The effect of the contaminant fungi Trichoderma sp. and Chaetomium olivacearum on the cultivation of the ABL 99/30 and ABL 04/49 isolates of A. blazei in two compost formulations made up with tyfton (Cynodom dactylon) and oat (Avena sativa) was evaluated. The experimental design was a totally randomized 2 x 2 x 3 factorial design with 6 repetitions. The experimental unit consisted of 12-12.5 kg of wet compost. During the spawning, 150 g of Trichoderma sp. and C. olivacearum were added to the compost. The experiment was carried out in a climatized room, with humidity between 75-90% and temperature of 28º C. The productivity averages of the ABL 99/30 isolate of A. blazei were higher than those of ABL 04/49 and Trichoderma sp. and C. olivacearum negatively influenced the production of A. blazei. The different composts (based on tyfton and oat straw) did not influence the production of basidiomata.

Huanglongbing (HLB) is associated with Candidatus Liberibacter spp., endogenous, sieve tube-restricted bacteria that are transmitted by citrus psyllid insect vectors. Transgenic expression in the phloem of specific genes that might affect Ca. Liberibacter spp. growth and development may be an adequate strategy to improve citrus resistance to HLB. To study specific phloem gene expression in citrus, we developed three different binary vector constructs with expression cassettes bearing the beta-glucuronidase (GUS) reporter gene (uidA) under the control of one of the three different promoters: Citrus phloem protein 2 (CsPP2), Arabidopsis thaliana phloem protein 2 (AtPP2), and Arabidopsis thaliana sucrose transporter 2 (AtSUC2). Transgenic lines of 'Hamlin', 'Pera', and 'Valencia' sweet oranges [Citrus sinensis (L.) Osbeck] were produced via Agrobacterium tumefaciens transformation. The epicotyl segments collected from in vitro germinated seedlings were used as explants. The gene nptII, which confers resistance to the antibiotic kanamycin, was used for selection. The transformation efficiency was expressed as the number of GUS-positive shoots over the total number of explants and varied from 1.54 to 6.08 % among the three cultivars and three constructs studied. Several lines of the three sweet orange cultivars analyzed using PCR and Southern blot analysis were genetically transformed with the three constructs evaluated. The histological GUS activity in the leaves indicates that the uidA gene was preferentially expressed in the phloem, which suggests that the use of the three promoters might be adequate for producing HLB-resistant transgenic sweet oranges. The results reported here conclusively demonstrate the preferential expression of GUS in the phloem driven by two heterologous and one homologous gene promoters. Nine marine fungi (Aspergillus sclerotiorum CBMAI 849, Aspergillus sydowii Ce19, Beauveria felina CBMAI 738, Mucor racemosus CBMAI 847, Penicillium citrinum CBMAI 1186, Penicillium miczynskii Ce16, P. miczynskii Gc5, Penicillium oxalicum CBMAI 1185, and Trichoderma sp. Gc1) catalyzed the asymmetric bioconversion of iodoacetophenones 1-3 to corresponding iodophenylethanols 6-8. All the marine fungi produced exclusively (S)-ortho-iodophenylethanol 6 and (S)-meta-iodophenylethanol 7 in accordance to the Prelog rule. B. felina CBMAI 738, P. miczynskii Gc5, P. oxalicum CBMAI 1185, and Trichoderma sp. Gc1 produced (R)-para-iodophenylethanol 8 as product anti-Prelog. The bioconversion of para-iodoacetophenone 3 with whole cells of P. oxalicum CBMAI 1185 showed competitive reduction-oxidation reactions.

Aspergillus fumigatus is a major opportunistic pathogen and allergen of mammals. Nutrient sensing and acquisition mechanisms, as well as the capability to cope with different stressing conditions, are essential for A. fumigatus virulence and survival in the mammalian host. This study characterized the A. fumigatus SebA transcription factor, which is the putative homologue of the factor encoded by Trichoderma atroviride seb1. The Delta sebA mutant demonstrated reduced growth in the presence of paraquat, hydrogen peroxide, CaCl2, and poor nutritional conditions, while viability associated with sebA was also affected by heat shock exposure. Accordingly, SebA:GFP (SebA:green fluorescent protein) was shown to accumulate in the nucleus upon exposure to oxidative stress and heat shock conditions. In addition, genes involved in either the oxidative stress or heat shock response had reduced transcription in the Delta sebA mutant. The A. fumigatus Delta sebA strain was attenuated in virulence in a murine model of invasive pulmonary aspergillosis. Furthermore, killing of the Delta sebA mutant by murine alveolar macrophages was increased compared to killing of the wild-type strain. A. fumigatus SebA plays a complex role, contributing to several stress tolerance pathways and growth under poor nutritional conditions, and seems to be integrated into different stress responses.

