carried out close to a community settled in the rural area in the south of Tocantins, being selected 9 of the most cited species (cajuí- Anacardium othonianum; inharé-Brosimum gaudichaudii; jatobá-Hymenaeae courbaril; jenipapo-Genipa americana, aroeira-Myracrodruon urundeuva; negramina-Siparuna guianensis; barbatimão- Stryphnodendron obovatum; assa peixe-Vernonia brasiliana, embaúba-Cecropia pachystachya). Crude foliar extracts were subjected to a preliminary phytochemical prospection and triage of secondary metabolites with antimicrobial activity of potential interest in health and familiar agriculture. Phenolic compounds, terpenes and flavonoids were detected in the extracts of most species, which suggests the presence of antimicrobial, antioxidant and anti-insect activities. It was evident the need to better know the LR as a reservoir of medicinal biomass in an area under ecological tension where 35% (610ha) of the property is LR and should be protected by law. Therefore, a forest inventory of live woody species was performed using the allometric or indirect method. This identified a rare remnant of Semidecidual Seasonal Forest amidst the largest world savannah, the Cerrado biome. An analysis of the forest average productivity per basal area (m².ha), aerial live biomass (ton.ha-1) and carbon stock was carried out. The forest fragment was considered relatively rich in species and diversity, although showing signs of disturbance and dominance by a few species. Its horizontal structure suggests biotic regeneration conditions. It is an important reservoir of medicinal plants. Of the families (57.5%) presenting medicinal species, 19 from a total of 33 are represented in the area and contain 44% (27) of the total species (61) and 63% (432) of the total individuals catalogued. Medicinal species have ecological importance for the equilibrium of the local flora and represent 80% of the 10 species with higher Importance Value Index (IVI): Tetragastris altissima, Chrysophyllum marginatum, Oenocarpus distichus, Sclerolobium paniculatum, Simarouba versicolor, Alibertia macrophylla, Siparuna guianensis, Maprounea guianensis, Licania parvifolia e Physocalymma scaberrimum. Medicinal productivity was high for this type of phytophysionomy: 183,2 ton. ha-1 of biomass and 91,51 ton. ha-1 of carbon representing 66% of the total biomass and carbon of this Cerrado forest. From this stage S. guianensis (Siparunaceae) was selected for performing bioassays in order to verify its biological activity against microorganisms of health and agricultural relevance. This is a native aromatic medicinal plant recommended as priority for conservation, with local popular medicinal validation and availability of medicinal feedstock (3300 Kg.ha-1), with the foliar fraction giving 38Kg/ha of crude extract and 5L/ha of essential oil. Foliar crude extracts and essential oil were obtained and tested in vitro using a disk diffusion bioassay. Different concentrations of these natural products were tested against gram-positive bacteria (Staphylococcus aureus ATCC 29213), gram-negative bacteria (Escherichia coli ATCC 25922 and ATCC 35218; Pseudomonas aeruginosa ATCC 10145) and fungi (Candida albicans ATCC 6258 e Fusarium oxysporum). The essential oil inhibited the growth of S. aureus in its crude concentration (380μg.mL-1), as well as diluted to half (190μg.mL-1) and a quarter strength (95μg.mL-1). It’s likely that such action is due to sesquiterpenes major components, such as bisabolol and bisabolene (10.35%), measured by gas chromatography (GC-MS, GC-FID). Extracts did not exhibit any antimicrobial activity against the microorganisms tested. The native medicinal plants prospective market is an alternative that favours the conservation of biodiversity while generating benefits for the development of sustainable family productive activities within local ecosystems instead of the current inappropriate uses. This strengthens conservation policies of Legal Reserve in rural settlements and is in agreement with public policy on global warming and climate changes.

Essential oils are used in Cosmetic, Perfumery, Food and Pharmaceutical Industries as flavours and/or medicines. However, part of the essential oil components that remains in the distillation water (hydrosol or distillate water) has been less studied both in chemical and biological terms. This research concerns the antioxidant activity, measured through several methods, of Lavandula officinalis L., Origanum majorana L., Rosmarinus officinalis L., Salvia officinalis L. and Thymus vulgaris L., Cinnamomum verum J. Presl. and Syzygium aromaticum (L.) Merrill and Perry hydrosols. The ability of hydrosols to prevent oxidation was checked by two main methods: prevention of lipid peroxidation through the measurement of malonaldehyde produced after degradation of hydroperoxides; and ability for scavenging free radicals including hydroxyl and superoxide anion radicals. The S. aromaticum and T. vulgaris hydrosols, predominantly constituted by eugenol and carvacrol, respectively, were the most effective as antioxidants, except for scavenging superoxide anion radical. In this case, L. officinalis hydrosol in which linalool prevailed was a stronger antioxidant. The worst hydrosol as antioxidant was that of S. officinalis, independent on the method checked.

