SHP1 is a cytosolic protein tyrosine phosphatase that contains two SH2 domains. It is highly expressed in hematopoietic cells and expressed in normal epithelium at lower levels. While SHP1 in hematopoietic cells is thought to be a negative regulator of cellular signaling by associating with and dephosphorylating various receptors and their downstream effectors after they become activated, its precise function in epithelium remains to be understood. The potential involvement of SHP1 in human tumorigenesis has been hypothesized from the findings that SHP1 can interact with, dephosphorylate, and regulate the activity of several protein tyrosine kinases (PTKs) implicated in human cancer. These PTKs include epidermal growth factor receptor (EGFR) and Src. Such speculation is also supported by the report that SHP1 is overexpressed in human ovarian cancers. ^ Here we report, for the first time, that the levels of SHP1 expression and activity are altered in human breast cancer cells in comparison with normal breast epithelium. In particular, SHP1 expression is nearly lost in the breast cancer cell lines MDA-MB231 and MDA-MB435. After the re-introduction of SHP1 both in wild type (wt) and enzymatically inactive (dn) forms, into the MDA-MB231 cells, we observed no changes in cellular proliferation. However, the overexpression of wt SHP1 led to increased anchorage-independent growth in the MDA-MB231 cells. SHP1 phosphatase activity is essential for such an increase since the overexpression of dn SHP1 had no effect. Enhanced turnorigenicity in nude mice was also observed in the MDA-MB231 cells overexpressing wt SHP1, but not dn SHP1, suggesting the crucial function of SHP1 enzymatic activity in this process. Our observations in this study indicate that SHP1 promotes tumorigenesis by a mechanism or mechanisms apart from enchancing angiogenesis. In addition, we have found no evidence that the overexpression of SHP1 could affect metastatic potential in the MDA-MB231 cells. ^ In the MDA-MB231 cells stably transfected with either wt or dn SHP1 the peak level of EGFR tyrosine phosphorylation induced by EGF, as well as the sensitivity to EGF stimulation, was not altered. However, the overexpression of wt SHP1 led to a slight increase in the kinetics of EGFR dephosphorylation, whereas the overexpression of dn SHP1 led to slightly delayed kinetics of EGFR dephosphorylation. The overexpression of either the wt or dn SHP1 did not lead to any significant increase in Src kinase activity. ^ In NIH3T3 cells, the transient overexpression of SHP1 led to no significant changes in MAP kinase (ERK2) activation by EGF or Akt activation by PDGF. In 3T3H4 cells, the transient overexpression of SHP1 led to no significant changes in MAP kinase (ERK2) activation by heregulin. The transient overexpression of wt SHP1 in the MDA-MB231 cells caused an apparent increase, ranging from 10% to 20%, in the G0/G1 population of the cells with a corresponding decrease in the S phase population. ^ In order to understand the mechanisms by which SHP1 exerts its positive effect on the tumorigenic potential of the MDA-MB231 cells, we employed two-dimensional electrophoresis in an attempt to identify cellular protein(s) with significantly altered tyrosine phosphorylation level upon wt SHP1 overexpression. The overexpression of wt SHP1 but not dn SHP1, leads increased tyrosine phosphorylation of a protein with a molecular weight of approximately 40 kDa and a pI between 5.9 to 6.6. ^

