Bulk chlorine concentrations and chlorine stable isotope compositions were determined for hydrothermally altered basalt (extrusive lavas and sheeted dikes) and gabbro samples (n = 50) from seven DSDP/ODP/IODP drill sites. These altered oceanic crust (AOC) samples span a range of crustal ages, tectonic settings, alteration type, and crustal depth. Bulk chlorine concentrations range from < 0.01 wt.% to 0.09 wt.%. In general, higher chlorine concentrations coincide with an increase in temperature of alteration and amphibole content. d37Cl values of whole rock AOC samples range from -1.4 to +1.8 per mil. High d37Cl values (>=~0.5 per mil) are associated with areas of higher amphibole content. This observation is consistent with theoretical calculations that estimate amphibole should be enriched in 37Cl compared to co-existing fluid. Negative to near zero d37Cl values are found in areas dominated by clay minerals. Chlorine geochemistry is a rough indicator of metamorphic grade and mineralogy. AOC is a major Cl host in the subducting oceanic lithospheric slab. Here we show that bulk chlorine concentrations are ~3 times higher than previous estimates resulting in a greater contribution of Cl to the mantle.

La recherche sur la conception de nouveaux matériaux, dits intelligents, est en constant progrès depuis plus de 30 ans. Historiquement, les premiers matériaux utilisés et transformés par l’homme étaient le bois, les minéraux et ses dérivés (pierre, métaux, etc.). C’est à la fin du 19e siècle que la synthèse des polymères organiques et inorganiques ainsi que leurs utilisations se développèrent. Ce progrès continue de nos jours. Ainsi, c’est dans cette direction que cette thèse fut rédigée, l’étude de polymères de coordination basés sur le cuivre(I). Elle s’orchestra en six principales sections de recherche. La première section, i.e. le chapitre 2 traite de la coordination de différents ligands monothioéthers sur du CuX (X = I, Br) pour former plusieurs types de polymères de coordination (CPs). Ainsi, le CP 1D [(Me[indice inférieur 2]S)[indice inférieur 3]{Cu[indice inférieur 2]([mu]-I)[indice inférieur 2}][indice inférieur n] est obtenu quand CuI et Sme[indice inférieur 2] réagissent ensemble dans le n-heptane, alors qu’ils entrainent la formation du CP 2D [(Me[indice inférieur 2]S) [indice inférieur 3] {Cu[indice inférieur 4]([mu]-I) [indice inférieur 4]}] [indice inférieur n] dans le MeCN. Ce dernier contient des unités de construction secondaire (SBU ; Secondary Building Units en anglais) en forme de cluster Cu[indice inférieur 4]I[indice inférieur 4] « cubane partiellement ouvert ». En faisant réagir le MeSEt avec du CuI, le CP 2D [(MeSEt) [indice inférieur 2]{Cu[indice inférieur 4] ([mu][indice inférieur 3]-I) [indice inférieur 2] ([mu][indice inférieur 2]-I) [indice inférieur 2]}(MeCN) [indice inférieur 2]] [indice inférieur n] contenant des SBUs de type Cu[indice inférieur 4]I[indice inférieur 4] « cubanes en escalier » a été isolé dans MeCN, alors qu’ils entrainent l’obtention du polymère 1D [(MeSEt) [indice inférieur 3]{Cu[indice inférieur 4] ([mu][indice inférieur 3]-I) [indice inférieur 4]}] [indice inférieur n] dans le n-heptane contenant quant à lui des clusters de types Cu[indice inférieur 4]I[indice inférieur 4] « cubanes fermés ». Alors que le traitement de MeSPr avec du CuI forme le CP 1D [(MeSPr) [indice inférieur 3]{Cu[indice inférieur 4] ([mu][indice inférieur 3]-I) [indice inférieur 4]}] [indice inférieur n], les composés [(L) [indice inférieur 4]{Cu[indice inférieur 4] ([mu][indice inférieur 3]-I) [indice inférieur 4]}] (L = EtSPr, Pr[indice inférieur 2]S) sont respectivement obtenus avec le EtSPr et le Pr[indice inférieur 2]S. À partir du [indice supérieur i]Pr[indice inférieur 2]S et de CuI, le cluster [([indice supérieur i]Pr[indice inférieur 2]S) [indice inférieur 6]{Cu[indice inférieur 8] ([mu][indice inférieur 3]-I) [indice inférieur 3]}([mu][indice inférieur 4]-I) [indice inférieur 2]}] est obtenu alors que l’on forme un CP 2D [(Cu[indice inférieur 3]Br[indice inférieur 3])(MeSEt) [indice inférieur 3]] [indice inférieur n] à partir de CuBr et MeSEt dans l’heptane. Ce dernier incorpore à la fois des Cu([mu][indice inférieur 2]-Br) [indice inférieur 2]Cu rhomboédriques et des SBUs de type Cu[indice inférieur 4]Br[indice inférieur 4] « cubanes ouverts ». Le MeSPr forme, quant à lui avec le CuBr dans l’heptane, le CP 1D [(Cu[indice inférieur 3]Br[indice inférieur 3])(MeSPr) [indice inférieur 3]] [indice inférieur n] qui, après recristallisation dans le MeCN, est converti en un CP 2D [(Cu[indice inférieur 5]Br[indice inférieur 5])([mu][indice inférieur 2]-MeSPr) [indice inférieur 3]] [indice inférieur n] incorporant des SBUs [(Cu[indice inférieur 5] ([mu][indice inférieur 4]-Br)([mu][indice inférieur 2]-Br)]. Les propriétés de stabilités thermiques et photophysiques de ces matériaux ont aussi été reportées.   