The volatiles from Coriandrum sativum L., Satureja montana L., Santolina chamaecyparissus L., and Thymus vulgaris L. were isolated by hydrodistillation (essential oil) and supercritical fluid extraction (volatile oil). Their effect on seed germination and root and shoot growth of the surviving seedlings of four crops (Zea mays L., Triticum durum L., Pisum sativum L., and Lactuca sativa L.) and two weeds (Portulaca oleracea L. and Vicia sativa L.) was investigated and compared with those of two synthetic herbicides, Agrocide and Prowl. The volatile oils of thyme and cotton lavender seemed to be promising alternatives to the synthetic herbicides because they were the least injurious to the crop species. The essential oil of winter savory, on the other hand, affected both crop and weeds and can be appropriate for uncultivated fields.

Supercritical fluid extraction (SEE) of the volatile oil from Thymus vulgaris L. aerial flowering parts was performed under different conditions of pressure, temperature, mean particle size and CO2 flow rate and the correspondent yield and composition were compared with those of the essential oil isolated by hydrodistillation (HD). Both the oils were analyzed by GC and GC-MS and 52 components were identified. The main volatile components obtained were p-cymene (10.0-42.6% for SFE and 28.9-34.8% for HD), gamma-terpinene (0.8-6.9% for SFE and 5.1-7.0% for HD), linalool (2.3-5.3% for SFE and 2.8-3.1% for HD), thymol (19.5-40.8% for SFE and 35.4-41.6% for HD), and carvacrol (1.4-3.1% for SFE and 2.6-3.1% for HD). The main difference was found to be the relative percentage of thymoquinone (not found in the essential oil) and carvacryl methyl ether (1.0-1.2% for HD versus t-0.4 for SFE) which can explain the higher antioxidant activity, assessed by Rancimat test, of the SFE volatiles when compared with HD. Thymoquinone is considered a strong antioxidant compound.

