Treatment of of (R,R)-N,N-salicylidene cyclohexane 1,2-diamine(H(2)L(1)) in methanol with aqueous NH(4)VO(3) solution in perchloric acid medium affords the mononuclear oxovanadium(V) complex [VOL(1)(MeOH)]-ClO(4) (1) as deep blue solid while the treatment of same solution of (R,R)-N,N-salicylidene cyclohexane 1,2-diamine(H(2)L(1)) with aqueous solution of VOSO(4) leads to the formation of di-(mu-oxo) bridged vanadium(V) complex [VO(2)L(2)](2) (2) as green solid where HL(2) = (R,R)-N-salicylidene cyclohexane 1,2-diamine. The ligand HL(2) is generated in situ by the hydrolysis of one of the imine bonds of HL(1) ligand during the course of formation of complex [VO(2)L(2)](2) (2). Both the compounds have been characterized by single crystal X-ray diffraction as well as spectroscopic methods. Compounds 1 and 2 are to act as catalyst for the catalytic bromide oxidation and C-H bond oxidation in presence of hydrogen peroxide. The representative substrates 2,4-dimethoxy benzoic acid and para-hydroxy benzoic acids are brominated in presence of H(2)O(2) and KBr in acid medium using the above compounds as catalyst. The complexes are also used as catalyst for C-H bond activation of the representative hydrocarbons toluene, ethylbenzene and cyclohexane where hydrogen peroxide acts as terminal oxidant. The yield percentage and turnover number are also quite good for the above catalytic reaction. The oxidized products of hydrocarbons have been characterized by GC Analysis while the brominated products have been characterized by (1)H NMR spectroscopic studies.

Since first reported in 2005, mononuclear ruthenium water oxidation catalysts have attracted a great deal of attention due to their catalytic performance and synthetic flexibility. In particular, ligands coordinated to a Ru metal centre play an important role in the catalytic mechanisms, exhibiting significant impact on catalyst efficiency, stability and activity towards water oxidation. This review focuses on finding possible correlations between the ligand effects and activity of mononuclear Ru aqua and non-aqua complexes as water oxidation catalysts. The ligand effects highlighted in the text include the electronic nature of core ligands and their substituents, the trans–cis effect, steric hindrance and the strain effect, the net charge effect, the geometric arrangement of the aqua ligand and the supramolecular effects, e.g., hydrogen bonding and influence of a pendant base. The outcome is not always obvious at the present knowledge level. Deeper understanding of the ligand effects, based on new input data, is mandatory for further progress towards a rational development of novel catalysts featuring enhanced activity in water oxidation.

Aims: The ATP-binding cassette transporters, ABCA1 and ABCG1, are LXR-target genes that play an important role in reverse cholesterol transport. We examined the effects of inhibitors of the cholesterol absorption (ezetimibe) and synthesis (statins) on expression of these transporters in HepG2 cells and peripheral blood mononuclear cells (PBMCs) of individuals with primary (and nonfamilial) hypercholesterolemia (HC). Materials & methods: A total of 48 HC individuals were treated with atorvastatin (10 mg/day/4 weeks) and 23 were treated with ezetimibe (10 mg/day/4 weeks), followed by simvastatin (10 mg/day/8 weeks) and simvastatin plus ezetimibe (10 mg of each/day/4 weeks). Gene expression was examined in statin- or ezetimibe-treated and control HepG2 cells as well as PBMCs using real-time PCR. Results: In PBMCs, statins and ezetimibe downregulated ABCA1 and ABCG1 mRNA expression but did not modulate NR1H2 (LxR-beta) and NR1H3 (LXR-alpha) levels. Positive correlations of ABCA1 with ABCG1 and of NR1H2 with NR1H3 expressions were found in all phases of the treatments. In HepG2 cells, ABCA1 mRNA levels remained unaltered while ABCG1 expression was increased by statin (1.0-10.0 mu M) or ezetimibe (5.0 mu M) treatments. Atorvastatin upregulated NR1H2 and NR1H3 only at 10.0 mu M, meanwhile ezetimibe (1.0-5.0 mu M) downregulated NR1H2 but did not change NR1H3 expression. Conclusion: Our findings reveal that lipid-lowering drugs downregulate ABCA1 and ABCG1 mRNA expression in PBMCs of HC individuals and exhibit differential effects on HepG2 cells. Moreover, they indicate that the ABCA1 and ABCG1 transcript levels were not correlated directly to LXR mRNA expression in both cell models treated with lipid-lowering drugs.