Dans la section 2, i.e. au chapitre 3, les réactions entre des ligands dithioétherbutanes (1,4-bis(phénylthio)butane et 1,4-bis(cyclohexylthio)butane) avec CuX (X = Br, I) ont été étudiées. En faisant réagir les CuX avec le 1,4-bis(cyclohexylthio)butane, dans le ratio (1:1), les CPs 1D, peu luminescents, isostructuraux [(Cu[indice inférieur 2]X[indice inférieur 2])([mu]-CyS(CH[indice inférieur 2]) [indice inférieur 4])SCy) [indice inférieur 2]] [indice inférieur n] (X = Br, I) sont obtenus. Inversement, quand CuI réagit avec 1,4-bis(phénylthio)butane, dans le ratio (2:1), il se forme le préalablement reporté CP 2D [(Cu[indice inférieur 4]I[indice inférieur 4])([mu]- PhS(CH[indice inférieur 2]) [indice inférieur 4])SPh) [indice inférieur 2]] [indice inférieur n], alors qu’avec le CyS(CH[indice inférieur 2]) [indice inférieur 4])SCy, un nouveau composé luminescent est obtenu, mais sa structure n’a pas pu être résolue.(1) Les caractérisations habituelles en photophysique et en stabilité thermique ont été menées sur ces matériaux. Dans la troisième section, i.e. dans le chapitre 4, les réactions de coordination de CuX (Br, I) sur les ligands dithioétherbutènes E- et Z-PhS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])SPh, E- et Z-pTolS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])S-pTol ont été comparées. Quand les sels CuX réagissent avec E-PhS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])SPh les CP 2D [Cu[indice inférieur 2]X[indice inférieur 2]{[mu]-E-PhS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])SPh}[indice inférieur 2]] [indice inférieur n] (X = I, Br), composés isostructuraux, sont obtenus. Incorporant une structure sans-précédente, ces réseaux sont formés à partir de couches 2D en alternance ABAB, contenants des SBUs Cu[indice inférieur 2] ([mu][indice inférieur 2]-X) [indice inférieur 2] rhomboédriques. Inversement, quand l’isomère Z-PhS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])SPh réagit avec des sels de CuX, deux structures différentes sont obtenues : le CP 2D [Cu[indice inférieur 4] ([mu][indice inférieur 3]-I) [indice inférieur 4] ([mu]-Z-PhS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])SPh}[indice inférieur 2]] [indice inférieur n] contenant des SBUs de type « cubane fermé » et le complexe 0D [Cu[indice inférieur 2]Br[indice inférieur 2]{[mu]-Z-PhS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])SPh}[indice inférieur 2]]. De par la réaction de E-pTolS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])S-pTol avec CuI, le CP 2D [{Cu([mu][indice inférieur 3]-I)} [indice inférieur 2] ([mu]-E-pTolS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])S-pTol)]n contenant des rubans parallèles en escalier est obtenu, alors que la structure issue de CuBr n’a pas pu être résolue. Finalement, quand CuX réagit avec Z-pTolS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])S-pTol, les CPs 2D iso-structuraux [Cu[indice inférieur 2]X[indice inférieur 2]{[mu]-Z-pTolS(CH[indice inférieur 2]CH=CHCH[indice inférieur 2])S-pTol}[indice inférieur 2]] (X = I, Br) sont formés. Dans ce cas, contrairement, aux premières structures obtenues, les couches de ces CPs sont composées de grilles incorporant des SBUs rhomboédriques Cu[indice inférieur 2] ([mu][indice inférieur 2]-X) [indice inférieur 2] dont les distances Cu···Cu sont identiques d’une couche à l’autre. Les caractérisations habituelles en photophysique et en stabilités thermiques ont été menées sur ces matériaux. De plus, des calculs théoriques ont été réalisés afin de mieux comprendre les propriétés photophysiques de ces composés. La quatrième section, i.e. le chapitre 5, traite des réactions de CuX (Br, I, Cl) sur des ligands dithioétherbutynes (1,4-bis(pTolthio)but-2-yne et 1,4-bis(benzylthio)but-2-yne. Quand CuBr réagit avec 1,4-bis(pTolthio)but-2-yne, le CP 1D [{Cu([mu][indice inférieur 2]-Br) [indice inférieur 2]Cu}([mu]-pTolSCH[indice inférieur 2]C≡CCH[indice inférieur 2]S-pTol) [indice inférieur 2]] [indice inférieur n] est obtenu, alors que le CP 2D [{Cu[indice inférieur 4] ([mu][indice inférieur 3]-I) [indice inférieur 4]}([mu]-pTolSCH[indice inférieur 2]C≡CCH[indice inférieur 2]S-pTol) [indice inférieur 2]] [indice inférieur n], préalablement reporté, est formé.