Le problème grandissant de l’antibiorésistance remet en question plusieurs pratiques reliées à l’utilisation des antibiotiques, dont l’usage à grande échelle pour des fins de promotion de la croissance chez les animaux de consommation. Depuis 2006, le retrait des antibiotiques dans les élevages de poulets de chair en Europe a été associé à la résurgence d’entérite nécrotique, une maladie intestinale causée par la bactérie Clostridium perfringens. Alors que la pathogénie de la maladie semblait bien comprise, le retrait des antibiotiques a montré que peu de solutions de remplacement sont disponibles afin de prévenir cette maladie. Très peu d’études se sont intéressées à mieux caractériser la dynamique des populations de C. perfringens dans les élevages avicoles et l’effet que pouvait avoir le retrait des antibiotiques sur l’évolution de ces populations. La présente étude a évalué l’impact du remplacement des antibiotiques promoteurs de croissance et des anticoccidiens pendant une période de 14 mois, dans huit élevages commerciaux de poulets de chair au Québec. Un protocole d’élevage alternatif, combinant des stratégies telles que des produits à base d’huiles essentielles, des acides organiques et inorganique, une vaccination contre la coccidiose ainsi qu’une amélioration des conditions de démarrage des poussins, a été utilisé en remplacement des antimicrobiens. Les performances zootechniques, la santé digestive ainsi que l’occurrence d’entérite nécrotique pour les lots de poulets soumis au protocole d’élevage alternatif ont été comparées avec celles de lots de poulets de chair recevant une ration conventionnelle. Des analyses moléculaires basées sur la PCR et le PFGE ont été utilisées afin de documenter l’impact de la mise en place du protocole d’élevage alternatif sur les populations de C. perfringens. Les résultats obtenus montrent qu’aucune différence entre les groupes soumis aux deux types de protocoles n'est observée en ce qui a trait à la viabilité en élevage, à l'âge d'abattage et aux taux de condamnations à l'abattoir. Toutefois, les lots soumis au traitement alternatif ont eu des performances moindres pour le poids moyen à l'abattage, le gain moyen quotidien et la conversion alimentaire. Un peu plus de 27% des lots soumis au protocole alternatif ont connu un épisode d’entérite nécrotique clinique. Les analyses ont aussi montré que l’un ou l’autre des protocoles ne semble pas exercer d’influence particulière sur la dynamique temporelle des populations de C. perfringens. Pour les lots soumis au protocole d’élevage alternatif, une forte diversité génétique pour C. perfringens a été liée à un risque augmenté de vivre un épisode d’entérite nécrotique. À l’opposé, les lots alternatifs ayant vécu des épisodes récurrents de la maladie ont montré une diminution significative de la diversité génétique, ainsi qu’une augmentation marquée du nombre de souches de C. perfringens transportant plusieurs gènes de virulence. La présente étude a permis de mieux documenter, d’un point de vue économique et scientifique, la production de poulets de chair élevés sans antibiotiques ni anticoccidiens au Québec. Cette étude est la seule du genre s’étant intéressée à la caractérisation et à la comparaison dans le temps des flores de C. perfringens retrouvées dans les deux types d’élevages. Elle a révélé que la production à grande échelle de poulets de chair élevés sans antibiotiques ni anticoccidiens est possible au Québec, mais que celle-ci présente des impacts sur les performances zootechniques des oiseaux, sur leur santé digestive et sur la dynamique des populations de C. perfringens. Heilkräuter sind während des Trocknungsprozesses zahlreichen Einflüssen ausgesetzt, welche die Qualität des Endproduktes entscheidend beeinflussen. Diese Forschungsarbeit beschäftigt sich mit der Trocknung von Zitronenmelisse (Melissa officinalis .L) zu einem qualitativ hochwertigen Endprodukt. Es werden Strategien zur Trocknung vorgeschlagen, die experimentelle und mathematische Aspekte mit einbeziehen, um bei einer adäquaten Produktivität die erforderlichen Qualitätsmerkmale im Hinblick auf Farbeänderung und Gehalt an ätherischen Ölen zu erzielen. Getrocknete Zitronenmelisse kann zurzeit, auf Grund verschiedener Probleme beim Trocknungsvorgang, den hohen Qualitätsanforderungen des Marktes nicht immer genügen. Es gibt keine standardisierten Informationen zu den einzelnen und komplexen Trocknungsparametern. In der Praxis beruht die Trocknung auf Erfahrungswerten, bzw. werden Vorgehensweisen bei der Trocknung anderer Pflanzen kopiert, und oftmals ist die Trocknung nicht reproduzierbar, oder beruht auf subjektiven Annäherungen. Als Folge dieser nicht angepassten Wahl der Trocknungsparameter entstehen oftmals Probleme wie eine Übertrocknung, was zu erhöhten Bruchverlusten der Blattmasse führt, oder eine zu geringe Trocknung, was wiederum einen zu hohen Endfeuchtegehalt im Produkt zur Folge hat. Dies wiederum mündet zwangsläufig in einer nicht vertretbaren Farbänderung und einen übermäßigen Verlust an ätherischen Ölen. Auf Grund der unterschiedlichen thermischen und mechanischen Eigenschaften von Blättern und Stängel, ist eine ungleichmäßige Trocknung die Regel. Es wird außerdem eine unnötig lange Trocknungsdauer beobachtet, die zu einem erhöhten Energieverbrauch führt. Das Trocknen in solaren Tunneln Trocknern bringt folgendes Problem mit sich: wegen des ungeregelten Strahlungseinfalles ist es schwierig die Trocknungstemperatur zu regulieren. Ebenso beeinflusst die Strahlung die Farbe des Produktes auf Grund von photochemischen Reaktionen. Zusätzlich erzeugen die hohen Schwankungen der Strahlung, der Temperatur und der Luftfeuchtigkeit instabile Bedingungen für eine gleichmäßige und kontrollierbare Trocknung. In Anbetracht der erwähnten Probleme werden folgende Forschungsschwerpunkte in dieser Arbeit gesetzt: neue Strategien zur Verbesserung der Qualität werden entwickelt, mit dem Ziel die Trocknungszeit und den Energieverbrauch zu verringern. Um eine Methodik vorzuschlagen, die auf optimalen Trocknungsparameter beruht, wurden Temperatur und Luftfeuchtigkeit als Variable in Abhängigkeit der Trocknungszeit, des ätherischer Ölgehaltes, der Farbänderung und der erforderliche Energie betrachtet. Außerdem wurden die genannten Parametern und deren Auswirkungen auf die Qualitätsmerkmale in solaren Tunnel Trocknern analysiert. Um diese Ziele zu erreichen, wurden unterschiedliche Ansätze verfolgt. Die Sorption-Isothermen und die Trocknungskinetik von Zitronenmelisse und deren entsprechende Anpassung an verschiedene mathematische Modelle wurden erarbeitet. Ebenso wurde eine alternative gestaffelte Trocknung in gestufte Schritte vorgenommen, um die Qualität des Endproduktes zu erhöhen und gleichzeitig den Gesamtenergieverbrauch zu senken. Zusätzlich wurde ein statistischer Versuchsplan nach der CCD-Methode (Central Composite Design) und der RSM-Methode (Response Surface Methodology) vorgeschlagen, um die gewünschten Qualitätsmerkmalen und den notwendigen Energieeinsatz in Abhängigkeit von Lufttemperatur und Luftfeuchtigkeit zu erzielen. Anhand der gewonnenen Daten wurden Regressionsmodelle erzeugt, und das Verhalten des Trocknungsverfahrens wurde beschrieben. Schließlich wurde eine statistische DOE-Versuchsplanung (design of experiments) angewandt, um den Einfluss der Parameter auf die zu erzielende Produktqualität in einem solaren Tunnel Trockner zu bewerten. Die Wirkungen der Beschattung, der Lage im Tunnel, des Befüllungsgrades und der Luftgeschwindigkeit auf Trocknungszeit, Farbänderung und dem Gehalt an ätherischem Öl, wurde analysiert. Ebenso wurden entsprechende Regressionsmodelle bei der Anwendung in solaren Tunneltrocknern erarbeitet. Die wesentlichen Ergebnisse werden in Bezug auf optimale Trocknungsparameter in Bezug auf Qualität und Energieverbrauch analysiert.