Given the importance of the inversion of seamount magnetic anomalies, particularly to the motion of the Pacific plate, it is important to gain a better understanding of the nature of the magnetic source of these features. Although different in detail, Ninetyeast Ridge is composed of submarine and subaerial igneous rocks that are similar to those found at many seamounts, making it a suitable proxy. We report here on the magnetic petrology of a collection of samples from Ninetyeast Ridge in the Indian Ocean. Our purpose is to determine the relationship between primary petrology, subsequent alteration, and magnetic properties of the recovered rocks. Such information will eventually lead to a more complete understanding of the magnetization of seamounts and presumably improvements in the accuracy of anomaly inversions. Three basement sites were drilled on Ninetyeast Ridge, with recovery of subaerial basalt flows at the first two (Sites 756 and 757) and submarine massive and pillow flows at the final one (Site 758). The three sites were distinctly different. Site 756 was dominated by ilmenite. What titanomagnetite was present had undergone deuteric alteration and secondary hematite was present in many samples. The magnetization was moderate and stable although it yielded a paleolatitude somewhat lower than expected. Site 757 was highly oxidized, presumably while above sea level. It was dominated by primary titanomagnetite, which was deuterically altered. Secondary hematite was common. Magnetization was relatively weak but quite stable. The paleolatitude for all but the lowermost flows was approximately 40° lower than expected. Site 758 was also dominated by primary titanomagnetite. There was relatively little oxidation with most primary titanomagnetite showing no evidence of high-temperature alteration. No secondary hematite was in evidence. This site had the highest magnetization of the three (although somewhat low relative to other seamounts) but was relatively unstable with significant viscous remanence in many samples. Paleolatitude was close to the expected value. It is not possible, at present, to confidently associate these rocks with specific locations in a seamount structure. A possible and highly speculative model would place rocks similar to Site 757 near the top of the edifice, Site 756 lower down but still erupted above sea level, and Site 758 underlying these units, erupted while the seamount was still below sea level.

The effects of desiccation on photochemical processes and nitrogenase activity were evaluated in Nostoc commune s.l. colonies in situ from a wet thufur meadow at Petuniabukta, Billefjorden, Central Svalbard, during the 2009 arctic summer. The colonies were collected in the fully hydrated state, and were subjected to slow desiccation at ambient temperatures (5 - 8°C) and low light (30 - 80 µmol/m**2/s). For each colony the weight, area, photochemical performance, and nitrogenase activity were determined at the beginning, as well as on every day during the first four days of the experiment; thereafter, on every second day until desiccation was complete. The photochemical performance was evaluated from variable chlorophyll fluorescence parameters (FV/FM, Phi(PSII) , qP, and NPQ), and the nitrogenase activity was estimated by an acetylene-ethylene reduction assay. A significant decrease in the photochemically active area was recorded from the third day, when the colony had lost approximately 40% of its original weight indicating some changes in the extracellular matrix, and stopped on the 14th to 18th day. No effects of the desiccation on the main photochemical parameters (FV/FM, Phi(PSII), qP) were observed up to the sixth to eighth days of desiccation. Slightly lower values of FV/FM and Phi(PSII) recorded in fully-hydrated colonies could be caused by impaired diffusion of CO2 into cells. The steep reduction of photochemical activity occurred between the eighth and tenth day of the experiment, when the colony had lost approximately 80% of its fully-hydrated weight. The nitrogenase activity was highest on the first day, probably due to improved diffusion of N2 into cells, then declined, but was detectable until the sixth day of the experiment. Since Nostoc commune s.l. colonies were capable of photosynthesis and nitrogen fixation to the level of ca. 60% of its fully-hydrated weight, even partly-hydrated colonies contribute substantially to carbon and nitrogen cycling in the High Arctic wet meadow tundra ecosystem.

The relatively fresh basement basaltic rocks cored at Sites 794 and 797 during ODP Legs 127 and 128 show compositional variations suggesting the following: (1) the aphyric rocks might be differentiated from compositional equivalents of the aphyric sample with the lowest FeO*/MgO (Sample 127-797C-12R-4, 35-37 cm); and (2) the plagioclase-phyric rocks (i.e., another constituent of the basement basaltic rocks from the sites) may be derivatives from the same parents; in this case, however, crystallized plagioclase was not effectively removed. Melting experiments were conducted for Sample 127-797C-12R-4, 35-37 cm, and the differentiation processes for the basement basaltic rocks were assessed. The high-pressure melting-phase relation can not account for the compositional variation of the aphyric rocks, suggesting that the variation was developed at relatively low pressure where olivine and plagioclase fractionation was followed by Ca-rich clinopyroxene fractionation. The density of Sample 127-797C-12R-4,35-37 cm, is comparable to that of plagioclase at some depth, but at still relatively low pressure, making it possible that the liquidus plagioclase was retained in the successive liquids to produce the plagioclase-phyric rocks. According to backtrack calculation assuming the olivine maximum fractionation, Sample 127-797C-12R-4, 35-37 cm, was differentiated from primary picritic high-Al basalt magma. The estimated primary magma composition was experimentally proved to coexist with harzburgite mantle at about 14 kbar, suggesting relatively shallow production (approximately 40-50 km below surface) of the rifting-related primary magma.

