C-type lectins are a superfamily of Ca2+ dependent carbohydrate-recognition proteins which play significant diverse roles in nonself-recognition and clearance of invaders. In the present study, a C-type lectin (CfLec-2) from Zhikong scallop Chlamys farreri was selected to investigate its functions in innate immunity. The mRNA expression of CfLec-2 in hemocytes was significantly up-regulated (P < 0.01) after scallops were stimulated by LPS. PGN or beta-glucan, and reached the highest expression level at 12h post-stimulation, which was 72.5-, 23.6- or 43.8-fold compared with blank group, respectively. The recombinant Cflec-2 (designated as rCfLec-2) could bind LPS, PGN, mannan and zymosan in vitro, but it could not bind beta-glucan. Immunofluorescence assay with polyclonal antibody specific for Cflec-2 revealed that CfLec-2 was mainly located in the mantle, kidney and gonad. Furthermore, rCfLec-2 could bind to the surface of scallop hemocytes, and then initiated cellular adhesion and recruited hemocytes to enhance their encapsulation in vitro, and this process could be specifically blocked by anti-rCfLec-2 serum. These results collectively suggested that CfLec-2 from the primitive deuterostome C. farreri could perform two distinct immune functions, pathogen recognition and cellular adhesion synchronously, while these functions were performed by collectins and selectins in vertebrates, respectively. The synchronous functions of pathogen recognition and cellular adhesion performed by CfLec-2 tempted us to suspect that CfLec-2 was an ancient form of C-type lectin, and apparently the differentiation of these two functions mediated by C-type lectins occurred after mollusk in phylogeny. The globular C1q-domain-containing (C1qDC) proteins are a family of versatile pattern recognition receptors via their globular C1q (gC1q) domain to bind various ligands including several PAMPs on pathogens. In this study, a new gC1q-domain-containing protein (AiC1qDC-1) gene was cloned from Argopecten irradians by rapid amplification of cDNA ends (RACE) approaches and expressed sequence tag (EST) analysis. The full-length cDNA of AiC1qDC-1 was composed of 733 bp, encoding a signal peptide of 19 residues and a typical gC1q domain of 137 residues containing all eight invariant amino acids in human C1qDC proteins and seven aromatic residues essential for effective packing of the hydrophobic core of AiC1qDC-1. The gC1q domain of AiC1qDC-1, which possessed the typical 10-stranded beta-sandwich fold with a jelly-roll topology common to all C1q family members, showed high homology not only to those of Cl qDC proteins in mollusk but also to those of C1qDC proteins in human. The AiC1qDC-1 transcripts were mainly detected in the tissue of hepatopancreas and also marginally detectable in adductor, heart, mantle, gill and hemocytes by fluorescent quantitative real-time PCR. In the microbial challenge experiment, there was a significant up-regulation in the relative expression level of AiC1qDC-1 in hepatopancreas and hemocytes of the scallops challenged by fungi Pichia pastoris GS115, Gram-positive bacteria Micrococcus luteus and Gram-negative bacteria Listonella anguillarum. The recombinant AiC1qDC-1 (rAiC1qDC-1) protein displayed no obvious agglutination against M. luteus and L. anguillarum, but it aggregated P. pastoris remarkably. This agglutination could be inhibited by D-mannose and PGN but not by LPS, glucan or D-galactose. These results indicated that AiC1qDC-1 functioned as a pattern recognition receptor in the immune defense of scallops against pathogens and provided clues for illuminating the evolution of the complement classical pathway. Peptidoglycan recognition protein (PGRP) is an essential molecule in innate immunity for both invertebrates and vertebrates, owing to its prominent ability in detecting and eliminating the invading bacteria. Several PGRPs have been identified from mollusk, but their functions and the underlined mechanism are still unclear. In the present study, the mRNA expression profiles, location, and possible functions of PGRP-S1 from Zhikong scallop Chlamys farreri (CfPG RP-St) were analyzed. The CfPGRP-S1 protein located in the mantle, gill, kidney and gonad of the scallops. Its mRNA expression in hemocytes was up-regulated extremely after PGN stimulation (P < 0.01), while moderately after the stimulations of LPS (P < 0.01) and beta-glucan (P < 0.05). The recombinant protein of CfPGRP-S1 (designated as rCfPGRP-S1) exhibited high affinity to PGN and moderate affinity to LPS, but it did not bind beta-glucan. Meanwhile, rCfPGRP-S1 also exhibited strong agglutination activity to Gram-positive bacteria Micrococcus luteus and Bacillus subtilis and weak activity to Gram-negative bacteria Escherichia coli. More importantly, rCfPGRP-S1 