Glial cell line-derived neurotrophic factor (GDNF) promotes survival of midbrain dopaminergic neurons and motoneurons. Expression of GDNF mRNA in cerebellum raises the possibility that cells within this structure might also respond to GDNF. To examine potential trophic activities of GDNF, dissociated cultures of gestational day 18 rat cerebellum were grown for < or = 21 days in the presence of factor. GDNF increased Purkinje cell number without affecting the overall number of neurons or glial cells. A maximal response (50% above control) was elicited with GDNF at 1 pg/ml. Effects of GDNF on Purkinje cell differentiation were examined by scoring the morphologic maturation of cells in treated and control cultures. GDNF increased the proportion of Purkinje cells that displayed relatively mature morphologies, characterized by dendritic thickening and the development of spines and filopodial extensions. Morphologic maturation of the overall neuronal population was unaffected. In sum, our data indicate that GDNF is a potent survival and differentiation factor for Purkinje cells, the efferent neurons of cerebellar cortex. Together with its other actions, these findings raise the possibility that GDNF might be a critical trophic factor at multiple loci in neuronal circuits that control motor function.

We present a 5.3-Myr stack (the ''LR04'' stack) of benthic d18O records from 57 globally distributed sites aligned by an automated graphic correlation algorithm. This is the first benthic delta18O stack composed of more than three records to extend beyond 850 ka, and we use its improved signal quality to identify 24 new marine isotope stages in the early Pliocene. We also present a new LR04 age model for the Pliocene-Pleistocene derived from tuning the delta18O stack to a simple ice model based on 21 June insolation at 65 N. Stacked sedimentation rates provide additional age model constraints to prevent overtuning. Despite a conservative tuning strategy, the LR04 benthic stack exhibits significant coherency with insolation in the obliquity band throughout the entire 5.3 Myr and in the precession band for more than half of the record. The LR04 stack contains significantly more variance in benthic delta18O than previously published stacks of the late Pleistocene as the result of higher resolution records, a better alignment technique, and a greater percentage of records from the Atlantic. Finally, the relative phases of the stack's 41- and 23-kyr components suggest that the precession component of delta18O from 2.7-1.6 Ma is primarily a deep-water temperature signal and that the phase of d18O precession response changed suddenly at 1.6 Ma.

We report geomagnetic directional paleosecular variation, relative paleointensity proxies and oxygen isotope data from the upper 88 m composite depth (mcd) at South Atlantic Ocean Drilling Program (ODP) Site 1089 (40°56.2?S, 9°53.64?E, 4620 m water depth). The age model is provided by high-resolution oxygen isotope stratigraphy, augmented by radiocarbon dates from the upper 8 mcd of nearby piston core RC11-83. Mean sedimentation rates at Site 1089 are in the range of 15 to 20 cm/kyr. Two intervals during the Brunhes Chron, at ?29.6 mcd (?190 ka) and at ?48 mcd (?335 ka), have component magnetization directions with positive (reverse polarity) inclination; however, the excursional directions are heavily overprinted by the postexcursional field. Magnetite is the dominant carrier of magnetic remanence, and occurs in the pseudosingle-domain (PSD) grain size. An additional higher-coercivity magnetic carrier, characterized by low unblocking temperatures (<350°C), is assumed to be authigenic pyrrhotite. A decrease in magnetization intensity down core is mirrored by a reduction in pore water sulfate, indicating diagenetic reduction of magnetite. Despite down-core changes in magnetic mineralogy, normalized intensity records from Site 1089 are comparable with high-resolution paleointensity records from the North Atlantic (e.g., ODP Sites 983 and 984). Sediment properties and sedimentation patterns within the Cape (Site 1089) and Iceland (Sites 983 and 984) Basins are distinctly different at both millennial and orbital timescales and therefore preclude lithologic variability from being the source of this correlation. Variations in normalized intensity from Site 1089 therefore appear to reflect changes in global-scale geomagnetic field intensity.

Correlation of Southern Ocean deep sea sediment core records with ice core records of polar climate delineates with unprecedented detail the relationship between high latitude climate and the ocean's thermohaline circulation over the last 80,000 years. Our observations suggest that, while North Atlantic Deep Water variability manifests itself clearly in Southern Ocean nutrient proxy records over periods as short as 500 yr, this deep water variability did not promote a direct link between climate variability in the high latitudes of the two hemispheres on millennial timescales. In particular, the proxy records indicate that, on average, northern hemisphere climate fluctuations lagged those of the southern hemisphere by 1500 yr.

Visual counts of ice-rafted debris (IRD), foraminifera, and radiolaria were made for ~1500 samples in Site 1094 spanning the last four climatic cycles (marine isotope stages 1-11). Most, but not all, of the IRD variability is captured by whole-core physical properties including magnetic susceptibility and Q-ray attenuation bulk density. Glacial periods are marked by high IRD abundance and millennial-scale variability, which may reflect instability of ice shelves in the Weddell Sea region. Each interglacial period exhibits low IRD and high foraminiferal abundance during the early part of the interglacial, indicating relatively warm sea-surface temperatures and reduced influence of sea ice. IRD increases and foraminiferal abundances decrease during the latter part of each interglacial, indicating a return to more glacial-like conditions. Glacial terminations I and V are each characterized by a step-wise reduction in ice-rafting punctuated by a brief pulse in IRD delivery and reversal in delta18O. The coarse fraction of the sediment is dominated by ash and radiolaria, and the relative abundance of these components is remarkably similar to the concentration of Na+ in Vostok. Each of these variables is believed to be controlled mainly by sea-ice cover, thereby providing a means for sediment-ice core correlation.

The strength and geometry of the Atlantic meridional overturning circulation is tightly coupled to climate on glacial-interglacial and millennial timescales, but has proved difficult to reconstruct, particularly for the Last Glacial Maximum. Today, the return flow from the northern North Atlantic to lower latitudes associated with the Atlantic meridional overturning circulation reaches down to approximately 4,000 m. In contrast, during the Last Glacial Maximum this return flow is thought to have occurred primarily at shallower depths. Measurements of sedimentary 231Pa/230Th have been used to reconstruct the strength of circulation in the North Atlantic Ocean, but the effects of biogenic silica on 231Pa/230Th-based estimates remain controversial. Here we use measurements of 231Pa/230Th ratios and biogenic silica in Holocene-aged Atlantic sediments and simulations with a two-dimensional scavenging model to demonstrate that the geometry and strength of the Atlantic meridional overturning circulation are the primary controls of 231Pa/230Th ratios in modern Atlantic sediments. For the glacial maximum, a simulation of Atlantic overturning with a shallow, but vigorous circulation and bulk water transport at around 2,000 m depth best matched observed glacial Atlantic 231Pa/230Th values. We estimate that the transport of intermediate water during the Last Glacial Maximum was at least as strong as deep water transport today.
