The mouse Rad51 gene is a mammalian homologue of the Escherichia coli recA and yeast RAD51 genes, both of which are involved in homologous recombination and DNA repair. To elucidate the physiological role of RAD51 protein, the gene was targeted in embryonic stem (ES) cells. Mice heterozygous for the Rad51 null mutation were intercrossed and their offspring were genotyped. There were no homozygous (Rad51-/-) pups among 148 neonates examined but a few Rad51-/- embryos were identified when examined during the early stages of embryonic development. Doubly knocked-out ES cells were not detected under conditions of selective growth. These results are interpreted to mean that RAD51 protein plays an essential role in the proliferation of cell. The homozygous Rad51 null mutation can be categorized in cell-autonomous defects. Pre-implantational lethal mutations that disrupt basic molecular functions will thus interfere with cell viability.

The scl gene encodes a basic-helix-loop-helix transcription factor which was identified through its involvement in chromosomal translocations in T-cell leukemia. To elucidate its physiological role, scl was targeted in embryonic stem cells. Mice heterozygous for the scl null mutation were intercrossed and their offspring were genotyped. Homozygous mutant (scl-/-) pups were not detected in newborn litters, and analysis at earlier time points demonstrated that scl-/- embryos were dying around embryonic day 9.5. The scl-/- embryos were pale, edematous, and markedly growth retarded after embryonic day 8.75. Histological studies showed complete absence of recognizable hematopoiesis in the yolk sac of these embryos. Early organogenesis appeared to be otherwise normal. Culture of yolk sac cells of wild-type, heterozygous, and homozygous littermates confirmed the absence of hematopoietic cells in scl-/- yolk sacs. Reverse transcription PCR was used to examine the transcripts of several genes implicated in early hematopoiesis. Transcripts of GATA-1 and PU.1 transcription factors were absent from RNA from scl-/- yolk sacs and embryos. These results implicate scl as a crucial regulator of early hematopoiesis.

En esta comunicación se presentan los estudios previos y las intervenciones realizadas en la iglesia de San Sebastián de Orihuela, Alicante (España) con el fin de subsanar los problemas estructurales que presentaba, con múltiples grietas en arcos y bóvedas y eliminar la humedad presente en sus fábricas, además de las sales aportadas por el agua. En el estudio histórico y arquitectónico se ha comprobado que la iglesia se construyó sobre una ermita gótica y se finalizó en 1743 pero ha sufrido numerosas intervenciones. Es una iglesia de nave única con capillas laterales cubierta mediante bóvedas. Su fachada y las pilastras interiores son de piedra vista. Para el diagnóstico se han empleado diversas técnicas (ultrasonidos, higrómetro…) que han permitido deducir las causas de las lesiones. Las intervenciones se han dirigido a reforzar la estructura mediante cosidos e inyecciones, así como a ventilar la base mediante un forjado sanitario. También se ha saneado la piedra vista, tanto en interior como en fachada y se han sacado a la luz los arcos de piedra ocultos bajo un estucado. También se han restaurado los restos arqueológicos de época gótica.
