Albumin concentrations in female Bronze turkeys were compared using agarose gel electrophoresis (AGE) and the bromocresol green (BCG) dye-binding method. The correlation coefficient observed for albumin in the BCG and AGE methods was low, and statistical differences were observed at paired t test (p < 0. 0001). Compared with electrophoresis, the BCG-binding method yielded significantly lower albumin values for female turkeys during laying season. © 2011 Springer-Verlag London Limited.

This case report describes a case of a mammary invasive pleomorphic lobular carcinoma in a female dog. Cytology revealed small cohesive clusters of cells and many scattered single large round cells with eccentric nuclei. Histologically, the neoplasm presented plasmacytoid cells with multiple, often bizarre nuclei and an invasive pattern with cells occasionally arranged in Indian files. Vascular invasion was consistently present. Tumour cells also revealed loss of E-cadherin immunoexpression and positive immunoreactivity for HER2/c-erbB2, oestrogen receptor alpha, cytokeratin 19, and Ki67 (52 %), and the tumour was characterised as a mammary invasive pleomorphic lobular carcinoma. This is an interesting case since this is a rare condition that is not widely recognised in dogs. © 2012 Springer-Verlag London Limited.

Several methods have been employed to quantify urinary glycosaminoglycans (GAGs), such as chromatography associated with electrophoresis and colorimetric methods, cheaper and faster ones, which employ mainly azure A and B, alcian blue, and dimethylmethylene blue (DMB). The purpose of this study was to standardize a reproducible and cheap method to measure total urinary GAGs in feline urine. Two colorimetric methods based on DMB were tested with chondroitin sulfate C as standard. Urine samples were obtained from 12 healthy cats and some modifications were made for the chosen method to be adequate. The modified technique using DMB acetate buffer carried out in this study can be used to measure feline urinary GAGs. © 2012 Springer-Verlag London.

Salmonella infection is responsible for major economic losses in poultry industry. Consequently, the development of new methods for fighting such disease is desirable, such as the use of acid-lactic bacteria. However, reference values of chicks in such conditions are dissimilar to those of other species. Leucometry reference values for chicks have not been reported. The aim of this article was to evaluate and determine the leucometric values of chicks inoculated with Salmonella Typhimurium or treated with Lactobacilli probiotics. In this study, 144 1-day-old birds were divided in three groups of 48 animals each (non-treated group, Salmonella Typhimurium (ST)-inoculated group, and Lactobacilli inoculated group). A total of four blood collections were made with the first one performed 3 h after inoculation with ST or treatment with Lactobacilli. Subsequent samples were obtained every 48 h for 7 days. Leucometric evaluation was performed immediately after each collection. All birds presented an initial decrease pattern in general leukocyte values, and the chicks inoculated with ST revealed lymphomonoheteropaenic leukopaenia, eosinophilia and basophilia in conjunction with convalescence after 96 h of inoculation. The animals inoculated with Lactobacilli revealed leucocytosis with monocytosis, lymphocytosis and marked eosinopaenia. We conclude that there is no efficient bone marrow response in 1-day-old chicks challenged with Salmonella Typhimurium; additionally, an immunostimulatory effect in 1-day-old chicks treated with Lactobacilli-modulated probiotics could be stated. © 2011 Springer-Verlag London Limited.

The purpose of this study was to compare albumin concentrations in ring-necked pheasants (Phasianus colchicus) using two different dye-binding methods: the bromocresol green (BCG) and bromocresol purple (BCP). High positive correlation was observed for albumin in BCG and BCP methods. Inappropriate reference limits increase the risks of unnecessary investigations and diagnostic failures. Species, breed, environment, handling, and physiologic stage can influence serum biochemical variables. This study aimed to establish serum biochemical reference intervals of Pêga donkeys and the influence of age and sex on these variables. Samples were taken from 110 donkeys (79 females and 31 males; 8 under 1 year, 33 between 1 and 3 years, and 69 over 3 years old). There were no differences for creatine kinase (CK), albumin, urea, and magnesium among age groups. Animals under 1 year old had the lowest aspartate aminotransferase (AST), creatinine, triglycerides, total calcium, and chloride means; lower indirect bilirubin, gamma-glutamyl transferase (GGT), alanine aminotransferase (ALT), and sodium levels than over 3-year-old group and the highest phosphorus, glucose, cholesterol, ionized calcium, and alkaline phosphatase (AP) values. Total protein increased with age. The group from 1 to 3 years had higher potassium than up to 1-year-old group. Animals over 3 years old had the highest means of direct and total bilirubin. ALT, CK, GGT, direct and total bilirubin, urea, triglycerides, sodium, potassium, and chloride levels did not differ between genders. Females had higher AST, total protein, total calcium, and indirect bilirubin levels. Males showed greater AP, albumin, phosphorus, glucose, creatinine, cholesterol, magnesium, and ionized calcium values. Discrepancies among present results and previous studies show influencing factors on biochemical profile of donkeys and reinforce the importance of establishment of specific reference intervals. This study is useful in clinical routine and as basis to other scientific researches with Pêga donkeys. © 2013 Springer-Verlag London.

