OBJECTIVE: This study aims to evaluate the citotoxic activity of two commonly used anti-depressants: paroxetine and bupropion. We also evaluated the in vitro natural killer activity (NKA) after incubating the blood samples with the antidepressants. METHODS: Peripheral blood samples from 15 healthy volunteers were collected and the mononuclear cells (PBMCs) were isolated and incubated for 24h with (or without = control cells) paroxetine and bupropion, in concentrations of 30, 100 and 1000 ng/ml. After the incubation period in both groups, the amount of dead cells was calculated using trypam blue technique. NKA was evaluated using the classic51Cr release assay. CONCLUSIONS: PBMCs dead cells occurred in both groups and in proportion to all pharmacological concentrations. Nevertheless, the NKA was not affected, even with the reduction in the number of effective cells.

OBJETIVO: Avaliar tecnicamente a monitorização ambulatorial de pressão arterial (MAPA) em adolescentes normais. MÉTODOS: Quarenta e cinco adolescentes eutróficos, entre 10-18 anos de idade, sendo 27 do sexo feminino. RESULTADOS: Verificaram-se, em média, 90% de medidas bem sucedidas: incômodo relacionado ao funcionamento do monitor em 30% dos casos; valores médios de descenso sistólico, diastólico e de diminuição de freqüência cardíaca, no sono noturno, respectivamente iguais a 13%, 23% e 24%; carga pressórica na vigília, no sexo masculino, de 25,4±27,7% e 11,8±14,6% e, no feminino, de 17,5±18,7% e 11,8±11,4% para pressão arterial sistólica (PAS) e diastólica (PAD), respectivamente; carga pressórica no sono, no sexo masculino, de 15,4±22,9% e 2,8±4,9% e, no feminino, de 10,5±18,2% e 1,8±2,7% para PAS e PAD; medidas diastólicas mais elevadas nas duas primeiras horas de monitorização; diferenças entre sono noturno e vespertino quanto aos parâmetros cardiovasculares estudados. CONCLUSÃO: A MAPA mostrou-se bem tolerada pela população adolescente. Os registros obtidos apresentaram-se tecnicamente adequados para análise.
Curva dose-resposta do exercício em hipertensos: análise do número de sessões para efeito hipotensor

FUNDAMENTO: O efeito do exercício na pressão arterial já é conhecido, entretanto a curva dose-resposta do efeito hipotensor do exercício em hipertensos ainda não está clara. OBJETIVO: Avaliar a curva dose-resposta do número de sessões necessárias para causar efeito hipotensor em indivíduos hipertensos. MÉTODOS: Participaram deste estudo 88 indivíduos, com 58 ± 11 anos, divididos em grupo experimental (GE) - composto de 48 integrantes de um programa de exercício físico (PEF) de 3 meses, 3 vezes por semana, com 40' de exercício aeróbio a 70% do VO2máx e exercícios musculares a 40% da CVM - e grupo-controle (GC) - 40 indivíduos que não realizaram PEF. As pressões arteriais sistólica (PAS) e diastólica (PAD) foram mensuradas antes de cada uma das 36 sessões no GE e avaliadas por MAPA no GC. Observaram-se as diferenças na PA, o índice de variação (D%) e o efeito hipotensor máximo (EHM%) entre as sessões. Os dados foram expressos por M ± DP, e usou-se teste t e correlação, considerando p < 0,05 significativo. RESULTADOS: No GC não houve diferença nos valores pressóricos. No GE, após o PEF, ocorreu uma queda importante de 15 mmHg na PAS e de 7 mmHg na PAD, e uma grande parte desse efeito ocorreu já na primeira sessão, e a maior parte até a quinta. Houve uma forte correlação inversa (R: -0,66) com o número de sessões. CONCLUSÃO: Na primeira sessão, já ocorreu efeito hipotensor importante, e observou-se que a curva dose-resposta pode ser abrupta e decrescente em vez de achatada.