(2) La réaction des sels CuI et CuCl avec 1,4-bis(benzylthio)but-2-yne engendre la formation de complexes isomorphes 0D [{Cu([mu][indice inférieur 2]-X) [indice inférieur 2]Cu}([mu]-PhCH[indice inférieur 2]SCH[indice inférieur 2]C≡CCH[indice inférieur 2]SCH[indice inférieur 2]Ph) [indice inférieur 2]] (X = I, Br). Contrairement à l’utilisation de CuCl, qui avec PhCH[indice inférieur 2]SCH[indice inférieur 2]C≡CCH[indice inférieur 2]SCH[indice inférieur 2]Ph forme le CP 2D [{Cu[indice inférieur 2] ([mu][indice inférieur 2]-Cl)([mu] [indice inférieur 3]-Cl)}([mu]-PhCH[indice inférieur 2]SCH[indice inférieur 2]C≡CCH[indice inférieur 2]SCH[indice inférieur 2]Ph)] [indice inférieur n]. Notons que ce CP présente des propriétés de photophysique peu communes pour un dérivé chloré, car il émet de la lumière autour de 600 nm. La cinquième section, i.e. le chapitre 6, traite des réactions de CuI avec PhS(CH[indice inférieur 2]) [indice inférieur 8]SPh et pTolS(CH[indice inférieur 2]) [indice inférieur 8]S-pTol qui génèrent respectivement les CPs luminescents 1D [Cu[indice inférieur 4]I[indice inférieur 4]{[mu][indice inférieur 2]-PhS(CH[indice inférieur 2]) [indice inférieur 8]SPh}[indice inférieur 2]] [indice inférieur n] et 2D [Cu8I8{[mu]2-pTolS(CH2)8S-pTol}3(MeCN)2]n. Le CP 2D [Cu8I8{[mu]2-pTolS(CH2)8S-pTol}3 (MeCN)2]n présente un réseau qui n’avait jamais été rencontré dans la littérature auparavant, c.-à-d., des couches de polymère construites à partir de deux cubanes fermés pontés ensemble par un rhomboèdre comme SBUs. Leurs propriétés physiques et de stabilités thermiques ont été étudiées et présentent quelques différences notables. La sixième section, i.e. le chapitre 7, traite des réactions entre CuI avec le ligand flexible pTolS(CH[indice inférieur 2]) [indice inférieur 8]S-pTol dans le MeCN ou EtCN et p-[indice supérieur t]BuC[indice inférieur 6]H[indice inférieur 4]S(CH[indice inférieur 2]) [indice inférieur 8]SC[indice inférieur 6]H[indice inférieur 4]-p-[indice supérieur t]Bu L2 dans EtCN. Les synthèses issues de pTolS(CH[indice inférieur 2]) [indice inférieur 8]S-pTol permettent l’obtention de CPs 2D [Cu[indice inférieur 8]I[indice inférieur 8]{pTolS(CH[indice inférieur 2]) [indice inférieur 8]S-pTol}[indice inférieur 3] (solvant) [indice inférieur 2]] [indice inférieur n] (1•MeCN et 1•EtCN) contenant des nœuds de connexion de type Cu[indice inférieur 8]I[indice inférieur 8]. Par opposition, l’utilisation du ligand p-[indice supérieur t]BuC[indice inférieur 6]H[indice inférieur 4]S(CH[indice inférieur 2]) [indice inférieur 8]SC[indice inférieur 6]H[indice inférieur 4]-p-[indice supérieur t]Bu dans EtCN entraine la formation d’un CP 1D [Cu[indice inférieur 4]I[indice inférieur 4]{p-[indice supérieur t]BuC[indice inférieur 6]H[indice inférieur 4]S(CH[indice inférieur 2]) [indice inférieur 8]SC[indice inférieur 6]H[indice inférieur 4]-p-[indice supérieur t]Bu}[indice inférieur 2] (EtCN) [indice inférieur 2]] [indice inférieur n] incorporant les SBUs de type cubane fermé. Les CPs 2D 1•MeCN et 1•EtCN, contrairement à 2•EtCN, présentent l’habilité de pouvoir perdre le solvant initialement incorporé dans leur structure sous vide et de le readsorber ou d’adsorber un autre solvant, chose qui peut être suivie à l’aide de la variation de la luminescence, la stabilité thermique, ou encore par diffraction des rayons X sur poudre. La septième section, i.e. le chapitre 8 traite des réactions, une fois encore, entre un ligand dithioéther, contenant un pont flexible butane (EtS(CH[indice inférieur 2]) [indice inférieur 4]SEt) et les sels CuX (X = I, Br). Dans ce cas, il se forme avec le CuI un CP luminescent 2D [Cu[indice inférieur 4]I[indice inférieur 4]{[mu]-EtS(CH[indice inférieur 2]) [indice inférieur 4]SEt}[indice inférieur 2]] [indice inférieur n], alors qu’il génère avec CuBr, le CP 3D [(Cu[indice inférieur 2]Br[indice inférieur 2]){[mu]-EtS(CH[indice inférieur 2]) [indice inférieur 4]SEt}][indice inférieur n] faiblement luminescent, construit sur des couches en parallèle pontées par les soufres doublement n-donneurs. Il est intéressant de remarquer qu’une migration de l’énergie d’excitation se produit dans le CP 3D (dérivé bromé) contrairement au CP 2D (dérivé iodé) sous excitation de haute intensité. Très peu d’exemples présentent ce type de processus parmi tous les CPs ( < 10). Pour conclure, les réactions entre les sels CuX (X = Cl, Br, I) avec des ligands thioéthers de types différents (mono-, di-thio, rigide ou flexible) peuvent offrir des matériaux de structures variables (CP 0D, 1D, 2D, 3D avec et sans cavités) présentant leurs propres spécificités (luminescence, stabilité thermique, adsorption de gaz, solvatochromisme, etc.). Le résultat le plus important à noter, en comparaison avec la littérature est, qu’il est très difficile, voire impossible, de pouvoir prédire la dimensionnalité, la structure et les propriétés dont résultera la coordination des ligands thioéthers sur des sels de CuX (X = Cl, Br, I). Par conséquent, de nombreuses combinaisons et études restent encore à être menées pour mieux comprendre ces matériaux et trouver la, ou les meilleures combinaisons possibles pour concevoir des MOFs luminescents à partir de CuX.