Increasing levels of anthropogenic carbon dioxide in the world's oceans are resulting in a decrease in the availability of carbonate ions and a drop in seawater pH. This process, known as ocean acidification, is a potential threat to marine populations via alterations in survival and development. To date, however, little research has examined the effects of ocean acidification on rare or endangered species. To begin to assess the impacts of acidification on endangered northern abalone (Haliotis kamtschatkana) populations, we exposed H. kamtschatkana larvae to various levels of CO2 [400 ppm (ambient), 800 ppm, and 1800 ppm CO2] and measured survival, settlement, shell size, and shell development. Larval survival decreased by ca. 40% in elevated CO2 treatments relative to the 400 ppm control. However, CO2 had no effect on the proportion of surviving larvae that metamorphosed at the end of the experiment. Larval shell abnormalities became apparent in approximately 40% of larvae reared at 800 ppm CO2, and almost all larvae reared at 1800 ppm CO2 either developed an abnormal shell or lacked a shell completely. Of the larvae that did not show shell abnormalities, shell size was reduced by 5% at 800 ppm compared to the control. Overall, larval development of H. kamtschatkana was found to be sensitive to ocean acidification. Near future levels of CO2 will likely pose a significant additional threat to this species, which is already endangered with extinction due in part to limited reproductive output and larval recruitment.

This paper explores intra-state disparity in access to electricity and examines the determinants of electrification at the village level in Bihar, one of the underdeveloped states in India. Our field survey of 80 villages in 5 districts conducted in 2008-09 found that 48 villages (60%) are electrified when using the definition of electrification that a village is electrified if any one household in the village is connected to electricity. The degrees of “electrification” in terms of the proportion of household connection and available hours of electricity remain by and large low, and at the same time differ across districts, villages and seasons. In the processes of electrification, approximately 40% of villages have been electrified in recent years. Based on the basic findings of the survey, this paper examines the electrification processes and how it has changed in recent years. The econometric analyses demonstrate that location is the most important determinant of a village’s electricity connection. Another important finding is that with the rapid progress of rural electrificationunder the recent government programme and the tendency to connect the villages which are easily accessible, the collective bargaining power of the village, which used to significantly affect the process of electrification, has lost influence. This adversely affects remote villages. In order to extend electricity supplies to remote and geographically disadvantaged villages, the government needs to consider seriously other options for sustainable electricity supply, such as decentralized distribution of electricity rather than the conventional connection through the national/local grids.

A study was conducted to determine the relationship between midday measurements of vine water status and daily water use of grapevines measured with a weighing lysimeter. Water applications to the vines were terminated on August 24th for 9 days and again on September 14th for 22 days. Daily water use of the vines in the lysimeter (ETLYS) was approximately 40 L vine−1 (5.3 mm) prior to turning the pump off, and it decreased to 22.3 L vine−1 by September 2nd. Pre-dawn leaf water potential (ΨPD) and midday Ψl on August 24th were −0.075 and −0.76 MPa, respectively, with midday Ψl decreasing to −1.28 MPa on September 2nd. Leaf g s decreased from ~500 to ~200 mmol m−2 s−1 during the two dry-down periods. Midday measurements of g s and Ψl were significantly correlated with one another (r = 0.96) and both with ETLYS/ETo (r = ~0.9). The decreases in Ψl, g s, and ETLYS/ETo in this study were also a linear function of the decrease in volumetric soil water content. The results indicate that even modest water stress can greatly reduce grapevine water use and that short-term measures of vine water status taken at midday are a reflection of daily grapevine water use