In this paper, we present the results of an experimental approach developed to study the macroscopic and microbiological alteration of bird and small mammal bones buried under a Cerrado biome. The first experiment evaluated the macroscopic alteration of cooked and fresh carcasses buried through the dry and rainy seasons. The second experiment analyzed the mycobiota associated to the decomposition of a complete bird that remained buried for almost a year. Results show that in tropical forest environments: 1) bone structure and pre-taphonomic factors determine its differential alteration by biochemical processes; 2) fungal populations associated to the decomposition of animal remains depend on soil chemistry and ecological dynamics; 3) even in a corrosive environment, bird bones are more capable of surviving to several mycological decomposition steps. Abstract Background There is an imperative necessity for alternative sources of energy able to reduce the world dependence of fossil oil. One of the most successful options is ethanol obtained mainly from sugarcane and corn fermentation. The foremost residue from sugarcane industry is the bagasse, a rich lignocellulosic raw material uses for the production of ethanol second generation (2G). New cellulolytic and hemicellulytic enzymes are needed, in order to optimize the degradation of bagasse and production of ethanol 2G. Results The ability to produce hemicellulases and related enzymes, suitable for lignocellulosic biomass deconstruction, was explored using 110 endophytic fungi and 9 fungi isolated from spoiled books in Brazil. Two initial selections were performed, one employing the esculin gel diffusion assay, and the other by culturing on agar plate media with beechwood xylan and liquor from the hydrothermal pretreatment of sugar cane bagasse. A total of 56 isolates were then grown at 29°C on steam-exploded delignified sugar cane bagasse (DEB) plus soybean bran (SB) (3:1), with measurement of the xylanase, pectinase, β-glucosidase, CMCase, and FPase activities. Twelve strains were selected, and their enzyme extracts were assessed using different substrates. Finally, the best six strains were grown under xylan and pectin, and several glycohydrolases activities were also assessed. These strains were identified morphologically and by sequencing the internal transcribed spacer (ITS) regions and the partial β-tubulin gene (BT2). The best six strains were identified as Aspergillus niger DR02, Trichoderma atroviride DR17 and DR19, Alternaria sp. DR45, Annulohypoxylon stigyum DR47 and Talaromyces wortmannii DR49. These strains produced glycohydrolases with different profiles, and production was highly influenced by the carbon sources in the media. Conclusions The selected endophytic fungi Aspergillus niger DR02, Trichoderma atroviride DR17 and DR19, Alternaria sp. DR45, Annulohypoxylon stigyum DR47 and Talaromyces wortmannii DR49 are excellent producers of hydrolytic enzymes to be used as part of blends to decompose sugarcane biomass at industrial level.

Sporotrichosis is a subcutaneous mycosis and is also a zoonosis (sapro- and anthropozoonosis). The objective of the present study was to determine the occurrence of sporotrichosis in domestic cats and in wild or exotic felines in captivity through the isolation of Sporothrix spp. from claw impressions in a culture medium. The samples included 132 felines, of which 120 (91.0 %) were domestic cats, 11 (8.3 %) were wild felines, and one (0.7 %) was an exotic felid. Twenty-one (17.5 %) were outdoor cats. Of the total, 89 (67.4 %) had contact with other animals of the same species. It was possible to isolate Sporothrix schenckii from the claws of one (0.7 %) of the felids probed; this animal exhibited generalised sporotrichosis and had infected a female veterinarian. The potential pathogenic agents Microsporum canis and Malassezia pachydermatis were isolated in 12.1 and 5.3 % of the animals, respectively. The following anemophilous fungi, which were considered to be contaminants, were also isolated: Penicillium sp. (28 or 21.2 %), Aspergillus sp. (13 or 9.8 %), Rhodotorula sp. (5 or 3.8 %), Candida sp. (5 or 3.8 %), Trichoderma sp. (1 or 0.7 %), and Acremonium sp. (1 or 0.7 %). Due to the low magnitude of occurrence (0.7 %) of Sporothrix in feline claws, the potential of the cats evaluated in this study to be sources of infection in the city of São Paulo is considerably low.