This thesis presents the achievements and scientific work conducted using a previously designed and fabricated 64 x 64-pixel ion camera with the use of a 0.35 μm CMOS technology. We used an array of Ion Sensitive Field Effect Transistors (ISFETs) to monitor and measure chemical and biochemical reactions in real time. The area of our observation was a 4.2 x 4.3 mm silicon chip while the actual ISFET array covered an area of 715.8 x 715.8 μm consisting of 4096 ISFET pixels in total with a 1 μm separation space among them. The ion sensitive layer, the locus where all reactions took place was a silicon nitride layer, the final top layer of the austriamicrosystems 0.35 μm CMOS technology used. Our final measurements presented an average sensitivity of 30 mV/pH. With the addition of extra layers we were able to monitor a 65 mV voltage difference during our experiments with glucose and hexokinase, whereas a difference of 85 mV was detected for a similar glucose reaction mentioned in literature, and a 55 mV voltage difference while performing photosynthesis experiments with a biofilm made from cyanobacteria, whereas a voltage difference of 33.7 mV was detected as presented in literature for a similar cyanobacterial species using voltamemtric methods for detection. To monitor our experiments PXIe-6358 measurement cards were used and measurements were controlled by LabVIEW software. The chip was packaged and encapsulated using a PGA-100 chip carrier and a two-component commercial epoxy. Printed circuit board (PCB) has also been previously designed to provide interface between the chip and the measurement cards.

A presença de plantas daninhas em plantios de eucalipto, especialmente nos dois primeiros anos, pode acarretar grandes prejuízos à produtividade, pois reduz a eficiência de aproveitamento dos recursos de crescimento pela cultura. Assim, objetivou-se com este trabalho avaliar os efeitos da interferência de plantas daninhas sobre o crescimento inicial de dois clones de Eucalyptus urophylla x Eucalyptus grandis e a concentração foliar de nutrientes na cultura e nas plantas daninhas. O experimento foi instalado em esquema fatorial 2 x 5 + 7, sendo dois clones de híbridos de Eucalyptus urophylla x Eucalyptus grandis, identificados como CNB001 e CNB016, em competição com cinco plantas daninhas Urochloa decumbens (capim-braquiária), Ipomoea nil (corda-de-viola), Commelina diffusa (trapoeraba), Spermacoce latifolia (erva-quente) e Panicum maximum (capim-colonião). Adicionalmente, foram cultivados os dois clones de eucalipto e as cinco plantas daninhas em monocultivo como padrão de comparação, no delineamento inteiramente casualizado, com quatro repetições. Foram avaliados eucalipto através da altura de plantas, o diâmetro do coleto, o número de ramos, a área foliar, a matéria seca e, o teor foliar de nutrientes do eucalipto, bem como o teor de nutrientes nas folhas das plantas daninhas. O clone CNB001 apresentou crescimento inicial superior ao clone CNB016, no entanto, livre da interferência de plantas daninhas, verificaram-se teores foliares semelhantes para a maioria dos nutrientes em ambos os genótipos. O clone CNB016 mostrou maior sensibilidade à interferência negativa das plantas daninhas que o clone CNB001, sendo seu crescimento inicial mais afetado por Ipomoea nil e a concentração de nutrientes reduzida pelas espécies Panicum maximum, Urochloa decumbens e Commelina diffusa. Panicum maximum apresentou maior interferência com o clone CNB001, enquanto Ipomoea nil pouco influenciou o crescimento e o teor de nutrientes deste híbrido. As plantas daninhas apresentaram elevada capacidade de extrair nutrientes do solo, mesmo em convivência com os clones de Eucalyptus urophylla x Eucalyptus grandis. A interferência imposta à cultura é dependente da espécie infestante e do genótipo de eucalipto.

Este trabalho teve como objetivo avaliar o efeito de indutores de florescimento em três cultivares de abacaxizeiro em Rio Branco-Acre. O delineamento experimental foi em blocos ao acaso, em esquema fatorial 3 x 2 x 2, com quatro repetições, sendo combinadas três cultivares de abacaxi (RBR-1, SNG-2 e SNG-3), dois indutores de florescimento (carbureto de cálcio-CaC2 e etefon-ácido 2-cloroetilfosfônico) e duas épocas de aplicação (aos 10 e 12 meses do plantio). As plantas induzidas com etefon aos 10 meses apresentaram maior percentagem de florescimento (96,25%) quando com CaC2 (85,42%) quando comparadas com as induzidas aos 10 meses (62,92%).

