Transglutaminase-catalyzed inactivation of glyceraldehyde 3-phosphate dehydrogenase and α-ketoglutarate dehydrogenase complex by polyglutamine domains of pathological length

Several adult-onset neurodegenerative diseases are caused by genes with expanded CAG triplet repeats within their coding regions and extended polyglutamine (Qn) domains within the expressed proteins. Generally, in clinically affected individuals n ≥ 40. Glyceraldehyde 3-phosphate dehydrogenase binds tightly to four Qn disease proteins, but the significance of this interaction is unknown. We now report that purified glyceraldehyde 3-phosphate dehydrogenase is inactivated by tissue transglutaminase in the presence of glutathione S-transferase constructs containing a Qn domain of pathological length (n = 62 or 81). The dehydrogenase is less strongly inhibited by tissue transglutaminase in the presence of constructs containing shorter Qn domains (n = 0 or 10). Purified α-ketoglutarate dehydrogenase complex also is inactivated by tissue transglutaminase plus glutathione S-transferase constructs containing pathological-length Qn domains (n = 62 or 81). The results suggest that tissue transglutaminase-catalyzed covalent linkages involving the larger poly-Q domains may disrupt cerebral energy metabolism in CAG/Qn expansion diseases.

Workshop on Schizophrenia

On November 29–30, 1995, the National Academy of Sciences and the Institute of Medicine brought together experts in schizophrenia and specialists in other areas of the biological sciences in a workshop aimed at promoting the application of the latest biological information to this clinical problem. The workshop paid particular attention to evidence of pathology in the brains of people with schizophrenia, and to the possibility that this reflects an abnormality in brain development that eventually leads to the appearance of symptoms. The participants were impressed with the complexity of the problem, and felt that multiple approaches would be required to understand this disease. They recommended that a major focus should be on the search for predisposing genes, but that there should be parallel research in many other areas.

Apolipoprotein E and Alzheimer disease.

Inheritance of specific apolipoprotein E (apoE) alleles determines, in large part, the risk and mean age of onset of late-onset familial and sporadic Alzheimer disease. The mechanism by which the apoE isoforms differentially contribute to disease expression is, however, unknown. Isoform-specific differences have been identified in the binding of apoE to the microtubule-associated protein tau, which forms the paired helical filament and neurofibrillary tangles, and to amyloid beta peptide, a major component of the neuritic plaque. These and other isoform-specific interactions of apoE give rise to testable hypotheses for the mechanism(s) of pathogenesis of Alzheimer disease. An unresolved issue of increasing importance is the relationship between the structural pathological lesions and the cellular pathogenesis responsible for the clinical disease phenotype, progressive dementia. The identification of apoE in the cytoplasm of human neurons and the characterization of isoform-specific binding of apoE to the microtubule-associated proteins tau and MAP-2 present the possibility that apoE may affect microtubule function in the Alzheimer brain.

¿Qué está pasando en el proceso de enseñanza-aprendizaje de Ciencias Básicas en nuestro sistema educativo?

Se ha creado una red multidisciplinar que agrupa profesorado de distintas etapas educativas con el objetivo de analizar debilidades en la enseñanza de asignaturas de Ciencias básicas y experimentales (en todas las etapas formativas) y proponer actuaciones de innovación docente que subsanen esas debilidades. El trabajo realizado hasta ahora se ha centrado en tres objetivos específicos: 1) Identificar debilidades y fortalezas en la docencia de Matemáticas, Químicas, Física, Biología y Geología. Para ello, se han analizado tanto las prácticas docentes más habituales como las innovaciones más recientes (TICs, programa EntusiasMAT, etc.); 2) Diseñar estrategias de comunicación y trabajo colaborativo que mejoren el flujo de comunicación e información entre docentes de Ciencias de distintas etapas educativas; 3) Proponer protocolos, estrategias y buenas prácticas para la mejora de la docencia en Ciencias en todas las etapas educativas (por ejemplo, se plantea el diseño e implementación de talleres para que los más pequeños aprendan cuestiones relativas a ciencias naturales mediante el juego). Entre las conclusiones obtenidas destacan: 1) se sigue empleando de forma mayoritaria la clase teórica magistral aún a pesar de las interesantes propuestas de innovación docente existentes; 2) el aprendizaje es más eficiente cuando se basa en experiencias prácticas.

Rosa gallica l. var. officinalis (hort. ex Andrews) Ser. en el norte de Alicante (Comunidad Valenciana)

Se ha localizado en el margen de la carretera comarcal CV 795 en su kilómetro 17 próximo a Ulls de Canals y dentro del término de Banyeres de Mariola, una buena población asilvestrada de Rosa gallica L. var. officinalis (hort. ex Andrews) Ser., que corresponde a la segunda población localizada en Alicante, puesto que se conocía una población pero situada próxima al litoral en la comarca de la Marina Alta. La llamada rosa de Provins o provincialis (de Provenza) es una de las rosas cultivadas en Europa desde más antiguo, probablemente la primera. Aparece dibujada en los frescos de Pompeya, fue también la rosa de las boticas (“Officinalis”) y se corresponde con la famosa rosa roja o Rosa de Lancanster, símbolo de este bando en la Guerra de civil inglesa llamada de “las dos rosas”. Su cultivo en España fue muy importante y era frecuentemente citada en la literatura botánica antigua, fue llamada “rosa de Castilla” pero con el tiempo y el abandono del campo, su presencia fue disminuyendo y de forma asilvestrada solo aparece en unas pocas provincias españolas, aunque se sigue manteniendo su cultivo a pequeña escala en colecciones particulares y rosaledas. Precisamente, en la Rosaleda “Pere Dot” del Jardín Botánico de Torretes (Ibi) mantenemos en cultivo dos de las variedades botánicas reconocidas y muchos de los cultivares e híbridos de este rosal, que hemos adquirido de colecciones extranjeras. El hallazgo de este material asilvestrado probablemente procedente de antiguos cultivos en la zona, tiene gran valor etnobotánico y puede tener valor genético. Aunque en su morfología no se han encontrado diferencias relevantes, se ha procedido a su multiplicación vegetativa y será próximamente incluido en la rosaleda del Jardín Botánico, junto con pliegos de herbario y semillas.