Mortality factors that act sequentially through the demographic transitions from seed to sapling may have critical effects on recruitment success. Understanding how habitat heterogeneity influences the causal factors that limit propagule establishment in natural populations is central to assess these demographic bottlenecks and their consequences. Bamboos often influence forest structure and dynamics and are a major factor in generating landscape complexity and habitat heterogeneity in tropical forests. To understand how patch heterogeneity influences plant recruitment we studied critical establishment stages during early recruitment of Euterpe edulis, Sloanea guianensis and Virola bicuhyba in bamboo and non-bamboo stands in the Brazilian Atlantic forest. We combined observational studies of seed rain and seedling emergence with seed addition experiments to evaluate the transition probabilities among regeneration stages within bamboo and non-bamboo stands. The relative importance of each mortality factor was evaluated by determining how the loss of propagules affected stage-specific recruitment success. Our results revealed that the seed addition treatment significantly increased seedling survivorship for all three species. E. edulis seedling survival probability increased in the addition treatment in the two stand types. However, for S. guianensis and V. bicuhyba this effect depended strongly on artificially protecting the seeds, as both species experienced increased seed and seedling losses due to post-dispersal seed predators and herbivores. Propagules of all three species had a greater probability of reaching subsequent recruitment stages when protected. The recruitment of large-seeded V. bicuhyba and E. edulis appears to be much more limited by post-dispersal factors than by dispersal limitation, whereas the small-seeded S. guianensis showed an even stronger effect of post-dispersal factors causing recruitment collapse in some situations. We demonstrated that E. edulis, S. guianensis and V. bicuhyba are especially susceptible to predation during early compared with later establishment stages and this early stage mortality can be more crucial than stand differences as determinants of successful regeneration. Among-species differences in the relative importance of dispersal vs. establishment limitation are mediated by variability in species responses to patch heterogeneity. Thus, bamboo effects on the early recruitment of non-bamboo species are patchy and species-specific, with successional bamboo patches exerting a far-reaching influence on the heterogeneity of plant species composition and abundance. © 2012 Perspectives in Plant Ecology, Evolution and Systematics.

The excision repair cross-complementation 1 (ERCC1) enzyme plays an essential role in the nucleotide excision repair pathway and is associated with resistance to platinum-based chemotherapy in different types of cancer. The aim of the present study was to evaluate the clinicopathological significance of ERCC1 expression in breast cancer patients. We analyzed the immunohistochemical expression of ERCC1 in a tissue microarray from 135 primary breast carcinomas and correlated the immunohistochemical findings with clinicopathological factors and outcome data. ERCC1 expression analysis was available for 109 cases. In this group, 58 (53.2%) were positive for ERCC1. ERCC1-positive expression was correlated with smaller tumor size (P=0.007) and with positivity for estrogen receptor (P=0.040), but no correlation was found with other clinicopathological features. Although not statistically significant, triple negative breast cancers were more frequently negative for ERCC1 (61.5% of the cases) compared to the non-triple negative breast cancer cases (41.5%). In conclusion, ERCC1 expression correlated significantly with favorable prognostic factors, such as smaller tumor size and ER-positivity, suggesting a possible role for ERCC1 as a predictive and/or prognostic marker in breast cancer. © 2013 Elsevier GmbH.

The expression of prognostic markers in cancer has become important in diagnostic routine and research. A high mitotic rate compromises the individual cell access to oxygen and nutrients, due to reduced blood supply. Cells undertake adaptive measures such as vascular endothelial growth factor (VEGF), expressed under the control of hypoxia-inducible factor-1α (HIF-1α). CD34 is an endothelial marker which can show the presence and distribution of blood vessels. This study evaluated the presence and relative expression of VEGF, HIF-1α and CD34 using immunohistochemistry of 60 breast tumors and double staining, correlating the findings with clinical and pathological variables. High VEGF expression was correlated with low cell proliferation, lymph node-negative, smaller tumor size and patients not receiving hormone therapy. High HIF-1α expression predominated in younger (<50-year) patients, subjected to neo-adjuvant therapy and in p53-negative tumors. Absence of metastasis, radiotherapy or hormone treatment, and estrogen receptor (ER)-positive tumors showed high CD34 immunoreactivity. We suggest that the angiogenic factors VEGF, HIF-1α and CD34 are important in breast cancer progression and their abundance in breast tumors has prognostic and predictive value. The gerbil is a rodent considered a good model for studies of prostatic morphophysiology under different experimental conditions. Studies involving castration and steroidal blockers of aged gerbils showed that the glandular epithelium persists after long-term therapy, preventing the organ atrophy. Thus, the objective of this study was to evaluate the phenotypic characteristics and behavior of prostatic epithelial cells that remained after different periods of hormone ablation in aged gerbils. The identification of elements that influenced the survival of this cell type was performed by morphometric, nuclear phenotypes, ultrastructural and immune histochemical analysis. The most significant responses to treatment, by analyzing morphometric features, were observed during the first three time points (day 1, day 3, and day 7), after which there appeared to be an adjustment of the gland to the hormone ablation. All treatments led to changes in the state of chromatin condensation, DNA methylation pattern and phenotypic changes indicated cell senescence. Additionally, an increase in the basal cells seemed to guarantee self-renewal properties to the epithelium. These data indicate that changes occur at many levels, including gene expression and nuclear architecture in the epithelial cells, when aging and steroidal blockade are associated. These aspects are important when considering castration-resistant prostate cancer, a malignant tumor posing difficult therapeutic intervention. © 2013 Elsevier GmbH. All rights reserved.