A study was conducted to examine the factorial validity of the Flinders Decision Making Questionnaire (Mann, 1982), a 31-item self-report inventory designed to measure tendencies to use three major coping patterns identified in the conflict theory of decision making (Janis and Mann, 1977): vigilance, hypervigilance, and defensive avoidance (procrastination, buck-passing, and rationalization). A sample of 2051 university students, comprising samples from Australia (n=262), New Zealand (n=260), the USA (n=475), Japan (n=359), Hong Kong (n=281) and Taiwan (n=414) was administered the DMQ. Factorial validity of the instrument was tested by confirmatory factor analysis with LISREL. Five different substantive models, representing different structural relationships between the decision-coping patterns had unsatisfactory fit to the data and could not be validated. A shortened instrument, containing 22 items, yielded a revised model comprising four identifiable factors-vigilance, hypervigilance, buck-passing, and procrastination. The revised model had adequate fit with data for each country sample and for the total sample, and was confirmed. It is recommended that the 22-item instrument, named the Melbourne DMQ, replace the Flinders DMQ for measurement of decision-coping patterns.

Bananas are susceptible to a diverse range of biotic and abiotic stresses, many of which cause serious production constraints worldwide. One of the most destructive banana diseases is Fusarium wilt caused by the soil-borne fungus, Fusarium oxysporum f. sp. cubense (Foc). No effective control strategy currently exists for this disease which threatens global banana production. Although disease resistance exists in some wild bananas, attempts to introduce resistance into commercially acceptable bananas by conventional breeding have been hampered by low fertility, long generation times and association of poor agronomical traits with resistance genes. With the advent of reliable banana transformation protocols, molecular breeding is now regarded as a viable alternative strategy to generate disease-resistant banana plants. Recently, a novel strategy involving the expression of anti-apoptosis genes in plants was shown to result in resistance against several necrotrophic fungi. Further, the transgenic plants showed increased resistance to a range of abiotic stresses. In this thesis, the use of anti-apoptosis genes to generate transgenic banana plants with resistance to Fusarium wilt was investigated. Since water stress is an important abiotic constraint to banana production, the resistance of the transgenic plants to water stress was also examined. Embryogenic cell suspensions (ECS) of two commercially important banana cultivars, Grand Naine (GN) and Lady Finger (LF), were transformed using Agrobacterium with the anti-apoptosis genes, Bcl-xL, Bcl-xL G138A, Ced-9 and Bcl- 2 3’ UTR. An interesting, and potentially important, outcome was that the use of anti-apoptosis genes resulted in up to a 50-fold increase in Agrobacterium-mediated transformation efficiency of both LF and GN cells over vector controls. Regenerated plants were subjected to a complete molecular characterisation in order to detect the presence of the transgene (PCR), transcript (RT-PCR) and gene product (Western blot) and to determine the gene copy number (Southern blot). A total of 36 independently-transformed GN lines (8 x Bcl-xL, 5 x Bcl-xL G138A, 15 x Ced-9 and 8 x Bcl-2 3’ UTR) and 41 independently-transformed LF lines (8 x Bcl-xL, 7 x BclxL G138A, 13 x Ced-9 and 13 x Bcl-2 3’ UTR) were identified. The 41 transgenic LF lines were multiplied and clones from each line were acclimatised and grown under glasshouse conditions for 8 weeks to allow monitoring for phenotypic abnormalities. Plants derived from 3 x Bcl-xL, 2 x Ced-9 and 5 x Bcl-2 3’ UTR lines displayed a variety of aberrant phenotypes. However, all but one of these abnormalities were off-types commonly observed in tissue-cultured, non-transgenic banana plants and were therefore unlikely to be transgene-related. Prior to determining the resistance of the transgenic plants to Foc race 1, the apoptotic effects of the fungus on both wild-type and Bcl-2 3’ UTR-transgenic LF banana cells were investigated using rapid in vitro root assays. The results from these assays showed that apoptotic-like cell death was elicited in wild-type banana root cells as early as 6 hours post-exposure to fungal spores. In contrast, these effects were attenuated in the root cells of Bcl-2 3’ UTR-transgenic lines that were exposed to fungal spores. Thirty eight of the 41 transgenic LF lines were subsequently assessed for resistance to Foc race 1 in small-plant glasshouse bioassays. To overcome inconsistencies in rating the internal (vascular discolouration) disease symptoms, a MatLab-based computer program was developed to accurately and reliably assess the level of vascular discolouration in banana corms. Of the transgenic LF banana lines challenged with Foc race 1, 2 x Bcl-xL, 3 x Ced-9, 2 x Bcl-2 3’ UTR and 1 x Bcl-xL G138A-transgenic line were found to show significantly less external and internal symptoms than wild-type LF banana plants used as susceptible controls at 12 weeks post-inoculation. Of these lines, Bcl-2 3’ UTR-transgenic line #6 appeared most resistant, displaying very mild symptoms similar to the wild-type Cavendish banana plants that were included as resistant controls. This line remained resistant for up to 23 weeks post-inoculation. Since anti-apoptosis genes have been shown to confer resistance to various abiotic stresses in other crops, the ability of these genes to confer resistance against water stress in banana was also investigated. Clonal plants derived from each of the 38 transgenic LF banana plants were subjected to water stress for a total of 32 days. Several different lines of transgenic plants transformed with either Bcl-xL, Bcl-xL G138A, Ced-9 or Bcl-2 3’ UTR showed a delay in visual water stress symptoms compared with