The common bean is affected by several pathogens that can cause severe yield losses. Here we report the introgression of resistance genes to anthracnose, angular leaf spot and rust in the `carioca-type` bean cultivar `Ruda`. Initially, four backcross (BC) lines were obtained using `TO`, `AB 136`, `Ouro Negro` and `AND 277` as donor parents. Molecular fingerprinting was used to select the lines genetically closer to the recurrent parent. The relative genetic distances between `Ruda` and the BC lines varied between 0.0% and 1.99%. The BC lines were intercrossed and molecular markers linked to the resistance genes were used to identify the plants containing the genes of interest. These plants were selfed to obtain the F(2), F(3) and F(4) plants which were selected based on the presence of the molecular markers mentioned and resistance was confirmed in the F(4) generation by inoculation. Four F(4:7) pyramid lines with all the resistance genes showed resistance spectra equivalent to those of their respective donor parents. Yield tests showed that these lines are as productive as the best `carioca-type` cultivars.

Different genes might be involved in Colletotrichum lindemuthianum resistance in leaves and stem of common bean. This work aimed to study the genetic mechanisms of the resistance in the leaf and stem in segregating populations from backcrosses involving resistant cultivar AN 910408 and susceptible cultivar Ruda inoculated with spore suspensions of C. lindemuthianum race 83. Our results indicate that two genes which interact epistatically, one dominant and one recessive, are involved in the genetic control of leaf anthracnose resistance. As for stem anthracnose resistance, two genes also epistatic, one dominant and one recessive, explain the resistance to C. lindemuthianum race 83. The recessive gene is the same for leaf and stem resistance; however, the dominant genes are distinct and independent from each other. The three independent resistance genes of AN 910408 observed in this work could be derived from Guanajuato 31.

The periaqueductal gray (PAG) has been reported as a potential site for opioid regulation of behavioral selection. Opioid-mediated behavioral and physiological responses differ between nulliparous and multiparous females. This study addresses the effects of multiple reproductive experiences on mu-, kappa- and delta-opioid receptor (Oprm1, Oprk1, and Oprd1 respectively) gene activity and mu, kappa and delta protein expression (MOR, KOR and DOR respectively) in the PAG of the female rats. This was done by evaluating the opioid gene expression using real-time (RT-PCR) and quantification of each protein receptor by Western blot analysis. The RT-PCR results show that multiple reproductive experiences increase Oprm1 and Oprk1 gene expression. Western blot analysis revealed increased MOR and KOR while DOR protein was decreased in multiparous animals. Taken together, these data suggest that multiple reproductive experiences influence both gene activity and opioid receptor expression in the PAG. Post-translational mechanisms seem particularly relevant for DOR expression. Thus, opioid transmission in the PAG might be modulated by different mechanisms of multiparity-induced plasticity according to the opioid receptor type.

Diseases and insect pests are major causes of low yields of common bean (Phaseolus vulgaris L.) in Latin America and Africa. Anthracnose, angular leaf spot and common bacterial blight are widespread foliar diseases of common bean that also infect pods and seeds. One thousand and eighty-two accessions from a common bean core collection from the primary centres of origin were investigated for reaction to these three diseases. Angular leaf spot and common bacterial blight were evaluated in the field at Santander de Quilichao, Colombia, and anthracnose was evaluated in a screenhouse in Popayan, Colombia. By using the 15-group level from a hierarchical clustering procedure, it was found that 7 groups were formed with mainly Andean common bean accessions (Andean gene pool), 7 groups with mainly Middle American accessions (Middle American gene pool), while 1 group contained mixed accessions. Consistent with the theory of co-evolution, it was generally observed that accessions from the Andean gene pool were resistant to Middle American pathogen isolates causing anthracnoxe, while the Middle American accessions were resistant to pathogen isolates from the Andes. Different combinations of resistance patterns were found, and breeders can use this information to select a specific group of accessions on the basis of their need.