En Colombia el manejo de pacientes en los diferentes servicios hospitalarios han presentado inconvenientes en el combate de infecciones por bacterias y la resistencia de las mismas; el Staphylococcus aureus Escherichia coli y Pseudomonas aeruginosa, han demostrado evolución en la creación de generaciones resistentes y también han participado junto con la Salmonella y el B.cereus en brotes por ETAS como principales microorganismos causales. En el presente estudio se analizó el aceite esencial de la Conobea scoparioides para evaluar su actividad frente a cinco cepas bacterianas. Se obtuvo el AE y se prepararon las bacterias aplicándose pruebas de sensibilidad y no paramétricas para determinar el porcentaje de inhibición, la evaluación de la MIC y comparar la efectividad del aceite vs la estreptomicina. El aceite esencial presentó actividad principalmente contra el B. cereus con el mayor % de inhibición y una MIC de 3.2 ug/ml, caso diferente presentó P. aeruginosa con un % de crecimiento por encima del 50% presentando una MIC de 16.7 ug/ml. Finalmente podemos concluir que se presentó mayor actividad frente a bacterias gram positivas como el caso del B. cereus que en gram negativas con MIC bajas. Estos resultados permite comparar la actividad de la conobea con estudios recientes bajo la misma modalidad que permiten identificar nuevas plantas con actividad biológica y percibir que la conobea es efectiva en mayor proporción frente a bacterias gram positivas.

A semiochemical based push-pull strategy for control of oilseed rape pests is being developed at Rothamsted Research. This strategy uses insect and plant derived semiochemicals to manipulate pests and their natural enemies. An important element within this strategy is an understanding of the importance of non-host plant cues for pest insects and how such signals could be used to manipulate their behaviour. Previous studies using a range of non-host plants have shown that, for the pollen beetle Meligethes aeneus (Coleoptera: Nitidulidae), the essential oil of lavender, Lavandula angustifolia (Lamiaceae), was the most repellent. The aim of this study was to identify the active components in L. angustifolia oil, and to investigate the behaviour of M. aeneus to these chemicals, to establish the most effective use of repellent stimuli to disrupt colonisation of oilseed rape crops. Coupled gas chromatography-electroantennography (GC-EAG) and gas chromatography-mass spectrometry (GC-MS) resulted in the identification of seven active compounds which were tested for behavioural activity using a 4-way olfactometer. Repellent responses were observed with (±)-linalool and (±)-linalyl acetate. The use of these chemicals within a push-pull pest control strategy is discussed.

