Water-ethanol mixtures exhibit many interesting anomalies, such as negative excess partial molar volume of ethanol, excess sound absorption coefficient at low concentrations, and positive deviation from Raoult's law for vapor pressure, to mention a few. These anomalies have been attributed to different, often contradictory origins, but a quantitative understanding is still lacking. We show by computer simulation and theoretical analyses that these anomalies arise from the sudden emergence of a bicontinuous phase that occurs at a relatively low ethanol concentration of x(eth) approximate to 0.06-0.10 (that amounts to a volume fraction of 0.17-0.26, which is a significant range!). The bicontinuous phase is formed by aggregation of ethanol molecules, resulting in a weak phase transition whose nature is elucidated. We find that the microheterogeneous structure of the mixture gives rise to a pronounced nonmonotonic composition dependence of local compressibility and nonmonotonic dependence in the peak value of the radial distribution function of ethyl groups. A multidimensional free energy surface of pair association is shown to provide a molecular explanation of the known negative excess partial volume of ethanol in terms of parallel orientation and hence better packing of the ethyl groups in the mixture due to hydrophobic interactions. The energy distribution of the ethanol molecules indicates additional energy decay channels that explain the excess sound attenuation coefficient in aqueous alcohol mixtures. We studied the dependence of the solvation of a linear polymer chain on the composition of the water-ethanol solvent. We find that there is a sudden collapse of the polymer at x(eth) approximate to 0.05-a phenomenon which we attribute to the formation of the microheterogeneous structures in the binary mixture at low ethanol concentrations. Together with recent single molecule pulling experiments, these results provide new insight into the behavior of polymer chain and foreign solutes, such as enzymes, in aqueous binary mixtures.

This work reports the measured spray structure and droplet size distributions of ethanol-gasoline blends for a low-pressure, multi-hole, port fuel injector (PFI). This study presents previously unavailable data for this class of injectors which are widely used in automotive applications. Specifically, gasoline, ethanol, and gasoline-ethanol blends containing 10%, 20% and 50% ethanol were studied using laser backlight imaging, and particle/droplet image analysis (PDIA) techniques. The fuel mass injected, spray structure and tip penetrations, droplet size distributions, and Sauter mean diameter were determined for the blends, at two different injection pressures. Results indicate that the gasoline and ethanol sprays have similar characteristics in terms of spray progression and droplet sizes in spite of the large difference in viscosity. It appears that the complex mode of atomization utilized in these injectors involving interaction of multiple fuel jets is fairly insensitive to the fuel viscosity over a range of values. This result has interesting ramifications for existing gasoline fuel systems which need to handle blends and even pure ethanol, which is one of the renewable fuels of the future. We carry out a series of long atomistic molecular dynamics simulations to study the unfolding of a small protein, chicken villin headpiece (HP-36), in water-ethanol (EtOH) binary mixture. The prime objective of this work is to explore the sensitivity of protein unfolding dynamics toward increasing concentration of the cosolvent and unravel essential features of intermediates formed in search of a dynamical pathway toward unfolding. In water ethanol binary mixtures, HP-36 is found to unfold partially, under ambient conditions, that otherwise requires temperature as high as similar to 600 K to denature in pure aqueous solvent. However, an interesting course of pathway is observed to be followed in the process, guided by the formation of unique intermediates. The first step of unfolding is essentially the separation of the cluster formed by three hydrophobic (phenylalanine) residues, namely, Phe-7, Phe-11, and Phe-18, which constitute the hydrophobic core, thereby initiating melting of helix-2 of the protein. The initial steps are similar to temperature-induced unfolding as well as chemical unfolding using DMSO as cosolvent. Subsequent unfolding steps follow a unique path. As water-ethanol shows composition-dependent anomalies, so do the details of unfolding dynamics. With an increase in cosolvent concentration, different partially unfolded intermediates are found to be formed. This is reflected in a remarkable nonmonotonic composition dependence of several order parameters, including fraction of native contacts and protein-solvent interaction energy. The emergence of such partially unfolded states can be attributed to the preferential solvation of the hydrophobic residues by the ethyl groups of ethanol. We further quantify the local dynamics of unfolding by using a Marcus-type theory.