It is generally accepted that two major gene pools exist in cultivated common bean (Phaseolus vulgaris L.), a Middle American and an Andean one. Some evidence, based on unique phaseolin morphotypes and AFLP analysis, suggests that at least one more gene pool exists in cultivated common bean. To investigate this hypothesis, 1072 accessions from a common bean core collection from the primary centres of origin, held at CIAT, were investigated. Various agronomic and morphological attributes (14 categorical and 11 quantitative) were measured. Multivariate analyses, consisting of homogeneity analysis and clustering for categorical data, clustering and ordination techniques for quantitative data and nonlinear principal component analysis for mixed data, were undertaken. The results of most analyses supported the existence of the two major gene pools. However, the analysis of categorical data of protein types showed an additional minor gene pool. The minor gene pool is designated North Andean and includes phaseolin types CH, S and T; lectin types 312, Pr, B and K; and mostly A5, A6 and A4 types alpha-amylase inhibitor. Analysis of the combined categorical data of protein types and some plant categorical data also suggested that some other germplasm with C type phaseolin are distinguished from the major gene pools.

Although the co-ordination of promotive root-sourced cytokinin (CK) and inhibitory shoot apex-sourced auxin (IAA) is central to all current models on lateral bud dormancy release, control by those hormones alone has appeared inadequate in many studies. Thus it was hypothesized that the IAA : CK model is the central control but that it must be considered within the relevant timeframe leading to lateral bud release and against a backdrop of interactions with other hormone groups. Therefore, IAA and a wide survey of cytokinins (CKs), were examined along with abscisic acid (ABA) and polyamines (PAs) in released buds, tissue surrounding buds and xylem sap at 1 and 4 h after apex removal, when lateral buds of chickpea are known to break dormancy. Three potential lateral bud growth inhibitors, IAA, ABA and cis-zeatin 9-riboside (ZR), declined sharply in the released buds and xylem following decapitation. This is in contrast to potential dormancy breaking CKs like trans-ZR and trans-zeantin 9-riboside 5'phosphate (ZRMP), which represented the strongest correlative changes by increasing 3.5-fold in xylem sap and 22-fold in buds. PAs had not changed significantly in buds or other tissues after 4 h, so they were not directly involved in the breaking of bud dormancy. Results from the xylem and surrounding tissues indicated that bud CK increases resulted from a combination synthesis in the bud and selective loading of CK nucleotides into the xylem from the root.

Although cytokinins (CKs) are widely thought to have a role in promoting shoot branching, there is little data supporting a causative or even a correlative relationship between endogenous CKs and timing of bud outgrowth. We previously showed that lateral bud CK content increased rapidly following shoot decapitation. However, it is not known whether roots are the source of this CK. Here, we have used shoot decapitation to instantaneously induce lateral bud release in chickpea seedlings. This treatment rapidly alters rate and direction of solvent and solute (including CK) trafficking, which may be a passive signalling mechanism central to initiation of lateral bud release. To evaluate changes in xylem transport, intact and decapitated plants were infiltrated with [H-3]zeatin riboside ([H-3]ZR), a water-soluble blue dye or [H-3]H2O by injection into the hypocotyl. All three tracers were recovered in virtually all parts of the shoot within I h of injection. In intact plants, solute accumulation in the lateral bud at node 1 was significantly less than in the adjacent stipule and nodal tissue. In decapitated plants, accumulation of [H-3]ZR and of blue dye in the same bud position was increased 3- to 10-fold relative to intact plants, whereas content of [H-3]H2O was greatly reduced indicating an increased solvent throughput. The stipule and cut stem, predicted to have high evapotranspiration rates, also showed increased solute content accompanied by enhanced depletion of [H-3]H2O. To assess whether metabolism modifies quantities of active CK reaching the buds, we followed the metabolic fate of [H-3]ZR injected at physiological concentrations. Within 1 h, 80-95% of [H-3]ZR was converted to other active CKs (mainly zeatin riboside-5'phosphate (ZRMP) and zeatin (Z)), other significant, but unconfirmed metabolites some of which may be active (O-acetylZR, O-acetylZRMP and a compound correlated with sites of high CK-concentrations) and inactive catabolites (adenosine, adenine, 5'AMP and water). Despite rapid metabolic degradation, the total active label, which was indicative of CK concentration in buds, increased rapidly following decapitation. It can be inferred that xylem sap CKs represent one source of active CKs appearing in lateral buds after shoot decapitation.

Intracellular inclusions in the pedicel and calyx-tube tissues of Chamelaucium uncinatum Schauer ( Myrtaceae) flowers are irregular in shape. They were shown, by polarised light and scanning electron microscopy, to be birefringent 8.9-29.5 mum druse (i.e. aggregate) crystals. Energy-dispersive X-ray spectroscopy showed that these crystals were predominantly composed of calcium. Histochemical and acid-solubility tests indicated that the crystals were calcium oxalate. Raman microprobe spectroscopy was used to confirm this chemical identity. The calcium oxalate crystals were located in xylem-vessel lumens and also in parenchyma cells adjacent to vascular tissues. Thus, the crystals may function to regulate soluble calcium concentrations in C. uncinatum tissues near sites where calcium is unloaded from the xylem.

