本研究采用平板式淀粉胶蛋白电泳技术对来自德钦县、巧家县、腾冲县的绵羊进行了 34个同功酶及血液蛋白的研究 ,共检测 36个遗传座位 ,其中有 8个座位显示多态 ,德钦绵羊多态百分率P =0 .194 4 ,平均杂合度H =0 .0 6 4 7,平均等位基因数A =1.2 2 2 2 ;昭通绵羊P =0 .2 2 2 2 ,H =0 .0 6 5 1;A =1.2 2 2 2 ;腾冲绵羊P =0 .16 6 7,H =0 .0 6 0 8;A =1.16 6 7。表明云南不同地区绵羊在蛋白水平上遗传多样性较为丰富。运用Phylip3.5软件包中的”GENETDIST”计算标准遗传距离 ,再结合以往研究的数据 ,运用该软件包中的”NEIGHBOR”和”UPGMA”法进行聚类分析 ,结果表明 ,云南绵羊起源于同一共同祖先 ,云南绵羊与印度绵羊、尼泊尔绵羊关系较近 ,而与西藏绵羊关系较远

采用水平式淀粉胶电泳技术 ,对云南龙陵黄山羊、宁蒗黑头山羊、马关无角山羊和路南圭山羊等 4个保种山羊的 1 2 0个个体共 3 9个基因座位的基因多态性进行了研究。结果显示 ,云南 4个保种山羊品种在AKP、CES - 1、ESD、GOI、LAP、MDH、ME和NP基因座位出现多态。多态座位基因在不同保种山羊中分布不同。多态基因座位百分比 (P)在 4个保种山羊中分别为 0 .2 0 51 ,0 .1 53 8,0 .1 2 82和 0 .1 53 8。平均杂合度 (H)分别为 0 .0 95,0 .0 61 4 ,0 .0 4 67和 0 .0 662。用UPGMA法对由基因频率计算得到的Nei氏标准遗传距离进行聚类分析 ,结果表明云南保种山羊具地理分布及品种特点 ,龙陵黄山羊和其它 3个品种的遗传距离最远。

采用水平式淀粉胶凝胶电泳技术和垂直式聚丙烯酰胺凝胶电泳技术检测了云南中 甸尼西鸡的血液蛋白多态,对33个样本的37个基因座位进行了分析,共有9个座位出现多态,分 别为ES-1、ES-2、AKP-1、AKP-2、LAP、PGM、CK、Tf及6PGD。多态位点百分比P=0.2432,平 均杂合度=0.1015,每个座位等位基因的平均数A=1.3784,结果表明尼西鸡的血液蛋白多态程度较 高,在蛋白质水平上的遗传多样性较为丰富。同时发现Amy-1座位只出现杂合子AB型。
GENETIC DIVERSITY IN THE CHINESE PANGOLIN (MANIS-PENTADACTYLA) INFERRED FROM PROTEIN ELECTROPHORESIS

We examined protein polymorphism of Chinese pangolins (Manis pentadactyla) from Yunnan Province of China, including two forms of three brown and nine dusky Chinese pangolins. Sixty-two genetic loci were screened; 12 loci were found to be polymorphic. The percentage of polymorphic loci (P) is 0.194, the mean individual heterozygosity (H) is 0.078, and the mean number of alleles (A) is 1.258. Furthermore, we calculated the genetic distance (D) between the two forms and found a low level of genetic divergence (D = 0.0206) between them, which indicates an almost-indistinguishable divergence at the level of proteins.

Background: Due to the advances of high throughput technology and data-collection approaches, we are now in an unprecedented position to understand the evolution of organisms. Great efforts have characterized many individual genes responsible for the interspecies divergence, yet little is known about the genome-wide divergence at a higher level. Modules, serving as the building blocks and operational units of biological systems, provide more information than individual genes. Hence, the comparative analysis between species at the module level would shed more light on the mechanisms underlying the evolution of organisms than the traditional comparative genomics approaches. Results: We systematically identified the tissue-related modules using the iterative signature algorithm (ISA), and we detected 52 and 65 modules in the human and mouse genomes, respectively. The gene expression patterns indicate that all of these predicted modules have a high possibility of serving as real biological modules. In addition, we defined a novel quantity, "total constraint intensity,'' a proxy of multiple constraints (of co-regulated genes and tissues where the co-regulation occurs) on the evolution of genes in module context. We demonstrate that the evolutionary rate of a gene is negatively correlated with its total constraint intensity. Furthermore, there are modules coding the same essential biological processes, while their gene contents have diverged extensively between human and mouse. Conclusions: Our results suggest that unlike the composition of module, which exhibits a great difference between human and mouse, the functional organization of the corresponding modules may evolve in a more conservative manner. Most importantly, our findings imply that similar biological processes can be carried out by different sets of genes from human and mouse, therefore, the functional data of individual genes from mouse may not apply to human in certain occasions.