This study provides histochemical data of the hepatopancreatic cells of adult female pink-shrimp (Farfantepenaeus brasiliensis) at two different developmental stages (those with developed gonads and those with exhausted gonads). The F. brasiliensis females were collected in seawater off the Guarapari coast, Espirito Santo, Brazil. Five cell types were identified in this digestive gland: B (vesicular), E (embryonic), F (fibrillar), M (basal) and R (resorptive). The digestive gland was stained with the following techniques: PAS/Alcian blue (for polysaccharides), bromophenol blue (for protein), von Kossa (for bound calcium) and Baker (for lipids). Acid glycoconjugates were found inside vacuoles in the R cells, while neutral polysaccharides were present in the B cells and near to the microvilli. In females with exhausted gonads polysaccharides were also seen in the intertubular spaces and inside the lumina of the tubules. The F and M cells were the most marked by the presence of large amounts of proteins observed in R cells and also inside the vacuoles of B cells. The bound calcium was mainly found in the F and M cells. The F cells showed strong positive staining for lipid while the R cell only stained weakly. The E cells did not react to any of the applied staining techniques. The similarities in the histochemical composition of these hepatopancreatic cells in females with developed gonads, compared to exhausted ones, is justified by the fact that transfer of these elements to the oocytes occurs, in significant quantity, only during the initial stages of gonadal development in F. brasiliensis. Also, they may be more related to the molt stage, as in the case of calcium salts. © 2013 Elsevier GmbH. All rights reserved.

A long-standing interest in cactus taxonomy has existed since the Linnaean generation, but an appreciation of the reproductive biology of cacti started early in the 1900s. Numerous studies indicate that plant reproductive traits provide valuable systematic information. Despite the extensive reproductive versatility and specializations in breeding systems coupled with the striking floral shapes, the reproductive biology of the Cactaceae has been investigated in approximately 10% of its species. Hence, the systematic value of architectural design and organization of internal floral parts has remained virtually unexplored in the family. This study represents the most extensive survey of flower and nectary morphology in the Cactaceae focusing on tribes Hylocereeae and Rhipsalideae (subfamily Cactoideae). Our objectives were (1) to conduct comparative morphological analyses of flowers and floral nectaries and (2) to compare nectar solute concentration in these two tribes consisting of holo- and semi-epiphytic species. Flower morphology, nectary types, and sugar concentration of nectar have strong taxonomic implications at the tribal, generic and specific levels. Foremost, three types of nectaries were found, namely chamber nectary (with the open and diffuse subtypes), furrow nectary (including the holder nectary subtype), and annular nectary. All Hylocereeae species possess chamber nectaries, in which the nectarial tissue has both trichomes and stomata. The Rhipsalideae are distinguished by two kinds of floral nectaries: furrow and annular, both nectary types with stomata only. The annular nectary type characterizes the genus Rhipsalis. Nectar concentration is another significant taxonomic indicator separating the Hylocereeae and Rhipsalideae and establishing trends linked to nectar sugar concentration and amount of nectar production in relation to flower size. There is an inverse relationship between flower size and amount of nectar production in the smaller Rhipsalideae flowers, in which nectar concentration is more than two-fold higher despite the smaller volume of nectar produced when compared to the large Hylocereeae flowers. Variability of nectary morphology and nectar concentration was also evaluated as potential synapomorphic characters in recent phylogenies of these tribes. In conclusion, our data provide strong evidence of the systematic value of floral nectaries and nectar sugar concentration in the Cactaceae, particularly at different taxonomic levels in the Hylocereeae and Rhipsalideae. © 2013 Perspectives in Plant Ecology, Evolution and Systematics.