Introduction: TLR-4 has also been identified as a receptor for endogenous alarmins, which are increased post transplantation. TLR-4 has also been associated with a polymorphism that could impact graft outcome. Objective: To assess the expression of TLR-4 in kidney transplant patients carrying or not a polymorphism. Methods: TLR-4 polymorphism (A299G/T399I) was studied in 200 renal transplant patients. Healthy volunteers were also enrolled as control group. The polymorphism analysis was performed using restriction enzymes technique (RFLP). Functionality of TLR-4 polymorphism was assessed in samples from controls by quantification of TNF-alpha after LPS stimulus. TLR-4 and -2 expressions were also analyzed by flow cytometry. Results: TLR-4 polymorphism was present in 8.5% of renal transplant patients. This polymorphism was associated with impairment in TNF-alpha secretion. In general, in renal transplant patients, TLR-4 expression in monocytes and in neutrophils was lower than in health volunteers. TLR-2 and TLR-4 expressions in healthy volunteers with A299G/T399I TLR-4 polymorphism was higher than in wild-type genotype healthy volunteers (p<0.01 and p<0.05, respectively), and also higher than A299G/T399I TLR-4 polymorphism renal transplant patients (p<0.05). TLR-2 expression on neutrophils in wild-type genotype renal transplant patients was higher compared to wild-type genotype healthy volunteers, and was also higher in relation to A299G/T399I kidney transplanted patients (p<0.01). Conclusion: Stable renal transplant patients with TLR-4 polymorphism have a lower expression of TLR-4 and TLR-2 receptors in peripheral mononuclear cells, which ultimately indicate a less responsiveness for alarmins. One of the early phases that lead to fibrosis progression is inflammation. Once this stage is resolved, fibrosis might be prevented. Bone marrow mononuclear cells (BMMCs) are emerging as a new therapy for several pathologies, including autoimmune diseases, because they enact immunosuppression. In this study we aimed to evaluate the role of BMMC administration in a model of kidney fibrosis induced by an acute injury. C57Bl6 mice were subjected to unilateral severe ischemia by clamping the left renal pedicle for 1 h. BMMCs were isolated from femurs and tibia, and after 6 h of reperfusion, 1 x 10(6) cells were administrated intraperitoneally. At 24 h after surgery, treated animals showed a significant decrease in creatinine and urea levels when compared with untreated animals. Different administration routes were tested. Moreover, interferon (IFN) receptor knockout BMMCs were used, as this receptor is necessary for BMMC activation. Labeled BMMCs were found in ischemic kidney on FACS analysis. This improved outcome was associated with modulation of inflammation in the kidney and systemic modulation, as determined by cytokine expression profiling. Despite non-amelioration of functional parameters, kidney mRNA expression of interleukin (IL)-6 at 6 weeks was lower in BMMC-treated animals, as were levels of collagen 1, connective tissue growth factor (CTGF), transforming growth factor-beta (TGF-beta) and vimentin. Protective molecules, such as IL-10, heme oxygenase 1 (HO-1) and bone morphogenetic 7 (BMP-7), were increased in treated animals after 6 weeks. Moreover, Masson and Picrosirius red staining analyses showed less fibrotic areas in the kidneys of treated animals. Thus, early modulation of inflammation by BMMCs after an ischemic injury leads to reduced fibrosis through modulation of early inflammation.