Poly(ether imine) dendritic macromolecules were undertaken to study the reversible dendrimer monomer-megamer assembly-disassembly in aqueous solutions. Synthesis of thiol functionalized poly(ether imine) (PETIM) dendrimers and their covalent aggregation behavior in the aqueous solution of ethanol/water (2:1) is demonstrated. The dendritic megamers were characterized using microscopic techniques. Kinetics of the aggregation behavior was followed using turbidity measurements, light-scattering and atomic force microscopic techniques. Inherent luminescence behavior of PETIM dendrimer monomers was retained in the dendrimer megamers also, which allowed visualization of the megamers through confocal microscopy. Extent of thiol functionalities that remained after the megamer assembly was estimated through Ellman's assay. Subsequent to megamer assembly, disassembly of megamers to dendrimer monomers was conducted, using dithiothreitol reagent. Water-insoluble sudan I dye was encapsulated in dendrimer megamer and subsequent release profile was assessed during the disassembly in aqueous solutions. The studies were conducted using first, second and third generations, representing 4, 8 and 16 sulfhydryl groups at their peripheries of dendrimers, respectively. (C) 2014 Elsevier Ltd. Here, we report the clean and facile synthesis of Pt and Pd nanoparticles decorated on reduced graphene oxide (rGO) by the simultaneous reduction of graphene oxide (GO) and the metal ions in Mg/acid medium. As-generated Pt and Pd nanoparticles serve as a heterogeneous catalyst for the further reduction of the rGO by the hydrogen spill-over process. The C/O ratio is much higher as compared to the rGO obtained by the reduction of GO by only Mg/acid. Overall, the process is rapid, facile and green that does not require any toxic chemical agent or any rigorous chemical reactions. We perform the catalytic reduction of 4-nitophenol (4-NP) to 4-aminophenol (4-AP) at room temperature by Pd@rGO and Pt@rGO. The reduction is complete within 35 s for Pd@rGO and 60 s for Pt@rGO when 50 mu g of hybrid catalyst is used for 0.5 ml of 1 mM of 4-NP. In case of ethanol oxidation, the current density for Pd@rGO is comparable to commercial Pt/C but is doubled for Pt@rGO. A Echinodorus macrophyllus (Alismataceae), conhecida como chapéu de couro no Brasil, é usada popularmente para tratar doenças reumáticas e inflamatórias. Neste trabalho, foram avaliados os efeitos antiinflamatórios do extrato aquoso de E. macrophyllus (EAEm) e suas frações etanólicas no modelo murino de air pouch. Para a obtenção das frações, 7 g do EAEm foram aplicadas em uma coluna cromatográfica aberta de sílica gel eluída com diferentes concentrações de etanol. Os cromatogramas do EAEm/frações foram obtidos usando um sistema de HPLC. Foram obtidas quatro frações, duas delas com maior rendimento. Resumidamente, a bolha de ar foi induzida pela injeção de 5 mL de ar estéril (s.c) no dorso de camundongos SW machos (25-35 g). Após 3 dias, mas 3 mL de ar estéril foram injetados para manter a bolha. No sexto dia, cada grupo (n = 4) foi tratado intraperitoneal (ip) ou oralmente (v.o) com EAEm (25 ou 250 mg/kg), Fr20 ou Fr40 (2,5, 25, 50 ou 100 mg/kg) e os controles com indometacina (10 mg/kg, v.o.) ou veículo (salina). Uma hora depois, 1 mL de salina ou de carragenina 1% estéril foi injetada dentro da bolha. Após 4 h, a cavidade foi lavada com NaCl 0,9%, EDTA 2 mM (1 mL), para a determinação do número de leucócitos, volume do exsudato e concentração de proteínas. Células do exsudato foram preparadas em citocentrífuga e coradas pelo método do Panótico para a contagem diferencial dos leucócitos. Cortes histológicos coletados dos diferentes grupos foram fixados com formol tamponado 10% (pH 7,4) por 7 dias, corados com HE e analisados em MO. A análise da expressão da iNOS e da COX-2 foi realizada em células do exsudato por RT-PCR. O acúmulo de nitrito (NO2−) no sobrenadante do cultivo de células RAW 264.7 foi determinado usando um ensaio colorimétrico baseado na reação de Griess. Os resultados foram expressos como média EP e comparados usando ANOVA seguido de teste de Dunnet. Os experimentos foram realizados em triplicata. No modelo air pouch, a injeção de carragenina 1% aumentou tanto a migração celular quanto a concentração de proteína no exsudato. Contudo, enquanto o pré-tratamento com a Fr40 aumentou a resposta inflamatória, o pré-tratamento com o EAEm e a Fr20, sobretudo por via i.p., inibiu esta resposta quando comparado ao grupo controle tratado apenas com o veículo. Assim, foram observadas as seguintes razões de inibição da migração de células: EAEm, i.p. a 25 mg/kg (66,44%) e a 250 mg/kg (87,27%) e Fr20 a 2,5 mg/kg (26,89%), 25 mg/kg (60,06%), 50 mg/kg (63,13%) e a 100 mg/kg (77,47%). Em relação à contagem diferencial, o EAEm e a Fr20 afetaram principalmente o número de neutrófilos, inibindo sua migração no exsudato. O EAEm e a Fr20 também reduziram a concentração total de proteínas no exsudato principalmente no tratamento i.p.; EAEm a 25 e 250 mg/kg mostrou 3,33 0,55 e 2,05 0,51 mg/mL, respectivamente, quando comparado aos grupos controles (Indometacina 2.88 0.64 mg/mL; Veículo 5.48 0.88 mg/mL). A Fr20 a 2,5, 25, 50 e 100 mg/kg mostrou 4,788 0,444, 1,417 0,519, 2,474 0,529 e 2,215 0, 361 mg/mL. A análise histológica mostrou infiltrado celular, principalmente composto de leucócitos polimorfonucleares ao longo da derme inflamada de animais tratados com veículo. O tratamento com o EAEm ou Fr20 reduziu a infiltração de leucócitos no tecido inflamado. Além disso, o tratamento com o EAEm e a Fr20 mostrou atividade supressora sobre a expressão de iNOS e COX-2, e mostrou efeitos inibitórios na produção de NO induzida por LPS. Concluindo, todos estes resultados confirmam o potencial antiinflamatório sugerido para esta planta e fornecem uma base para a compreensão de seus mecanismos moleculares de ação. Contudo, outros estudos devem ser realizados para melhor elucidar as vias pelas quais o EAEm e a Fr20 exercem seus efeitos antiinflamatórios. Além disso, estudos fitoquímicos devem ser realizados para identificar os compostos ativos no EAEm e na Fr20.