To determine the contribution of polar auxin transport (PAT) to auxin accumulation and to adventitious root (AR) formation in the stem base of Petunia hybrida shoot tip cuttings, the level of indole-3-acetic acid (IAA) was monitored in non-treated cuttings and cuttings treated with the auxin transport blocker naphthylphthalamic acid (NPA) and was complemented with precise anatomical studies. The temporal course of carbohydrates, amino acids and activities of controlling enzymes was also investigated. Analysis of initial spatial IAA distribution in the cuttings revealed that approximately 40 and 10% of the total IAA pool was present in the leaves and the stem base as rooting zone, respectively. A negative correlation existed between leaf size and IAA concentration. After excision of cuttings, IAA showed an early increase in the stem base with two peaks at 2 and 24h post excision and, thereafter, a decline to low levels. This was mirrored by the expression pattern of the auxin-responsive GH3 gene. NPA treatment completely suppressed the 24-h peak of IAA and severely inhibited root formation. It also reduced activities of cell wall and vacuolar invertases in the early phase of AR formation and inhibited the rise of activities of glucose-6-phosphate dehydrogenase and phosphofructokinase during later stages. We propose a model in which spontaneous AR formation in Petunia cuttings is dependent on PAT and on the resulting 24-h peak of IAA in the rooting zone, where it induces early cellular events and also stimulates sink establishment. Subsequent root development stimulates glycolysis and the pentosephosphate pathway

El propósito del presente proyecto fue seleccionar la configuración de fabricación de probetas obtenidas mediante el proceso de Modelado por Deposición Fundida (FDM) con el termoplástico acrilonitrilo butadieno estireno (ABS), que optimice las propiedades mecánicas de las probetas y el ahorro de material de apoyo. Se aplicaron técnicas de caracterización física y mecánica y de microscopia electrónica de barrido (SEM). Los resultados indicaron que las probetas verticales presentaron aproximadamente el 6 % de pérdida de material frente cerca de un 40% de las probetas horizontales. La rotura de los cordones se produjo longitudinalmente en el borde de las probetas horizontales mientras que en el borde de las probetas verticales fueron por despegue de los cordones de ABS. La rotura de los cordones en el interior de ambas probetas fue en la dirección de los cordones. ABSTRACT The purpose of this project was to select the manufacture design in test specimens obtained using Fused Deposition Modeling (FDM) with Acrylonitrile Butadiene Styrene (ABS), thus optimizing the mechanical properties of the test specimens and saving the support material. The study was carried out by mean of mechanical and physical characterization techniques as well as Scanning Electron Microscopy (SEM). The results indicated that the horizontal test specimen showed approximately 40% of material loss compared to the vertical test specimen showed a loss 8%. The ABS filament breakage occurred longitudinally on the edge of the horizontal test specimen while the ABS filament breakage was transversely by the separation of the ABS filament on the edge of the vertical tests. The breakage of the filament inside both test specimens was in the direction of the filaments.