Apocynin has been extensively used as an inhibitor of NADPH oxidase (NOX) in many experimental models using phagocytic and non-phagocytic cells. Currently, there is some controversy about the efficacy of apocynin in non-phagocytic cells, but in phagocytes the reported results are consistent, which could be due to the presence of myeloperoxidase in these cells. This enzyme has been proposed as responsible for activating apocynin by generating its dimer, diapocynin, which is supposed to be the active compound that prevents NADPH oxidase complex assembly and activation. Here, we synthesized diapocynin and studied its effect on inhibition of gp91(phox) RNA expression. We found that diapocynin strongly inhibited the expression of gp91(phox)mRNA in peripheral blood mononuclear cells (PBMC). Only at a higher concentration, apocynin was able to exert the same effect. We also compared the apocynin and diapocynin efficacy as inhibitors of tumor necrosis factor-alpha (TNF-alpha) and interleukin-10 (IL-10) production in response to lipopolysaccharide (LPS)-activated PBMC. Although apocynin did inhibit TNF-alpha production, diapocynin had a much more pronounced effect, on both TNF-alpha and IL-10 production. In conclusion, these findings suggest that the bioconversion of apocynin to diapocynin is an important issue not limited to enzymatic activity inhibition, but also for other biological effects as gp91(phox) mRNA expression and cytokine production. Hence, as diapocynin can be easily prepared from apocynin, a one-step synthesis, we recommend its use in studies where the biological effects of apocynin are searched. (C) 2010 Elsevier Inc. Background & aim: To compare the effect of fish oil-based (FO) lipid emulsions (LE) for parenteral administration with standard LE and a new FO containing LE composed of four different oils on the antigen presentation and inflammatory variables. Methods: Phytohemagglutinin (PHA) activated human mononuclear leukocytes were cultured with different LE - Control: without LE; SO: soybean oil; SO/FO: soybean and FO (4:1); MCT/SO: medium chain triglycerides and SO (1:1); MCT/SO/FO: MCT/SO and FO (4:1) and SMOF: a new LE containing FO. Cytokine production was evaluated by ELISA, the expression of antigen-presenting and co-stimulatory surface molecules were analyzed by flow cytometry and lymphocyte proliferation was assessed by H(3)-Thymidine incorporation, after tetanus toxoid-induced activation. Results: All LE decreased the HLA-DR and increased CD28 and CD152 expression on monocytes/macrophages and lymphocytes surface (p < 0.05). SO/FO and MCT/SO/FO decreased lymphocyte proliferation (p<0.05). All LE decreased IL-2 product ion, but this effect was enhanced with MCT/SO/FO and SMOF (p < 0.05). MCT/SOTO decreased IL-6 and increased IL-10, whereas SO had the opposite effect (p < 0.05). Conclusion: FO LE inhibited lymphocyte proliferation and had an anti-inflammatory effect. These effects seem to be enhanced when FO is mixed with MCT/SO. SMOF had a neutral impact on lymphocyte proliferation and IL-6 and IL-10 production.

In many hemolytic disorders, such as malaria, the release of free heme has been involved in the triggering of oxidative stress and tissue damage. Patients presenting with severe forms of malaria commonly have impaired regulatory responses. Although intriguing, there is scarce data about the involvement of heme on the regulation of immune responses. In this study, we investigated the relation of free heme and the suppression of anti-inflammatory mediators such as PGE(2) and TGF-beta in human vivax malaria. Patients with severe disease presented higher hemolysis and higher plasma concentrations of Cu/Zn superoxide dismutase (SOD-1) and lower concentrations of PGE(2) and TGF-beta than those with mild disease. In addition, there was a positive correlation between SOD-1 concentrations and plasma levels of TNF-alpha. During antimalaria treatment, the concentrations of plasma SOD-1 reduced whereas PGE(2) and TGF-beta increased in the individuals severely ill. Using an in vitro model with human mononuclear cells, we demonstrated that the heme effect on the