Two varieties of adzuki beans (Vigna angularis), Bloodwood and Erimo, were stored at temperatures of 10, 20 or 30degreesC, and relative humidities (RH) 40 or 65%, and samples were analysed at 0, 1.5, 3 and 6 months. Storage at 30degreesC for > 1.5 months caused a significant decrease in the a(star) and b(star) colour values and darkening of the seed coat. Beans stored at 65% RH had lower L-star but higher a(star) and b(star) colour values than those stored at 40% RH. Bloodwood and Erimo samples showed similar trends in colour during storage. The best storage conditions for the preservation of the adzuki colour were 10degreesC and 65% RH. The Australian beans had lower L-star, a(star) and b(star) colour values than Japanese Erimo-shouzu beans and storage increased the difference.

Two varieties of adzuki grown in Australia, Bloodwood and Erimo, were stored for up to 6 months at three temperatures (10, 20 and 30 degreesC), and two relative humidities (RH; 40 and 65%). The amount of cell wall material increased with time under all storage conditions. This increase was greatest at 30 degreesC and 40% RH. Storage time and conditions did not affect the total pectin levels in the cell wall. Erimo constantly exhibited a higher total pectin level than Bloodwood. The Bloodwood soluble pectin, Ca++ and Mg++ and Erimo Ca++ in the cell wall remained stable during storage, while the Erimo soluble pectin and Mg++ exhibited a slight decrease at 20 and 30 degreesC after 3 months of storage. O feijoeiro (Phaseolus vulgaris L.) é um dos principais alimentos consumidos no Brasil, rico em vitaminas, carboidratos e minerais. É crescente o número de pesquisas que integram desde o melhoramento de plantas e o manejo da adubação para incrementar nutrientes nas partes comestíveis, desenvolvendo plantas com maiores teores de vitaminas e micronutrientes. A introdução de alimentos biofortificados como o feijão, complementa a nutrição humana que sofre com desnutrição. Sendo assim, é importante conhecer como as condições de cultivo influenciam na qualidade do grão e na importância para obtenção de um alimento com maior valor nutricional. Além de conhecer até que ponto a influência de planta daninha afeta a produção e a absorção de nutrientes pelo feijoeiro. O objetivo deste trabalho foi analisar as características agronômicas e nutricionais influenciadas pela interferência de planta daninha, efeito da adubação e do solo, nas cultivares de feijão. Foi avaliado o teor de clorofila, número de folhas, diâmetro do caule, número de vagem por planta, índice de colheita por planta, massa seca da trapoeraba, relação grão e lócus, peso médio de sementes e análise de ferro e zinco nas folhas e nos grãos. Os resultados obtidos no estudo apontaram que a competição com trapoeraba afetou algumas características agronômicas, devido à competição por nutriente. O solo para cultivo também interferiu na produção, o solo eutrófico proporcionou melhores resultados das cultivares. A adubação não interferiu nos teores de ferro e zinco nos grãos do feijoeiro. As cultivares BRS Agreste, BRS Ametista e BRS Estilo apresentaram melhores resultados na maioria das características analisadas. Por fim, conclui-se que a produção do feijoeiro, necessita de um solo com uma boa nutrição, um bom teor de matéria orgânica, manejo da adubação e de plantas daninhas para aumentar a produção.

A consorciação de culturas anuais com o cafeeiro é realizada desde a introdução da cafeicultura no Brasil. Entretanto, os últimos resultados disponíveis foram obtidos com cultivares antigos e espaçamentos ultrapassados. Com o objetivo de avaliar o efeito do número de linhas de plantas e da dose de adubação NPK do feijoeiro intercalado ao cafeeiro, foram conduzidos dois experimentos de campo na Universidade Federal de Lavras, um em lavoura em formação (cv. Topázio) e outro em lavoura recepada (cv. Acaiá Cerrado), em duas épocas de plantio (das águas e da seca). O delineamento experimental foi blocos casualizados, com três repetições, sendo os tratamentos distribuídos em esquema fatorial 4 x 4 + 1, envolvendo quatro números de linhas intercalares de feijoeiro (uma, duas, três e quatro) e quatro doses de adubação (0, 50, 100 e 150% da adubação NPK recomendada para o monocultivo), mais um tratamento adicional (o monocultivo do cafeeiro ou do feijoeiro). Concluiu-se que o aumento do número de linhas de feijoeiro elevou o rendimento de grãos da leguminosa em cultivo intercalar ao cafeeiro, independentemente da dose de adubação, do tipo de manejo da lavoura (formação ou recepa) e da época de plantio, mas reduziu o incremento no diâmetro do caule do cafeeiro em formação, a emissão de nós e o crescimento dos ramos plagiotrópicos, tanto no cafeeiro em formação quanto no recepado. O emprego da adubação NPK em até 150% do equivalente à dose recomendada para o monocultivo não influenciou o rendimento de grãos do feijoeiro intercalado ao cafeeiro.