the wild-type control plants. These plants all began producing new growth from the pseudostem following daily rewatering for one month. In an attempt to determine whether the protective effect of anti-apoptosis genes in transgenic banana plants was linked with reactive oxygen species (ROS)-associated programmed cell death (PCD), the effect of the chloroplast-targeting, ROS-inducing herbicide, Paraquat, on wild-type and transgenic LF was investigated. When leaf discs from wild-type LF banana plants were exposed to 10 ìM Paraquat, complete decolourisation occurred after 48 hours which was confirmed to be associated with cell death and ROS production by trypan blue and 3,3-diaminobenzidine (DAB) staining, respectively. When leaf discs from the transgenic lines were exposed to Paraquat, those derived from some lines showed a delay in decolourisation, suggesting only a weak protective effect from the transgenes. Finally, the protective effect of anti-apoptosis genes against juglone, a ROS-inducing phytotoxin produced by the causal agent of black Sigatoka, Mycosphaerella fijiensis, was investigated. When leaf discs from wild-type LF banana plants were exposed to 25 ppm juglone, complete decolourisation occurred after 48 hours which was again confirmed to be associated with cell death and ROS production by trypan blue and DAB staining, respectively. Further, TdT-mediated dUTP nick-end labelling (TUNEL) assays on these discs suggested that the cell death was apoptotic. When leaf discs from the transgenic lines were exposed to juglone, discs from some lines showed a clear delay in decolourisation, suggesting a protective effect. Whether these plants are resistant to black Sigatoka is unknown and will require future glasshouse and field trials. The work presented in this thesis provides the first report of the use of anti-apoptosis genes as a strategy to confer resistance to Fusarium wilt and water stress in a nongraminaceous monocot, banana. Such a strategy may be exploited to generate resistance to necrotrophic pathogens and abiotic stresses in other economically important crop plants.

The Raman and infrared spectrum of the antimonate mineral stibiconite Sb3+Sb5+2O6(OH) were used to define aspects of the molecular structure of the mineral. Bands attributable to water, OH stretching and bending and SbO stretching and bending were assigned. The mineral has been shown to contain both calcium and water and the formula is probably best written (Sb3+,Ca)ySb5+2-x(O,OH,H2O)6-7 where y approaches 1 and x varies from 0 to 1. Infrared spectroscopy complimented with thermogravimetric analysis proves the presence of water in the stibiconite structure. The mineral stibiconite is formed through replacement of the sulphur in stibnite. No Raman or infrared bands attributable to stibnite were identified in the spectra.

The objective of this investigation was to compare the acute effects of exercise and diet manipulations on energy intake, between dietary restrained and unrestrained females. Comparisons of two studies using an identical 2 x 2 repeated-measures design (level of activity (rest or exercise) and lunch type (high-fat or low-fat)) including thirteen dietary unrestrained and twelve restrained females were performed. Energy expenditure during the rest session was estimated and the energy cost of exercise was measured by indirect calorimetry. Relative energy intake was calculated by subtracting the energy expenditure of the exercise session from the energy intake of the test meal. Post-meal hedonic ratings were completed after lunch. Energy intake and relative energy intake increased during high-fat conditions compared with the low-fat, independently of exercise (P < 0.001). There was a positive relationship between dietary restraint scores and energy intake or relative energy intake in the rest conditions only (r 0.54, P < 0.01). The decrease of relative energy intake between the rest and exercise conditions was higher in restrained than in unrestrained eaters (P < 0.01). These results confirm that a high-fat diet reversed the energy deficit due to exercise. There was no energy compensation in response to an acute bout of exercise during the following meal. In restrained eaters, exercise was more effective in creating an energy deficit than in unrestrained eaters. Exercise may help restrained eaters to maintain control over appetite.

Fusarium wilt, caused by Fusarium oxysporum f. sp. cubense (Foc), is one of the most devastating diseases of banana (Musa spp.). Apart from resistant cultivars, there are no effective control measures for the disease. We investigated whether the transgenic expression of apoptosis-inhibition related genes in banana could be used to confer disease resistance. Embryogenic cell suspensions of the banana cultivar, ‘Lady Finger’, were stably transformed with animal genes that negatively regulate apoptosis, namely Bcl-xL, Ced-9 and Bcl-2 3’ UTR, and independently transformed plant lines were regenerated for testing. Following a 12 week exposure to Foc race 1 in small-plant glasshouse bioassays, seven transgenic lines (2 x Bcl-xL, 3 x Ced-9 and 2 x Bcl-2 3’ UTR) showed significantly less internal and external disease symptoms than the wild-type susceptible ‘Lady Finger’ banana plants used as positive controls. Of these, one Bcl-2 3’ UTR line showed resistance that was equivalent to that of wild-type Cavendish bananas that were included as resistant negative controls. Further, the resistance of this line continued for 23 weeks post-inoculation at which time the experiment was terminated. Using TUNEL assays, Foc race 1 was shown to induce apoptosis-like features in the roots of wild-type ‘Lady Finger’ plants consistent with a necrotrophic phase in the lifecycle of this pathogen. This was further supported by the observed reduction of these effects in the roots of the resistant Bcl-2 3’ UTR transgenic line. This is the first report on the generation of transgenic banana plants with resistance to Fusarium wilt.