functioned as a bactericidal amidase to degrade PGN and strongly inhibit the growth of E. coli and Staphyloccocus aureus in the presence of Zn2+. These results indicated that CfPGRP-S1 could not only serve as a pattern recognition receptor recognizing bacterial PGN and LPS, but also function as a scavenger involved in eliminating response against the invaders. Serine protease inhibitors, critical regulators of endogenous proteases, are found in all multicellular organisms and play crucial roles in host physiological and immunological effector mechanisms. The first mollusk serine proteinase inhibitor (designated AISPI) cDNA was obtained from the bay scallop Argopecten irradians by randomly sequencing a whole tissue cDNA library and rapid amplification of cDNA ends (RACE). The full-length cDNA of the scallop serine protease inhibitor was 1020 bp, consisting of a 5'-terminal untranslated region (UTR) of 39 bp, a 3'-terminal UTR of 147 bp with a canonical polyadenylation signal sequence AATAAA and a poly(A) tail, and an open reading frame of 834 bp. The AISPI cDNA encoded a polypeptide of 278 amino acids with a putative signal peptide of 22 amino acids and a mature protein of 256 amino acids. The deduced amino-acid sequence of AISPI contained six tandem and homologous domains similar to that of Kazal-type serine protease inhibitors, including the conserved sequence C-X(7)-C-X(6)-Y-X(3)-C-X(2,3)-C and six cysteine residues responsible for the formation of disulfide bridges, indicating that the AISPI protein from bay scallop should be a member of the Kazal-type serine protease inhibitor family. The temporal expression of AISPI was measured by semi-quantitative RT-PCR after injury or bacterial challenge. After the adductor muscle was wounded or injected with Vibrio anguillarum, the expression of AISPI mRNA in hemolymph was up-regulated and reached the maximum level at 8 and 16 h, respectively, and then progressively dropped back to the original level. The results indicated that AISPI could play an important role in injury healing and immune response in mollusks as it could be induced by injury and bacterial challenge. (c) 2005 Elsevier Ltd. The loess-paleosol sequences in China are among the best continental records of paleoclimate changes. Although numerous sedimentological and geochemical studies have contributed greatly to the understanding of past climate changes during this period, it is still necessary to decipher more details through investigating these sequences using various approaches including biological analyses. In this study, we analyze the mollusk fossil assemblages preserved in the upper part of the Xifeng section, from the fifth loess layer (L5) to the Holocene soil (S0), with the sampling interval of 10 cm. The main results and conclusions obtained are as follows: 1. A continuous terrestrial mollusk fossil record, covering the past 500 ka, has been obtained from the Xifeng loess-paleosol sequence, which provides important biological data for the study of paleoenvironmental changes in the Loess Plateau and its comparison with marine record during this period. A total of 475 mollusk assemblages were studied, and twenty-one species have been identified among the 210,000 mollusk individuals counted. Among these species, most have modern representatives and are found in previous terrestrial mollusk studies of Chinese loess-paleosol sequences. Thus, they can be grouped into cold-aridiphilous, thermo-humidiphilous, oriental, and cool-humidiphilous ecological groups, as defined by previous studies. 2. Comparison of mollusk assemblages between the last five glacials and four interglacials and Holocene shows very different climate conditions. The warmest period occurred at MIS 11, MIS 5e, and Holocene, respectively. The coldest period is the Last Glacial Maximam, rather than the MIS 12. 3. Our mollusk record provides insight into the climate conditions in the Loess Plateau during the MIS 11, interpreted as the closest analog to the present interglacial. S4 paleosol, equivalent of MIS 11, developed under two major different climate regimes: ranging from the very warm–humid early phase to the mild-cool late interval. Furthermore, a cooling spell has been documented at the interglacial optimum, reflecting unstable climate conditions. The early warm–humid conditions lasted over 30 ka, supporting that MIS 11 is a unique long interglacial in the Quaternary climate history. 4. Comparison of MIS 11 and Holocene climates based on the mollusk species compositions indicates major differences. The climate at the early part of MIS 11 was warmer and more humid than during the Holocene optimum period, but the conditions during the late part of MIS 11 were similar to or cooler than late Holocene. Our study indicates that the extent of warming during the Holocene might be significantly less than the conditions that prevailed during the early part of MIS 11 interglacial period. 