El objeto de la investigación es analizar la situación de las plataformas logísticas y caracterizar las mismas, sus áreas funcionales y los parámetros de diseño que se utilizan para su planificación y desarrollo. La investigación se ha realizado sobre las plataformas logísticas existentes en España, si bien se ha contrastado la situación nacional con otras experiencias internacionales. Para ello se ha procedido al estudio del estado del arte a nivel internacional, determinando la terminología, caracterización y clasificación que aporta la comunidad científica al respecto sobre las plataformas logísticas. La investigación se ha centrado en el estudio de un elevado número de plataformas logísticas ubicadas en distintas Comunidades Autónomas, que representan en torno al cuarenta por ciento de las que se encuentran en estos momentos en activo, analizando sus datos básicos, su clasificación en lo que respecta a su grado de centralidad, su intermodalidad, su accesibilidad, y la información urbanística relativa a superficies de áreas funcionales, edificabilidades, usos principales, complementarios y no admitidos, red viaria, zonas verdes, y parámetros máximos y mínimos de ordenación. El análisis, valoración e interpretación de los resultados obtenidos, ha permitido concluir en la heterogeneidad existente sobre el término, proponiéndose por parte del autor un nuevo concepto que englobe y defina de forma genérica pero clara la tipología de plataformas logísticas presentes en el panorama nacional. Del mismo modo, y como consecuencia de la investigación realizada, ha sido posible la caracterización de una “plataforma logística tipo”, estableciendo unos parámetros de diseño estándar para desarrollos posteriores. Finalmente se proponen varias líneas de investigación. En primer lugar, analizar el conjunto del transporte de mercancías, para determinar y valorar su desequilibrio, para de este modo potenciar la intermodalidad e internacionalizar la misma; y, en segundo lugar, establecer una regulación global de las competencias y legislación en materia de planificación de centros logísticos, para racionalizar el desarrollo de estos elementos. The aim of this research is to analyze the situation of logistics platforms and to characterize them, their functional areas and the design parameters that are used in their planning and development. This study was conducted on logistics platforms in Spain, although the situation in Spain has been compared to other international experiences. For this purpose, the international state-of-the-art has been examined, determining the terminology, characterization and classification provided by the scientific community in regard to logistics platforms. The research focuses on the study of a large number of logistics platforms located in different Autonomous Regions, representing approximately 40% of those currently in operation. An analysis has been made of the fundamental data, classification in regard to the degree of centrality, intermodality, accessibility and the urban planning information relating to functional area spaces, as well as buildable potential, main, complementary and prohibited uses, the road network, green zones and maximum and minimum development planning parameters. The analysis, evaluation and interpretation of the results obtained has led to the conclusion of the heterogeneous nature of the term, and the author proposes a new concept that would cover and define, in a generic yet clear way, the typology of logistics platforms present in the national scenario. In a similar manner, and as a result of the research conducted, it has been possible to define a “logistics platform type”, establishing standard design parameters for future developments. Lastly, various lines of research are proposed. In the first place, to analyze overall goods transport to determine and evaluate its imbalances and, in this way, to promote its intermodality and internationalization. In the second place, to establish a global regulation of competencies and legislation with regard to the planning of logistics centers, to rationalize the development of these elements.

The α- and β-karyopherins (Kaps), also called importins, mediate the nuclear transport of proteins. All α-Kaps contain a central domain composed of eight approximately 40 amino acid, tandemly arranged, armadillo-like (Arm) repeats. The number and order of these repeats have not changed since the common origin of fungi, plants, and mammals. Phylogenetic analysis suggests that the various α-Kaps fall into two groups, α1 and α2. Whereas animals encode both types, the yeast genome encodes only an α1-Kap. The β-Kaps are characterized by 14–15 tandemly arranged HEAT motifs. We show that the Arm repeats of α-Kaps and the HEAT motifs of β-Kaps are similar, suggesting that the α-Kaps and β-Kaps (and for that matter, all Arm and HEAT repeat-containing proteins) are members of the same protein superfamily. Phylogenetic analysis indicates that there are at least three major groups of β-Kaps, consistent with their proposed cargo specificities. We present a model in which an α-independent β-Kap progenitor gave rise to the α-dependent β-Kaps and the α-Kaps.

