The contribution of two blue-green algae species, Anabaeria flos-aquae and Microcystis aeruginosa, to the formation of trihalomethanes (THMs) and haloacetic acids (HAAs) was investigated. The experiments examined the formation potential of these disinfection by-products (DBPs) from both algae cells and extracellular organic matter (EOM) during four algal growth phases. Algal cells and EOM of Anabaena and Microcystis exhibited a high potential for DBP formation. Yields of total THMs (TTHM) and total HAAs (THAA) were closely related to the growth phase. Reactivity of EOM from Anabaena was slightly higher than corresponding cells, while the opposite result was found for Microcystis. Specific DBP yields (yield/unit C) of Anabaena were in the range of 2-11 mu mol/mmol C for TTHM and 217 mu mol/mmol C for THAA, while those of Microcystis were slightly higher. With regard to the distributions of individual THM and HAA compounds, differences were observed between the algae species and also between cells and EOM. The presence of bromide shifted the dominant compounds from HAAs to THMs. (C) 2009 Elsevier Ltd. All rights reserved.

In nature, living creatures are affected by several stimuli simultaneously. The response of living creatures to stimuli is called taxis. In order to reveal the principles of taxis behavior in response to complex stimuli, we simultaneously applied photostimulation and electric stimulation perpendicularly to a Volvox algae solution. The probability distribution of the swimming direction showed that a large population of swimming cells moved in a direction that was the result of the composition of phototaxis and electrotaxis. More surprisingly, we uncovered the coupling of signs of taxis, i.e., coupling of phototaxis and electrotaxis induced positive electrotaxis, which did not emerge in the single stimulation experiments. We qualitatively explained the coupling of taxis based on the polarization of the swimming cells induced by the simultaneous photo- and electric stimulation.

This study develops a simplified model describing the evolutionary dynamics of a population composed of obligate sexually and asexually reproducing, unicellular organisms. The model assumes that the organisms have diploid genomes consisting of two chromosomes, and that the sexual organisms replicate by first dividing into haploid intermediates, which then combine with other haploids, followed by the normal mitotic division of the resulting diploid into two new daughter cells. We assume that the fitness landscape of the diploids is analogous to the single-fitness-peak approach often used in single-chromosome studies. That is, we assume a master chromosome that becomes defective with just one point mutation. The diploid fitness then depends on whether the genome has zero, one, or two copies of the master chromosome. We also assume that only pairs of haploids with a master chromosome are capable of combining so as to produce sexual diploid cells, and that this process is described by second-order kinetics. We find that, in a range of intermediate values of the replication fidelity, sexually reproducing cells can outcompete asexual ones, provided the initial abundance of sexual cells is above some threshold value. The range of values where sexual reproduction outcompetes asexual reproduction increases with decreasing replication rate and increasing population density. We critically evaluate a common approach, based on a group selection perspective, used to study the competition between populations and show its flaws in addressing the evolution of sex problem.

RNA isolation is essential to study gene expression at the molecular level. However, RNA isolation is difficult in organisms (plants and algae) that contain large amounts of polysaccharides, which co-precipitate with RNA. Currently, there is no commercial kit available, specifically for the isolation of high-quality RNA from these organisms. Furthermore, because of the large amounts of polysaccharides, the common protocols for RNA isolation usually result in poor yields when applied to algae. Here we describe a simple method for RNA isolation from the marine red macroalga Gracilaria tenuistipitata var. liui Zhang et Xia (Rhodophyta), which can be applied to other plants and algae.