The regional distribution of an ancient Y-chromosome haplogroup C-M130 (Hg C) in Asia provides an ideal tool of dissecting prehistoric migration events. We identified 465 Hg C individuals out of 4284 males from 140 East and Southeast Asian populations. We genotyped these Hg C individuals using 12 Y-chromosome biallelic markers and 8 commonly used Y-short tandem repeats (Y-STRs), and performed phylogeographic analysis in combination with the published data. The results show that most of the Hg C subhaplogroups have distinct geographical distribution and have undergone long-time isolation, although Hg C individuals are distributed widely across Eurasia. Furthermore, a general south-to-north and east-to-west cline of Y-STR diversity is observed with the highest diversity in Southeast Asia. The phylogeographic distribution pattern of Hg C supports a single coastal 'Out-of-Africa' route by way of the Indian subcontinent, which eventually led to the early settlement of modern humans in mainland Southeast Asia. The northward expansion of Hg C in East Asia started similar to 40 thousand of years ago (KYA) along the coastline of mainland China and reached Siberia similar to 15 KYA and finally made its way to the Americas. A novel peptide inhibitor (OGTI) of serine protease with a molecular weight of 1949.8, was purified from the skin secretion of the frog, Odorrana grahami. Of the tested serine proteases, OGTI only inhibited the hydrolysis activity of trypsin on synthetic chromogenic substrate. This precursor deduced from the cDNA sequence is composed of 70 amino acid residues. The mature OGTI contains 17 amino acid residues including a six-residue loop disulfided by two half-cysteines (AVNIPFKVHFRCKAAFC). In addition to its unique six-residue loop, the overall structure and precursor of OGTI are different from those of other serine protease inhibitors. While conducting experiments to investigate antimicrobial peptides of amphibians living in the Yunnan-Sichuan region of southwest China, a new family of antimicrobial peptides was identified from skin secretions of the rufous-spotted torrent frog, Amolops loloensis. Members of the new peptide family named amolopins are composed of 18 amino acids with a unique sequence, for example, NILSSIVNGINRALSFFG. By BLAST search, amolopins did no show similarity to any known peptides. Among the tested microorganisms, native and synthetic peptides only showed antimicrobial activities against Staphylococcus aureus ATCC2592 and Bacillus pumilus, no effects on other microorganisms. The CD spectroscopy showed that it adopted a structure of random combined with beta-sheet in water, Tris-HCl or Tris-HCl-SDS. Several cDNAs encoding amolopins were cloned from the skin cDNA library of A. loloensis. The precursors of amolopin are composed of 62 amino acid residues including predicted signal peptides, acidic propieces, and mature antimicrobial peptides. The preproregion of amolopin precursor comprises a hydrophobic signal peptide of 22 residues followed by an 18 residue acidic propiece which terminates by a typical prohormone processing signal Lys-Arg. The preproregions of precursors are very similar to other amphibian antimicrobial peptide precursors but the mature amolopins are different from other antimicrobial peptide families. The remarkable similarity of preproregions of precursors that give rise to very different antimicrobial peptides in distantly related frog species suggests that the corresponding genes form a multigene family originating from a common ancestor. The Ag5 proteins are the most abundant and immunogenic proteins in the venom secretory ducts of stinging insects. An antigen 5-like protein (named tabRTS) composed of 221 amino acid residues was purified and characterized from the salivary glands of the horsefly, Tabanus yao (Diptera, Tabanidae). Its cDNA was cloned from the cDNA library of the horsefly's salivary gland. TabRTS containing the SCP domain (Sc7 family of extracellular protein domain) was found in insect antigen 5 proteins. More interestingly, there is an Arg-Thr-Ser (RTS) disintegrin motif at the C-terminus of tabRTS. The RTS motif is positioned in a loop bracketed by cysteine residues as those found in RTS-disintegrins of Crotalidae and Viperidae snake venoms, which act as angiogenesis inhibitors. Endothelial Cell Tube formation assay in vitro and chicken chorioallantoic membrane (CAM) angiogenesis assay in vivo were performed as to investigate the effect of tabRTS on angiogenesis. It was found that tabRTS could significantly inhibit angiogenesis in vitro and in vivo. Anti-alpha(1)beta(1) monoclonal antibody could dose-dependently inhibit the anti-angiogenic activity of tabRTS. This result indicated that tabRTS possibly targets the alpha(1)beta(1) integrin to exert the anti-angiogenic activity as snake venom RTS-/KTS-disintegrins do. The current work revealed the first angiogenesis inhibitor protein containing RTS motif from invertebrates, a possible novel type of RTS-disintegrin. (C) 2009 Elsevier Ltd. All rights reserved.