The combined action of the plant-derived volatile, S-carvone, and mild heat treatment on the food-borne pathogen, Listeria monocytogenes, was evaluated. The viability of exponential phase cultures grown at 8 °C could be reduced by 1·3 log units after exposure to S-carvone (5 mmol l−1) for 30 min at 45 °C, while individual treatment with S-carvone or exposure to 45 °C for 30 min did not result in a loss in viability. Other plant-derived volatiles, namely carvacrol, cinnamaldehyde, thymol and decanal, were also found to reduce the viability of L. monocytogenes in combination with the same mild heat treatment at concentrations of 1·75 mmol l−1, 2·5 mmol l−1, 1·5 mmol l−1 and 2 mmol l−1, respectively. These findings show that essential oil compounds can play an important role in minimally processed foods, and can be used in the concept of Hurdle Technology to reduce the intensity of heat treatment or other individual hurdles.

The use of semiochemicals for the manipulation of the pollen beetle, Meliethes aeneus (Fabricius) (Coleoptera: Nitidulidae), is being investigated for potential incorporation into a push-pull strategy for this pest, which damages oilseed rape, Brassica napus L. (Brassicaceae), throughout Europe. Previous laboratory behavioural studies using volatiles from non-host plants showed that M. aeneus is repelled by the odour of lavender, Lavendula angustifolia Mill. (Lamiaceae), essential oil. This article reports on semi-field and field trials to investigate this behaviour under more realistic conditions. Semi-field experiments were conducted to assess the relative importance of olfaction at different points in host location behaviour by M. aeneus. The results showed that oilseed rape plants treated with lavender odour were less colonised by M. aeneus in comparison with an untreated control, but that the treatment effect was much reduced if the lavender odour was applied after colonisation. The field experiment demonstrated that lavender odour caused a significant reduction in the number of adultM. aeneus infesting the oilseed rape plants in the treatment plots compared to the control plots. Overall, these findings are very encouraging for the future development of a push-pull pest control system.

Ethnopharmacological importance: Many species of plants in the Brazilian cerrado (savanna) are widely used in ethnomedicine. However, the safety and effectiveness of medicinal plants used in communities with little or no access to manufactured drugs should be evaluated. Aim of the study: Evaluate the antimicrobial and cytotoxic activities of extracts from eight plant species, obtained using Brazilian cachaca as the extractor liquid. Materials and methods: The extracts were tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans, Candida parapsilosis, promastigote forms of Leishmania amazonensis, and poliovirus. In addition, cytotoxic activity was assayed in Vero cells and in human erythrocytes. Results: The plant species Curatella americana, Sclerolobium aureum, and Plathymenia reticulata showed the best activity against yeasts, especially the crude extract of C. americana and its ethyl-acetate fraction. Kielmeyera lathrophyton showed a minimum inhibitory concentration of 250 mu g/ml against S. aureus, and was inactive against Gram-negative bacteria. The extract obtained from Annona coriacea showed the best activity against the promastigote forms of Leishmania amazonensis (IC(50) = 175 mu g/ml). Only C. americana showed potential for antipoliovirus activity. The concentrations of the crude extracts that showed toxicity to VERO cells had CC(50) between 31 and 470 mu g/ml, and the lyophilized Brazilian cachaca showed a CC(50) of 307 mu g/ml. None of the extracts showed toxicity against human erythrocytes. Conclusions: Among the plant species studied. C americana proved to be effective against microorganisms, especially as an antifungal. The results will help in the search for alternative drugs to be used in pharmacotherapy, and will contribute to establish safe and effective use of phytomedicines in the treatment of infectious diseases. A pH indicator film based on cassava starch plasticized with sucrose and inverted sugar and incorporated with grape and spinach extracts as pH indicator sources (anthocyanin and chlorophyll) has been developed, and its packaging properties have been assessed. A second-order central composite design (2(2)) with three central points and four star points was used to evaluate the mechanical properties (tensile strength, tensile strength at break, and elongation at break percentage), moisture barrier, and microstructure of the films, and its potential as a pH indicator packaging. The films were prepared by the casting technique and conditioned under controlled conditions (75% relative humidity and 23 degrees C), at least 4 days before the analyses. The materials were exposed to different pH solutions (0, 2, 7, 10, and 14) and their color parameters (L*, a*, b*, and haze) were measured by transmittance. Grape and spinach extracts have affected the material characterization. Film properties (mechanical properties and moisture barrier) were strongly influenced by extract concentration presenting lower results than for the control. Films containing a higher concentration of grape extract presented a greater color change at different pH`s suggesting that anthocyanins are more effective as pH indicators than chlorophyll or the mixture of both extracts.