FUNDAMENTO: A sedação para a realização de cateterismo cardíaco tem sido alvo de preocupação. Benzodiazepínicos, agonistas alfa-2 adrenérgicos e opioides são utilizados para esse fim, entretanto, cada um destes medicamentos possui vantagens e desvantagens. OBJETIVO: Avaliar a eficácia do sufentanil e da clonidina como sedativos em pacientes submetidos a cateterismo cardíaco, observando o impacto dos mesmos sobre os parâmetros hemodinâmicos e respiratórios, a presença de efeitos colaterais, além da satisfação do paciente e do hemodinamicista com o exame. MÉTODOS: Trata-se de um ensaio clínico prospectivo, duplo-cego, randomizado e controlado, que envolveu 60 pacientes que receberam 0,1 µg/kg de sufentanil ou 0,5 µg/kg de clonidina antes da realização do cateterismo cardíaco. O escore de sedação segundo a escala de Ramsay, a necessidade de utilização de midazolam, os efeitos colaterais, os parâmetros hemodinâmicos e respiratórios foram registrados, sendo os dados analisados em 06 diferentes momentos. RESULTADOS: O comportamento da pressão arterial, da frequência cardíaca e da frequência respiratória foi semelhante nos dois grupos, entretanto, no momento 2, os pacientes do grupo sufentanil (Grupo S) apresentaram menor escore de sedação segundo a escala de Ramsay, e a saturação periférica da oxihemoglobina foi menor que o grupo clonidina (Grupo C) no momento 6. Os pacientes do Grupo S apresentaram maior incidência de náusea e vômito pós-operatório que os pacientes do Grupo C. A satisfação dos pacientes foi maior no grupo clonidina. Os hemodinamicistas mostraram-se satisfeitos nos dois grupos. CONCLUSÃO: O sufentanil e a clonidina foram efetivos como sedativos em pacientes submetidos a cateterismo cardíaco.

Der vorliegende Praxisleitfaden soll die Anlage von Säumen und Feldrainen in produktiven Agrarlandschaften und im urbanen Bereich erleichtern. Es werden wertvolle Hinweise zur Planung von Ansaatmischungen mit Wildpflanzen, zur Bodenvorbereitung, zur Umsetzung der Maßnahme (Ansaatzeitpunkt, Ansaatmethode, Ansaatstärke) und zur Entwicklungs- und Folgepflege gegeben. Darüber hinaus werden Fördermöglichkeiten und vergaberechtliche Aspekte erörtert. Mit 10 Praxisbeispielen werden bereits erfolgreich umgesetzte Maßnahmen aus Niedersachsen und Sachsen-Anhalt vorgestellt. Das Kapitel "Aus Fehlern lernen" zeigt kurz und prägnant, wie Misserfolge vermieden werden können. Insgesamt dokumentiert der Leitfaden, wie monotone Grasstreifen nach einer intensiven Bodenstörung, einer Ansaat mit zertifizierten, gebietseigenen Wildpflanzen und einer angepassten Entwicklungs- und Folgepflege erfolgreich in ausdauernde, blütenreiche Bestände umgewandelt werden können, die vielfältige Ökosystemdienstleistungen erfüllen und die Biodiversität erhöhen. Der Praxisleitfaden entstand im Rahmen des Projektes "ProSaum", das vom Bundesministerium für Bildung und Forschung gefördert wurde.

Knowledge of the spatial distribution of hydraulic conductivity (K) within an aquifer is critical for reliable predictions of solute transport and the development of effective groundwater management and/or remediation strategies. While core analyses and hydraulic logging can provide highly detailed information, such information is inherently localized around boreholes that tend to be sparsely distributed throughout the aquifer volume. Conversely, larger-scale hydraulic experiments like pumping and tracer tests provide relatively low-resolution estimates of K in the investigated subsurface region. As a result, traditional hydrogeological measurement techniques contain a gap in terms of spatial resolution and coverage, and they are often alone inadequate for characterizing heterogeneous aquifers. Geophysical methods have the potential to bridge this gap. The recent increased interest in the application of geophysical methods to hydrogeological problems is clearly evidenced by the formation and rapid growth of the domain of hydrogeophysics over the past decade (e.g., Rubin and Hubbard, 2005).