Lo studio “Lotta biologica a Fusarium solani f.sp. cucurbitae su zucchino” si colloca nell’ambito della difesa integrata delle colture orticole dalle fitopatie fungine, in particolare quelle causate da patogeni ad habitat terricolo nei confronti dei quali è sempre più frequente il ricorso a mezzi di lotta diversi dai prodotti chimici. Interessante e innovativa appare la prospettiva di utilizzare microrganismi adatti a svilupparsi nel suolo, competenti per la rizosfera delle piante e con spiccate caratteristiche d’antagonismo verso i patogeni tellurici e di stimolazione delle difese sistemiche della pianta. Il marciume del colletto delle cucurbitacee, causato da diversi patogeni tra cui Fusarium solani f.sp. cucurbitae, rappresenta la principale malattia fungina di tipo tellurica che colpisce lo zucchino ed il melone nella Pianura Padana e che può portare a consistenti perdite produttive. Indagini condotte dal 2004 da parte del Diproval nell’areale bolognese, hanno evidenziato un’elevata frequenza del patogeno su zucchino coltivato soprattutto in tunnel. Considerata la carenza di conoscenze locali di F. solani f.sp. cucurbitae e di mezzi chimici di lotta efficaci, la ricerca svolta ha inteso approfondire la diagnosi della malattia e le caratteristiche biologiche degli isolati locali, e valutare la possibilità di utilizzare metodi biologici di lotta contro questo patogeno, nonché di studiare alcune delle possibili modalità d’azione di microrganismi antagonisti. Sono state pertanto prelevate, da zone diverse del Bolognese, campioni di piante di zucchino che presentavano sintomi di marciume del colletto ed è stato isolato il patogeno, che in base alle caratteristiche morfologiche macro e microscopiche, alle prove di patogenicità su diversi ospiti e a saggi biomolecolari, è stato identificato come Fusarium solani f. sp. cucurbitae W.C. Snyder & H.N. Hansen razza 1. Dagli isolati di campo sono state realizzate un centinaio di colture monosporiche venti delle quali sono state utilizzate per la prosecuzione delle prove. I venti ceppi sono stati saggiati per la loro patogenicità inoculandoli in terriccio sterile e con trapianto di giovani piante di zucchino. E’ risultata un’elevata variabilità del livello di virulenza tra i ceppi, stimata da 39% a 83% riguardo la gravità della malattia e da 61 a 96% per la frequenza di malattia. Sono state condotte prove di accrescimento miceliare che hanno evidenziato differenze tra i ceppi e tra gli esperimenti condotti a tre diverse temperature (17, 23 e 28°C) alla luce ed al buio. La crescita maggiore complessivamente è stata ottenuta a 23°C. I venti ceppi hanno anche mostrato di produrre, in vitro, vari livelli di enzimi di patogenesi quali cellulasi, poligalatturonasi, pectin liasi e proteasi. E’ stata evidenziata unan correlazione significativa tra attività cellulasica e pectin liasica con frequenza e gravità della malattia dei venti ceppi del patogeno. Le prove relative al contenimento della malattia sono state condotte in cella climatica. Sono stati considerati prodotti commerciali (Remedier, Rootshield, Cedomon, Mycostop, Proradix, Clonotry) a base rispettivamente dei seguenti microrganismi: Trichoderma harzianum ICC012 + T. viride ICC080, T. harzianum T22, Pseudomonas chlororaphis MA342, Streptomyces griseoviridis K61, P. fluorescens proradix DSM13134 e T. harzianum + Clonostachys rosea). I prodotti sono stati somministrati sul seme, al terreno e su seme+terreno (esperimenti 1 e 2) e in vivaio, al trapianto e vivaio+trapianto (esperimenti 3 e 4), riproducendo situazioni di pratico impiego. L’inoculazione del patogeno (un ceppo ad elevata patogenicità, Fs7 ed uno a bassa patogenicità, Fs37) è stata effettuata nel terreno distribuendo uno sfarinato secco di semi di miglio e cereali