Four species of marine benthic algae (Laurencia filiformis, L. intricata, Gracilaria domingensis and G. birdiae) that belong to the phylum Rhodophyta were collected in Espirito Santo State, Brazil and investigated concerning their biochemical composition (fatty acid, total lipid, soluble proteins, amino acid and ash). The total content of lipid (% dry weight) ranged from 1.1% to 6.2%: fatty acid from 0.7% to 1.0%: soluble protein from 4.6% to 18.3%, amino acid from 6.7% to 11.3% and ash from 22.5% to 38.4%. judging from their composition, the four species of algae appear to be potential sources of dietary proteins, amino acids, lipids and essential fatty acids for humans and animals. This paper reports experiments involving the electrochemical combustion of humic acid (HA) and removal of algae from pond water. An electrochemical flow reactor with a boron-doped diamond film anode was used and constant current experiments were conducted in batch recirculation mode. The mass transfer characteristics of the electrochemical device were determined by voltammetric experiments in the potential region of water stability, followed by a controlled current experiment in the potential region of oxygen evolution. The average mass transfer coefficient was 5.2 x 10(-5) m s(-1). The pond water was then processed to remove HA and algae in the conditions in which the reaction combustion occurred under mass transfer control. To this end, the mass transfer coefficient was used to estimate the initial limiting current density applied in the electrolytic experiments. As expected, all the parameters analyzed here-solution absorbance at 270 nm, total phenol concentration and total organic carbon concentration-decayed according to first-order kinetics. Since the diamond film anode successfully incinerated organic matter, the electrochemical system proved to be predictable and programmable.

Little is known about the microbial diversity associated with marine macroorganisms, despite the vital role microorganisms may play in marine ecosystems. The aim of the present study was to investigate the diversity of bacteria and fungi isolated from eight marine invertebrate and one algae samples. Data derived from ARDRA and sequencing analyses allowed the identification of marine-derived microorganisms isolated from those samples. Microbial strains identified up to the genus level revealed 144 distinct ribotypes out of 256 fungal strains and 158 distinct ribotypes out of 181 bacterial strains. Filamentous fungi were distributed among 24 different genera belonging to Ascomycota, Zygomycota and Basidiomycota, some of which had never been reported in the literature as marine invertebrate-inhabiting fungi (Pestalotiopsis, Xylaria, Botrysphaeria and Cunnninghamella). Bacterial isolates were affiliated to 41 different genera, being Bacillus, Ruegeria, Micrococcus, Pseudovibrio and Staphylococcus the most abundant ones. Results revealed an unexpected high microbial diversity associated to the macroorganisms which have been collected and suggested the selection of certain microbial taxonomic groups according to the host. The combined data gathered from this investigation contribute to broaden the knowledge of microbial diversity associated to marine macroorganisms, including as a promising source for the discovery of new natural products. The Brazilian Northeast is the most vulnerable region to climatic variability risks. For the Brazilian semi-arid is expected a reduction in the overall rates of precipitation and an increase in the number of dry days. These changes predicted by the IPCC (2007) will intensify the rainfall and droughts period that could promote the dominance of cyanobacteria, thus affecting the water quality of reservoirs, that are most used for water supply, in the semi-arid. The aim of this study was to evaluate the effects of increasing temperature combined with nutrient enrichment on the functional structure of the phytoplankton community of a mesotrophic reservoir in the semi-arid, in the worst case scenario of climate change predicted by the IPCC (2007). Two experiments were performed, one in a rainy season and another in the dry season. In the water sampled, nutrients (nitrate and orthophosphate) were added in different concentrations. The microcosms were submitted to two different temperatures, five-year average of air temperature in the reservoir (control) and 4°C above the control temperature (warming). The results of this study showed that warming and nutrient enrichment benefited mainly the functional groups of cyanobacteria. During the rainy season it was verified the increasing biomass of small functional groups of unicellular and opportunists algae such as F (colonial green algae with mucilage) and X1 (nanoplanktonic algae of eutrophic lake systems). It was also observed an increasing in total biomass, in the richness and diversity of the community. In the dry season experiment there was a greater contribution in the relative biomass of filamentous algae, with a replacement of the group S1 (non-filamentous cyanobacteria with heterocytes) for H1 (filamentous cyanobacteria with heterocytes) in nutrient- enriched treatments. Moreover, there was also loss in total biomass, species richness and diversity of the community. The effects of temperature and nutrients manipulation on phytoplankton community of reservoir Ministro João Alves provoked changes in species richness, the diversity of the community and its functional composition, being the dry period which showed the highest susceptibility to the increase in the contribution of potentially toxic cyanobacteria with heterocytes

Acanthonyx scutiformis, an endemic species in the Brazilian coast, is commonly found in intertidal rocky-shore algal communities. This study analyzes the population biology of A. scutiformis from Ubatuba region. A total of 371 specimens were collected over one year. Size range was 4.2[long dash]12.7 mm CW (carapace width) for females and 3.7[long dash]15.8 mm CW for males. Females predominated in intermediate size classes, whereas males prevailed in the largest ones. The estimated size when 50% crabs were mature was 10.7 mm CW for males and 8.9 mm CW for females. Sex ratio varied among the demographic groups. The processes that influence A. scutiformis population structure can be related to the different times males and females reach sexual maturity and probably to the distinct predation pressures on each sex during the adult phase.