impairment of the production of PGE(2) and TGF-beta partially involves heme binding to CD14 and depends on the production of SOD-1. Aside from furthering the current knowledge about the pathogenesis of vivax malaria, the present results may represent a general mechanism for hemolytic diseases and could be useful for future studies of therapeutic approaches. Four new ternary complexes of copper(I) with thiosaccharin and phosphanes were prepared. The reaction of [Cu(4)(tsac)(4)(CH(3)CN)(2)] (1) (tsac: thiosaccharinate anion) with PPh(3) in molar ratios Cu(I)/PPh(3) 1:075 and 1:2 gave the complexes [Cu(4)(tsac)(4)(PPh(3))(3)] center dot CH(3)CN (2) and Cu(tsac)(PPh(3))(2) (3), respectively. The reaction of 1 with Ph(2)PCH(2)PPh(2) (dppm) in molar ratios Cu(I)/dppm 2:1 and 1:1 gave the complexes [Cu(4) (tsac)(4)(dppm)(2)] center dot 2CH(2)Cl(2) (4) and [Cu(2)(tsac)(2)(dppm)(2)] center dot CH(2)Cl(2) (5), respectively. All the compounds have been characterized by spectroscopic and X-ray crystallographic methods. Complex 2 presents a tetra-nuclear arrangement with three metal centers in distorted tetrahedral S(2)NP environments, the fourth one with the Cu(I) ion in a distorted trigonal S(2)N coordination sphere, and the tsac anions acting as six electron donor ligands in mu(3)-S(2)N coordination forms. Complex 3 shows mononuclear molecular units with copper(I) in a distorted trigonal planar coordination sphere, built with the exocyclic S atom of a mono-coordinated thiosaccharinate anion and two P-atoms of triphenylphosphane molecules. With dppm as secondary ligand the structures of the complexes depends strongly on the stoicheometry of the preparation reaction. Complex 4 has a centrosymmetric structure. Two triply bridged Cu(2)(tsac)(2)(dppm) units are joined together by the exocyclic S-atoms of two tsac anions acting effectively as bridging tridentate ligands. Complex 5 is conformed by asymmetric dinuclear moieties where the two dppm and one tsac ligands bridge two Cu(I) atoms and the second tsac anion binds one of the metal centers through its exocyclic S-atom. Sabe-se que o refluxo crônico pode induzir lesão mucosa, estimular a proliferação de células e promover tumorigênese no esôfago distal. Ainda não é sabido por que apenas uma parcela dos pacientes com refluxo esofágico progrediram para uma metaplasia intestinal (Esôfago de Barrett) e adenocarcinoma. O estresse oxidativo parece exercer um papel importante nessa progressão . Assim sendo, examinamos o padrão de expressão da enzima Heme Oxigenase-1 (HO-1), enzima indutora do estresse oxidativo, em peças de esôfago obtidos de um estudo experimental com ratos que avaliou o papel do refluxo gástrico e duodenoesofágico na carcinogênese esofágica. Métodos: Uma amostra de três (3) peças de esôfago de cada grupo de ratos submetidos a tratamentos diferentes tiveram a expressão da enzima Heme Oxigenase-1 avaliada através de imunohistoquímica. Os ratos foram divididos nos seguintes grupos: (1) cardioplastia para induzir refluxo predominantemente gástrico, (2) anastomose esofagoduodenal para induzir refluxo duodenal, (3) sem tratamento, (4) cardioplastia+dietilnitrosamina (DEN), (5) anastomose esofagoduodenal +DEN, (6) DEN. Resultados: Não houve desenvolvimento de câncer ou metaplasia intestinal nos animais que não foram expostos ao refluxo de conteúdo duodenal. A expressão de HO-1 foi observada apenas em ratos submetidos à anastomose esofagoduodenal (Grupos 2 e 5) e a análise da média de intensidade de fluorescência demonstrou uma diferença significante de expressão de HO-1 (4,8 e 4,6 vezes respectivamente) comparando-se ao controle (Grupo 3) (p<0,05). O alvo principal para expressão da HO-1 foram as células inflamatórias dentro do tumor ou em áreas subepiteleiais. Os ratos expostos ao refluxo gástrico não desenvolveram tumores ou expressaram a HO-1. Conclusões: A esofagite de refluxo induzida por refluxo esofágico com conteúdo duodenal provocou estresse oxidativo considerável e pode desempenhar um papel importante na carcinogênese esofágica. O refluxo gástrico não foi suficiente para induzir estresse oxidativo neste modelo experimental.