colonizzati dal patogeno. La malattia è stata valutata come intensità e gravità. I prodotti sono stati impiegati in situazioni di particolare stress, avendo favorito particolarmente la crescita del patogeno. Complessivamente i risultati hanno evidenziato effetti di contenimento maggiore della malattia nel caso del ceppo Fs37, meno virulento. La malattia è stata ridotta quasi sempre da tutti i formulati e quello che l’ha ridotta maggiormente è stato Cedomon. Il contenimento della malattia somministrando i prodotti solo nel terreno di semina o di trapianto è stato in generale quello più basso. Il contenimento più elevato è stato ottenuto con la combinazione di due tipologie di trattamento, seme+terreno e vivaio+trapianto. Le differenze tra i prodotti sono risultate più evidenti nel caso del ceppo Fs7. Per quanto riguarda lo studio di alcune delle modalità d’azione dei microrganismi contenuti nei formulati più efficaci, è stato verificato che tutti sono stati in grado di inibire, se pur in vario modo, la crescita del patogeno in vitro. Gli antagonisti più efficaci sono stati S. griseoviridis K61 (Mycostop) e P. fluorescens proradix DSM13134). I ceppi di Trichoderma, ed in particolare T.harzianum T22 (Rootshield), sono risultati i più attivi colonizzatori del substrato. Riguardo il fenomeno dell’antibiosi, il batterio P. fluorescens proradix DSM13134 ha mostrato di produrre i metaboliti non volatili più efficaci nel ridurre lo sviluppo del patogeno. Nelle condizioni sperimentali adottate anche i due ceppi di T. viride ICC080 e T. harzianum ICC012 hanno dimostrato di produrre metaboliti efficaci. Tali risultati, anche se relativi a prove in vitro, possono contribuire alla comprensione dei meccanismi dei microrganismi sul contenimento dell’infezione relativamente al rapporto diretto sul patogeno. E’ stato inoltre verificato che tutti i microrganismi saggiati sono dotati di competenza rizosferica e solo i batteri di endofitismo. Si conclude che, nonostante l’elevata pressione infettiva del patogeno che ha certamente influito negativamente sull’efficacia dei microrganismi studiati, i microrganismi antagonisti possono avere un ruolo nel ridurre l’infezione di F. solani f.sp. cucurbitae razza 1.

Ziel der Untersuchungen war, Pilze aus geschädigtem und ungeschädigtem Wurzelmaterial konventionell und ökologisch bewirtschafteter Weinbergsböden zu isolieren und diese auf ihre Durchsetzungsfähigkeit gegenüber den anderen Arten bzw. deren Pilzmetabolitsuspensionen unter unterschiedlichen Nahrungsbedingungen zu prüfen und eine eventuelle substratabhängige Verhaltensänderung bei den Spezies in Interaktion festzustellen. Zudem wurde in weiteren In-vitro- Versuchen das pathogene Potenzial der gefundenen Arten gegenüber Vitis spp. getestet. Hintergrund dieser Untersuchungen war die Hypothese, dass Absterbeerscheinungen in Rebanlagen nicht durch die Reblaus per se verursacht werden, sondern dass ein Zusammenhang zwischen der Bewirtschaftungsmethode und dem Schadbild in reblausbefallenen Rebanlagen besteht und dessen Entstehung auf pathogenkonduktive und –suppressive Eigenschaften des Bodens zurückgeführt werden kann. Aus rund 2400 Wurzelproben konnten insgesamt 49 Pilzarten isoliert und bestimmt und mehr als die Hälfte davon in Wurzeln beider Versuchsflächen nachgewiesen werden. Ein Großteil der Pilze wurde sowohl in geschädigten als auch in ungeschädigten Wurzelgeweben identifiziert. Darunter waren Arten, die in der Literatur als Parasiten und Saprobier beschrieben werden, aber auch Arten, die eine andere Lebensweise pflegen oder deren Lebensweise nicht bekannt ist. Mit Hilfe von Interaktionsversuchen auf unterschiedlichen Nährmedien (einem Voll- und einem Mangelmedium) konnte bei den untersuchten Arten teilweise starke substratabhängige