In order to support intelligent transportation system (ITS) road safety applications such as collision avoidance, lane departure warnings and lane keeping, Global Navigation Satellite Systems (GNSS) based vehicle positioning system has to provide lane-level (0.5 to 1 m) or even in-lane-level (0.1 to 0.3 m) accurate and reliable positioning information to vehicle users. However, current vehicle navigation systems equipped with a single frequency GPS receiver can only provide road-level accuracy at 5-10 meters. The positioning accuracy can be improved to sub-meter or higher with the augmented GNSS techniques such as Real Time Kinematic (RTK) and Precise Point Positioning (PPP) which have been traditionally used in land surveying and or in slowly moving environment. In these techniques, GNSS corrections data generated from a local or regional or global network of GNSS ground stations are broadcast to the users via various communication data links, mostly 3G cellular networks and communication satellites. This research aimed to investigate the precise positioning system performances when operating in the high mobility environments. This involves evaluation of the performances of both RTK and PPP techniques using: i) the state-of-art dual frequency GPS receiver; and ii) low-cost single frequency GNSS receiver. Additionally, this research evaluates the effectiveness of several operational strategies in reducing the load on data communication networks due to correction data transmission, which may be problematic for the future wide-area ITS services deployment. These strategies include the use of different data transmission protocols, different correction data format standards, and correction data transmission at the less-frequent interval. A series of field experiments were designed and conducted for each research task. Firstly, the performances of RTK and PPP techniques were evaluated in both static and kinematic (highway with speed exceed 80km) experiments. RTK solutions achieved the RMS precision of 0.09 to 0.2 meter accuracy in static and 0.2 to 0.3 meter in kinematic tests, while PPP reported 0.5 to 1.5 meters in static and 1 to 1.8 meter in kinematic tests by using the RTKlib software. These RMS precision values could be further improved if the better RTK and PPP algorithms are adopted. The tests results also showed that RTK may be more suitable in the lane-level accuracy vehicle positioning. The professional grade (dual frequency) and mass-market grade (single frequency) GNSS receivers were tested for their performance using RTK in static and kinematic modes. The analysis has shown that mass-market grade receivers provide the good solution continuity, although the overall positioning accuracy is worse than the professional grade receivers. In an attempt to reduce the load on data communication network, we firstly evaluate the use of different correction data format standards, namely RTCM version 2.x and RTCM version 3.0 format. A 24 hours transmission test was conducted to compare the network throughput. The results have shown that 66% of network throughput reduction can be achieved by using the newer RTCM version 3.0, comparing to the older RTCM version 2.x format. Secondly, experiments were conducted to examine the use of two data transmission protocols, TCP and UDP, for correction data transmission through the Telstra 3G cellular network. The performance of each transmission method was analysed in terms of packet transmission latency, packet dropout, packet throughput, packet retransmission rate etc. The overall network throughput and latency of UDP data transmission are 76.5% and 83.6% of TCP data transmission, while the overall accuracy of positioning solutions remains in the same level. Additionally, due to the nature of UDP transmission, it is also found that 0.17% of UDP packets were lost during the kinematic tests, but this loss doesn't lead to significant reduction of the quality of positioning results. The experimental results from the static and the kinematic field tests have also shown that the mobile network communication may be blocked for a couple of seconds, but the positioning solutions can be kept at the required accuracy level by setting of the Age of Differential. Finally, we investigate the effects of using less-frequent correction data (transmitted at 1, 5, 10, 15, 20, 30 and 60 seconds interval) on the precise positioning system. As the time interval increasing, the percentage of ambiguity fixed solutions gradually decreases, while the positioning error increases from 0.1 to 0.5 meter. The results showed the position accuracy could still be kept at the in-lane-level (0.1 to 0.3 m) when using up to 20 seconds interval correction data transmission.