Este estudo incluiu 523 suínos, entre três e quatro meses de idade, com sinais clínicos sugestivos de circovirose. A maioria (471) desses animais foi recebida viva no Setor de Patologia Veterinária – SPV/UFRGS, onde foram avaliados clinicamente, eutanasiados e necropsiados. Os principais sinais observados foram caquexia, crostas eritematosas circulares na pele, alterações articulares, palidez ou icterícia de mucosas, diarréia, sinais respiratórios e sinais nervosos. Os principais achados macroscópicos foram linfadenomegalia, pulmões não colapsados, consolidações pulmonares, hiperqueratose do quadrilátero esofágico, esplenomegalia, rins pálidos ou amarelados com pontos brancos, ascite, hidropericárdio e hidrotórax. Fragmentos de pele, linfonodo inguinal, linfonodo mesentérico, tonsila, baço, íleo, cólon, fígado, rim, pulmão, coração e cérebro foram coletados e processados convencionalmente para histologia. As principais alterações microscópicas incluíram infiltrado linfoistiocitário e depleção centrofolicular em órgãos linfóides, infiltrado linfoistiocitário nos intestinos, pneumonia intersticial, hepatite portal mononuclear, nefrite intersticial e hiperplasia centrofolicular, edema e dilatação de vasos linfáticos dos intestinos e linfonodos. Amostras de cólon e íleo de 111 suínos que apresentavam conteúdo de consistência fluida foram coletadas e processadas para bacteriologia. Em 27 desses animais foram detectados agentes bacterianos patogênicos. E. coli foi o mais prevalente, mas Salmonella sp. e Brachyspira sp. também foram detectadas. Cortes de linfonodos mesentéricos, pulmões, rins e intestinos de 56 animais foram submetidos à imunoistoquímica com anticorpo policlonal anti-PCV2. Destes, amostras de 50 foram positivas. Os achados clínicos e macroscópicos desses animais foram compatíveis com circovirose, mas a associação de lesões histológicas características da doença com a presença do PCV2, demonstrada imunoistoquímicamente, foi diagnóstica.

O transplante de ilhotas humanas, utilizado como reposição das células produtoras de insulina em pacientes portadores de diabetes mellitus tipo 1, está se tornando uma importante prática clínica. Entretanto, eventos inflamatórios não específicos presente nas ilhotas, são responsáveis pela vulnerabilidade das mesmas, e contribuem à diminuição do número celular durante o processo de isolamento e posterior transplante. CD40 é um membro da família do receptor de necrose tumoral, descrito em uma variedade de células. Em condições fisiológicas, o CD40 presente nas células apresentadoras de antígenos participa como molécula co-estimulatória na ativação dos linfócitos T. Porém, o CD40 também foi descrito em condições patológicas, como psoríase, aterosclerose e fibrose cística, onde sua expressão está envolvida em eventos crônicos inflamatórios. É interessante ressaltar que, o CD40 também tem sido descrito em neurônios, células que apresentam uma variedade de moléculas similares às expressas nas células M pancreáticas. Em vista desses achados, tentou-se determinar se a células M também poderiam expressar o receptor de CD40, e se presente, determinar possíveis conseqüências próinflamatórias após a sua ativação. Utilizaram-se diversas técnicas como RT-PCR, western blot, citometria de fluxo, imuno-histoquímica assim como imunofluorescência, para detectar a expressão de CD40 em ilhotas de camundongo, macaco e humano, e também na linhagem de células M NIT-1. Determinaram-se as vias de transdução de sinais de CD40 por western blot e ensaios com gene repórter. Foi determinada por tecnologia luminex, a secreção de citocinas e quimiocinas dependente de CD40 em ilhotas humanas, estimuladascom a proteína recombinante CD40L e em alguns casos confirmada por RT-PCR e imunofluorescência. Os resultados demonstram a expressão de CD40 nas células M, que pode ser aumentada pela ação de citocinas pró-inflamatórias, cuja ativação induz a secreção de mais citocinas e quimiocinas (IL-6, IL-8, MCP-1 e MIP-1M) dependentes das vias de transdução de sinais Raf/MEK/ERK e NF-VB. A interação CD40-CD40L aumentou a expressão de ICAM-1 e a induziu morte celular nas células M pancreáticas. Nesse sentido, a ativação de CD40 induz a secreção de mediadores solúveis próinflamatórios que podem comprometer a viabilidade das células M. O cenário próinflamatório sustentando pela ação de CD40 sugere que o mesmo poderia ter um papel ativo orquestrando um processo inflamatório