Verhaltensänderung in Interaktion mit bestimmten Pilzkolonien festgestellt und auf die Verfügbarkeit von organischem Kohlenstoff zurückgeführt werden. Starke Konkurrenz um organischen Kohlenstoff und dadurch entstehende fungistatische und antibiotische Effekte können in diesem Zusammenhang pathogenkonduktive bzw. pathogensuppressive Bodeneigenschaften fördern oder hemmen. Weiterhin konnte gezeigt werden, dass alle 15 in vitro an Vitis spp. inokulierten Pilze (Absidia glauca, Acremonium kiliense, Aspergillus ustus, Cylindrocarpon magnusianum, Cylindrocarpon sp., Fusarium culmorum, F. detonianum, F. oxysporum, F. sacchari, F. semitectum, Gliocladium roseum, Leptosphaeria coniothyrium, Penicillium expansum, Trichoderma harzianum, T. pseudokoningii), unter denen sich auch als Saprobier bekannte Arten befanden (P. expansum, T. harzianum), selbst bei Verfügbarkeit organischer Kohlenstoffverbindungen im Substrat, gegenüber Vitis spp. ein fakultativ pathogenes Potenzial besitzen. Diese aus In-vitro-Interaktionsversuchen gewonnenen Erkenntnisse geben Hinweise darauf, welchen Einfluss die Bewirtschaftung, insbesondere die Versorgung der Weinbergsböden mit organischem Kohlenstoff, auf fakultativ pathogene Sekundärparasiten in Form von Bodenpilzen und folglich auf die Entwicklung von Schadbildern an durch die Reblaus prädispositionierten Rebpflanzen in vivo haben kann.
Untersuchungen zur Esca-Erkrankung von Weinpflanzen : Pilz-Pflanze und antagonistische Interaktionen

Ziel dieser Arbeit war es, ein besseres Verständnis für die molekularen und biochemischen Grundlagen der Esca-Krankheit an Reben zu erarbeiten. Hierzu wurden sechs Teilprojekte durchgeführt. Die beiden Pilze Pm. chlamydospora und Pa. aleophilum wurden submers kultiviert um die gewonnenen Extrakte auf phytotoxische Substanzen zu untersuchten. Dabei wurden Metabolite isoliert, die zum ersten mal mit der Esca-Krankheit assoziiert werden konnten, wie Siderophoren und Kaempferol-3-O-Glukosid. Diese können das Verständnis der Pilz-Pflanze-Interaktion um neue Aspekte ergänzen. Die Glykosylierung von Kaempferol durch Pm wurde erstmals nachgewiesen und die hier dargelegten Ergebnisse deuten darauf hin, dass der Pilz in der Lage ist, die von der Pflanze als Abwehrantwort produzierte Substanz, seinerseits zu nutzen. Dies bedeutet, dass Pm einen pflanzlichen Abwehrstoff durch Glykosylierung umwandelt und für die Wirtspflanze toxifiziert. Die durchgeführten Injektionstests in vivo in Weinpflanzen zeigten zudem, dass Esca-ähnliche Symptome durch K3G hervorgerufen werden können. Abseits dieser neuen Erkenntnisse konnten auch literaturbekannte Phytotoxine in den Extrakten identifiziert werden.rnIm zweiten Teilprojekt dieser Arbeit wurden Polyketidsynthase-Gene der beiden Pilze Pm und Pa inaktiviert. Zusätzlich wurde der veränderte Sekundärstoffwechsel bzw. die Reaktion der Insertionsmutanten auf abiotischen Stress und Infektionsfähigkeit von Vitis-Pflanzen untersucht. Damit ist es in dieser Arbeit erstmals gelungen selektiv Gene der Esca-assoziierten Pathogene zu inaktivieren. Es zeigte sich, dass die Mutanten sensitiver als die Wildtyp-Stämme bezüglich Salz-, Trocken- und oxidativen Stress reagieren. Die Infektionsfähigkeit bleibt jedoch unverändert.rnDie Analyse der Infektionsmechanismen sollte auch bei der Transformation der Pilze mit Fluoreszenzgenen im Fokus stehen. Im Rahmen dieser Arbeit gelang es GFP-Mutanten der Pilze Pm, Pa und El zu generieren um das Wachstum nach Infektion von Weinpflanzen zu erfassen. Mit Hilfe der Fluoreszenzmutanten war es möglich ein neues Verfahren zur Suche nach potentiellen Antagonisten zu erstellen. Mit dem Vitis Shoot Assay konnten die Esca-assoziierten Pilze und Fungizide, endophytische Pilze, Trichoderma spp. und Bakterien-Stämme