Jerdonitin is a P-II class snake venom metalloproteinase comprising metalloproteinase and disintegrin domains. In this study, we established a high-level expression system in Pichia pastoris and developed a purification strategy for the recombinant Jerdonitin. This recombinant Jerdonitin degraded fibrinogen at a level of activity comparable with its wild type. The effects of recombinant Jerdonitin on inhibiting ADP-induced human platelet aggregation were in a dose-dependent manner with an IC50 of 248 nM. In addition, we reported here that Jerdonitin can significantly inhibit the growth of several cell lines, including human liver cancer cells (Bel7402), human leukemia cells (K562) and human gastric carcinoma cells (BGC823). This study offers recombinant Jerdonitin that will be valuable for further functional and structural studies of Jerdonitin. (C) 2009 Elsevier Ltd. All rights reserved.

A controllable silver nanoparticle aggregate system has been synthesized by adding different amounts of ethanol to cetyltrimethylammonium bromide (CTAB) capped silver nanoparticles (Ag-nps), which could be used as highly efficient surface-enhanced Raman scattering (SERS) active substrates. This ethanol-induced aggregation can be attributed to preferential dissolution of CTAB into ethanol, which leads a partial removal of the protective CTAB layer on Ag-nps. The optical and morphological properties of these aggregates under various volumes of ethanol were explored via UV-vis spectroscopy and atomic force microscopy.