Even though titanium dioxide photocatalysis has been promoted as a leading green technology for water purification, many issues have hindered its application on a large commercial scale. For the materials scientist the main issues have centred the synthesis of more efficient materials and the investigation of degradation mechanisms; whereas for the engineers the main issues have been the development of appropriate models and the evaluation of intrinsic kinetics parameters that allow the scale up or re-design of efficient large-scale photocatalytic reactors. In order to obtain intrinsic kinetics parameters the reaction must be analysed and modelled considering the influence of the radiation field, pollutant concentrations and fluid dynamics. In this way, the obtained kinetic parameters are independent of the reactor size and configuration and can be subsequently used for scale-up purposes or for the development of entirely new reactor designs. This work investigates the intrinsic kinetics of phenol degradation over titania film due to the practicality of a fixed film configuration over a slurry. A flat plate reactor was designed in order to be able to control reaction parameters that include the UV irradiance, flow rates, pollutant concentration and temperature. Particular attention was paid to the investigation of the radiation field over the reactive surface and to the issue of mass transfer limited reactions. The ability of different emission models to describe the radiation field was investigated and compared to actinometric measurements. The RAD-LSI model was found to give the best predictions over the conditions tested. Mass transfer issues often limit fixed film reactors. The influence of this phenomenon was investigated with specifically planned sets of benzoic acid experiments and with the adoption of the stagnant film model. The phenol mass transfer coefficient in the system was calculated to be km,phenol=8.5815x10-7Re0.65(ms-1). The data obtained from a wide range of experimental conditions, together with an appropriate model of the system, has enabled determination of intrinsic kinetic parameters. The experiments were performed in four different irradiation levels (70.7, 57.9, 37.1 and 20.4 W m-2) and combined with three different initial phenol concentrations (20, 40 and 80 ppm) to give a wide range of final pollutant conversions (from 22% to 85%). The simple model adopted was able to fit the wide range of conditions with only four kinetic parameters; two reaction rate constants (one for phenol and one for the family of intermediates) and their corresponding adsorption constants. The intrinsic kinetic parameters values were defined as kph = 0.5226 mmol m-1 s-1 W-1, kI = 0.120 mmol m-1 s-1 W-1, Kph = 8.5 x 10-4 m3 mmol-1 and KI = 2.2 x 10-3 m3 mmol-1. The flat plate reactor allowed the investigation of the reaction under two different light configurations; liquid and substrate side illumination. The latter of particular interest for real world applications where light absorption due to turbidity and pollutants contained in the water stream to be treated could represent a significant issue. The two light configurations allowed the investigation of the effects of film thickness and the determination of the catalyst optimal thickness. The experimental investigation confirmed the predictions of a porous medium model developed to investigate the influence of diffusion, advection and photocatalytic phenomena inside the porous titania film, with the optimal thickness value individuated at 5 ìm. The model used the intrinsic kinetic parameters obtained from the flat plate reactor to predict the influence of thickness and transport phenomena on the final observed phenol conversion without using any correction factor; the excellent match between predictions and experimental results provided further proof of the quality of the parameters obtained with the proposed method.

We examined the effects of progressive resistance training (PRT) and supplementation with calcium-vitamin D(3) fortified milk on markers of systemic inflammation, and the relationship between inflammation and changes in muscle mass, size and strength. Healthy men aged 50-79 years (n = 180) participated in this 18-month randomized controlled trial that comprised a factorial 2 x 2 design. Participants were randomized to (1) PRT + fortified milk supplement, (2) PRT, (3) fortified milk supplement, or (4) a control group. Participants assigned to PRT trained 3 days per week, while those in the supplement groups consumed 400 ml day(-1) of milk containing 1,000 mg calcium plus 800 IU vitamin D(3). We collected venous blood samples at baseline, 12 and 18 months to measure the serum concentrations of IL-6, TNF-alpha and hs-CRP. There were no exercise x supplement interactions, but serum IL-6 was 29% lower (95% CI, -62, 0) in the PRT group compared with the control group after 12 months. Conversely, IL-6 was 31% higher (95% CI, -2, 65) in the supplement group compared with the non-supplemented groups after 12 and 18 months. These between-group differences did not persist after adjusting for changes in fat mass. In the PRT group, mid-tibia muscle cross-sectional area increased less in men with higher pre-training inflammation compared with those men with lower inflammation (net difference similar to 2.5%, p < 0.05). In conclusion, serum IL-6 concentration decreased following PRT, whereas it increased after supplementation with fortified milk concomitant with changes in fat mass. Furthermore, low-grade inflammation at baseline restricted muscle hypertrophy following PRT.

Video-based training combined with flotation tank recovery may provide an additional stimulus for improving shooting in basketball. A pre-post controlled trial was conducted to assess the effectiveness of a 3 wk intervention combining video-based training and flotation tank recovery on three-point shooting performance in elite female basketball players. Players were assigned to an experimental (n=10) and control group (n=9). A 3 wk intervention consisted of 2 x 30 min float sessions a week which included 10 min of video-based training footage, followed by a 3 wk retention phase. A total of 100 three-point shots were taken from 5 designated positions on the court at each week to assess three-point shooting performance. There was no clear difference in the mean change in the number of successful three-point shots between the groups (-3%; ±18%, mean; ±90% confidence limits). Video-based training combined with flotation recovery had little effect on three-point shooting performance.

Researchers examined the sun-protective intentions and behavior of young, Caucasian, Australian sportswomen aged between 17 and 35 years (N = 100). The study adopted a 2 x 2 experimental design, comparing group norms (supportive vs. non-supportive) and image norms (tanned vs. pale) related to sun protection and taking into account group identification with friends and peers in the sport. While no significant findings emerged involving image norms, regression analyses revealed a significant two-way interaction for group norm x identification on recreational sportswomen's intentions to engage in sun protection in the next fortnight. Participants identifying strongly with their group had stronger intentions to engage in sun protection when exposed to a norm reflecting fellow recreational sportswomen engaging in sun-protective actions in comparison to those exposed to a non-supportive group. In addition, while prior intentions to engage in sun protection were not significantly related to sun-protection behavior, post-manipulation intentions after exposure to the sun-protective information that was provided were significantly related to follow-up behavior. Overall, the findings supported the importance of group-based social influences, rather than tanned media images, on sun-protective decisions among young recreational sportswomen and provided a targeted source for intervention strategies encouraging sun safety among this at-risk group for repeated sun exposure.