in einem Testverfahren auf Schnittwunden untersucht werden. Dabei konnten erste biologische Wundschutzmittel auf Basis ganzer Organismen identifiziert werden. Es zeigte sich, dass der durchgeführte Antagonistentest und das hier beschriebene innovative Vitis Shoot Assay nicht immer äquivalente Ergebnisse generierten und dass sich die beiden Verfahren ergänzen.rnDes Weiteren wurde eine Interaktionsstudie der vier Esca-assoziierten Pilze unter Stressbedingungen und verschiedenen Temperaturen durchgeführt um den Einfluss äußere biotischer und abiotischer Faktoren auf das Zusammenwirken dieser Erreger zu untersuchen. Die Pilze reagieren sehr unterschiedlich in den durchgeführten Stresstests und die Dominanz einzelner Erreger hängt stark von der Temperatur und den osmotischen Verhältnissen ab. Es konnte gezeigt werden, dass veränderte klimatische Bedingungen zur veränderten Zusammensetzung des Erregerspektrums führen kann.rnDie in dieser Arbeit generierten Ergebnisse geben neue Einblicke in die Esca Erkrankung von Weinreben und ermöglichen die weitere Forschung einiger weiterer Aspekte.

Secondary metabolites are produced by numerous organisms and can either be benign to humans or harmful. Genes involved in the synthesis and transport of these secondary metabolites are frequently found in gene clusters, which are often located in subtelomeric regions of the chromosome. These clusters are often coordinately regulated, being almost exclusively dependent on transcription factors that are located within the clusters themselves. Secondary metabolites are also regulated by a variety of factors, including nutritional factors, environmental factors and developmental processes. Gliotoxin, which is produced by a variety of Aspergillus species, Trichoderma species, and Penicillium species, exhibits immunosuppressive properties and has therefore been the subject of research for many laboratories. There have been a few proteins shown to regulate the gliotoxin cluster, most notably GliZ, a Zn2Cys6 binuclear finger transcription factor that lies within the cluster, and LaeA, a putative methyltransferase that globally regulates secondary metabolism clusters within numerous fungal organisms, although no study has demonstrated the direct binding of any protein to a promoter region in the gliotoxin cluster. I report here two novel proteins, GipA, a C2H2 transcription factor and GipB, a hybrid sensor kinase, which are involved in regulating the gliotoxin biosynthetic cluster. GipA plays an important role in gliotoxin production, as high-copy expression of gipA induces gliotoxin biosynthesis and loss of gipA reduces gliotoxin biosynthesis by 50%. GipB is also involved in regulating gliotoxin production, as high-copy expression of gipB induces gliotoxin biosynthesis, but only during certain stages of asexual development. Furthermore, loss of gipB reduces gliotoxin biosynthesis by 10%. Based on data obtained from this project, I propose a model for the regulation of gliA, the efflux pump of the gliotoxin cluster, which involves GipB signaling through both GliZ and GipA. I propose that GliZ and GipA are interdependent, as mutation of the GipA DNA binding site in the gliA promoter negatively affects both GliZ-mediated and GipA-mediated induction of gliA. This is further supported by the fact that GliZ cannot fully induce gliA in the absence of GipA and vice versa. This is the first time that anyone has shown evidence of a protein directly binding to the gliotoxin cluster. Even though biosynthetic clusters are often coordinately regulated, my model raises the possibility that gliA is independently regulated, as the layout of the binding site in the gliA promoter is not present upstream of any other genes in the gliotoxin cluster, except for gliZ.
