A remoção da smear layer é um procedimento importante para a regeneração periodontal e para o tratamento da sensibilidade dentinária. Diversos tratamentos tem sido realizados para sua remoção, porém, mostrando resultados conflitantes ou insatisfatórios. No presente estudo nos propomos a analisar através do microscópio eletrônico de varredura a eficiência dos detergentes: lauril sulfato de sódio, Plax e de mamona assim como o EDTA 24% na remoção da smear layer. Todos os produtos testados foram aplicados com bolinha de algodão, renovada a cada 30 segundos, a qual era esfregada na superfície radicular por diferentes períodos. As fotomicrografias foram analisadas através da aplicação de um índice de remoção da smear layer por três examinadores calibrados e os resultados analisados estatisticamente. De acordo com nossos resultados pudemos chegar as seguintes conclusões: 1- a instrumentação com curetas de Gracey provocou a formação da smear layer; 2- o detergente lauril sulfato de sódio não removeu a smear layer, tendo sido o pior resultado entre os detergentes testados; 3- Os detergentes Plax e de mamona mostraram resultados semelhantes entre si e superiores ao controle negativo e ao lauril sulfato de sódio, porém, não removeram a smear layer satisfatoriamente; 4- O EDTA 24% neutro mostrou ser eficiente para a remoção da smear layer.

AimTo evaluate the antibiofilm activity of sodium hypochlorite (NaOCl) and chlorhexidine (CHX) solutions associated with cetrimide (CTR), and QMiX using confocal laser scanning microscopy.MethodologyEnterococcus faecalis (ATCC- 29212) biofilms were induced on bovine dentine blocks for 14days. The dentine blocks containing biofilm were immersed for 1min in the following solutions: 2.5% NaOCl; 2.5% NaOCl+0.2% CTR; 2% CHX; 2% CHX+0.2% CTR; 0.2% CTR; QMiX. After contact with the solutions, the dentine blocks were stained with Live/Dead((R)) BacLight for analysis of the remaining biofilm using confocal laser scanning microscope. Images were evaluated using the BioImage_L software to determine the total biovolume (m(3)), the green biovolume (live cells) (m(3)) and the percentage of substrate coverage (%). The data were subjected to nonparametric statistical test using Kruskal-Wallis and Dunn's tests at 5% significance level.ResultsAfter exposure to irrigants, the total biovolume observed for CHX, CHX+CTR, CTR, QMiX was similar to distilled water (P>0.05). NaOCl and NaOCl+CTR had the lowest total and green biovolume. The CTR and QMiX had intermediate green biovolume, with greater antibacterial activity than CHX and CHX+CTR (P<0.05). The NaOCl and NaOCl+CTR solutions were associated with microorganism removal and substrate cleaning ability.ConclusionsNaOCl and NaOCl+CTR solutions were effective on microorganism viability and were able to eliminate biofilm. The addition of cetrimide did not influence antibacterial activity.

Diuron (3-(3,4-Dichlorophenyl)-1,1-dimethylurea) is a substituted urea herbicide widely used on agricultural crops such as soy, cotton and sugar cane. In a previous long-term study this herbicide exerted carcinogenic activity on the urinary bladder and renal pelvis mucosa of Wistar rats and breast of mice. Also, it was shown to be carcinogenic to the mice skin in a initiation-promotion assay. In 1997, the northamerican EPA evaluated Diuron as a “known/likely” carcinogen for humans (USEPA, 2004). In a previous study developed at this laboratory, male Wistar rats treated with Diuron 2500 ppm during 20 weeks presented increased indices of cell proliferation and incidences of simple urothelial hyperplasia (HS) in the urinary bladder. Under scanning electron microscopy (SEM) severe urothelial necrosis and hyperplasia were observed. However, in that study the urinary bladders of animals exposed to lower doses of Diuron were not examined under SEM. Therefore, the possible dose-response influence of Diuron on the urothelium under SEM is not known. The present study aimed to analyze under SEM the urinary bladder of male Wistar rats exposed to 125 ppm, 500 ppm and 2500 ppm doses of Diuron through diet during 20 weeks and to compare to the previous histological findings in the same material. Under SEM, 125 ppm and 2500 ppm groups presented significantly (p<0,05) increased incidences of simple hyperplasia, i.e., 7/10 and 8/10 respectively, compared to control group and the 500 ppm group The sensitivity of SEM was higher since it detected a 45% incidence of hyperplasiaswhile the histological analysis found only 27%. Considering SEM as the gold-standard, histology showed a 44% sensitivity, 86.4% specificity, a positive predictive value of 72,7% and negative predictive value of 65,5% and accuracy of 67,5%. This project developed microstructural characterization technics of commercial dualphase and multi-phasic (TRIP) steels that were provided by the automotive industry and are currently used as the raw material for the production of automobiles. Inserted in this context, there is the development of the advanced high strength steels in consonance with the ULSABAVC project, which aims the production of safe, economically viable and efficient in terms of fuel consumption vehicles for the 21st century. The micro-structural characterization of biphasic and multiphase steels was done by the identification and quantification of the coexistent phases. In this item, a special attention was given to the technics that were performed using optic microscopy and scanning electron microscopy. An important contribution to this work was the utilization of different alternative chemical reagents (Beraha, Heat-Tinting technics) in addition to the classical ones (Nital and LePera)already used conventionally by the UNESP's Group of Mechanical, Microstructural and Fractografic Characterization of Materials. The revealed microstructures were correlated with the materials' mechanical properties determined through traction tests, such as ultimate tensile strengths, yield strength and stretching important since the material has structural application in the automotive industry. As a result, it was observed the superiority concerning the studied mechanical properties for the biphasic and multiphasic steels when compared to the conventional carbon steels. Besides, it was perceived a large potential for the industrial scale utilization of the Heat Tinting technics in this field, seen its differentiation of the existent phases and easy reproducibility