5. Two strong summer monsoon events were observed during the MIS 12 and MIS 10. They correspond to the maximam values of insolation gradient between low and high latitudes, suggesting a causal linkage. 6. Our study, combined with the previously investigated Luochuan land snail record, reveals that the climate in the Loess Plateau during MIS 3 experienced three stages: relatively warm, humid climate prevailed during MIS 3c, relatively cold, dry climate during MIS 3b, and relatively warm-humid period during MIS 3a. Climate at this time fluctuated frequently in Luochuan, and changed from warm-cool to cold-dry in Xifeng. Our results reveal that the relatively warm-humid climate during MIS 3c may be resulted from an increasing insolation gradient controlled by obliquity. Our result also reveals that obvious regional difference existed in the Loess Plateau during MIS 3. A greater climate gradient occurred during this time compared with today’s climate pattern in the Loess Plateau.

In this study, 172 mollusk assemblages from the Weinan loess section in the southeast of Loess Plateau, China, were identified quantitatively at relative high resolution. The results show: 1) the history and processes of paleoclimatic and paleoenvironmental changes in Weinan since the last 70 ka; 2) the characteristics of climatic changes during the period of the last glacial maximum (LGM); 3) the spatial pattern of paleoclimate variations at the south and middle parts of the Loess Plateau during the LGM period; 4) the timing of the last deglaciation and the return event of rapid climate change during the last deglaciation. The main conclusions are as follows: 1) 172 mollusk samples taken from the uppermost 9 m deposits cover the past 70 ka, which were sampled at the internals of 5 cm for S_0, 3 cm for L_(1-1) and L_(1-2), and 10 cm for L_(1-3), L_(1-4) and L_(1-5). Author analyzed quantitatively all individuals including broken pieces of snail shells, percentages of 15 species identified from 172 samples. Three main groups were determined according to the ecological requirement of each taxon. Based on the variations of three ecological groups and typical ecological species, The author intended to reconstruct the history of and processes of climate and environment since the last 70 ka in the Weinan region. The climate and environment in this region experienced the following changes: relative warm and humid stage from 67.5-20.3 cal. ka B.P., a period of forest-steppe or steppe developed; cold and arid stage from 20.3-15.5 cal. ka B.P., a dry steppe period, later wetter and colder; cold and humid period once time from 15.5 to 12.3 cal. ka B.P., a typical steppe or forest-steppe stage; cold and humid again from 12.3 cal. ka B.P. to 8.2 cal. ka B.P., a tropical steppe stage; warm and humid climate, a forest-steppe developed. 2) The climate during the period of the last glacial maximum (LGM) in Weinan was characterized by a general cold-humid condition, represented by occurrence of a number of the cool-humidiphilous mollusk species such as Gastocopta amigerella and Vallonia cf. pulchella in the section. 3) Comparison of the variations in abundance of Puncture orphana at Weinan with those at Luochuan and Changwu sections suggests that the summer monsoon intensity influenced differently at the three regions during the LGM period. The Weinan was weaker summer monsoon impact during all the period, the Luochuan was influenced occasionally, and Changwu was only a very short time affected, which indicated it might be located at the western margin of the summer monsoon influence during that period. 4) The ratio of thermo-humidiphilous mollusk group to cold-aridiphilous one shows an increase tendency at about 15 cal. ka B.P., reflecting the climate warming after the deglaciation in Weinan, which is approximately corresponding to the timing of warming period of the last deglaciation, found in the East Atlantic Ocean, the South China Sea and the Loess Plateau (indicated by the phytolith study). 5) A remarkable decrease in the number of thermo-humidiphilous and cool-humidiphilous mollusk species from 12.7 - 11.6 cal. ka B.P. indicates a cooling in climate and might be the reflection of the Younger Dryas event in Weinan. 6) Variations in the ratios of thermo-humidiphilous mollusk species to cold-aridiphilous ones reflect the climate instability in Holocene. There were four warm-humid periods (10-8.1 cal. ka B.P., 6.9-6.1 cal. ka B.P., 5.2-2.6cal. ka B.P., 1.6cal. ka B.P. to the present ) and three relative cold-arid periods (8.1-6.9 cal. ka B.P., 6.1-5.2 cal. ka B.P., 2.6-1.6 cal. ka B.P.), showing about a 1,000 year climatic oscillation.