The phase and morphology variations of titania prepared in ethanol/acetic acid mixture solvents have been systematically investigated. X-ray diffraction results and microscopy observations reveal that pure anatase aggregates consisted of small nanoparticles, pure rutile microspheres comprised of nanofibers, and their mixtures could be obtained by varying ratios of ethanol to acetic acid under solvothermal conditions. The contents of anatase and rutile in the mixed phases also vary with the ratios of ethanol to acetic acid. Field emission scanning electron microscopy and high resolution transmission electron microscopy results show that the two phases are separated from each other in final products and form aggregates with morphologies resembling to their pure phase products obtained under favorable conditions. The as-produced rutile nanofibers, either in pure phase or in mixed phases, tend to grow into hollow microspheres.

A new sensitive assay for aspartate aminotransterase (AST) and alanine aminotransferase (ALT) activities in biofluids was developed, based on the separation and detection of alanine, glutamate, and aspartate using capillary electrophoresis (CE) with electrochemiluminescence (ECL) detection. The three amino acids were separated in 5 mM phosphate of pH 2.1 as background electrolyte, and detected on a 500 mu m platinum disk electrode at 1.2 V (versus Ag/AgCl) in the presence of 10 mM tris(2,2'-bipyridyl)ruthenium(II) dissolved in 80 mM phosphate of pH 10.5. A mass detection limit of 37.3 fmol (or 81.5 fmol) for glutamate, corresponding to the product in the enzyme reaction catalyzed by 1.24 x 10(-9) U AST (or 2.72 x 10(-9) U ALT) in a 30 min reaction period, was achieved. This assay was applied to investigate the cytotoxicity effect of ethanol on HepG2 cells and differentiating nonalcoholic steatohepatitis (NASH) from alcoholic liver disease, indicating that the technique is promising for the application in the cell biological and clinical fields.

Different mechanisms for the formation of acetaldehyde and ethanol on the Rh-based catalysts were investigated by the TPR (temperature programmed reaction) method, and the active sites were studied by CO-TPD, TPSR (temperature programmed surface reaction of preadsorbed CO by H-2) and XPS techniques. The TPR results indicated that ethanol and acetaldehyde might be formed through different intermediates, whereas ethanol and methanol might result from the same intermediate. Results of CO-TPD, TPSR, and XPS showed that on the Rh-based catalyst, the structure of the active sites for the formation of C-2-oxygenates is ((RhxRhy+)-Rh-0)-O-Mn+ (M=Mn or Zr, x>>y, 2 less than or equal ton less than or equal to4). The tilt-adsorbed CO species is the main precursor for CO dissociation and the precursor for the formation of ethanol and methanol. Most of the linear and geminal adsorbed CO species desorbed below 500 K. Based on the suggested model of the active sites, detailed mechanisms for the formation of acetaldehyde and ethanol are proposed. Ethanol is formed by direct hydrogenation of the tilt-adsorbed CO molecules, followed by CH2 insertion into the surface CH2-O species and the succeeding hydrogenation step. Acetaldehyde is formed through CO insertion into the surface CH3-Rh species followed by hydrogenation, and the role of the promoters was to stabilize the intermediate of the surface acetyl species. In the present work, Nafion (R) membrane porosity changes were determined in aqueous ethanol solutions with different concentrations by weighing vacuum-dried and ethanol aqueous solution equilibrated membranes at room temperature. The ethanol crossover rate through Nafion (R)-115 membrane at different temperatures and different concentrations had been investigated in a fuel cell test apparatus by using membrane gets higher as ethanol solution gas chromatography analysis. The experimental results show that the swelling degree of Nafion (R) concentration increases. The ethanol crossover rate increases with ethanol concentration and temperature increment. The single direct ethanol fuel cell (DEFC) tests were carried out to investigate the effect of ethanol concentration on ethanol crossover and consequently, on the open circuit voltage and the cell performance of DEFC. It can be found that ethanol crossover presented a negative effect on the OCV and the cell performance of DEFC. It can also be found that an improved DEFC performance was obtained as temperature increased although the ethanol crossover rate increased with temperature increment. (c) 2005 Elsevier B.V. The principal objective of this thesis was to investigate the ability of reversible optical O2 sensors to be incorporated into food/beverage packaging systems to continuously monitor O2 levels in a non-destructive manner immediately postpackaging and over time. Residual levels of O2 present in packs can negatively affect product quality and subsequently, product shelf-life, especially for O2-sensitive foods/beverages. Therefore, the ability of O2 sensors to continuously monitor O2 levels present within food/beverage packages was considered commercially relevant in terms of identifying the consequences of residual O2 on product safety and quality over time. Research commenced with the development of a novel range of O2 sensors based on phosphorescent platinum and palladium octaethylporphyrin-ketones (OEPk) in nano-porous high density polyethylene (HDPE), polypropylene (PP) polytetrafluoroethylene (PTFE) polymer supports. Sensors were calibrated over a temperature range of -10°C to +40°C and deemed suitable for food and beverage packaging applications. This sensor technology was used and demonstrated itself effective in determining failures in packaging containment. This was clearly demonstrated in the packaging of cheese string products. The sensor technology was also assessed across a wide range of packaged products; beer, ready-to-eat salad products, bread and convenience-style, muscle-based processed food products. The O2 sensor technology performed extremely well within all packaging systems. The sensor technology adequately detected O2 levels in; beer bottles prior to and following pasteurisation, modified atmosphere (MA) packs of ready-to-eat salad packs as respiration progressed during product storage and MA packs of bread and convenience-style muscle-based products as mycological growth occurred in food packs over time in the presence and absence of ethanol emitters. The use of the technology, in conjunction with standard food quality assessment techniques, showed remarkable usefulness in determining the impact of actual levels of O2 on specific quality attributes. The O2 sensing probe was modified, miniaturised and automated to screen for the determination of total aerobic viable counts (TVC) in several fish species samples. The test showed good correlation with conventional TVC test (ISO:4833:2003), analytical performance and ruggedness with respect to variation of key assay parameters (probe concentration and pipetting volume). Overall, the respirometric fish TVC test was simple to use, possessed a dynamic microbial range (104-107 cfu/g sample), had an accuracy of +/- one log(cfu/g sample) and was rapid. Its ability to assess highly perishable products such as fish for total microbial growth in <12 hr demonstrates commercial potential.