Este trabalho foi realizado com o objetivo de avaliar a resposta da cultura do feijoeiro (Phaseolus vulgaris L), cv. Carioca Precoce, à adubação com nitrogênio (N) e fósforo (P), cultivado no norte de Minas Gerais. O experimento foi realizado na safra do inverno-primavera de 2008, com delineamento experimental de blocos casualizados e três repetições. Os tratamentos foram organizados segundo esquema fatorial 4x4, sendo quatro doses de N: 0, 70, 140 e 210 kg ha-1 e quatro doses de P: 0, 100, 200 e 300 kg ha-1 de P2O5. A ureia e o superfosfato simples foram utilizados como fontes de N e P, respectivamente. O incremento das doses do fertilizante nitrogenado reduziu linearmente o estande inicial de plantas de feijão, sem influência das doses de fósforo. O número de vagens por planta foi o único componente de rendimento influenciado pela aplicação dos fertilizantes, unicamente para doses de N, com número máximo na dose de 108 kg ha-1 de N. Houve resposta quadrática da produtividade de grãos às diferentes doses de N e P, sendo a máxima produtividade (1.528 kg ha-1) alcançada com 98 kg ha-1 de N e 201 kg ha-1 de P, sem haver interação entre eles.

O Zabrotes subfasciatus (Boheman, 1833) (Coleoptera: Bruchidae) é considerado a principal praga do feijão armazenado. O método de controle mais utilizado tem sido o uso de inseticidas; no entanto outros métodos podem ser empregados, como, por exemplo, a resistência de plantas. Este trabalho teve por objetivo determinar os tipos de resistência em 12 genótipos de feijoeiro ao ataque dessa praga. O experimento foi conduzido no Laboratório de Resistência de Plantas a Insetos, do Departamento de Fitossanidade da FCAV\UNESP, Jaboticabal - SP, no período de março a novembro de 2009. Foram realizados testes de não preferência para oviposição, com e sem chance de escolha, e de alimentação, sem chance de escolha. Concluiu-se que os genótipos Dor 391, Dor 476, IAPAR-MD 806 e Raz 59 apresentam resistência do tipo não preferência para oviposição por Z. subfasciatus, em teste com chance de escolha; no teste sem chance de escolha todos os genótipos foram igualmente ovipositados pelo inseto; no teste com chance de escolha, machos e fêmeas foram igualmente atraídos por todos os genótipos testados; e Raz 56, Arc 2, Raz 55, Raz 49 e Raz 59 apresentaram resistência dos tipos não preferência para alimentação e,ou, antibiose.

O conhecimento da época e da dose de aplicação de dessecante, em pré-colheita do feijoeiro, é de extrema importância para se evitarem perdas na produtividade da cultura. Diante disso, o objetivo deste trabalho foi avaliar a produtividade de feijoeiro de inverno, após aplicação do dessecante paraquat em diferentes épocas (30; 35; 40 e 45 dias após o início do florescimento - DAF) e das doses (0; 200; 400 e 600 g ha-1), em pré-colheita. O experimento foi conduzido no período de outono/inverno de 2010, em Selvíria, Mato Grosso do Sul. Os tratamentos foram dispostos em delineamento experimental de blocos ao acaso, em esquema fatorial 4 x 4 (épocas de aplicação x doses do herbicida), com quatro repetições. O aumento das doses de paraquat, até os 35 DAF, aumentou de forma linear o percentual de sementes defeituosas, enquanto as sementes tiveram sua massa reduzida linearmente, até os 40 DAF. A produtividade do feijoeiro não foi afetada pela aplicação de paraquat a partir dos 35 DAF, ao contrário do incremento da dose, que provocou redução linear desse parâmetro.