During blood banking, erythrocytes undergo storage lesions, altering or degrading their metabolism, rheological properties, and protein content. Carbonylation is a hallmark of protein oxidative lesions, thus of red blood cell oxidative stress. In order to improve global erythrocyte protein carbonylation assessment, subcellular fractionation has been established, allowing us to work on four different protein populations, namely soluble hemoglobin, hemoglobin-depleted soluble fraction, integral membrane and cytoskeleton membrane protein fractions. Carbonylation in erythrocyte-derived microparticles has also been investigated. Carbonylated proteins were derivatized with 2,4-dinitrophenylhydrazine (2,4-DNPH) and quantified by western blot analyses. In particular, carbonylation in the cytoskeletal membrane fraction increased remarkably between day 29 and day 43 (P<0.01). Moreover, protein carbonylation within microparticles released during storage showed a two-fold increase along the storage period (P<0.01). As a result, carbonylation of cytoplasmic and membrane protein fractions differs along storage, and the present study allows explaining two distinct steps in global erythrocyte protein carbonylation evolution during blood banking. PIKfyve is a kinase encoded by pip5k3 involved in phosphatidylinositols (PdtIns) pathways. These lipids building cell membranes have structural functions and are involved in complex intracellular regulations. Mutations in human PIP5K3 are associated with François-Neetens mouchetée fleck corneal dystrophy [Li, S., Tiab, L., Jiao, X., Munier, F.L., Zografos, L., Frueh, B.E., Sergeev, Y., Smith, J., Rubin, B., Meallet, M.A., Forster, R.K., Hejtmancik, J.F., Schorderet, D.F., 2005. Mutations in PIP5K3 are associated with François-Neetens mouchetee fleck corneal dystrophy. Am. J. Hum. Genet. 77, 54-63]. We cloned the zebrafish pip5k3 and report its molecular characterization and expression pattern in adult fish as well as during development. The zebrafish PIKfyve was 70% similar to the human homologue. The gene encompassed 42 exons and presented four alternatively spliced variants. It had a widespread expression in the adult organs and was localized in specific cell types in the eye as the cornea, lens, ganglion cell layer, inner nuclear layer and outer limiting membrane. Pip5k3 transcripts were detected in early cleavage stage embryos. Then it was uniformly expressed at 10 somites, 18 somites and 24 hpf. Its expression was then restricted to the head region at 48 hpf, 72 hpf and 5 dpf and partial expression was found in somites at 72 hpf and 5 dpf. In situ on eye sections at 3 dpf showed a staining mainly in lens, outer limiting membrane, inner nuclear layer and ganglion cell layer. A similar expression pattern was found in the eye at 5 dpf. A temporal regulation of the spliced variants was observed at 1, 3 and 5 dpf and they were also found in the adult eye.

The optimal size-to-age at maturity depends on growth and mortality rates, which vary with environment. Therefore, organisms in spatially or temporaly changing environments should develop adaptative phenotypic plasticity for this trait. Experimental work by Alm (1959) on several fish species shows a dome-shape norm of reaction for size-to-age at maturity: size at maturity is smaller in both fast-growing and slow-growing fishes, than it is in fish with a medium growth rate. Using computer simulations, we show that such a dome-shaped norm of reaction is optimal when assuming a finite life span and a negative relationship between production and survival rates. This latter assumption is supported by empirical data, as well as by physiological and emographic arguments.

PURPOSE OF REVIEW: The mechanisms involved in the formation of red blood cell (RBC) microparticles in vivo as well as during erythrocyte storage are reviewed, and the potential role of microparticles in transfusion medicine is described. RECENT FINDINGS: Microparticles release is an integral part of the erythrocyte ageing process, preventing early removal of RBCs. Proteomics analyses have outlined the key role of band 3-ankyrin anchoring complex and the occurrence of selective RBC membrane remodelling mechanisms in microparticles formation. The presence of several RBC antigens, expressed on microparticles, has been demonstrated. The potential deleterious effects of RBC microparticles in transfused recipients, including hypercoagulability, microcirculation impairment and immunosuppression, are discussed. SUMMARY: Formation and role of RBC microparticles are far from being completely understood. Combining various approaches to elucidate these mechanisms could improve blood product quality and transfusion safety. Implementation of RBC microparticles as biomarkers in the laboratory routine needs to overcome technical barriers involved in their analysis.

As part of CHD NSF implementation, a pilot project is being undertaken under the auspices of the National Screening Committee to test the practical implications and outcomes of implementing a systematic programme of cardiovascular risk reduction in primary care, initially identifying those at high risk due to pre-existing cardiovascular disease or diabetes. To assist in assessing the magnitude of the challenge faced by the pilot programmes, the UK National Screening Committee (NSC) commissioned a review of current practice based on recent cardiovascular and diabetes audits in the UK. This report details the findings of the review, providing 6 key recommendations for future cardiovascular audits.