Biofilm production is an important step in the pathogenesis of Staphylococcus epidermidis polymer-associated infections and depends on the expression of the icaADBC operon leading to the synthesis of a polysaccharide intercellular adhesin. A chromosomally encoded reporter gene fusion between the ica promoter and the beta-galactosidase gene lacZ from Escherichia coli was constructed and used to investigate the influence of both environmental factors and subinhibitory concentrations of different antibiotics on ica expression in S. epidermidis. It was shown that S. epidermidis biofilm formation is induced by external stress (i.e., high temperature and osmolarity). Subinhibitory concentrations of tetracycline and the semisynthetic streptogramin antibiotic quinupristin-dalfopristin were found to enhance ica expression 9- to 11-fold, whereas penicillin, oxacillin, chloramphenicol, clindamycin, gentamicin, ofloxacin, vancomycin, and teicoplanin had no effect on ica expression. A weak (i.e., 2.5-fold) induction of ica expression was observed for subinhibitory concentrations of erythromycin. The results were confirmed by Northern blot analyses of ica transcription and quantitative analyses of biofilm formation in a colorimetric assay.

The solubility of water in the hydrophobic 1-alkyl-3-methylimidazolium hexafluorophosphate (alkyl = butyl, hexyl, and octyl) ionic liquids, can be significantly increased in the presence of ethanol as a co-solute. 1-Hexyl-3-methylimidazolium hexafluorophosphate and 1-octyl-3-methylimidazolium hexafluorophosphate are completely miscible with ethanol, and immiscible with water, whereas 1-butyl-3-methylimidazolium hexafluorophosphate is totally miscible with aqueous ethanol only between 0.5-0.9 mole fraction ethanol at 25degreesC. At higher and lower mole fraction of ethanol, the aqueous and IL components are only partially miscible and a biphasic system is obtained upon mixing equal volumes of the IL and aqueous ethanol. The observation of a large range of total miscibility between water and the IL in the three-component system has important implications for purifications and separations from IL.