In this study, 260 mollusk fossil samples from a Red Clay sequence at Xifeng, Gansu province, in the northern China were analyzed quantitatively. 12 fossil species and four fossil zones have been identified. Three main ecological groups were determined based on ecological requirement of each mollusk taxon. According to fossil composition and succession of three ecological groups, the author discussed the origin and sedimentary environment of the red clay deposits, and the process of ecological environmental changes as well as the variations of the East Asia monsoons during 6.2-2.4 Ma in the Loess Plateau. A preliminary study on periodicity of paleoclimatic changes was also conducted by using spectral analysis method. The main results and conclusions are presented as follows:A continuous land mollusk fossil sequence of 6.2-2.4 Ma from Xifeng Red Clay Formation has been established, which provided a basic data for studying the environmental changes during late Miocene to Pliocene.The study of composition and preservation condition of mollusk fossils reveals a terrestrial in situ ecological population in the Red Clay Formation. All of identifiable mollusk species are composed of terrestrial taxa, which support the view that the Red Clay is an eolian origin, similar to the overlying Quaternary loess deposits.The mollusk record reveals the processes of ecological and environmental changes during 6.2-2.4 Ma in the Loess Plateau. Climatic changes experienced cold and dry from 6.2-5.4 Ma, warm and wet during 5.4-4.5 Ma, mild and moderate from 4.5-3-4 Ma, to rapid cooling and drying after 3.4 Ma. From '5.4- 2.4 Ma, climate was stepwise cooling. The cooling trend is in good agreement with a general1 0global cooling trend during this period, as documented by marine 5 0 records.4. Three remarked ecological shifts took place in mollusk assemblages from 6.2-2.4 Ma, focused on about 5.4, 4.5 and 3.4 Ma. The warming shift around 5.4 Ma was probably related to the rising of the global temperature. The cooling shifts around 4,5 and 3.4 Ma however might be closely linked to the uplift of Tibet Plateau and the development of Northern Hemisphere ice sheet.The succession in mollusk ecological groups also recorded the variability of the East Asian winter and summer monsoon. The winter monsoon dominated two periods from 6.2-5.4 Ma and from 3.4-2.4 Ma, while the summer monsoon was strong during 5.4-4.5 Ma. The variations in winter and summer monsoons were in phase during 4.5-3.4 Ma. Monsoon regimes changed with the duration about 1 Ma, which roughly corresponds to the cycle driven by tectonic activity on the time scales of ICP-IO7 years. In addition, mollusk fossils recorded the large amplitude and high frequency fluctuations overlapped on 105-107 years climate cycle.The maximum entropy spectral analysis and filter-band analysis of total mollusk individuals and three typical ecological groups suggest that the climate changes controlled mainly by solar insolation had periods about 70 ka and 40 ka on the time scales of 105 during late Miocene-Pliocene. Climatic periodicity intensified from 4.0 Ma, which reflected strengthened forcing by high latitude ice volume.

It is an open question how animals find food in dynamic natural environments where they possess little or no knowledge of where resources are located. Foraging theory predicts that in environments with sparsely distributed target resources, where forager knowledge about resources’ locations is incomplete, Lévy flight movements optimize the success of random searches. However, the putative success of Lévy foraging has been demonstrated only in model simulations. Here, we use high-temporal-resolution Global Positioning System (GPS) tracking of wandering (Diomedea exulans) and black-browed albatrosses (Thalassarche melanophrys) with simultaneous recording of prey captures, to show that both species exhibit Lévy and Brownian movement patterns. We find that total prey masses captured by wandering albatrosses during Lévy movements exceed daily energy requirements by nearly fourfold, and approached yields by Brownian movements in other habitats. These results, together with our reanalysis of previously published albatross data, overturn the notion that albatrosses do not exhibit Lévy patterns during foraging, and demonstrate that Lévy flights of predators in dynamic natural environments present a beneficial alternative strategy to simple, spatially intensive behaviors. Our findings add support to the possibility that biological Lévy flight may have naturally evolved as a search strategy in response to sparse resources and scant information.