This study aimed to evaluate the effect of Er:YAG (L) and diamond drills (DD) on: 1) the microshear bond strength (MPa); 2) the adhesive interface of two-step (TS) – Adper Scotchbond Multipurpose and one-step (OS) adhesives – Adper EasyOne, both from 3M ESPE. Material and methods: According to the preparation condition and adhesives, the samples were divided into four groups: DD_TS (control); DD_OS; L_TS and L_OS. 60 bovine incisors were randomly divided into experimental and groups: 40 for microshear bond strength (n = 10) and 20 for the adhesive interface morphology [6 to measure the thickness of the hybrid layer (HL) and length of tags (t) by CLSM (n = 3); 12 to the adhesive interface morphology by SEM (n = 3) and 2 to illustrate the effect of the instruments on dentine by SEM (n = 1)]. To conduct the microshear bond strength test, four cylinders (0.7 mm in diameter and 1 mm in height with area of adhesion of 0.38 mm) were constructed with resin composite (Filtek Z350 XT – 3M ESPE) on each dentin surface treated by either L or DD and after adhesives application. Microshear bond strength was performed in universal testing machine (EMIC 2000) with load cell of 500 kgf and a crosshead speed of 0.5 mm / min. Adhesive interface was characterized by thickness of hybrid layer (HL) and length of tags (t) in nm, with the aid of UTHSCSA ImageTool software. Results: Microshear bond strength values were: L_TS 34.10 ± 19.07, DD_TS 24.26 ± 9.35, L_OS 33.18 ± 12.46, DD_OS 21.24 ± 13.96. Two-way ANOVA resulted in statistically significant differences only for instruments (p = 0.047). Mann-Whitney identified the instruments which determined significant differences for HL thickness and tag length (t). Concerning to the adhesive types, these differences were only observed for (t). Conclusion: It can be concluded that 1) laser Er:YAG results in higher microshear bond strength values regardless of the adhesive system (TS and OS); 2) the tags did not significant affect the microshear bond strength; 3) the adhesive interface was affected by both the instruments for cavity preparation and the type of adhesive system used.

Introduction The aim of this study was to compare the effect of QMix, BioPure MTAD, 17 % EDTA, and saline on the penetrability of a resin-based sealer into dentinal tubules using a confocal laser scanning microscope (CLSM) and to describe the cleaning of root canal walls by SEM. Methods Eighty distobuccal roots from upper molars were selected and randomly divided into four groups (n=20) before root canal preparation according to the solution used in the final rinse protocol (FRP): QG (QMix), MG (BioPure MTAD), EG (17 % EDTA), and CG (control group: saline). Ten roots of each group were prepared for SEM, and images (×2000) from the canal walls were acquired. The remaining canals were filled with a single gutta-percha cone and AH Plus with 0.1 % Rhodamine B. The specimens were horizontally sectioned at 4 mm from the apex, and the slices were analyzed in CLSM (×10). Sealer penetration was analyzed with Adobe Photoshop software. Results QG and EG presented similar amounts of sealer penetration (P>.05). MG and CG presented the lowest penetrability values (P<.05). The best results for smear layer removal of the apical third of the root canal were achieved by the QG and EG groups when compared with MG and CG (P<.05). Conclusions Seventeen percent EDTA and QMix promoted sealer penetration superior to that achieved by BioPure MTAD and saline. Clinical relevance Despite studies have not confirmed the relationship between sealing ability of endodontic sealers and their penetration in dentinal tubules, sealer penetration assumes importance, since endodontic sealers, unlike guttapercha, are able to penetrate in dentinal tubules, isthmus, and accessory canals, filling the root canal system.

Recent bonding systems have been advocated as multi-purpose bonding agents. The aim of this study was to determine if some of these bonding systems could be associated to composite resins from different manufacturers. This investigation was conducted to test lhe shear bond strength of three bonding systems: Scotchbond Multi-Purpose (3M Dental Products), Optibond Light Cure (Kerr) and Optibond Dual Cure (Kerr), when each of them was associated to lhe composite resins: Z1 00 (3M Dental Products), Prisma - APH (Dentsply) and Herculite XRV (Kerr). Seventy-two flat dentin bonding sites were prepared to 600 grit on human premolars mounted using acrilic resins. The teeth were assigned at random to 9 groups of 8 samples each. A split die with a 3mm diameter was placed over lhe surface of lhe dentin treated with one of lhe adhesive systems, and lhe selected composite resin was inserted and light cured. The split mold was removed and all samples were termocycled and stored in 37ºC water for 24 hours before testing. Shear bond strength was determined using an lnstron Universal testing machine. Some failures were examined under lhe S.E.M. Data was analysed by one-way analysis of variance, that demonstrated a significant difference (p<0,05) in the mean shear bond strength among Optibond Light Cure (15,446 MPa), Scotchbond Multi-Purpose (13,339 MPa) and Optibond Dual Cure (10,019 MPa). These values did not depend on the composite resin used. The association between bonding system/composite resin was statistycally significant (p<0,05) and the best results were obtained when the composite resins Z100 and Herculite were used with the adhesive system Optibond Light Cure, and when the composite resin APH was used with the adhesive system Scotchbond Multi-Purpose