We describe here the cloning and characterization of a cDNA encoding a protein kinase that has high sequence homology to members of the mitogen-activated protein kinase (MAPK) kinase kinase (MAPKKK or MEKK) family; this cDNA is named cATMEKKI (Arabidopsis thaliana MAP kinase or ERK kinase kinase 1). The catalytic domain of the putative ATMEKK1 protein shows approximately 40% identity with the amino acid sequences of the catalytic domains of MAPKKKs (such as Byr2 from Schizosaccharomyces pombe, Ste11 from Saccharomyces cerevisiae, Bck1 from S. cerevisiae, MEKK from mouse, and NPK1 from tobacco). In yeast cells that overexpress ATMEKK1, the protein kinase replaces Ste11 in responding to mating pheromone. In this study, the expression of three protein kinases was examined by Northern blot analyses: ATMEKK1 (structurally related to MAPKKK), ATMPK3 (structurally related to MAPK), and ATPK19 (structurally related to ribosomal S6 kinase). The mRNA levels of these three protein kinases increased markedly and simultaneously in response to touch, cold, and salinity stress. These results suggest that MAP kinase cascades, which are thought to respond to a variety of extracellular signals, are regulated not only at the posttranslational level but also at the transcriptional level in plants and that MAP kinase cascades in plants may function in transducing signals in the presence of environmental stress.

Recent developments in multidimensional heteronuclear NMR spectroscopy and large-scale synthesis of uniformly 13C- and 15N-labeled oligonucleotides have greatly improved the prospects for determination of the solution structure of RNA. However, there are circumstances in which it may be advantageous to label only a segment of the entire RNA chain. For example, in a larger RNA molecule the structural question of interest may reside in a localized domain. Labeling only the corresponding nucleotides simplifies the spectrum and resonance assignments because one can filter proton spectra for coupling to 13C and 15N. Another example is in resolving alternative secondary structure models that are indistinguishable in imino proton connectivities. Here we report a general method for enzymatic synthesis of quantities of segmentally labeled RNA molecules required for NMR spectroscopy. We use the method to distinguish definitively two competing secondary structure models for the 5' half of Caenorhabditis elegans spliced leader RNA by comparison of the two-dimensional [15N] 1H heteronuclear multiple quantum correlation spectrum of the uniformly labeled sample with that of a segmentally labeled sample. The method requires relatively small samples; solutions in the 200-300 microM concentration range, with a total of 30 nmol or approximately 40 micrograms of RNA in approximately 150 microliters, give strong NMR signals in a short accumulation time. The method can be adapted to label an internal segment of a larger RNA chain for study of localized structural problems. This definitive approach provides an alternative to the more common enzymatic and chemical footprinting methods for determination of RNA secondary structure.

A photoactivatable derivative of neurotoxin II from Naja naja oxiana containing a 125I-labeled p-azidosalicylamidoethyl-1,3'-dithiopropyl label at Lys-25 forms a photo-induced cross-link with the delta subunit of the membrane-bound Torpedo californica nicotinic acetylcholine receptor (AChR). The cross-linked radioactive receptor peptide was isolated by reverse-phase HPLC after tryptic digestion of the labeled delta subunit. The sequence of this peptide, delta-(260-277), and the position of the label at Ala-268 were established by matrix-assisted laser-desorption-ionization mass spectrometry based on the molecular mass and on post-source decay fragment analysis. With the known dimensions of the AChR molecule, of the photolabel, and of alpha-neurotoxin, finding the cross-link at delta Ala-268 (located in the upper part of the channel-forming transmembrane helix M2) means that the center of the alpha-neurotoxin binding site is situated at least approximately 40 A from the extracellular surface of the AChR, proximal to the channel axis.

Characteristics of six operating hot spring recreational facilities were reviewed to identify opportunities for integrating a range of sustainable design and operation approaches into a community park. Potential operating cost savings were evaluated for a conceptual landscape project, bathhouse project, and swimming pool project that reduced the use of electricity, natural gas, water, the discharge of water, and solid waste generation. The projects showed a combined cost savings of approximately $40,000 per year by adopting passive solar design, energy efficient lighting, native vegetation, water efficient fixtures, and a natural swimming pool. The greatest potential operating cost reductions were observed for cutbacks in the use of swimming pool water and reductions in natural gas needed for building and swimming pool heating.