We present the first marine reservoir age and Delta R determination for the island of St. Helena using marine mollusk radiocarbon dates obtained from an historical context of known age. This represents the first marine reservoir a.-c and Delta R determination in the southern Atlantic Ocean within thousands of kilometers of the island. The depletion of C-14 in the shells indicates a rather larger reservoir age for that portion of the surface Atlantic than models indicate. The implication is that upwelling old water along the Namibian coast is transported for a considerable distance, although it is likely to be variable on a decadal timescale. An artilleryman's button, together with other artifacts found in a midden, demonstrate association of the mollusk shells with a narrow historic period of AD 1815-1835.

Specimens of the polyplacophoran mollusk 'Helminthochiton' thraivensis Reed from the Upper Ordovician of southwest Scotland provide rare examples of complete valve series preserved in near life position, albeit as external molds. Application of high-resolution X-ray microtomography to one such specimen has revealed the exceptional preservation of its last meal, which included elements of a crinoid column, in its intestine. The interaction was either predatory or scavenging; extant chitons are not known to be crinoidivorous. This is the earliest direct record of predation or scavenging on crinoids in the fossil record. It is also the first indication that the broad axial canal of primitive crinoids may have contained nutritious tissues. The predatory or scavenging habit of H. thraivensis is consistent with its inferred phylogenetic position as a stem-group aplacophoran and provides new data suggesting an origin of carnivory early in the evolution of this clade.

Marine radiocarbon bomb-pulse time histories of annually resolved archives from temperate regions have been underexploited. We present here series of Delta C-14 excess from known-age annual increments of the long-lived bivalve mollusk Arctica islandica from 4 sites across the coastal North Atlantic (German Bight, North Sea; Tromso, north Norway; Siglufjordur, north Icelandic shelf; Grimsey, north Icelandic shelf) combined with published series from Georges Bank and Sable Bank (NW Atlantic) and the Oyster Ground (North Sea). The atmospheric bomb pulse is shown to be a step-function whose response in the marine environment is immediate but of smaller amplitude and which has a longer decay time as a result of the much larger marine carbon reservoir. Attenuation is determined by the regional hydrographic setting of the sites, vertical mixing, processes controlling the isotopic exchange of C-14 at the air-sea boundary, C-14 content of the freshwater flux, primary productivity, and the residence time of organic matter in the sediment mixed layer. The inventories form a sequence from high magnitude-early peak (German Bight) to low magnitude-late peak (Grimsey). All series show a rapid response to the increase in atmospheric Delta C-14 excess but a slow response to the subsequent decline resulting from the succession of rapid isotopic air-sea exchange followed by the more gradual isotopic equilibration in the mixed layer due to the variable marine carbon reservoir and incorporation of organic carbon from the sediment mixed layer. The data constitute calibration series for the use of the bomb pulse as a high-resolution dating tool in the marine environment and as a tracer of coastal ocean water masses.

Mollusk shells are frequently radiocarbon dated and provide reliable calibrated age ranges when the regional marine reservoir correction is well-established. For mollusks from an estuarine environment the reservoir correction may be significantly different than the regional marine reservoir correction due to the input of bedrock or soil derived carbonates. Some mollusk species such as oysters are tolerant of a significant range of salinities which makes it difficult to determine which reservoir correction is appropriate. A case study is presented of an anomalous radiocarbon age for an oyster shell paint dish found in the fabric of the ruined nave walls of St Mary's Church, Shoreham-by-Sea, West Sussex, England. Stable isotopes (delta O-18 and delta C-13) were used to establish the type of environment in which the oyster had lived. Paired marine and terrestrial samples from a nearby medieval site were radiocarbon dated to provide an appropriate reservoir correction.