The responses of relative growth rate (% day-1) and pigment content (chlorophyll a, phycocyanin and phycoerythrin) to temperature, irradiance and photoperiod were analyzed in culture in seven freshwater red algae: Audouinella hermannii (Roth) Duby, Audouinella pygmaea (Kützing) Weber-van Bosse, Batrachospermum ambiguum Montagne, Batrachospermum delicatulum (Skuja) Necchi et Entwisle, 'Chantransia' stages of B. delicatulum and Batrachospermum macrosporum Montagne and Compsopogon coeruleus (C. Agardh) Montagne. Experimental conditions included temperatures of 10, 15, 20 and 25°C and low and high irradiances (65 and 300 μmol photons m-2 s-1, respectively). Long and short day lengths (16:8 and 8:16 LD cycles) were also applied at the two irradiances. Growth effects of temperature and irradiance were evident in most algae tested, and there were significant interactions among treatments. Most freshwater red algae had the best growth under low irradiance, confirming the preference of freshwater red algae for low light regimens. In general there was highest growth rate in long days and low irradiance. Growth optima in relation to temperature were species-specific and also varied between low and high irradiances for the same alga. The most significant differences in pigment content were related to temperature, whereas few significant differences could be attributed to variation in irradiance and photoperiod or interactions among the three parameters. The responses were species-specific and also differed for pigments in distinct temperatures, irradiances and photoperiods in the same alga. Phycocyanin was generally more concentrated than phycoerythrin and phycobiliproteins were more concentrated than chlorophyll a. The highest total pigment contents were found in two species typical of shaded habitats: A. hermannii and C. coeruleus. The expected inverse relationship of pigment with irradiance was observed only in C. coeruleus. In general, the most favorable conditions for growth were not coincident with those with highest pigment contents.

Fourteen culture isolates of freshwater acrochaetioid algae from distinct regions around the world were analysed, including the reddish species Audouinella hermannii, the dubious blue-greenish species A. pygmaea, and Chantransia stages from distinct taxonomic origins in the Batrachospermales sensu lato (Batrachospermaceae, Lemaneaceae and Thoreaceae). Four isolates (two 'Chantransia' stages and two species of Audouinella, A. hermannii and A. pygmaea) were tested under experimental conditions of temperature (10-25°C), irradiance (65 and 300 μmol photons m-2 s-1) and photoperiod (16:8 h and 8:16 h light/dark cycles). Plant colour is proposed as the only vegetative character that can be unequivocally applied to distinguish Audouinella from 'Chantransia', blue-greenish representing Chantransia stages and reddish applying to true Audouinella species (also forming reproductive structures other than monosporangia, e.g. tetrasporangia). Some isolates of A. pygmaea were proven to be unequivocally 'Chantransia stages owing either to production of juvenile gametophytes or to derivation from carpospores. No association of the morphology of A. pygmaea was found with any particular species, thus it should be regarded as a complex involving many species of the Batrachospermales sensu lato, as is also the case with A. macrospora. We therefore recommend that all blue-greenish acrochaetioid algae in freshwater habitats be considered as Chantransia stages of members of the Batrachospermales, and that the informal descriptors pygmaea and macrospora be used to distinguish the two discernable morphologies. Induction of gametophytes occurred under much wider conditions than previously reported, reinforcing the conclusion that requirements are probably species-specific. Although phenotypic plasticity was in evidence, with temperature, irradiance and photoperiod affecting morphology, no alga showed variation outside the limits based on traditional taxonomic studies. No overall trend was observed for vegetative or reproductive characters in response to temperature, irradiance and photoperiod for all the algae tested, only for specific algae or characters. Effects of temperature and irradiance on morphological characters were more evident, as well as strong interactions between these variables, whereas few differences were generally found in response to photoperiod and irradiance.
